Pathological findings of paraquat poisoning in mice, rats and rabbits (마우스, 랫트 및 토끼에서 paraquat 중독의 병리학적 관찰)

  • This study was carried out to investigate the pathological changes with paraquat(1.1'-dimethyl-4.4'-dipyrildiylium dichloride) administered by intraperitoneally, orally, skin applied in mice, rats and rabbits. Results were obtained as follows; In 2 days after paraquat administration clinical signs anorexia, depression, tachypnea, and tachycardia, respiratory failure, coma and death were observed in mice, rats and rabbits. Compared toxicity of paraquat with mouse were observed toward to paraquat that resistance was strong than rats and rabbits. Also, mortality of skin application were found the low than intraperitoneal and high than oral administration. In the case of gross observation were appear lips moisture in orally administered rats and rabbits by skin application. Lung of all laboratory animals were observed congestion and haemorrhage, swelling or atrophy. In the case of microscopic findings were severe congestion and haemorrhage, interstitial pneumonia of lung. Congestion and haemorrhage of liver, congestion and haemorrhage, renal tubule epithelium necrosis of kidney were observed in mice, rats and rabbits. Skin application group of mice, rats and rabbits showed infiltration of inflammatory cells and folliculitis of epidermis and dermis. Also, in oral administration group showed congestion and haemorrhage, tachment, necrosis of alimentary tract mucosa.

Effect of Antioxidant and Skin Whitening of Ethanol extracts from Ultrasonic Pretreated Abeliophyllum distichum Nakai (초음파 처리 미선나무 에탄올 추출물의 항산화 및 미백효과)

  • This research evaluate antioxidant and skin-whitening effect of Abeliophyllum distichum Nakai by extraction processes. First, antioxidant effects were follows: EE (70% ethanol extract) showed higher DPPH scavenging activity of 69.66% than WE (hot water exract) 59.13% at $0.3mg/m{\ell}$, also UE's (70% ethanol extract by sonication process) higher than EE. Reducing power was that also EE showed higher than WE, and it was the highest value with UE's because of ultrasonic pretreatment. Next, the whitening effect tyrosinase inhibition activity was measured that EE was 23.88%, WE's was 16.69%, and UE was 23.34%. Ultrasonic pretreatment did not influence to tyrosinase inhibition activity. Cell viability showed low cell toxicity in all groups. UE's inhibited melanin synthesis, 55.1%, that is higher than EE and WE, 52.7% and 39.5%, respectively. As a result, we confirmed that antioxidant activities and skin-whitening effect by extraction process. Also, this results confirmed that the Abeliophyllum distichum Nakai extracts worth as cosmetic materials.

Evaluation of Acute Toxicity of Plant Extracts, Lavender, Lemon Eucalyptus and Cassia Essential Oil (식물 추출물 라벤더, 레몬 유칼립투스 및 계피 오일의 급성독성평가)

  • The acute toxicity test of high bio-active plant essential oils was conducted with Lavender, Lemon eucalyptus and Cassia oils selected to develop environment-friendly insecticides. The results of acute oral toxicity using rats showed that $LD_{50}$ of over 2,000 mg/kg bw for Lavender, Lemon eucalyptus and Cassia oils. The calculated acute dermal $LD_{50}$ value of all testing materials was over 4,000 mg/kg bw. The Skin irritation test indicated that Lavender and Lemon eucalyptus oil have no irritation while Cassia oil has a moderate irritation. For the Eye Irritation test, the result showed no irritation for Lavender and Lemon Eucalyptus oil and irritation for Cassia oils. However, the irritation was not showed for Eye Irritationwashing test of Cassia oil. Consequently, the Lavender and Lemon eucalyptus oils were showed to be low in toxicity whereas Cassia oil indicated to cause a moderate irritation on the skin and eyes.

