Identification of Candidate SNP (Single Nucleotide Polymorphism) for Growth and Carcass Traits Related to QTL on Chromosome 6 in Hanwoo (Korean Cattle)

  • As genetic markers, single nucleotide polymorphisms (SNP) are very appropriate for the development of genetic tests for economic traits in livestock. Several microsatellite markers have been identified as useful markers for the genetic improvement of Hanwoo. Among those markers, ILSTS035 was recently mapped at a similar position with four SNPs (AH1_11, AH1_9, 31465_446, and 12273_165) in a linkage map of EST-based SNP in BAT6. Among the four SNPs, two SNPs (31465_446 and 12273_165) were analyzed using BLAST at the NCBI web site. The sequences including the 12273_165 SNP were identified at the intron region within the LOC534614 gene on the gene sequence map (Bos taurus NCBI Map view, build 3.1). The LOC534614 gene represents a protein similar to myosin heavy chain, fat skeletal muscle, embryonic isoform 1 in the dog, and myosin_1 (Myosin heavy chain D) in Macaca mulatta. In cattle, the myosin heavy chain was associated with muscle development. The phenotypic data for growth and carcass traits in the 415 animals were analyzed by the mixed ANCOVA (analysis of covariance) linear model using PROC GLM module in SAS v9.1. By the genotyping of Hanwoo individuals (n = 415) to evaluate the association of SNP with growth and carcass traits, it was shown that the 12273_165 SNP region within LOC534614 may be a candidate marker for growth. The results of the statistical analyses suggested that the genotype of the 12273_165 SNP significantly affected birth weight, weight of the cattle at 24 months of age, average daily gain and carcass cold weight (p<0.05). Consequently, the 12273_165 SNP polymorphisms at the LOC534614 gene may be associated with growth in Hanwoo, and functional validation of polymorphisms in LOC534614 should be performed in the future.

확장 13-Element EPSAR 안테나를 사용한 7×7 MIMO 시스템 (7×7 MIMO System Using Extended 13-Element ESPAR Antenna)

  • MIMO 시스템은 통신 시스템의 채널 용량을 증가시키기 위해서 다양한 분야에서 사용된다. 그러나 이러한 통신 시스템은 다수의 RF 단을 필요로 하기 때문에 안테나 소형화에 문제가 있다. 게다가 다수의 RF 단으로 인하여 아날로그 회로에서의 전력 소모가 증가하고, RF 단의 간섭으로 인하여 통신 효율이 심각하게 저하된다. 이러한 이유 때문에 단일 RF 단을 가지면서도 MIMO 통신이 가능한 BS-MIMO 통신 방식이 제안되었다. BS-MIMO 시스템은 기본적으로 ESPAR 안테나를 사용한다. 기존의 ESPAR 안테나는 5-element 구조를 가지고 있으며, $3{\times}3$ MIMO 전송 까지만 가능하다. 그러므로 MIMO 차원을 확장시키기 위해서는 ESPAR 안테나의 구조 확장이 필수적이다. 본 논문에서는 단일 원 형태의 ESPAR 안테나의 구조 확장을 통해서 BS-MIMO 의 차원을 기존 MIMO 기술처럼 증가시킬 수 있음을 보였다. 설계의 예로써, 13-ESPAR 안테나를 사용하면 $7{\times}7$ BS-MIMO 전송이 가능함을 보였다. 또한 기생 배열안테나의 수가 2개씩 증가할수록 전송 가능한 MIMO 차원이 일씩 증가한다.

