입술은 구조적으로 일반 피부와 다른 특유의 각질, 보습 특성을 갖는다. 특히, 각질의 턴오버 주기가 빠르고 최외곽 각질층이 얇아 비교적 각질의 발생량이 많고, 부속기관이 존재하지 않아 수분 유지에 매우 취약한 환경을 가지고 있다. 본 연구에서는 새롭게 개발될 립케어 제품의 타겟을 각질-보습-장벽 세 가지로 설정하고 각 맞춤 소재의 개선 가능성을 확인하는 것을 목표로 하여 각질 박리 효능이 있는 소재 3 종에 대해, 각질, 보습, 장벽 개선 효능을 검증하였다. 구체적으로 Bacillus clausii extract에 의한 각질 개선, 글루코노락톤에 의한 피부 수분 보유량 개선, 세린에 의한 장벽 기능 강화 효능을 확인하였다. 립케어의 인체 효능 검증을 위해 각 효능 성분의 자극이 없는 범위의 농도를 설정하여 립밤 제조 및 인체 효능 평가를 진행하였고, 입술 각질·보습·탄력·주름 개선의 효능이 있음을 확인할 수 있었다. 결과적으로 입술의 각질-보습-장벽 케어를 통해 입술의 미용, 건강 지표가 개선될 수 있음을 확인하고 해당 소재들을 립세린 제품에 적용하였다.
호장은 마디풀과에 속하는 다년초로 우리나라를 포함한 동아시아에서 잘 자생한다. 호장근은 호장의 뿌리로 항염증과 진경제로 사용되어 왔으며 유효성분으로 emodin을 포함하고 있다. 피부의 표피는 자극, 해로운 물질을 차단하고 수분 증발을 방지함으로써 체내를 보호하는 중요한 역할을 한다. 본 연구에서는 호장근과 그 유효성분인 emodin이 피부 장벽과 보습능에 미치는 영향에 대해 평가하고자 하였다. 먼저 호장근은 ABTS+ radicals을 우수하게 제거함으로써 항산화 효능이 뛰어남을 확인하였다. 다음으로, 실시간 중합연쇄효소반응을 통해 각질형성세포의 분화에 중요한 역할을 하는 filaggrin의 유전자 발현을 비교한 결과, 호장근과 emodin에 의해 농도-의존적으로 filaggrin mRNA 발현이 증가하였다. 또한, 호장근과 emodin은 히알루론산 합성에 중요한 역할을 하는 HAS-2 mRNA 발현을 유의하게 증가시키는 것으로 나타났다. 종합적으로, emodin을 유효성분으로 포함하는 호장근은 피부 장벽 강화와 보습능 증강을 위한 기능성 화장품 소재로서 활용될 수 있을 것으로 기대된다.
Atopic dermatitis (AD) is a chronic inflammatory skin disease associated with cutaneous hyperreactivity to environmental triggers. The clinical phenotype that characterizes AD is the product of interactions between susceptible genes, the environmental factors, defective skin barrier function, and immunologic responses. This review summarizes recent progress in our understanding of the immunopathophysiology of AD and the implications for mouse models of AD in drug discovery from medicinal plants.
