Effects of hyperbaric oxygen and α-tocopherol on full-thickness skin grafts in rats (쥐(rats)의 full-thickness skin grafts에 대한 hyperbaric oxygen과 α-tocopherol의 효과)

  • To document that effects of hyperbaric oxygen(HBO) and ${\alpha}-tocopherol$ on full-thickness skin grafts in rat, we performed full-thickness skin grafts bilaterally on each rats. The HBO-treated rats were received HBO twice daily for 90 minutes at 2 ATA. Surgical control rats were not treated with HBO. ${\alpha}-tocopherol$ treated rats were received the agent via oral gastric tube daily for 3 days preoperative and a fourth dose 1 to 2 hours postoperative. HBO plus ${\alpha}-tocopherol$ treated rats were received HBO and ${\alpha}-tocopherol$ as mentioned above. Biopsy specimens were taken from each rat at the time of grafting and on days 2, 4, 7, 10, 14, 21, and 28, then were processed for tissue-concentration of total glutathione(GSHt), oxidized/reduced glutathione level, and thiobarbituric acid-reactive substance(TBARS) levels. The percentage of viable graft on day 10 ranged from 67 to 93%, and was not significantly different among the each other groups. The percentage of viable graft were, however, higher in HBO plus ${\alpha}-tocopherol$ treated rats(78.6%) than in HBO alone treated rats(59.1%), ${\alpha}-tocopherol$ alone treated rats(66.7%) and surgical control rats(58.2%). TBARS concentration had a significant increase from preoperative concentration at day 2, and peak concentration at day 4(p<0.01). Concentration then decreased to preoperative concentration at day 28. GSHt concentration of free skin graft had a similar patteren of change in four groups and decreased significantly from preoperative concentration at day 2, returning to preoperative concentration by day 7(surgical control, HBO-treated, and ${\alpha}-tocopherol-treated$, alone) and 28(HBO plus ${\alpha}-tocopherol-treated$). Percentage of the concentration of reduced glutathione decreased in surgical control, HBO-treated and, ${\alpha}-tocopherol-treated$(p<0.05), and HBO plus ${\alpha}-tocopherol-treared$(p<0.01) on day 7 after surgery, whereas the concentration of oxidized increased significantly in HBO-treated(p<0.05), ${\alpha}-tocopherol-treated$(p<0.05), and HBO plus ${\alpha}-tocopherol-treated$(p<0.01).

Acute Oral Toxicity and Skin Irritation Studies on Lamia-Kill$^{(R)}$ Composed of Benzalkonium Chloride and Citric Acid (염화벤지코늄과 구연산을 주성분으로 하는 살균 소독제 라미아-킬에 대한 급성경구독성 및 피부자극성 시험에 관한 연구)

  • This test was performed to evaluate the acute oral toxicity and skin irritation of Lamia-Kill$^{(R)}$, disinfectant, containing 20% benzalkonium chloride and 10% citric acid. In acute oral toxicity, Lamia-Kill$^{(R)}$ was orally administered at dose levels of 2,000, 1,000, 500, 250 and 0 mg/kg body weight. After single oral administration to both sexes of SD rats, the rats were observed for 14 days. In primary skin irritation test, New Zealand white rabbits were dermally treated with Lamia-Kill$^{(R)}$ for 24 hr and observed for 3 days. All rats treated with Lamia-Kill$^{(R)}$ were induced no toxic signs in mortalities, clinical findings, body weights and gross findings. Also, the disinfectant did not induce any adverse reactions such as erythema and edema on intact skin sites for the most part rabbits, but on abraded skin sites, some rabbits showed very slight erythema on 24 hr after topical application. With the results of this study, Lamia-Kill$^{(R)}$ have no effect on acute toxicity and side effect in SD rats and was classified as a practically non-irritating material based on the score 0.50 of primary irritation index.

Entrance Skin Dose and Image Quality Evaluation According to Use Grid Radiography for the Extremity in FPD System (FPD System에서 상.하지 촬영 시 격자에 따른 환자 선량 및 화질 평가)

  • By accessing the current status of FPD system use in the hospitals located in Seoul and Gyeonggi Province as well as the entrance skin dose and the image quality evaluation realized by C-D Phantom, and the image assessment by the medical professionals regarding the radiography for the extremity, the following results were derived. 1. According to the evaluation made in the actual use of FPD system (12 machines), the grid ratio varied from 8:1 to 13:1, and 6 machines used the grid ratio with 12:1, realizing the largest number. Among the machines, there were 8 machines that allowed a removable grid while 3 machines did use a removable grid (25.0%). 2. When it came to the equipments used for the experiment, it showed that the amount of the entrance skin dose increased from 4.13 times up to 4.79 times with the grid use. 3. The difference in the entrance skin dose depending on the changes in the exposure condition(0.5times or 2.0times) was not significantly different regardless of the patients' thickness. 4. In terms of the image quality depending on C-D Phantom, the grid use was distinguished well. However, the images were well distinguishable as the exposure condition got increased. 5. In the clinical assessment, the grid use was less effective for the Hand PA, which was considered to shoot a thin body part. It was evaluated that the grid use was preferred for the Knee AP, which was shooting for a relatively thick body part. Nonetheless, 3 out of 5 people said that they would not use the grid if the entrance skin dose to reduced.

