  • Objective: A Case Report on Reduction of Inflammation Using Nutritional Therapy Methods: The patient is a Korean male aged 62 years. The wound has developed inflammation and is not healing while the patient complains of pain. Results: After nutritional therapy, inflammation diminished. Conclusion: Nutritional therapy can be beneficial for inflammation, for which even skin transplantation was advised.

Anti-Allergic Effect of Ulmus davidiana Cortex on Contact Dermatitis Induced by Dinitrofluoro-Benzene in Mice

  • Objective: The root bark of Ulmus davidiana var. Japonica (Ulmi Radicis cortex, URC) is a medicinal herb used for promoting diuresis and treating dampness. In Korea, URC has long been used as an efficacious therapy for inflammation, burns, frostbite and skin diseases such as eczema and psoriasis. Methods: In the present study, we used 1-fluoro-2,4-dinitrofluorobenzene (DNFB)-induced contact dermatitis (CD) mouse model to investigate the anti-allergic and the anti-inflammatory effects of URC on skin lesion, histopathological changes and specific antibody production. Results: URC treatment, 10 mg/mL, effectively inhibited skin lesions induced by repeated paintings with DNFB. In the histopathological observation, topical application of URC inhibited spongiosis. In addition, URC lowered the production levels of total immunoglobulin and IgG2a in serum. Conclusion: These data indicate that URC has an anti-inflammatory effect that produces an improvement of skin lesions in CD mice.

해양조류 유래 Porphyra-334의 UV 흡수능에 의한 피부세포 보호 효과 (Effect of sun screen utilizing Porphyra-334 derived from Ocean Algae for skin protection)

  • 광노화의 주요 요인인 자외선으로부터 피부를 보호하는 것은 피부노화를 막는 중요한 해결책 중 하나이다. 이를 위해 자연친화적 친환경적인 다양한 형태의 소재개발이 필요한 실정이다. 이에 본 연구는 미세조류(Chlamydomonas hedleyi.)로부터 Mycosporine-like amino acid (MAA)를 분리 정제 하여 인간 피부세포에서 UV protection 효과를 확인하였다. MAAs 중 porphyra-334를 분리하여 피부세포에서 UV protection 및 항염 효능을 시험하였고 그 결과 porphyra-334가 UV로부터 피부세포를 보호하여 cell viability가 약 26% 정도 증가하였고 COX-2 발현 40% 이상 저해함을 통해 항염 효능을 확인하였다. 이를 통해 porphyra-334의 자외선 차단용 소재뿐 아니라 항염 관련 소재로서의 가능성을 확인할 수 있었다.

Effects of Egg Shell Membrane Hydrolysates on Anti-Inflammatory, Anti-Wrinkle, Anti-Microbial Activity and Moisture-Protection

  • This study was conducted to examine the effects of eggshell membrane hydrolysates (ESMH) on the anti-inflammatory, anti-wrinkle, anti-microbial activity, and moisture-protection for cosmetic use. Whole ESMH (before fractionation), and fraction I (>10 kDa), fraction II (3-10 kDa), and fraction III (<3 kDa) of the hydrolysates were assessed in this experiment. As lipopolysaccharide (LPS) and IFN-${\gamma}$ caused the inflammation on Raw264.7 cell, whole ESMH and fraction I showed to be effective in inhibiting the induction of cell inflammation depending on the concentration, and also showed outstanding effect to suppress the skin inflammation. Fraction I inhibited collagenase and elastase activities to a greater extent than the other fractions, while all fractions had antibiotic effects at concentrations of 10 mg/disc and 20 mg/disc. In addition, it showed the moisture protection effects of skin on the holding amount and losing amount of moisture in upper-inner arm of the human body with a relatively low loss rate in skin, which confirmed that the hydrolyzed fractions of ESM helps to form the superior protective layer of moisture. It was concluded that ESMH fractions with different molecular weights, especially the 10 kDa fraction, have anti-lipopolysaccharide, anti-IFN-${\gamma}$-induced inflammation, anti-collagenase and elastase activities, and thus can be used as a cosmetic agent to protect skin.

