  • In order to prevent the blackening and texture softening of heated shrimp, the pH was adjusted by soaking shrimps in acidic and alkali solutions, and their qualities were evaluated. The lightness of shrimps pretreated with 0.2% (w/w) citric acid and 0.05% (w/w) ascorbic acid solution increased by 20% compared to that of the control. The strength of mechanical hardness of shrimps soaked in acetic acid and phosphate solution (pH 6.0) was significantly higher ($1209g_f$) compared to that of untreated shrimp ($801g_f$; p<0.05), and the overall preference of texture was 0.4 points higher than that of the control in the descriptive sensory evaluation (p<0.05). In contrast, soaking in solution of pH 8 exhibited a weak texture hardening effect ($855g_f$). Additionally, the hardness of the heated shrimp after soaking at an adjusted pH of 4.0 increased to $4046g_f$, but the yield based on weight decreased to 38% compared to that of untreated shrimp (70%; p<0.05).

  • Land shrimp farms can control the growth environment more stably than coastal ones, making them advantageous for high-quality, large-scale production. In order to maintain an optimal shrimp growth environment, various factors such as water circulation, maintaining appropriate water temperature, oxygen supply, and feed supply must be managed. In particular, failure to properly manage water quality can lead to the death of shrimp, making it difficult to have people stationed at the farm 24 hours a day to continuously manage them. In this paper, to solve this problem, we design an integrated monitoring system for land farms that can be operated with minimal manpower. The proposed design plan uses IoT technology to collect real-time images of land farms, pump status, water quality data, and energy usage and transmit them to the server. Through web interfaces and smartphone apps, administrators can check the status of the farm stored on the server anytime, anywhere in real time and take necessary measures. Therefore, it is possible to significantly reduce field work hours without the need for managers to reside in the farm.

  • Diagnostic monitoring in fish farms with land-based tanks and netpen cases were conducted in eastern, western, southern and Jeju island of Korea during summer of 2007~2011. In total, 2413-fish samples of 4 marine fish species were tested for the detection of bacteria and parasite. Fish species tested were olive flounder (Paralichthys olivaceus), black rockfish (Sebastes schlegeli), red sea bream (Pagrus major), pacific white shrimp (Litopenaeus vannamei). During the diagnostic monitoring from 2007 to 2011, the infection rates by single infection of bacterial or parasitic pathogens were relatively higher than the mixed infections. The main bacterial pathogens in olive flounder, black rockfish and pacific white shrimp were Vibrio spp. (V. harveyi, V. ichthyoenteri, Vibrio sp.). The main bacterial pathogens in red sea bream were also Vibrio sp. and Photobacterium damselae subsp. damselae. The main parasitic pathogens were both Miamiensis avidus and Trichodina sp. in olive flounder, Microcotyle sebastes in black rockfish, Microcotyle tai in red sea bream and Zoothamnium sp. in pacific white shrimp.

  • The present study examined the effect of the dietary administration of a halophilic bacterium Bacillus sp. Mk22 on the growth improvement of black tiger shrimp, Penaeus monodon, and reduced shrimp infection by Vibrio parahaemolyticus and white spot syndrome virus (WSSV). The shrimp were fed 45 days using three experimental diets: no addition (control), commercial probiotic, and Bacillus sp. Mk22. The shrimp treated with the halophilic bacterium Mk22 showed a significant improvement of growth (7.1 ± 0.21 g), survival (94.3 ± 0.58%), weight gain (178 ± 4.93 g), and reduced feed conversion rate (0.8 ± 0.03 g) compared with the shrimp in the other groups. The shrimp treated with Bacillus sp. Mk22 also showed a lower Vibrio count (0.02 ± 0.01 × 102 CFU/ml) in the shrimp culture water compared with the other groups. The shrimp in the three groups were challenged with either Vibrio or WSSV. For the Vibrio infection, no mortality was observed from water infection or oral feeding infection in the commercial probiotic group and Mk 22 group. For the WSSV infection, a 68% survival rate from water infection and 20% survival rate from oral feeding infection was observed on day 45 in the Mk22 group, while 100% mortalities were recorded at a much earlier time in both the control and commercial probiotic groups. The antioxidant enzyme activities, indicators of oxidative stress, such as catalase and superoxide dismutase, significantly decreased in both the Vibrio and WSSV-infected Mk22 groups compared with the other groups, indicating that Bacillus sp. Mk22 was effective in reducing oxidative stress, possibly due to the reduced infection.