Synthesis and Crystal Structure of a New Pentanary Thiophosphate, K0.5Ag0.5Nb2PS10

  • Single crystals of the new pentanary thiophosphate, $K_{0.5}Ag_{0.5}Nb_2PS_{10}$ has been prepared through reactions of the elements with halide fluxes. The structure of $K_{0.5}Ag_{0.5}Nb_2PS_{10}$ has been analyzed by single crystal X-ray diffraction technique. The structure of $K_{0.5}Ag_{0.5}Nb_2PS_{10}$ is made up of one-dimensional ${^{\infty}_1}[Nb_2PS_{10}]$ chains along the [001] direction and these chains are separated from one another by $Ag^+$ and disordered $K^+$ ions. This chain is basically built up from bicapped trigonal prismatic [Nb2S12] units and tetrahedral [PS4] groups. The [$Nb_2S_{12}$] units are connected together to form the linear chain, ${^{\infty}_1}[Nb_2S_9]$ by sharing the S-S prism edge. Short (2.885(2) $\AA$ or 2.888(2) $\AA$) and long (3.743(1) $\AA$) Nb-Nb distances are alternating along the chain, and the $S_2{^2-}]\;and\;S^{2-}$anionic species are observed. The distorted octahedral coordination around the $Ag^+$ ion can be described as [2+4] bonding interaction.

Rapid Origin Determination of the Northern Mauxia Shrimp (Acetes chinensis) Based on Allele Specific Polymerase Chain Reaction of Partial Mitochondrial 16S rRNA Gene

  • Acetes chinensis is an economically important shrimp that belongs to the Sergestidae family; following fermentation, A. chinensis' economic value, however, is low in China, and much of the catch in China is exported to Korea at a low price, thus leading to potential false labeling. For this reason, we developed a simple method to identify A. chinensis' origin using allele-specific polymerase chain reaction (PCR). Ten single nucleotide polymorphisms (SNPs) were identified from partial (i.e., 570 bp) DNA sequence analysis of the mitochondrial 16s rRNA gene in 96 Korean and 96 Chinese individual shrimp. Among 10 SNP sites, four sites were observed in populations from both countries, and two sites located in the middle with SNP sites at their 3'-ends were used to design allele-specific primers. Among the eight internal primers, the C220F primer specific to the Chinese A. chinensis population amplified a DNA fragment of 364 bp only from that population. We were able to identify the A. chinensis population origin with 100% accuracy using multiplex PCR performed with two external primers and C220F primers. These results show that the 16S rRNA gene that is generally used for the identification of species can be used for the identification of the origin within species of A. chinensis, which is an important finding for the fair trade of the species between Korea and China.

  • Application methods of chemicals were investigated to minimize strength reduction of corrugated boards under the high humidity environment encountered in the cold chain system. Starch insolubilizers were introduced in the starch solution preparation of the Stein hall method and their insolubilization effect of starch binder were estimated. The performance of water repellent agents(WRA) and moisture proof agents(MPA) were evaluated in terms of water and moisture resistance. And effects of the combination of the chemicals and the coating method were also examined. Addition of the polyamine polyamide insolubilizer to the main part in the Stein hall process improved the binding force and water resistance of starch, which contributed to minimize the strength reduction of paper under the high humidity environment. AZC and Glyoxal type insolubilizers could not be used in the experiment due to an excessively increased viscosity of starch solution and the poor stability. Conventional WRA treatment to the base paper enhanced water and moisture resistance very slightly even though water repellency of the paper reached R10 by the treatment. MPA showed excellent performance than WRA not only in water and moisture resistance but in water repellency. Double coating on paper with MPA was more effective than the single coating at the same coating weight. A newly developed MPA showed excellent performance and runnability only by a single coating instead of a double coating.

Mitochondrial Efficiency-Dependent Viability of Saccharomyces cerevisiae Mutants Carrying Individual Electron Transport Chain Component Deletions

