The Secretion Optimization of Oligopeptide with His-Pro Repeats in Bacillus subtilis and Its Anti-Diabetic Effects (고초균에서 His-Pro 반복서열을 갖는 Oligopeptide의 분비 최적화 및 항당뇨 효과)

  • To verify anti-diabetic effect of oligopeptide with His-Pro repeats (mHP peptide), the oligopeptide was first secreted and optimized using the secretion vector, pRBAS with alkaline protease gene promoter and the signal sequence in Bacillus subtilis and directly the anti-diabetic effect of the mHP peptide was investigated in insulinoma cell, RINm5F cell line. The oligopeptide gene was obtained by annealing oligonucleotides with repeated His-Pro sequence and finally was constructed as 18 dipeptides (108 bp and 4.0 kDa) coding gene, named oligopeptide with His-Pro repeats (mHP peptide) to make cyclo(His-Pro) known to be anti-diabetic effects. The region encoding the oligopeptide gene was subcloned into the pRBAS secretion vector (E.coli-Bacillus shuttle vector) after PCR amplification using the designed primers including initiation and termination codons and His tag, named pRBAS-mHP (6.56 kb). To optimize secretion of the oligopeptide, various culture conditions were investigated in Bacillus subtilis LKS. As a result, the secreted oligopeptide was maximally measured (approximately $59.6{\mu}g/mL$) in 3 L batch culture and the highest secretion was achieved at $30^{\circ}C$, PY medium, and carbon sources (particularly barley and glycerol). In the RINm5F cells treated with 2 mM STZ, the oligopeptide treatment (0.1 mg/mL) restored the cell viability (10%) and reduced the nitric oxide (NO) generation (35%) and DNA fragmentation (90%). And also, insulin secretion level was increased to 17% higher than in STZ-treated RINm5F cells. These results suggest that the oligopeptide with His-Pro repeats could be a candidate material for anti-diabetic agent against STZ-induced diabetes.

  • In this paper, we present the Modulator Channel Card and VLSI chip for the Radio Transceiver Unit (RTU) of direct sequence code division multiple access (DS-CDMA) Wireless Local Loop (WLL) System. The Modulator Channel Card is designed and implemented using ASIC's, FPGA's and DSP's. The ASIC, compliance with Common Air Interface specification proposed by ETRI, has 40K gates which is designed to operate at 32MHz, and is fabricated using $0.6\mu\textrm{m}$ CMOS process. The ASIC carries out for I- or Q- phase data channel signal processing at a time, where each data channel processing consists of channel coding, block interleaving, scrambling, Walsh modulation, Pseudo-Noise (PN) spreading, and baseband filtering. The Modulator Channel Card has been integrated as a part of RTU of WLL system and is confirmed that it meets all functional and performance requirements.

A "GAP-Model" based Framework for Online VVoIP QoE Measurement

  • Increased access to broadband networks has led to a fast-growing demand for voice and video over IP(VVoIP) applications such as Internet telephony(VoIP), videoconferencing, and IP television(IPTV). For pro-active troubleshooting of VVoIP performance bottlenecks that manifest to end-users as performance impairments such as video frame freezing and voice dropouts, network operators cannot rely on actual end-users to report their subjective quality of experience(QoE). Hence, automated and objective techniques that provide real-time or online VVoIP QoE estimates are vital. Objective techniques developed to-date estimate VVoIP QoE by performing frame-to-frame peak-signal-to-noise ratio(PSNR) comparisons of the original video sequence and the reconstructed video sequence obtained from the sender-side and receiver-side, respectively. Since processing such video sequences is time consuming and computationally intensive, existing objective techniques cannot provide online VVoIP QoE. In this paper, we present a novel framework that can provide online estimates of VVoIP QoE on network paths without end-user involvement and without requiring any video sequences. The framework features the "GAP-model", which is an offline model of QoE expressed as a function of measurable network factors such as bandwidth, delay, jitter, and loss. Using the GAP-model, our online framework can produce VVoIP QoE estimates in terms of "Good", "Acceptable", or "Poor"(GAP) grades of perceptual quality solely from the online measured network conditions.