  • Whole cure shrimp, eviscerated common squid and little squid meats were irradiated with gamma radiation from 60Co using varying dose up to one Mrad. Changes in quality were assessed by organoleptic, chemical (VRS and TMA-N), biochemical (hypoxanthine) and microbiological tests; and the effect of radiation on prolonging of the storage life at 2-4 C was studied. With the cure shrimp, 0.12-0.20 Mrad appears to be optimal with 8 days of storage life, whereas the unirradiated control samples had a storage life of 3 days. With the common squid meat, 0.20-0.30 Mrad appears to be optimal with 14-21 days of storage life, where-as the unirradiated control samples had a storage life of 8 days. And for the little squid meat, 0.12-0.20 Mrad appears to be optimal with 10 days of storage, where-as the unirradiated control samples had a storage life of 5 to 6 days.

  • This work was performed in order to examine the effect of nitrite, thiocyanate and ascorbic acid on formation of N-nitrosamine(NA) in kimchi, fermented anchovy and shrimp under simulated gastric digestion, in vitro. The contents of nitrate were 10.7~24.5 mg/kg in kimchi, 1.5~5.6 mg/kg in fermented anchovy, 1.0~2.0 mg/kg in fermented shrimp and those of nitrite were average 0.3 mg/kg in all analyzed samples. Dimethylamine and trirmethylarnine contents were 4.9~l5.4 mg/kg, 0.6~0.8 mg/kg in kimchi, 3.3~4.0 mg/kg, 1.9~2.8 mg/kg in fermented anchovy, 30.3~177.9 mg/kg, 4.4~21.3 mg/kg in fermented shrimp, respectively. The contents of N-nitrosodime -thylamine(NDMA) were in the range of 0.8~6.9 $\mu\textrm{g}$/kg in kimchi, 0~l.2 $\mu\textrm{g}$/kg in fermented anchovy and 0~0.9 $\mu\textrm{g}$/kg in fermented shrimp. After simulated gastric digestion, NDMA was increased about 1.5 times in all sample. In every nitrite added samples, the contents of NDMA were increased by 183.1 times in fermented shrimp and were detected 192.4 $\mu\textrm{g}$/kg and 220.9 $\mu\textrm{g}$/kg when it was treated with 4 mM and 8 mM of nitrite, respectively. NDMA, when above samples were added 8 mM nitrite and 6.4 mM thiocyanate, was increased about 1.5 times than control samples. The formation of NDMA was inhibited by 49.9~92.4% in all samples added 12.8 mM ascorbic acid compared with the control sample.

  • Photosynthetic bacteria (PSB) attract considerable interest as useful microorganisms; nevertheless, a generalized culture technique has not been previously reported owing to difficulty in their cultivation. Therefore, a simple culture technique suitable for public use was investigated. Among the PSB tested, the strain Rhodobacter azotoformans EBN-7 was the most suitable for scale-up production because it showed the highest specific growth rate (0.20 h-1) on basal medium. In scale-up cultivation (500 L), R. azotoformans EBN-7 showed 4.50 × 1010 colony-forming units mL-1 (number of viable cells), dry cell weight of 26.8 g/L, and a specific growth rate of 0.15 h-1. Cultivation using this final culture broth (as seed culture) in a 15 L simple reactor was successful, with maintenance of cell activity evident. For use as seed culture, the maximum allowable preservation period of R. azotoformans EBN-7 at 4℃ was 3 months. When R. azotoformans EBN-7 cultivated in a simple technique was applied to shrimp aquaculture water, NH4+-N was reduced from 0.61 mg/L to 0.24 mg/L (by 60.7%) in 4 days in comparison with the control. Thus, this simple culture technique using R. azotoformans EBN-7 has the potential for a good removal efficiency of NH4+-N, making seed culture easier and suitable for public use.