  • Mitochondria play a crucial role in eukaryotic cells; the mitochondrial electron transport chain (ETC) generates adenosine triphosphate (ATP), which serves as an energy source for numerous critical cellular activities. However, the ETC also generates deleterious reactive oxygen species (ROS) as a natural byproduct of oxidative phosphorylation. ROS are considered the major cause of aging because they damage proteins, lipids, and DNA by oxidation. We analyzed the chronological life span, growth phenotype, mitochondrial membrane potential (MMP), and intracellular ATP and mitochondrial superoxide levels of 33 single ETC component-deleted strains during the chronological aging process. Among the ETC mutant strains, 14 ($sdh1{\Delta}$, $sdh2{\Delta}$, $sdh4{\Delta}$, $cor1{\Delta}$, $cyt1{\Delta}$, $qcr7{\Delta}$, $qcr8{\Delta}$, $rip1{\Delta}$, $cox6{\Delta}$, $cox7{\Delta}$, $cox9{\Delta}$, $atp4{\Delta}$, $atp7{\Delta}$, and $atp17{\Delta}$) showed a significantly shorter life span. The deleted genes encode important elements of the ETC components succinate dehydrogenase (complex II) and cytochrome c oxidase (complex IV), and some of the deletions lead to structural instability of the membrane-$F_1F_0$-ATP synthase due to mutations in the stator stalk (complex V). These short-lived strains generated higher superoxide levels and produced lower ATP levels without alteration of MMP. In summary, ETC mutations decreased the life span of yeast due to impaired mitochondrial efficiency.

  • 본 논문은 TV 브라운관과 컴퓨터 모니터에 사용되는 마그네트(Magnet)에 일정한 자력을 갖도록 자화 시키는 착자기를 제어하며, 검사공정을 자동화하는 제어 시스템용의 집적회로를 설계하여 개발한 것에 관한 것으로써, 착자기의 콘트롤 모듈과 프로토콜 모듈의 주변기기 제어회로 부분을 0.8um CMOS SOG 기술로 설계하여 ETRI에서 공정하여 칩(Chip)을 완성시켜 동작을 확인하였다. 본 논문에서는 개별 셀(Single cell)의 지연 예측에 사용되었던 기존의 프로파게이션/램프 지연 모델(Propagation/ramp delay model)을 분석, 문제점을 보완 수정한 LODECAP(LOgic DEsign CAPture)의 인버터 선형 지연 모델을 응용하여 타이밍 콘트롤 블록 내의 지연 체인(Delay chain)을 설계 할 수 있는 새로운 "지연 예측 수식"을 제안하였다. 본 논문은 추출된 수식에 의거하여 타이밍 콘트롤 블록의 설계, 시스템에 적용하였으며, 나머지 블록들을 설계한 기법에 대하여도 상술하였다.여도 상술하였다.

Association of single-nucleotide polymorphisms in dual specificity phosphatase 8 and insulin-like growth factor 2 genes with inosine-5'-monophosphate, inosine, and hypoxanthine contents in chickens

  • Objective: This study aimed to identify the single-nucleotide polymorphisms (SNPs) in the dual-specificity phosphatase 8 (DUSP8) and insulin-like growth factor 2 (IGF2) genes and to explore their effects on inosine-5'-monophosphate (IMP), inosine, and hypoxanthine contents in Korean native chicken -red-brown line (KNC-R Line). Methods: A total sample of 284 (males, n = 127; females n = 157) and 230 (males, n = 106; females, n = 124) aged of 10 weeks old KNC-R line was used for genotyping of DUSP8 and IGF2 genes, respectively. One SNP (rs313443014 C>T) in DUSP8 gene and two SNPs (rs315806609A/G and rs313810945T/C) in IGF2 gene were used for genotyping by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and KASP methods, respectively. The Two-way analysis of variance of the R program was used to associate DUSP8 and IGF2 genotypes with nucleotide contents in KNC-R chickens. Results: The DUSP8 (rs313443014 C>T) was polymorphic in KNC-R line and showed three genotypes: CC, CT, and TT. The IGF2 gene (rs315806609A/G and rs313810945T/C) was also polymorphic and had three genotypes per SNP, including GG, AG, and AA for the SNP rs315806609A/G and genotypes: CC, CT, and TT for the SNP rs313810945T/C. Association resulted into a strong significant association (p<0.01) with IMP, inosine, and hypoxanthine. Moreover, the significant effect of sex (p<0.05) on nucleotide content was also observed. Conclusion: The SNPs in the DUSP8 and IGF2 genes might be used as genetic markers in the selection and production of chickens with highly flavored meat.

