Downregulatory Effect of AGI-1120 $({\alpha}-Glucosidase Inhibitor)$ and Chaga Mushroom (Inonotus obliquus) on Cellular $NF-{\kappa}B$ Activation and Their Antioxidant Activity (AGI-1120과 차가버섯의 $NF-{\kappa}B$ 활성화 억제 및 항산화 효과)

  • Effect of AGI $({\alpha}-Glucosidase\;Inhibitor)-1120$, pine (Pinus densiflora) bark extract and Chaga mushroom (Inonotus obliquus) - and Chaga mushroom mycelium extracts on cellular $NF-{\kappa}B$ activation in malignant human keratinocytes (SCC-13) were evaluated to elucidate the possible correlation of $NF-{\kappa}B$ with antioxidant activity. The antioxidant activities of these natural products were examined in three different evaluation methods, i.e., lipid peroxidation value (POV) evaluation test, and 1,1diphenyl-2-picrylhydrazyl radical (DPPH) and nitric oxide (NO) scavenging test. In a cell-based $NF-{\kappa}B$ monitoring assay systern, all samples revealed the downregulatory profiles on the cellular $NF-{\kappa}B$ activity. AGI -1120 (1, 2 mg) and Chaga mushroom extract (0.05, 0.1 mg) downregulated the $NF-{\kappa}B$ activity in a dose-dependent manner. Chaga mushroom mycelium extract (5 mg) significantly inhibited the $NF-{\kappa}B$ activity (p<0.05). Although AGI-1120 and Chaga mushroom mycelium extract exhibited no antioxidant activities evaluated in pay, Chaga mushroom extract showed antioxidant in a dose-dependent manner at concentrations of $0.05{\sim}1$ mg. While AGI-1120 and Chaga mushroom extract possessed a relatively potential DPPH radical scavenging activity, the NO scavenging activity of Chaga mushroom extract $(SC_{50}:47\;{mu}g)$ was higher than the known antioxidant, vitamin C $(SC_{50}:77\;{mu}g)$. These results suggest that AGI-1120 and Chaga mushroom- and Chaga mushroom mycelium extracts may serve as an useful radical scavenging antioxidant agents with $NF-{\kappa}B$ inhibitory effect in human skin.

Reconstruction with Radial Forearm Free Flap after Ablative Surgery for Oral Cavity and Oropharyngeal Cancers (구강암과 구인두암의 절제술 후 전완유리피판술을 이용한 재건술)

  • Background and Objectives: Surgical ablation of tumors in the oral cavity and the oropharynx results in a three dimensional defect because of the needs to resect the adjacent area for the surgical margin. Although a variety of techniques are available, radial forearm free flap has been known as an effective method for this defect, which offers a thin, pliable, and relatively hairless skin and a long vascular pedicle. We report the clinical results of our 54 consecutive radial forearm free flaps used for oral cavity and oropharynx cancers. Materials and Methods: We reviewed the medical records of patients who were offered intraoral reconstruction with a radial forearm free flap after ablative surgery for oral cavity and oropharyngeal cancers from August 1994 to February 2003 and analyzed surgical methods, flap survival rate, complication, and functional results. Among these, 20 cases were examined with modified barium swallow to evaluate postoperative swallowing function and other 8 cases with articulation and resonance test for speech. We examined recovery of sensation with two-point discrimination test in 15 cases who were offered sensate flaps. Results: The primary sites were as follows : mobile tongue (18), tonsil (17), floor of mouth (4), base of tongue (2), soft palate (2), retromolar trigone (3), buccal mucosa (1), oro-hypopharynx (6), and lower lip (1). The paddles of flaps were tailored in multilobed designs from oval shape to tetralobed design and in variable size according to the defects after ablation. This procedures resulted in satisfactory flap success rate (96.3%) and showed good swallowing function and social speech. Eight of 15 cases (53.3%) who had offered sensate flap showed recovery of sensation between 1 and 6 postoperative months (average 2.6 month). Conclusion: The reconstruction with radial forearm free flap might be an excellent method for the maximal functional results after ablative surgery of oral cavity and oropharyngeal cancers that results in multidimensional defect.

Evaluation on In-plane Shear Strength of Lightweight Composite Panels (경량 복합패널의 면내 전단 성능 평가)

  • The number of natural disasters in Korea, such as earthquakes, is increasing. As a result, there is growing need for temporary residences or shelters for disaster conditions. The aim of this study was to produce post-disaster refugees housing differentiated from existing shelters using lightweight composite panels. To accomplish this, the structural performance of lightweight composite panels was validated, and an in-plane shear strength test was conducted according to the ASTM E72 criteria among the performance test methods for panels. As a result of the experiment, the maximum load for each specimen under an in-plane shear load was determined. All the experiments ended with the tear of the panel's skin section. The initial stiffness of the specimens was consistent with that predicted by the calculations. On the other hand, local crushing and tearing, as well as the characteristics of the panel, resulted in a decrease in stiffness and final failure. Specimens with an opening showed a difference in stiffness and strength from the basic experiment. The maximum load and the effective area were found to be proportional. Through this process, the allowable shear stress of the specimens was calculated and the average allowable shear stress was determined. The average ultimate shear stress of the lightweight composite panels was found to be $0.047N/mm^2$, which provides a criterion of judgement that could be used to expect the allowable load of lightweight composite panels.

