  • 북가주에 있는 삼나무 수종에서 cadinane과 acorane류의 sesquiterpenoids를 chromatography를 사용하여 분리한 후, 화학적인 방법과 분광학적인 방법을 이용하여 분리된 성분의 화학구조를 규명하였다. 그 sesquiterpenoids의 생합성 경로를 정량적인 상관성에 의하여 밝혔다. 또한, 여러 삼나무 수종에 존재하는 sesquiterpenoids의 분류학적 차이에 대하여 논하였다.

  • Antibiotic HS-2 was purified from the culture broth of Streptomyces plicatosporus which was isolated from soil, by solvent extraction, silica gel column chromatography and gel filtration. Through the analysis of UV, $^{1}$H-NMR, $^{13}$C-NMR spectrum, HS-2 was identified as rubiginone B$_{2}$. It was confirmed that HS-2 enhanced the cytotoxicity of colchicine against multidrug-resistant tumor cells.

Inhibition of Xanthine Oxidase by Seaweed Extracts (해조류 추출물의 Xanthine Oxidase 저해작용)

  • Inhibition of xanthine oxidase by seaweed extracts obtained from Undaria pinnatifida, Ecklonia stolonifera, Ecklonia cava, Laminaria japonica, Sargassum, Codiumfragile, Enteromorpha compressa and Porphyra tenera were investigated. Extracts of E. stolonifera and E. mua remarkably inhibited xanthine oxidase activity compared to those of other seaweed. The xanthine oxidase inhibitory activity of E. cava was higher than that of E. stolonifera. Diethyl ether extract from E. cava was more effective in the inhibition of xanthine oxidase than other solvent extracts. Two xanthine oxidase inhibitors(A-1 and A-2) from diethyl ether extract were isolated and purified by silica gel column chromatography, thin layer chromatography and high performance liquid chromatography. Xanthine oxidase inhibitory activities of these compounds were 27.8 and 48.1% per 0.4mg, respectively. The active compound A-2 had absorption peak at 420nm, 456nm and 467nm, which can be considered as siphonaxanthine.

  • In our investigation on chemical constituents of edible mushrooms, seven compounds I-VII have been isolated from the methanolic extract of the fruit body of Sarcodon aspratus. The methanolic extract was separated by solvent partition, silica gel column chromatography and Sephadex LH-20 column chromatography, and compounds I-VII were finally purified by preparative HPLC or TLC. Their structures were assigned as 4-hydroxybenzoic acid methyl ester, 4-hydroxybenzaldehyde, cyclo(Ala-Pro), adenosine, nicotinamide, BL V, linoleic acid, respectively, on the basis of mainly $^1H\;NMR$ spectra.

  • The fruits of tomato (Lycopersicon esculentum) were extracted with 70% aqueous methanol (MeOH) and the concentrates were partitioned into ethyl acetate (EtOAc), n-butanol (n-BuOH), and water (H2O) fractions. The repeated silica gel (SiO2) and octadecyl silica gel column chromatographies for the EtOAc fraction, whose activity was confirmed, led to isolation of one flavone compound. Nuclear magnetic resornance, infrarad spectroscopy, and mass spectroscopy (MS) revealed the chemical structure of the isolated compound, 5,7,3'-trihydroxy-6,4',5'-trimethoxyflavone (1). LC-MS/MS analysis determined the content level of compounds 1 in the MeOH extract to be 4.02±0.12 ㎍/mg and in the TME-10 fraction to be 0.96±0.03 ㎍/mg. Through this study, the antioxidantive capacity was confirmed by demonstrating that the L. esculentum extract and their fractions showing an increase in glutathione mean and a decrease in glutathione heterogeneity uniformly raises the intracellular glutathione level.

Purification and Physical Proerties of Biosurfactant Produced from Rhodotorula muciloginosa (Rhodotorula muciloginosa G-1에서 생산되는 biosurfactant의 정제 및 물리적 성질)

  • The surface tension-decreasing biosurfactant was purified from Rhodotorula muciloginosa G-1. The purification procedure was the solvent extraction of culture broth. To ensure complete extraction, the sample was extracted twice with equal volume of ethylacetate. The crude solution was washed with n-hexane to remove unconsumed soybean oil. The crude sample of biosurfactant was applied to Silica gel column chromatography equilibrated with chloroform, and eluted with chloroform : methanol gradient. Serveral solvent system was used to developed the thin layer chromatography (TLC). The purified biosurfactant sample gave one spot (Rf 0.78). It was estimated that biosurfactant was glycolipid about having M.W.1,500 with standard of polyethyleneglycol by Sephadex LH-20 column chromatography.

  • The essential oils of Lycopus lucidus have been studied. The essential oil obtained from the dried herba by steam distillation followed by fractionation by column chromatography was analysed by gas chromatography-mass spectrometry(GC-MS). The compounds identified by GC-MS were carvacrol, 2,5-dimethoxy-p-cymene, trans-caryophyllene, spathurenol and $trans-{\beta}-farnesene$. Two compounds, thymol and caryophyllene were isolated by silica gel column chromatography and analyzed by TLC, IR, Mass and NMR. And the weak diuretic effects of essential oil and water extract from the dried drugs were observed in rats.

  • A strain of Penicillium sp. extracellularly produced an inhibitory substance for lipoxygenase. These purification procedures were followed : ethanol treatment, chromatographies on Dowex 50W, Sephadex G-25, silica gel column and HPLC. The inhibitor was stable in pH range from 3.0 to 5.0 at $25^{\circ}C$, and a treatment at 10$0^{\circ}C$ for 2 hours didn't diminish its original activity. The purified inhibitor was charred at temperature near 22$0^{\circ}C$~23$0^{\circ}C$ and decomposed. Molecular weight of the inhibitor was estimated to be approximately 270 by Sephadex G-25 column chromatography. The inhibitor rapidly formed EI complex with lipoxygenase and inhibited enzyme activity.

  • The volatile components of Korean and Chinese Inulae flos were identified. It is made of dried Inulae flos from the components were collected by simultaneous steam distillation extract method. Those were analyzed by combined gas chromatography(GC) and gas chromatography-mass spectrometrt(GC/MS). Sixty two components, including 10 hydrocarbons, 7 aldehydes, 18 alcohols, 3 ketones, 5 esters, 13 acids and 6 miscellaneous components were identified. Volatile components in Inulae flos were fractinated into one hydrocarbon fraction and two oxygenated hydrocarbon fractions by using silica gel column chromatography. The volatile components consisted of 16.85% hydrocarbon and 83.15% oxygenated hydrocarbons in Korean Inulae flos, 23.46% hydrocarbon and 76.53% oxygenated hydrocarbons in Chinese Inulae flos.

  • Isolation and identification of pathogens from slaughter and meat processing plant were investigated. Antimicrobial activity of Amaranthus lividus against isolated pathogens such as Aeromonas sobria, Escherichia coli, Escherichia coli O157, Listeria monocytogenes, Salmonella spp., and Staphylococcus aureus was investigated. Among the chloroform, ethyl acetate and buthanol fraction of amaranthus lividus showed inhibitory effect against Aeromonas sobria CLFM1 and Escherichia coli CLFM2. Antimicrobial substance in chloroform fraction was isolated by silica gel adsorption column chromatography, sephadex LH-20 column chromatography and silica gel partition column chromatography. The antimicrobial compound of amaranthus lividus was identified as diethyl phtalate by HPLC, GC-MS, H-NMR and C-NMR.