  • Efficiency study of chamomile and Thyme essential oil about the basal layer & sebaceous gland in dry-skin induced by surfactant is as following : 1. Formal observation of outer skin layer and basal layer There was refreshing effects about Surfactant induction skin inflammation in Chamomile, Thyme essential oil processing group but effect in surface layer appeared by higher thing in Chamomile essential oil and it could know that thing which do not use Thyme essential oil in case Thyme essential oil is ultraviolet rays allergy at skin care because Melanocyte appear on the surface is bigger than Chamomile essential oil that it was usefully reaction in basal layer. 2. Formal observation result of sebaceous glands. There are few damages in all group but there is recuperative through excessive water damage by excessive a sebaceous gland made of mulberry bark secretion because Thyme essential oil application group cause chapped skin phenomenon after surface-active agent processing but is seen that difference does not exist greatly and Thyme essential oil that general skin is strong in permeation is effective, but Surfactant induction skin inflammation estimates that Chamomile essential oil is effective. Therefore, it was considered that use of Chamomile essential oil is effectiveness than Thyme essential oil by essential oil used for recovery after damage of inflammation etc..

Inhibition of Contact Dermatitis in Animal Models and Suppression of Proinflammatory Gene Expression by Topically Applied Flavonoid, Wogonin

  • Wogonin (5,7-dihydroxy-8-methoxyflavone) is a down-regulator of cyclooxygenase-2 and inducible nitric oxide synthase expression, contributing to anti-inflammatory activity in vivo. For further characterization of modulatory activity on ploinflammatory gene expression in vivo, the effect of wogonin was examined in this experiment using animal models of skin inflammation. By topical application, wogonin inhibited an edematic response as well as ploinflammatory gene expression against contact dermatitis In mice. Wogonin inhibited ear edema ($19.4-22.6\%$) at doses of $50-200\;{\mu}g$/ear and down-regulated interleukin-$1{\beta}$ induction ($23.1\%$) at $200{\mu}g$/ear in phenol-induced simple irritation. Wogonin ($2{\times}50-2{\times}200{\mu}g$/ear) also inhibited edematic response ($51.2-43.9\%$) and down-regulated ploinflammatory gene expression of cyclooxygenase-2, interleukin-$1{\beta}$, interferon-$\gamma$, intercellular adhesion molecule-1 and inducible nitric oxide synthase with some different sensitivity against picryl chloride-induced delayed hypersensitivity reaction. All these results clearly demonstrate that wogonin is a down-regulator of ploinflammatory gene expression in animal models of skin inflammation. Therefore, wogonin may have potential for a new anti-inflammatory agent against skin inflammation.

NF-κB 조절을 통한 오매추출물의 항염효과 및 작용기작에 관한 연구 (Study on the Anti-inflammatory Effect and Mechanism of Prunus mume Extract Regarding NF-κB)

  • NF-${\kappa}B$ is a transcriptional factor which is involved in many biological processes including immunity, inflammation, and cell survival. Many investigators studied on the mechanism involved in activation of NF-${\kappa}B$ signalling pathway via ubiquitination and degradation of $I{\kappa}B$ regarding skin disease. Some specific molecules including Akt, MEK, p38 MAP Kinase, Stat3, et al. represent convergence points and key regulatory proteins in signaling pathways controlling cellular events such as growth and differentiation, energy homeostasis, and the response to stress and inflammation. Ultraviolet (UV) irradiation has many adverse effects on skin, including inflammation, alteration in the extracellular matrix, cellular senescence, apoptosis and skin cancer. Prunus mume, a naturally derived plant extract, has beneficial biological activities as blood fluidity improvement, anti-fatigue action, antioxidative and free radical scavenging activities, inhibiting the motility of Helicobacter pyolri. Previous reports on various beneficial function prompted us to investigate UVB-induced or other immunostimulated biological marker regarding P. mume extract. P. mume extract suppresses UVB-induced cyclooxygenase-2 (COX-2) expression in mouse skin epidermal JB6 P+ cells. The activation of activator protein-1 and nuclear factor-${\kappa}B$ induced by UVB was dose-dependently inhibited by P. mume extract treatment. This results suggest that P. mume extracts might be used as a potential agents for protection of inflammation or UVB induced skin damage.