  • Several equalizers are applied to the DVD-ROM(Digital Versatile Disc-Read Only Memory) system. Because imperfect writing process may cause nonlinear distortion in the reply signal, neural equalizers which have strong nonlinear mapping capability are applied to the system to compensate the nonlinear distortion. Experimental results to verify that the combination of decision-feedback type equalizers and modulation code is formidable are also given. The experimental results shwo that the SNR gain of the neural equalizers over the conventional equalizers becomes much as the nonlinearity in the channel increases. Furthermore, the decision-feedback type equalizers outperform the equalizers which have no decision-feedback in eliminating ISI(Intersymbol Interference) of random data sequence but there is no performance gain of the decision-feedback type equalizers over the equalizers without decision-feedback when these are applied to compensate the ISI of modulation-encoded data sequence.

High Level Production of Glycoprotein H of HSV-1 (F) Using HcNPV Vector System

  • The Herpes simplex virus type 1 (HSV-1) strain F glycoprotein H (gH) gene in the pHLB-4 plasmid was recombinated into a baculovirus expression vector (lacZ-HcNPV) to construct a recombinant virus GH-HcNPV expressing gH. The sequences of gH and its expression were analyzed. The gH gene was located in the 6.41 kb BglII fragment. The open reading frame (ORF) of the gH gene was 2,517 bp and codes 838 amino acid residues. Insect cells infected with this recombinant virus synthesized a high level of the matured and gX-gH fusion protein with approximately 112 kDa. The fusion gH protein was localized on the membrane of the insect cells as seen by using immunofluorescence assay and accumulated in the cultured media by the SDS-PAGE and immunoprecipitation assays. The amino acid sequence presents additional characteristics compatible with the structure of a viral glycoprotein: signal peptide, putative glycosylation sites and a long C-terminal transmembrane sequence. Antibodies raised in mice to this recombinant protein recognized viral gH and neutralized the infectivity of HSV-1 in vitro. These results demonstrate that it is possible to produce a mature protein by gene transfer in eukaryotic cells, and indicate the utility of the HcNPV-insect cell system for producing and characterizing eukaryotic proteins. Furthermore, the neutralizing antibodies would appear to protect mice against HSV; accordingly, this particular recombinant protein may be useful in the development of a subunit vaccine.

Characterization of T7 RNA Polymerase Transcription Elongation Complex in Sequence-specific Transcription Termination (염기서열 특이적 전사종결부위에서 T7 RNA 중합효소 전사연장복합체 특성에 관한 연구)

  • T7 RNA polymerase is a single subunit RNA polymerase able to accomplish whole transcription process without auxiliary factors. In order to study transcription elongation mechanism of phage T7 RNA polymerse, stepwise walking of RNA polymerase was established by immobilizing biotinylated DNA template with streptavidin bead, series of active and stable elongation complexes were obtained, Transcripts were radio isotope labeled at the 16thm 17th and 18th nucleotide residues so stable elongation transcription complex of T7 RNA polymerase containing 22-40 nucleotide residues could be identified. We identified the positions of stablely formed transcription elongation complexes of termination site in intrinsic hairpin-independent PTH terminator sequence through the established stepwise walking of wild-type of mutant R173C T7 RNA polymerases. The results suggest that stable elongation transcription complexes were at the site of passing PTH terminator signal by mutant R173C RNA polymerase.

  • T7 RNA polymerase is a single subunit RNA polymerase able to accomplish whole transcription process without auxiliary factors. T7 phage lysozyme involcing in destruction of host cell wall repress T7 transcription and affects transcription termination process. Therefore expression vector pT7lys containing T7 phage lysozyme gene was constructed and expressed. T7 phage lysozyme protein was purified to homogeneity by Ni-NTA column chromatography. Also amidase activity of the purified lysozyme was identified. In order to understand the effect of the lysozyme on the sequence specific transcription termination. T7 transcription elongation complexes at the site rrnB T1 transcription termination signal were made in the presence the lysozyme. The results shows that the transcription elongation complexes are unstable in the presence of T7 phage lysozyme.