  • In this study, nylon mash and silicone tube mainly used as shrimp farm equipment were contaminated with Enterocytozoon hepatopenaei (EHP) which is the cause of Hepatopancreatic microsporidiosis (HPM), and were treated with calcium hypochlorite or UV-C disinfection methods for EHP eradication. As a result, similar with the control group (not disinfected), EHP was detected on the nested PCR until the 10 days in the UV-C single treated group. On the other hand, EHP was not detected from 7 days in calcium hypochlorite single treated group (total concentration 200 ppm as available chlorine), and combination of calcium hypochlorite and UV-C treated group revealed no detection of EHP from 3 days. It is appropriate that treated with UV-C and calcium hypochlorite for 3 days or single treated with calcium hypochlorite for 7 days to eradicate EHP on contaminated instrument used in shrimp farms. In contrast, disinfection effect of only using UV-C is very low.

  • Doxycycline is a semi-synthetic broad-spectrum antibiotic, and it has been used to get rid of bacteria in animals and humans. The use of antibiotics has greatly contributed to the aquaculture production although its misuse sometimes presents public health problems. This study was performed to investigate the toxic effects of doxycycline on whiteleg shrimp (Litopenaeus vannamei) administered for possible infection treatments. The shrimp were allocated into four groups and doxycycline was fed three times a day for 7 days at 0, 20, 50 and 100 mg/kg to each group. After 24 hr following the 7-day treatment, hemolymph and hepatopancreas were used for blood and biochemical analysis: Total hemocyte counts, Total protein, Total cholesterol, Gluscose, Glutamic pyruvic transaminase, Glutamic oxaloacetic transaminase, Glutathione peroxidase, Superoxide dismutase, Glutathione-s-transferases, Total antioxidant capacity colorimetric and Acid phosphatase. In addition, histopathological examination was performed on the hepatopancreas and muscle. It was observed that body weight gain was significantly retarded in 100 mg/kg doxycycline group. Doxycycline was found to induce biochemical or functional disorders at 100 mg/kg as observed many of the blood and biochemical parameters were significantly reduced. In conclusion, it was judged that there will be no major toxicity problems with doxycycline when used for shrimp aquaculture at regular doses.

  • Vibrio harveyi is one of the most serious pathogens causing vibriosis in larval and grow-out shrimp culture. This study was performed to investigate the bactericidal effect of a monopersulfate compound against V. harveyi and its toxicity in Litopenaeus vannamei. The monopersulfate compound was prepared at 0, 0.15, 0.3, 0.6, 1.2, and 2.4 ppm for the bactericidal efficacy study, and then V. harveyi was added at a rate of 1×106 CFU/mL. Subsequently, five shrimps/dose were added to the water bath. The other groups of shrimp were exposed to monopersulfate, but not to the bacterium. None of the shrimps exposed to any of the monopersulfate treatment doses without bacteria died, and no changes in their movement were detected for 7 days. However, shrimps exposed to bacteria without monopersulfate showed decreased movement and lethargy, but no death. The total number of other bacteria and V. harveyi at the different concentrations decreased significantly by 4-6 log values compared to that in the bacterial control group. The monopersulfate compound completely inhibited V. harveyi growth beginning 1 h after exposure to 2.4 ppm. These results show that the monopersulfate compound is an efficient disinfectant against naturally occurring marine bacteria and V. harveyi without being toxic to shrimp.