Experiences of Latent Tuberculosis Infection Treatment for the North Korean Refugees

  • Background: Tuberculosis (TB) is increasing in immigrants. We aimed to investigate the current status of latent tuberculosis infection (LTBI) treatment for North Korean Refugees (NKR) compared to South Koreans Contacts (SKC). Methods: TB close contacts in a closed facility of SKC and NKR who underwent LTBI screening in a settlement support center for NKR were analyzed retrospectively. Results: Among tuberculin skin test (TST) ${\geq}10mm$ (n=298) reactors, the males accounted for 72.2% in SKC (n=126) and 19.5% in NKR (n=172) (p<0.01). The mean age was higher in South Korea ($42.8{\pm}9.9years$ vs. $35.4{\pm}10.0years$, p<0.01). Additionally, the mean TST size was significantly bigger in NKR ($17.39{\pm}3.9mm$ vs. $16.57{\pm}4.2mm$, p=0.03). The LTBI treatments were initiated for all screened NKR, and LTBI completion rate was only 68.0%. However, in NKR, LTBI treatment completion rate was significantly increased by shorter 4R regimen (odds ratio [OR], 9.296; 95% confidence interval [CI], 4.159-20.774; p<0.01) and male (OR, 3.447; 95% CI, 1.191-9.974; p=0.02). Conclusion: LTBI treatment compliance must be improved in NKR with a shorter regimen. In addition, a larger study regarding a focus on LTBI with easy access to related data for NKR should be conducted.

A Study on the Influence of Blending Ratio of Powder and Oil on the Stability of Talc-Free Pressed Powder Formulation (파우더와 오일의 배합 비율이 탈크 프리 프레스드 파우더 제형의 안정성에 미치는 영향에 관한 연구)

  • The main stability factors of the pressed powder include hardness and drop stability. In general, for pressed powder with talc, the hardness and drop stability are evenly met and the skin texture is excellent. Recently, more than ever customers are looking for a replacement due to asbestos issue of talc. Therefore, it is urgent to develop a pressed powder formulation without talc that maintains stability and does not lose its sense of use. In this study, experiments were conducted to find the optimal ingredients mixing ratio to make talc-free pressed powder. The characteristics of raw materials used mainly in powder were checked and the ratio was changed, and the lightness and hardness were measured and drop test was conducted. If the natural mica ratio was higher than the synthetic mica or non-coated silica was used instead of the coated silica, the hardness and drop stability were lower than the content containing talc, but the lightness was similar. Conversely, if the synthetic mica ratio was equal to or higher than the natural mica ratio and the coated silica ratio was equal to or higher than the non-coated silica ratio, the hardness and drop stability of the content containing talc were similar, but the lightness was low. It was found that the hardness was higher than the content containing talc, but the drop stability was lower. Therefore, further study of the correlation between hardness and fall stability is also thought to be necessary.

Analysis of Antioxidant Activity and Total Phenol Content and Flavonoid Content Through the Synergistic Effect of Rosa multiflora Extracts and Ascorbic Acid (찔레꽃(Rosa multiflora) 추출물과 Ascorbic Acid 의 시너지 효과를 통한 항산화 활성과 총 페놀함량 및 플라보노이드 함량 분석)

  • In this study, Rosa multiflora was sampled through extraction, concentration, and freezing. The antioxidant efficacy of a mixture of R. multiflora extract and ascorbic acid was compared with a single ascorbic acid. The purpose of this study was to analyze the antioxidant efficacy through the synergistic effect of R. multiflora extract and ascorbic acid. The experiment measured electron donating ability, ABTS+ radical scavenging, SOD-like activity, xanthine oxidase inhibition activity, reducing power, FRAP, polyphenolic content measurement, and flavnoid content measurement. As a result, the complex showed higher antioxidant activity than the control group (ascorbic acid) in the electron donating ability test and the xanthine oxidase inhibitory activity test. In the case of the flavonoid content experiment, it was confirmed that RMW+A (272.1 mg QE/g), RME+A (90.6 mg QE/g), and RMW+A (79.4 mg QE/g) at a concentration of 100 ㎍/mL had a higher flavonoid content than the control group (19.0 mg QE/g). Therefore, the extract and ascorbic acid complex exhibited a synergistic effect compared to the single use. Therefore, it is thought that this complex will contribute to product stabilization and skin aging inhibition when used in antioxidant cosmetics.