  • Centella asiatica (Linn.) Urban (Umbelliferae) which is also known as 'pegaga' is highly consumed and eaten raw as 'ulam' in Malaysia. C. asiatica is used in traditional medicines to treat various health conditions such as rheumatism, inflammation, syphilis, skin diseases and diarrhoea. Various reports exhibited that the crude extracts and isolated bioactive compounds of C. asiatica possessed a broad range of pharmacological activities such as anti-oxidant, anti-diabetic, anti-tumor, wound healing, anti-microbial, anti-inflammatory, immunomodulatory, hepatoprotective and memory enhancing properties. The pharmacological validation on anti-microbial and immunomodulatory of C. asiatica is very limited and several existence review papers related for this plant had not been focused for both activities. This review therefore attempts to combine the existing literature to offer immense scope for researchers engaged in validation of the traditional claims and bioactivities of this plant related with anti-microbial and immunomodulatory potential.

Utilization of School Health Room and Health Problems among Elementary and Secondary School Students, and Nursing Performance of School Health Teachers in a City (일 도시 초·중등 학생의 보건실 이용률과 건강문제 및 보건교사의 간호수행도)

  • The purpose of this study was to identify the utilization of school health room and the health problems among elementary and secondary school students as years go by sex, school locations, and to analyze the frequency and difficulty of nursing interventions performance that school health teachers was available to resolve students' health problems at school health room. Participants were fifty nine school health teachers with more than 3 years of career. As the years go by, utilization of school health room has gradually increased. Utilization of female than male was higher than 200%, i.e. Elementary school girls in urban area were the highest rate, and secondary school boys in urban area were the lowest. In male, utilization of school health room was in order the skin and subcutaneous, musculoskeletal, and digestive problems, and also was in order the skin and subcutaneous, digestive, and musculoskeletal problems in female. Utilization of in urban elementary school was higher on patricular health problem than in suburban areas. Nursing interventions of most often using were self-care management & education and counselling. Nursing intervention was the highest degree of difficulty in self-harm, seizure, circulatory health problems. In conclusions, utilization of school health room has gradually increased; therefore workload of school health teacher aggravated. To perform a heath teacher to effectively operate the general and emergency health issues, the expansion of qualified school health teachers is needed, and the standardized guidelines and refresher training programs is required.

  • UV irradiation causes skin-aging involving coarse wrinkles, thickening, dyspigmentation, and rough skin surface. These phenomena in complex skin tissue is controlled with receptor of cell surface growth factor and cytokine receptors. The activation of receptors induces multiple downstream signaling pathways including expression of MMPs (matrix metalloproteinases). This study was aimed to elucidate the mechanism for anti-wrinkle activity of Korean red ginseng, Torilis fructus and Corni fructus mixture (KTNG0345). In this animal study, we have investigated decreasing effects of Korean red ginseng mixture on MMP-3 synthesis through diminishing $TNF-{\alpha}$ signaling that express MMP-1, -3, and -9. c-Jun and c-fos as a component of transcription factor AP-1 (activator protein-1) were analyzed the expression level using real time PCR and western blotting. c-Jun was decreased dose dependent manner both gene and protein level where as cfos was not changed. In upstream, JNK and PAK was not changed, but p38 was decreased in down stream. MMP-3, final product in this pathway was significantly decreased in dose dependent manner. These results suggest that Korean red ginseng mixture have a anti-wrinkle activity through $TNF-{\alpha}$ mediated MMPs expression pathway.

The Skin Improvement Effect of Facial mask pack using Chickpea natto fermented with Bacillus subtilis natto (Bacillus subtilis natto 균주로 발효한 병아리콩 낫토를 이용한 mask pack의 피부 개선 효과)

  • In order to utilize health food chick bean as a raw material for cosmetics, the skin improvement effect of a mask pack made of chickpea Natto fermented with Bacillus subtilis Natto strain was studied. Powder obtained by lyophilization of chickpea Natto was applied to a facial mask pack as a paste formulation. This Natto pack was applied to subjects in their 50s and 60s to investigate their effect on skin improvement. As the number of treatments of Natto pack increased, the skin was improved and the improvement effect was remarkable after 4 treatments. After four times treatments, moisture content and sebaceous secretion increased by $8.4{\pm}3.6%p$ and $4.0{\pm}2.3%p$, respectively. Skin pores and wrinkles were also decreased by $1.8{\pm}0.3%p$ and $1.8{\pm}0.9%p$, respectively. Skin pigmentation decreased by $1.3{\pm}0.2%p$ and skin tone also increased from 55.2% to 55.9%. These results indicate that Natto product obtained by fermenting chickpea has the effect of improving skin such as moisturizing, pore, wrinkle, pigmentation, sebum secretion and skin tone, and can be used as a raw material for various functional cosmetics.