자외선으로 손상된 마우스 피부에 대한 라벤더 오일 효과 (Effect of Lavender Oil on the Ultraviolet-Damaged Mice Skin)

  • In this study, the backs with a hair cut of 6-week-old healthy ICR male mice were once exposed to a dose of $400\;mJ/cm^2$ UVB. An acute dermal inflammation was observed, and the certified 100% pure and natural lavender essential oil were applied to the UVB-exposed mice skin twice a day. It was observed that the mice exposed to UVB resulted in an acute inflammation, and when treated with lavender oil the degree of inflammation was much alleviated, and the inflamed skins of both the control and lavender oil-treatment groups were cured almost completely after 6 days of the UVB exposure. At 24 hours after UVB exposure, the epidermal keratinocytes in the control group showed a cell-membrane damage with the destruction of intercellular junctions, agglutination of tonofilaments within the cytoplasm and nucleus damage, while the lavender oil-treatment group had much less cell damage than the control group. While the control group showed a significant increase (p<0.05) in the activity of XO up to 144 hours, the lavender oil-treatment group did not show any significant increase except for 48 hours after the UVB exposure. Both the control and lavender essential oil-treatment groups had a significant decrease in the activities of CAT and SOD up to 96 hours. Particularly, the CAT activity was significantly lower(p<0.05) in the lavender oil-treatment group than the control group up to 48 hours, and higher than the control group at and after 96 hours. The GST activity was significantly decreased in both the control and lavender oil-treatment groups up to 96 hours after the UVB exposure except for the control group at 24 hours, and that of the lavender oil-treatment group was higher than the control group at and after 96 hours. Therefore, it is assumed that the application of the lavender oil to the ultraviolet-damaged mice skin can be effective in treatment for the damaged skin.

천연물 도포가 UVB 파로 손상된 C57BL/6 mouse 피부의 색소침착과 염증생성에 미치는 영향 (Effects of Natural Extracts on UVB-induced Pigmentation and Inflammation in C57BL/6 Mouse Skin)

  • Ultraviolet(UV) radiation causes a variety of biological effects on the skin, including inflammation, pigmentation, photoaging and cancer. Free radicals are involved in inflammatory skin reactions induced by UVB radiation. In this study, we investigated the effects of antioxidants(Tea, Korean red ginseng, Ginkgo biloba extract) on UVB-induced skin damage. Tea, KRG and EGb 761 were topically treated to dorsal skin of ICR mouse. The mice were also treated soon after IMED ($1.4KJ/m^{2}$) of UVB irradiation. Skin pigmentation of irradiated mouse was observed by a chromameter after 2 weeks. Topical application of Tea, KRG and EGb 761 for 2 weeks decreased skin pigmentation compared to DVB control group(p<.05). Tea, KRG and EGb 761 also reduced UVB-induced infiltration of inflammatory cells. These results showed that Tea, KRG and EGb 761 as a topical application may have preventive effect against UVB-induced skin damage.

Oxidative Stress and Skin Diseases: Possible Role of Physical Activity

  • Background: The skin is the largest body organ that regulates excretion of metabolic waste products, temperature, and plays an important role in body protection against environmental physical and chemical, as well as biological factors. These include agents that may act as oxidants or catalysts of reactions producing reactive oxygen species (ROS), reactive nitrogen species (RNS), and other oxidants in skin cells. An increased amount of the oxidants, exceeding the antioxidant defense system capacity is called oxidative stress, leading to chronic inflammation, which, in turn, can cause collagen fragmentation and disorganization of collagen fibers and skin cell functions, and thus contribute to skin diseases including cancer. Moreover, research suggests that oxidative stress participates in all stages of carcinogenesis. We report here a summary of the present state of knowledge on the role of oxidative stress in pathogenesis of dermatologic diseases, defensive systems against ROS/RNS, and discuss how physical activity may modulate skin diseases through effects on oxidative stress. The data show duality of physical activity actions: regular moderate activity protects against ROS/RNS damage, and endurance exercise with a lack of training mediates oxidative stress. These findings indicate that the redox balance should be considered in the development of new antioxidant strategies linked to the prevention and therapy of skin diseases.