  • ApolipophorinIII (apoLp-III) is a protypical exchangeable apolipoprotein that is abundant in hemolymph of many insect species. Its function lies in the stabilization of low-density lipophorin particles (LDLp) crossing the hemocoel in phases of high energy consumption to deliver lipids from the fat body to the flight muscle cells. But, recent studies with naive Galleria mellonella-apoLp-III gave first indication of an unexpected role of that protein in insect immune activation. In this research, we cloned a cDNA encoding putative apoLp-III from the silkworm, Bombyx mori injected with E. coli and characterized its role. We constructed a cDNA library using whole bodies of B. mori larvae injected with E. coli, carried out the differential screening, and selected the up-regulated clones. Among these clones, we focused on a cDNA showing a high sequence similarity to the apolipophorinIII from other insects and analyzed the nucleotide and deduced amino acid sequences. The pupative B. mori Jam123 apoLp-III cDNA contained 1,131 bp encoding 186 amino acid residues. Phylogenetic analysis revealed that the nucleotide and amino acid sequences of the B. mori apoLp-III cDNA formed a highly inclusive subgroup with Bombycidae. But, it was interesting that B. mori Jam123 is closer to B. mandarina than B. mori P50 and B. mori N4. Northern blot analysis showed a signal in the fat body, posterior silkgland and midgut.

Molecular Cloning and Characterization of Attacin from the Swallowtail Butterfly, Papilio xuthus

  • Attacin is an insect antibacterial protein that plays an important role in immune response to injury and infection. In this report, we have isolated and characterized of cDNA encoding for the attacin from the immunized larvae of swallowtail butterfly, $Papilio$ $xuthus$. A full length cDNA of $P.$ $xuthus$ attacin was obtained by employing annealing control primer (ACP)-based differential display PCR and 5' RACE. The complete $P.$ $xuthus$ attacin cDNA was comprised of 949 bp encoding a 250 amino acid precursor. It contains a putative 18 amino acid signal peptide sequence, a 42 amino acid propeptide sequence, and a 190 amino acid mature protein with a theoretical molecular mass of 19904.01 and a pI of 9.13. The putative mature protein of $P.$ $xuthus$ attacin showed 48-52% and 24-30% identity in amino acid sequences with that of lepidopteran and dipteran insects, respectively. Semiquantitive RT-PCR results revealed that the transcript of $P.$ $xuthus$ attacin gene was up-regulated at significant levels after injection with bacterial lipopolysaccharide (LPS). We sub-cloned cDNA fragment encoding mature $P.$ $xuthus$ attacin into the expression vector, highly expressed in $E.$ $coli$ BL21 cells, and its antibacterial activity was analyzed. Recombinant $P.$ $xuthus$ attacin evidenced considerably antibacterial activity against Gram-negative bacteria, $E.$ $coli$ ML 35 and $Klebsiella$ $pneumonia$.

Cloning and Characterization of Bombyx mori Cyclophilin A

  • Cyclophilins are originally identified as cytosolic binding protein of the immunosuppressive drug cyclosporine A. They have an activity of peptidyl prolyl cis/trans-isomerases (PPIase), which may play important roles in protein folding, trafficking, assembly and cell signaling. In this study, we report the cloning and characterization of a Bombyx mori cyclophilin A (bCypA) cDNA. The full-length cDNA of bCypA consist of 947 nucleotides with a polyadenylation signal sequence AATAAA and contain an open reading frame of 498 nucleotides encoding a polypeptide of 166 amino acids. The deduced amino acid sequence of bCypA shares a central peptidyl prolyl cis/trans-isomerase and a cyclosporin-A-binding domain with other cyclophilin sequences. Relative quantification real-time (RT) PCR analysis shows that mRNA transcripts of bCypA are detected in all the investigated tissues and highest expression level in the skin of 3-day-old 5 instar larva. Also, bCypA had PPIase activity on the proline-containing peptides. Accordingly, we suggest that bCypA is a new member of the cyclophilin A (CyPA) family and will be useful for quality control of bioactivity recombinant proteins with proline-containing peptides.