천연보습인자(NMF, natural moisturizing factor)는 정상 피부 내의 각질층에 존재하는 수용성, 친수성 성분을 총칭하는 말로 외부로부터 수분을 흡수하고 각질층의 수분보유능을 증가시키며 피부가 건조해지는 것을 방지하여 피부의 유연성과 가수성을 유지하는 기능을 한다. NMF는 주로 아미노산과 아미노산의 대사산물로 구성되며 이들은 필라그린(filaggrin)이 분해되면서 생성된다. 본 연구에서는 피부 각질층의 22종의 개별 아미노산의 함량분석을 통하여 피부보습능과 개개의 아미노산과의 상관관계를 연구하였다. 15명의 건강한 지원자들의 전박과 이마로부터 tape stripping한 각질 시료 중 22종의 아미노산 함량을 UPLC-PDA를 이용하여 동시 분석하였다. 그 결과 각질 표면보다는 안쪽에서의 아미노산들의 함량이 높음을 확인하였다. 또한 신체 부위별 비교에서 전박이 이마에 비해 아미노산 함량이 1.5배 정도 높음을 확인하였다. 전박부위의 아미노산 총 함량과 보습능(TEWL)과의 상관 관계분석을 통해서는 보습능이 클수록 총아미노산 함량이 높은 것으로 확인되었으며 특히 Ser, Glu, Gly, Ala 및 Thr은 각질 내 존재하는 주요 아미노산 성분으로 보습에 긍정적인 영향을 주는 성분으로 확인되었다. 이상의 결과로써 NMF의 주성분인 아미노산의 함량은 각질 생리기능과 밀접한 관련을 가지고 있음을 확인할 수 있었으며 본 연구를 통해 얻은 각질층의 아미노산의 통합적인 분석법은 보습, 노화, 미백, 피부 염증질환 등 다양한 피부 상태와 아미노산류들의 변화와의 관계를 깊이 이해하고 피부 기능과 관련된 새로운 분자적 타겟을 발굴하고 증명하는데 기초적인 방법으로써 사용될 수 있을 것으로 사료된다.
리소좀(lysosome)은 진핵세포에서 에너지 대사 및 세포 내 소화 작용에 관여하는 세포 소기관으로 protease, nuclease, glycosidase, lipase, phosphatase 들이 다수 존재한다. 우리는 선행 연구결과들을 통해 난백 리소좀의 멜라닌 색소 탈색능을 보고하였다[8]. 그러나 B16F10 melanocyte 세포주에서 난백 리소좀에 의한 멜라닌 함량 변화 및 피부장벽 조절 연구는 거의 보고되지 않았다. 따라서 우리는 계란 난백(egg white)으로부터 추출한 lysosome-related organelle extract (LOE)에 의한 세포 내 멜라닌 함량 변화 및 피부장벽 강화 효과를 규명하고자 하였다. 먼저 LOE의 미백 효능을 확인하기 위해 B16F10 세포주를 이용하여 세포독성 평가를 진행하였다. B16F10 세포주에서 LOE에 의한 세포독성은 0에서 20 mg/mL 농도에서 관찰되지 않았으나, 40 mg/mL 부터 세포독성이 관찰되어 이후 모든 실험에서 최대 농도값을 20 mg/mL로 설정하였다. 먼저 LOE를 이용한 melanin contents assay 결과, 음성 대조군인 α-MSH 처리군 대비 LOE 처리군 5, 10, 20 mg/mL 농도에서 61.5 ± 4.0%, 61.4 ± 7.3%, 58.3 ± 8.3%로 세포 내 멜라닌 함량이 감소되는 것을 확인하였고, 20 mg/mL 농도 조건에서 MITF 발현 억제도 관찰하였다. LOE의 피부 장벽에 미치는 영향을 관찰하기 위해 각질형성세포주(HaCaT)를 이용하여 TEER (trans-epithelial electrical resistance) assay를 수행한 결과, LOE에 의해 농도 의존적으로 TEER 저항값이 증가하여 LOE가 피부장벽 강화에도 효과가 있음을 알 수 있었다. 또한 피부 염증 유발을 위한 TNF-α 처리조건에서도 LOE는 TEER 저항값을 증가시켜 염증 유발 조건에서도 LOE에 의해 피부장벽이 정상적으로 회복되었음을 알 수 있었다. 마지막으로 cell migration assay를 통해 LOE에 의한 세포이동 촉진 효과를 관찰한 결과, LOE는 세포분열 및 세포이동을 촉진시켰다. 위 결과들을 통해 LOE는 미백 기능 뿐 아니라 피부재생 및 피부장벽 강화에도 효과를 나타내는 소재이며, 효소안정화 및 제형화 기술이 접목된다면 향후 새로운 미백 기능성 화장품 소재로도 개발될 수 있을 것이다.