The Attractant Effects of LED Lights on Aphidius colemani (콜레마니진디벌에 대한 LED 광원의 유인효과)

  • In this study, we sought to determine an appropriate LED light source that can be applied for the quality control of Aphidius colemani. Four LED light sources (385, 405, 450, and 660 nm) were evaluated in a Y-tube experiment (choice test) using a 5,000-k white LED as a control. The 385 nm LED light source was found to have attractant effect on A. colemani. In a no-choice test (Y-tube experiment) evaluating the remaining three LED light sources (385, 405, and 450 nm), the attractiveness of these light sources for A. colemani was high in the order 450 nm > 385 nm > 405 nm light, with no statistically significant differences between 450 nm and 385 nm. Given that 385 nm LED illumination may cause skin damage, we selected 450 nm LEDs for application in a device used for the quality control of A. colemani, which we subsequently demonstrated to have an attraction rate of 87.4%. We believe the findings of this study will make a valuable contribution to the quality control of natural enemies.

Experimental study on the vertical bearing behavior of nodular diaphragm wall in sandy soil based on PIV technique

  • The nodular diaphragm wall (NDW) is a novel type of foundation with favorable engineering characteristics, which has already been utilized in high-rise buildings and high-speed railways. Compared to traditional diaphragm walls, the NDW offers significantly improved vertical bearing capacity due to the presence of nodular parts while reducing construction time and excavation work. Despite its potential, research on the vertical bearing characteristics of NDW requires further study, and the investigation and visualization of its displacement pattern and failure mode are scant. Meanwhile, the measurement of the force component acting on the nodular parts remains challenging. In this paper, the vertical bearing characteristics of NDW are studied in detail through the indoor model test, and the displacement and failure mode of the foundation is analyzed using particle image velocimetry (PIV) technology. The principles and methods for monitoring the force acting on the nodular parts are described in detail. The research results show that the nodular part plays an essential role in the bearing capacity of the NDW, and its maximum load-bearing ratio can reach 30.92%. The existence of the bottom nodular part contributes more to the bearing capacity of the foundation compared to the middle nodular part, and the use of both middle and bottom nodular parts increases the bearing capacity of the foundation by about 9~12% compared to a single nodular part of the NDW. The increase in the number of nodular parts cannot produce a simple superposition effect on the resistance born by the nodular parts since the nodular parts have an insignificant influence on the exertion and distribution of the skin friction of NDW. The existence of the nodular part changes the displacement field of the soil around NDW and increases the displacement influence range of the foundation to a certain extent. For NDWs with three different nodal arrangements, the failure modes of the foundations appear to be local shear failures. Overall, this study provides valuable insights into the performance and behavior of NDWs, which will aid in their effective utilization and further research in the field.

  • The bedsore prevention cushion serves to prevent the skin on the buttocks of a wheelchair user from being damaged through friction with the wheelchair seat. This is because it absorbs some of the weight applied to the wheelchair seat and allows the weight to be applied evenly to the entire contact surface. For the impact test, which is part of the performance testing of bedsore prevention cushions, a hip model that represents the sitting posture of a wheelchair user is required. In this study, a hip model was manufactured for impact testing of bedsore prevention cushions for wheelchairs. Performance tests for pressure bedsore prevention cushions for wheelchairs include KSP 0236, a Korean standard, and KS P ISO 16840-2, an ISO international standard. The hip model proposed in KS P ISO 16840-2 was more suitable for impact testing of bedsore prevention cushions for wheelchairs. However, the guidelines for making hip models proposed by international standards need to be modified to reflect the advancement of model making technology and use easier methods. We propose a new hip model production method that produces a hip model out of plastic all at once and additionally attaches SS-41 steel plates processed into the shape of the hip to make the mass of the model similar to the human body.

Comparative analysis of blood glucose test results on the forearm, finger, and vein (팔, 손가락, 정맥에서 채취한 혈액의 혈당검사결과 비교 분석)

  • Capillary blood sampling on the forearm reduces pain caused by skin puncture. The present study compared the blood glucose test results performed at different sampling sites of the forearm, finger, and vein to evaluate clinical validity of this alternative site blood sampling technique. Subjects numbered 555 including 61 diabetic patients participated to measure the glucose concentration on the finger ($G_F$) and the forearm ($G_A$) with a portable glucometer under overnight fasting state. Then, the venous glucose concentration ($G_V$) was measured in 514 subjects in less than 1 hour. The test results were analyzed by simple linear regression, intraclass correlation, and Passing-Bablok regression techniques. $G_A$ was highly correlated with $G_F$ or $G_V$ showing the correlation coefficients (r) of approximately 0.97 (P<0.0001) in the normal group. The patient group also resulted similarly high correlation with only slightly lower r value. The mean differences in glucose concentration were less than ${\pm}10mg/dL$ regardless of the sampling sites. Intraclass correlation coefficients were slightly smaller than r but very much similar in value in both groups. The 95% confidence intervals of the slope as well as the intercept in the Passing-Bablok regression analysis were < ${\pm}20%$ and < ${\pm}20mg/dL$, respectively, which were within the clinically acceptable ranges. These three statistical techniques introduced in the present study well demonstrated the consistency of $G_A$ with $G_F$ and $G_V$. Therefore, the forearm blood glucose test could be considered as clinically valid under fasting condition.