Toxicity Assessment and Establishment of Acceptable Daily Intake of Fungicide Isotianil (살균제 Isotianil의 독성평가와 일일섭취허용량 설정)

  • Isotianil is a fungicide which has prevention effects against rice blast disease. In order to register this new pesticide, the series of toxicity data on animal testing were reviewed to evaluate its hazards to consumers and to determine its acceptable daily intake. Isotianil was almost excreted by urine and feces. It has low acute oral toxicity while has no skin toxicity and ocular irritation. Its skin sensitization was evaluated as slight. Genotoxicity of parent compound and metabolite was negligible. Chronic toxicity tests on rats and dogs showed changes of hematology, clinical biochemistry and liver weight. It had no reproductive and teratogenic effects. The estimation of Acceptable Daily Intake(ADI) is based on the lowest no-observed adverse effect level (NOAEL). The lowest NOAEL of 2.83 mg/kg bw/day was found in the twelve-months rats study. The NOAEL was based on increased liver weight and treatment-related effect on clinica chemistry finding at the nest higher dose level of 2.83 mg/kg bw/day. Therefore, it is considered appropriated to apply an uncertainty factor of 100 to the NOAEL 2.83 mg/kg bw/day from the rat study, resulting in an ADI of 0.028 mg/kg bw/day.

Acute Toxicity Evaluation of Loess-sulfur Complex in different pH (황토유황합제의 pH차이에 따른 급성독성평가)

  • Loess-sulfur complex has been widely used as an environmental friendly organic materials for insecticides and fungicides in crop cultivation. However, there are high concerns about skin and eye irritation for farm workers due to the high alkaline properties of loess-sulfur complex. The acute toxicity evaluation was conducted with three samples of loess-sulfur complex in different pH (pH 9, 10, 11) in order to supply the evidentiary data for selecting the optimal product among the test materials. The results of acute oral toxicity using rats showed $LD_{50}$ of over 2,000 mg/kg b.w. for all three samples of loess-sulfur complex. The calculated acute dermal $LD_{50}$ of all tested materials was over 4,000 mg/kg b.w.. The Skin and eye irritation indicated that all tested materials have no irritation. Consequently, it was suggested that loess-sulfur complex be low in acute toxicity at all different pH values (pH 9~11).

Evaluation of Acute Toxicity of Black Pepper extracts, Clove bud, Rosemary and Origanum Essential oils (식물추출물 후추, 클로브버드, 로즈마리 및 오리가늄오일의 급성독성평가)

  • Highly bio-active plant essential extracts and oils from pepper, clove bud, rosemary and origanum which are selected to develop environment-friendly insecticides was studied for their acute toxicity. The results of acute oral toxicity using rats showed $LD_{50}$ value of over 2,000 mg/kg bw for pepper, clove bud, rosemary and origanum oils. The calculated acute dermal $LD_{50}$ value of pepper was over 4,000 mg/kg bw and anther testing materials was over 4,000 mg/kg bw. The skin irritation test showed that pepper, clove bud and rosemary oils had no irritation while origanum oil had a moderate irritation. According to the eye irritation test, it showed that there was no irritation for pepper and rosemary oils, while there were irritation for clove bud and origanum oils. Consequently, pepper and rosemary oils were shown to be low in toxicity whereas clove bud oil was indicated to cause a mild eye irritability and origanum oil, causing a moderate skin and eye irritability.

Comparison study of dermal cell toxicity and zebrafish brain toxicity by humidifier sterilizer chemicals (PHMG, PGH, CMIT/MIT) (가습기 살균제 성분(PHMG, PGH, CMIT/MIT)의 사람 피부세포 독성 및 제브라피쉬 뇌신경 독성 비교 연구)

  • Toxicities to many organs caused by humidifier disinfectants have been reported. Recently, humidifier disinfectants have been reported to cause cardiovascular, embryonic, and hepatic toxicities. This study was designed to investigate the toxic mechanism of humidifier disinfectants and compare toxicity in a cellular model and a zebrafish animal model. Because brain toxicity and skin toxicity have been less studied than other organs, we evaluated toxicity in a human dermal cell line and zebrafish under various concentrations of humidifier disinfectants that included polyhexamethyleneguanidine phosphate (PHMG), oligo-[2-(2-ethoxy)-ethoxyethyl-guanidinium-chloride] (PGH) and methylchloroisothiazolinone/methylisothiazolinone (CMIT/MIT). A human dermal fibroblast cell line was treated with disinfectants (0, 2, 4, 6, 8, and 16 mg L-1) to compare their cytotoxicity. The fewest PHMG-treated cells survived (up to 33%), while 49% and 40% of the PGH- and CMIT/MIT-treated cells, respectively, survived. The quantification of oxidized species in the media revealed that the PHMG-treated cells had the highest MDA content of around 28 nM, while the PGH- and CMIT/MIT-treated cells had 13 and 21 nM MDA, respectively. As for brain toxicity, treatment of the zebrafish tank water with CMIT/MIT (final 40 mg L-1) for 30 min resulted in a 17-fold higher production of reactive oxygen species (ROS) than in the control. Treatment with PGH or PHMG (final 40 mg L-1) resulted in 15- and 11-fold higher production, respectively. The humidifier disinfectants (PHMG, PGH, and CMIT/MIT) showed severe dermal cell toxicity and brain toxicity. These toxicities may be relevant factors in understanding why some children have language disorders, motor delays, and developmental delays from exposure to humidifier disinfectants.