아토피 피부염을 설명하는 데는 두 가지 패러다임이 있는데, 첫 번째는 outside-inside paradigm이고 두 번째는 inside-ouside paradigm이다. Outside-inside paradigm에 의하면 아토피 피부염을 치료하기 위한 가장 좋은 방법은 피부 장벽 기능을 회복시키는 것이다. 장벽 기능은 각질층의 특징적인 구조에 의해 유지되는데, 각질층은 층상구조의 inter-cellular lipids와 corneocyte로 구성되어 있다. 아토피 피부염 환자, 층판상 어린선 환자의 각질층 내 층상구조는 hexagonal 구조가 많이 존재하고 있음을 보였으며, 이는 Ceramide 1 타입과 long chain fatty acid의 감소 때문이라고 보여진다. 이러한 이유에서 라멜라구조를 가지며 intercellular long periodicity phase 기능을 복원할 수 있는 보습제는 각질층의 intercellular lipid의 층상구조를 복원시킬 수 있을 것이다. 또한 세라마이드와 유사세라마이드를 함유한 보습제는 손상된 피부장벽, 피부질환, 아토피 피부염을 위한 유용한 치료제로 사용할 수 있다. Inside-outside paradigm에 따르면, 아토피 피부염 등이 보조 T cell, lg E, eosinophil 등을 포함한 면역반응과 관계되는데, 특히 Th1/Th2 imbalance의 복원이 아토피 피부염을 치료하는데 효과적일 것이다. 현재 여러 종류의 면역억제제가 소개되고 있지만, 이들은 고유 면역기능을 감소시킬 우려를 가지고 있다. 따라서, 세라마이드 대사산물인 SPC와 S1P는 면역조절 기능 및 상처치유의 기능을 가짐으로써 향후 관심이 집중될 것이다.

Patients with diabetes mellitus (DM) often suffer from diverse skin disorders, which might be attributable to skin barrier dysfunction. To explore the role of lipid alterations in the epidermis in DM skin disorders, we quantitated 49 lipids (34 ceramides, 14 free fatty acids (FFAs), and cholesterol) in the skin epidermis, liver, and kidneys of db/db mice, a Type 2 DM model, using UPLC-MS/MS. The expression of genes involved in lipid synthesis was also evaluated. With the full establishment of hyperglycemia at the age of 20 weeks, remarkable lipid enrichment was noted in the skin of the db/db mice, especially at the epidermis and subcutaneous fat bed. Prominent increases in the ceramides and FFAs (>3 fold) with short or medium chains ($LXR{\alpha}/{\beta}$ and $PPAR{\gamma}$, nuclear receptors promoting lipid synthesis, lipid synthesis enzymes such as elongases 1, 4, and 6, and fatty acid synthase and stearoyl-CoA desaturase were highly expressed in the skin and livers of the db/db mice. Collectively, our study demonstrates an extensive alteration in the skin and systemic lipid profiles of db/db mice, which could contribute to the development of skin disorders in DM.

Monoolein-based cubic liquid crystalline systems were formulated for the local delivery of oregonin and hirsutanonol for the treatment of atopic dermatitis. The liquid crystalline phase and its nanodispersion containing drugs were prepared. The skin permeation and deposition properties of the drugs were examined in normal and delipidized rat skin. The proportion of oregonin (%) deposited in normal skin after topical administration of the drugs in the form of aqueous solution, cubic phase or cubic nanodispersions were $1.53\;{\pm}\;0.46$, $3.62\;{\pm}\;0.17$ and $5.13\;{\pm}\;0.73$, and those of hirsutanonol were $2.46\;{\pm}\;0.02$, $5.44\;{\pm}\;0.27$ and $17.28\;{\pm}\;2.19$, respectively. The greater lipophilicity and thus greater skin affinity of hirsutanonol than oregonin contributed the greater amount of skin deposition. The monoolein-based liquid crystalline phases significantly increased the amount of both drugs permeated and deposited. Approximately 3.2, 2.1 and 3.0 times greater amount of oregonin, and 3.4, 2.1 and 2.2 times greater amount of hirsutanonol were deposited in delipidized skin after administration of each drug in the form of aqueous solution, cubic phase and cubic nanodispersions system, respectively, because of lowered barrier function of the delipidized skin. In this study, the effects of drug property, vehicles type and skin condition on skin deposition and permeation properties of drug were examined and concluded that monoolein-based liquid crystalline systems would be a promising formulation for the local delivery of drugs.