Safety Evaluation of Black Garlic Extract for Development of Cosmeceutical Ingredients -Skin irritation and Sensitization Studies- (화장품 소재로서의 흑마늘 추출물에 대한 안전성 평가 -1차 피부자극 실험 및 감작성 중심으로-)

  • We evaluated the anti-aging potential and safety of black garlic extract for cosmeceutical ingredient. Black garlic was made by spontaneous fermentation for 40 days at $60{\sim}70^{\circ}C$, 85~95% RH without any additives. The 10% black garlic extract had sweet odor, antioxidant activities and inhibitory activities of skin againg enzymes such as tyrosinase and elastase. The skin safety was performed to evaluate of potential toxicity using the primary irritation test and skin sensitization test. The black garlic extract did not show any adverse reactions such as erythema and edema on intact skin sites at primary irritation test, but on abraded sites, some experimental animals showed very slight erythema. So, the black garlic extract was classified as a practically non-irritating material based on the score 0.23 of primary irritation index. The skin sensitization study was tested by the guinea pig maximization test (GPMT) and Freund's complete adjuvant (FCA) with intradermal injection of 10% black garlic extract. The skin sensitization test showed no skin sensitization. The allergic sensitization depends on tumor necrosis factor-$\alpha$ (TNF-$\alpha$) and interleukin-6 (IL-6). The concentration of IL-6 on challenged tissue of treated with black garlic extract was not significantly different with negative control group (saline treated group). Based on this study, the potential for black garlic as a cosmeceutical ingredient was proven.

The Development of Diagnosis for Atopic Dermatitis by Evaluating the Expression of Skin Barrier Proteins Using a Non-Invasive Method (비침습적 피부장벽단백질 측정을 통한 아토피 피부염의 진단 및 유용성)

  • Atopic dermatitis is a chronic inflammatory skin disease caused by a variety of genetic and environmental factors, dysregulation of immunological response, as well as dysfunction of the skin barrier proteins. The purpose of this study is to develop an ELISA kit suitable for evaluating the expression of skin barrier proteins. Proteins were obtained from the skin via AriNo and D-Squame patches. The efficiency of protein collection from the skin, using the Arino patch, was shown to be more effective than using D-Squame; while the efficiency of lysis using 0.1% Triton-X100 was higher than that of other lysis solutions, including 0.1 M Tris-HCL, 0.1% Tween-20, and 5 mM KOH. Recombinant skin barrier proteins, such as filaggrin and involucrin, were produced by molecular biological methods. Monoclonal antibodies against filaggrin and involucrin were produced by immunization of mice, fusion of spleen cells and myeloma cells, as well as a selection of antibody-producing hybridoma cells. The filaggrin expression in the skin of subjects suffering from atopic dermatitis was lower than that in normal mice. Involucrin expression was not altered between normal individuals and subjects with atopic dermatitis. These findings contribute to an elucidation of the importance of the skin barrier protein expression in atopic dermatitis and the development of a diagnostic kit for atopic dermatitis.

Influencing Factors and Interactions among the Skin Microbiomes in Affecting Detrimental Bacteria (피부 마이크로바이옴의 요인과 상호작용이 유해균에 미치는 영향에 대한 연구)

  • This study was conducted to empirically analyze the effects and interactions among beneficial bacteria, commensal bacteria, and acne bacteria, which are factors in the skin microbiomes, on detrimental bacteria by 289 people, who are 20 to 49 years old among Koreans. As a result of multiple regression models using bio big data of skin microbiomes, when the difference in skin microbiomes according to the sex and age of the subjects was controlled, the beneficial bacteria showed a negative (-) effect on the detrimental bacteria, while the commensal and acne bacteria showed a positive (+) effect. Particularly, the negative (-) effect of beneficial bacteria on detrimental bacteria was different through interaction with acne bacteria according to the level of commensal bacteria. These results demonstrate that the activation of beneficial bacteria inhibits detrimental bacteria, and the effect of skin microbiomes on detrimental bacteria is balanced with skin microbiomes through interaction with independent influence. Therefore, it is suggested that when studying skin microbiomes products to help the proliferation of beneficial bacteria and to create a skin environment that inhibits detrimental bacteria in the personalized cosmetics manufacturing industry, it is necessary to consider the independent effects and interactions among skin microbiome factors together.
