Secretory Production of Biologically Active Human Thrombopoietin by Baculovirus Expression System

  • Human thrombopoietin (hTPO) was expressed to high levels in insect cells using the baculovirus expression system. Full-length hTPO cDNA containing a native signal peptide sequence was amplified by PCR from a human fetal liver cDNA library and cloned into the Autographa californica nuclear polyhedrosis virus (AcNPV) expression vector. Immunoblot analysis with antiserum against hTPO indicated that an approximately 55 kDa protein was produced in recombinant AcNPV infected insect cells. Recombinant hTPO was produced 4-fold higher in Trichoplusia ni (Tn5) cells than in Spodoptera frugiperda (Sf9) cells. with most of the hTPO produced in Tn5 cells secreted into the culture medium. Addition of tunicamycin in the culture medium resulted in the reduction of the size of hTPO to 35-38 kDa, and most of the protein remained within the cell. These results suggest that N-glycosylation of hTPO is required for the secretion of the protein into the culture medium in insect cells. hTPO produced in insect cells induced proliferation and maturation of megakaryocyte progenitors, indicating that it is in a biologically active form.

Isolation and Differential Expression of an Acidic PR-1 cDNA Gene from Soybean Hypocotyls Infected with Phtophthora sojae f. sp. glycines

  • Using differential display techniques, a new acidic pathogenesis-related (PR) protein-1 cDNA (GMPRla) gene was isolated from a cDNA library of soybean (Glycinemax L.Merr, cultivar Jangyup) hypocotyls infected by Phytophthora sojae f. sp. glycines. The 741 bp of fulllength GMPRla clone contains an open reading frame of 525 nucleotides encoding 174 amino acid residues (pI 4.23) with a putative signal peptide of 27 amino acids in the N-terminus. Predicted molecular weight of the protein is 18,767 Da. The deduced amino acid sequence of GMPRla has a high level of identity with PR-1 proteins from Brassica napus, Nicotiana tabacum, and Sambucus nigra. The GMPRla mRNA was more strongly expressed in the incompatible than the compatible interaction. The transcript accumulation was induced in the soybbean hypocotyls by treatment with ethephon or DL-$\beta$-amino-n-butyric acid, but not by wounding. In situ hybridization data showed that GMPRIa mRNAs were usually localized in the vascular bundle of hypocotyl tissues, especially phloem tissue. Differences between compatible and incompatible interactions in the timing of GMPRla mRNA accumulation were remarkable, but the spatial distribution of GMPRla mRNA was similar in both interactions. However, more GMPRla mRNA was accumulated in soybean hypocotyls at 6 and 24 h after inoculation.

  • In this paper, we deal with implementing design for a correlator access code generator module which they are used for setting up a connection between units, a packet decision, a clock syncronization, by FPGA. The orrelator module which is composed of the Wallace Tree's CSA and threshold value decision device decides useful a packet and syncronizes a clock, after it correlates an input signal of 1 Mbps transmission rate by a sliding window. An access code generator module which is composed of a BCH (Bose-Chadhuri-Hocquenghem) cyclic encoder and control device was designed according as a four steps' generation process proposed in the bluetooth standard. The pseudo random sequence which solves syncronization problem saved a voluntary device Proposed the module was designed by VHDL. An simulation and test are inspected by Xilinx FPGA.

  • It is recognized that the magnetization reverses with a sequence of discrete and jerky jumps, known as the Barkhausen effect. Recently, interest in the Barkhausen effect has grown as it is a good example of dynamical critical behavior, evidenced by experimental observation of a power law distribution of the Barkhausen jump size. So far, most experimental studies have been carried out on bulk samples using a classical inductive technique, which is difficult to apply to thin film samples mainly due to the ]ow signal intensity. For this reason, very few experiments have been done on two-dimensional ferromagnetic thin films. In this talk, we report a direct domain observation of Barkhausen avalanche at criticality in Co and MnAs thin films investigated by means of a magnetooptical microscope magnetometer (MOMM), capable of time-resolved domain observation with an image grabbing rate of 30 frames/s in real time. In Fig. 1, we demonstrate a series of six representative domain-evolution patterns of 25-nm Co film observed successively by means of the MOMM, where one can directly witness Barkhausen avalanche.

Analysis of Channel Flow Low During Fuelling Operation of Selected Fuel Channels at Wolsong NPP

  • Wolsong NPP are CANDU6 type reactors and there are 4 CANDU6 type reactors in commercial operation. CANDU type reactors require on-power refuelling by two remote controlled F/Ms (Fuelling Machines). Most of channels, fuel bundles are float by channel coolant flow and move toward downstream, however in about 30% of channels the coolant flow are not sufficient enough to carry fuel bundles to downstream. For those channels a special device, FARE (Flow Assist Ram Extension) device, is used to create additional force to push fuel bundles. It has been showing that during fuelling operation of some channels the channel coolant flow rate is reduced below specified limit (80% of normal), and consequently trip alarm signal turns on. This phenomenon occurs on selected channels that are instrumented for the channel flow and required to use the FARE device for refuelling. Hence it is believed that the FARE device causes the problem. It is also suspected that other channels that do not use the FARE device for refuelling might also go into channel flow low state. The analysis revealed that the channel How low occurs as the FARE device is introduced into the core and disappears as the FARE device is removed from the core. This paper presented the FARE device behavior, detailed fuelling operation sequence with the FARE device and effect on channel flow low phenomena. The FARE device components design changes are also suggested, such as increasing the number or now holes in the tube and flow slots in the ring orifice.