The Protective Effect of Spirulina-derived Phycocyanin on Dermal Fibroblasts Induced by UV Rays (자외선으로 손상을 유도한 피부섬유아세포에서 스피룰리나 유래 피코시아닌의 보호 효과)

  • UV induce oxidative stress and increase matrix metalloproteinase (MMP) expression, resulting in skin aging. Thus, preventing skin damage from ultraviolet B (UVB)-induced skin damage can attenuate skin aging. Spirulina is comprised of prokaryotes a powerful antioxidant. This study aimed to investigate the photoprotective effects of spirulina-derived phycocyanin (PC) against UVB radiation using human skin fibroblast. As a results, PC showed no toxicity at concentrations of 5-40 ㎍/mL in terms of fibroblast viability. Survival rate of UVB-irradiated fibroblast incresased to 73.5% from 50.5% with PC treatment. UVB treatment increased MMP-1 and MMP-9 expression whereas PC treatment decreased it. The results indicate that PC might reduce or prevent skin aging by reducing UVB irradiation-induced skin wrinkles and free radicals.

A Subchronic Toxicity Study of DA-5018 Creams in Rats (DA-5018 cream의 랫드에 대한 경피투여 아급성독성시험)

  • A 13-week dermal toxicity test was conducted to assess the toxicity of DA-5018, a capsaicin derivative. Three groups of Sprague-Dawley rats (10-15 males and 10-15 females) were treated with DA-5018 cream daily by dermal application at concentrations of 0.1%, 0.3% or 0.9% as 500 mg/kg for 13 weeks. One further group of rats (15 males and 15 females) received cream base at 500 mg/kg/day and acted as controls. One male receiving 0.3% DA-5018 cream died during the treatment period. But the animal did not show any signs of treatment-related toxicity until death. There were no local skin reaction of application site and systemic reaction to the treatment of DA-5018 creams in all experimental groups throughout treatment and recovery period. Weight gain and food consumption in animals that received DA-5018 creams appeared to be comparable to that of the controls. Laboratory analyses (hematology, urinalysis and opthalmoscopic examination) did not revealed pathological values. In biochemical investigations, an increase of glucose level associated with increased food consumption and some other significant changes were noted in the animals of both sexes received DA-5018 creams. But these changes were not considered to be of toxicological importance. Postmortem examination did not show macroscopic or histological alterations attributable to the DA-5018 treatments. Based on these results, NOAEL(no-observable-adverse-effect level) of DA-5018 cream if estimated to be over 500 mg/tg/day as 0.9% cream.

  • Amiodarone is a potent antiarrythmic agent used in the treatment of refractory tachyarrythmias and premature ventricular contractions. Amiodarone may be responsible for the frequent and various side effects including corneal deposits, abnormal liver function test, hyperthyriodism or hypothyroidism, bluish discoloration of the skin, neuropathies, and the others. However, pulmonary toxicity is most serious adverse reaction limiting the clinical efficacy. Rescently, we experienced a case of pulmonary toxicity induced by low dose 10-month amiodarone treatment for atrial fibrilation with rapid ventricular response. Lung biopsy reveals interstitial inflammation, fibrosis, hyperplasia of pneumocytes, and foamy macrophages. Respiratory symptoms and abnormal chest X-ray findings were nearly complete cleared after using steroid and withdrawal of amiodarone. We report a case of amiodarone-induced pulmonary toxicity with literature review.