대장균의 alkaline phosphatase가 융합된 anti-DR4 single-chain Fv (ScFv) 항체의 개발 (The development of anti-DR4 single-chain Fv (ScFv) antibody fused to Escherichia coli alkaline phosphatase)

  • 항체의 특이적 결합을 분석하는 효소면역분석법은 항원의 탐지를 위해 주로 horseradish peroxidase (HRP) 또는 alkaline phosphatase (AP) 등의 효소를 사용한다. 이때 효소를 주로 화학적으로 항체에 결합시켜 사용하게 되는데 이 과정이 복잡하며 불규칙하게 일어나서 항체 및 효소의 기능을 감소시키게 된다. 또한 대부분의 효소면역분석법에서는 주로 일차 항체의 항원결합을 탐지하기위해 이차 항체를 사용하는데, 즉 이차 항체에 결합한 효소의 기질발색에 의해 일차 항체의 항원결합을 탐지하므로 이차 항체가 요구 되어질 뿐만 아니라 이차 항체의 일차 항체에 대한 반응을 위한 부가적인 배양시간이 필요하다. 더욱 더 중요한 것은 이차 항체만의 비특이적 항원 결합 역시 제거되어져야 한다. 본 연구에서는 대장균의 genomic DNA로부터 PCR을 통해 alkaline phosphatase 유전자(Sadeghi et al., 2008)를 증폭 분리한 다음 이를 TRAIL (tumor necrosis factor ${\alpha}$ related apoptosis induced ligand) receptor인 death receptor 4 (DR4)에 특이적으로 결합하는 hAY4 single-chain Fv (ScFv)에 융합시킨 재조합 ScFv-AP 형태로 대장균에서 발현시켜 정제하였다. 정제된 hAY4 ScFv-AP는 SDS-PAGE에서 단량체(monomer) 분자량인 73.8 kDa을 나타내었다. 그러나 size-exclusion chromatography(SEC)에서는 147.6 kDa을 나타내는 결과를 통해 hAY4 ScFv-AP는 AP의 자연적인 비공유결합에 의해 이량체(dimeric form)형성이 유도되어짐을 확인하였다. 또한 ELISA, Western blot 그리고 immunocytochemistry에서 이차 항체 없이 일차 항체 hAY4 ScFv에 직접 융합된 AP의 기질발색에 의해 ScFv 일차 항체의 특이적 항원결합을 나타내었다. 요약하면 hAY4 ScFv와 대장균의 alkaline phosphatase 유전자를 융합시켜 대장균에서 수용성 형태로 성공적으로 정제하였으며 정제된 ScFv-AP 융합단백질은 ELISA, Western blot 및 immunocytochemistry에서 항원결합력을 나타냈으며 또한 구매에 따른 고비용, 부가적인 배양시간 및 비특이적 결합에 의한 오류 등의 문제점을 갖는 이차 항체를 사용하지 않고 직접적인 항원결합력을 나타내었다.

  • Single-index models have found applications in econometrics and biometrics, where multidimensional regression models are often encountered. Here we propose a nonparametric estimation approach that combines wavelet methods for non-equispaced designs with Bayesian models. We consider a wavelet series expansion of the unknown regression function and set prior distributions for the wavelet coefficients and the other model parameters. To ensure model identifiability, the direction parameter is represented via its polar coordinates. We employ ad hoc hierarchical mixture priors that perform shrinkage on wavelet coefficients and use Markov chain Monte Carlo methods for a posteriori inference. We investigate an independence-type Metropolis-Hastings algorithm to produce samples for the direction parameter. Our method leads to simultaneous estimates of the link function and of the index parameters. We present results on both simulated and real data, where we look at comparisons with other methods.