Small-scale structures in the dust cloud associated with 17P/Holmes outburst

  • A short-period comet, 17P/Holmes, is one of the most outstanding comets because of the outburs in 2007. It orbits the sun at the distance between 2.1AU and 5.2 AU with the orbital period of 6.9 year. On 2007 October 23, its brightness was suddenly increased by about a million times from 17 mag to 2.5 mag. We made observations of 17P/Holmes soon after the outburst on October 25, 27 and 28, using a 105cm telescope at the Ishigakijima Astronomical observatory, Japan. We took the images with V, R and I-band filters simultaneously. Total exposure times are 15 (October 25), 69 (October 27), and 37 (October 28) minute in each filter. The composite images provide good signal to noise ratio and help us to recognize faint structures embedded in the dust cloud. We examined a sequence of images using a digital filter that enhances the small-scale structures. As the result of the data analysis, we confirm (1) the radial expanded structure coming out from the nucleus of comet, and (2) dozens of blobs that moved radially away from the nucleus. In this presentation, we introduce the observations and the data reductions, and consider the origins of these fine structure.

  • Appearance of a cloned AP(Access Point) causes MS(Mobile Station) to break an association with a normal AP(Access Point). If signal power of the cloned AP is stronger than that of the normal AP, MS associates with the cloned AP. Therefore, MS is easily exposed to attackers who installed the cloned AP. In this paper, we distinguish cloned AP from normal AP by using the association time and frame sequence number between normal AP and MS, then isolates the cloned AP. The simulation by NS-2 shows that our mechanism isolates efficiently a cloned AP and builds safer wireless LAN environment.

  • Plasma cell granuloma is a tumor-like disease characterized by non-neoplastic polyclonal proliferation of plasma cells and other mononuclear cells. This disease occurs most frequently in the lung and upper respiratory tract, while the involvement of the central nervous system is very rare. A 44-year-old female patient presented with nausea and progressive visual disturbance. Brain magnetic resonance imaging (MRI) revealed the mass along the right tentorium with low signal intensity in the T2 weighted image (T2WI) and fluid-attenuated inversion recovery (FLAIR) sequence, and an isosignal intensity in T1 weighted image (T1WI), the latter of which was enhanced after administration of gadolinium-diethylenetriamine penta-acetic acid (Gd-DTPA). The thickest portion of the tentorium was partially excised via the combined suboccipital and infratentorial approach. The histopathological examination indicated a diagnosis of plasma cell granuloma. Postoperative steroid therapy was administered for remnant tumor control. Although a follow up MRI scan taken 20 months after the operation showed a slight decrease in tumor size, the lesion had extended to the falx and left frontal convexity along with parenchymal edema at 32 months after the operation and the clinical status was aggravated. The mass was removed from the left frontal convexity. Radiation therapy was given, together with steroid administration.

IL-2-enhanced NK Cell Cytotoxicity is Regulated by Adiponectin from Hypothalamo-pituitary-adrenal Axis (Adiponectin에 의한 IL-2 증가 자연살해세포 독성의 조절)

  • Background: The Hypothalamo-Pituitary-Adrenal (HPA) axis is an important regulator for the body's stress response. As a primary stress responsive system, HPA-axis secretes various neurotransmitters, hormones, and cytokines, which regulates the immune system. Natural killer (NK) cell which is plays an important role in the innate immune response, is specially decreased their numbers and loose cytolytic activity in response to stress. However, the effect of HPA-axis secreted proteins on NK cell activity has not been defined. Herein, we studied the effect of adrenal secreted adiponectin on NK cell cytotoxicity. Adiponectin which is well-known metabolic control protein, plays important roles in various diseases, including hypertension, cardiovascular diseases, inflammatory disorders, and cancer. Methods: Signal sequence trap was used to find stress novel secretory protein from HP A-axis. Selected adiponectin was treated mouse mature primary NK cells and then examined the effect of adiponectin to NK cell cytotoxicity and cytokine expression level. Results: We found that adiponectin which is secreted from adrenal gland, suppress IL-2 induced NK cell cytotoxicity. And also investigated cytolytic cytokines are suppressed by adiponectin. Conclusion: These data suggest that adiponectin inhibites NK cell cytotoxicity via suppression of cytotoxicity related target gene.

  • A key issue toward mobile multimedia communications is to create technologies for broadband signal transmission that ran support high quality services. Such a broadband mobile communications system should be able to overcome severe distortion caused by time-varying multi-path fading channel, while providing high spectral efficiency and low power consumption. For these reasons, an adaptive suboptimum decision feedback equalize. (DFE) for the single-carrier short-burst transmissions system is considered as one of the feasible solutions. For the performance improvement of the system with the short-burst format including the short training sequence, in this paper, the multiple-training least mean square (MTLMS) based DFE scheme with soft decision feedback is proposed and its performance is investigated in mobile wireless channels throughout computer simulation.
