Effect of Alcohol Fermented Feed on Lactating Performance, Blood Metabolites, Milk Fatty Acid Profile and Cholesterol Content in Holstein Lactating Cows

  • A feeding experiment with 40 lactating Holstein cows and 4 dietary treatments was conducted to investigate supplementation with different levels of alcohol fermented feed to the TMR on lactating performance, blood metabolites, milk fatty acid profile and cholesterol concentration of blood and milk. Forty Holstein lactating cows ($106{\pm}24$ d post-partum; mean${\pm}$SD) were distributed into four groups and randomly assigned to one of four treatments with each containing 10 cows per treatment. The treatment supplemented with TMR (DM basis) as the control (CON), and CON mixed with alcohol-fermented feeds (AFF) at a level of 5%, 10% and 15% of the TMR as T1, T2 and T3, respectively. Dry matter intake and milk yield were not affected by supplementation of AFF. An increased 4% FCM in the milk occurred in cows fed T3 diet compared with CON, while T1 and T2 diets decreased 4% FCM in a dose dependent manner. Supplementation of AFF increased the concentration of albumin, total protein (TP), ammonia, and high density lipoprotein-cholesterol in serum compared with CON. In contrast, supplementation with AFF clearly decreased concentration of blood urea nitrogen (BUN) and total cholesterol (TC) compare with CON. AFF supplementation increased the proportion of C18:1n9 and C18:2n6 compared to CON. A decrease in the concentration of saturated fatty acid (SFA) for T1, T2 and T3 resulted in an increased unsaturated fatty acid (USFA) to SFA ratio compared to CON. Concentration of cholesterol in milk fat was reduced in proportion to the supplemental level of AFF. Feeding a diet supplemented with a moderate level AFF to lactating cows could be a way to alter the feed efficiency and fatty acid profile of milk by increasing potentially human consumer healthy fatty acid without detrimental effects on feed intake and milk production. A substantially decreased cholesterol proportion in milk induced by supplementation AFF suggests that alcohol fermented feed may improve milk cholesterol levels without any negative effects in lactating cows.

Bioequivalence of Samchundang Lercanidipine Tablet 10 mg to Zanidip Tablet (Lercanidipine Hydrochloride 10 mg) by Liquid Chromatography with Tandem Mass Spectrometry

  • The purpose of the present study was to evaluate the bioequivalence of two lercanidipine hydrochloride tablets, Zanidip tablet (LG Life Sciences Ltd., Korea, reference drug) and Samchundang Lercanidipine tablet 10 mg (Sam Chun Dang Pharm. Co. Ltd., Korea, test drug), according to the guidelines of Korea Food and Drug Administration (KFDA). After adding an internal standard (amlodipine maleate) to human serum, serum samples were extracted using hexan-isoamyl alcohol (100:1, v/v). Compounds were analyzed by liquid chromatography/tandem mass spectrometry. This method showed linear response over the concentration range of 0.05-20 ng/mL with correlation coefficient of 0.9999. The lower limit of quantitation using 0.5 mL of serum was 0.05 ng/mL which was sensitive enough for pharmacokinetic studies. Thirty healthy male Korean volunteers received each medicine at the lercanidipine hydrochloride dose of 20 mg in a $2\;{\times}\;2$ crossover study. There was a one-week washout period between the doses. Serum concentrations of lercanidipine were monitored by an LC/MS/MS fer over a period of 24 hr after the administration. $AUC_t$ (the area under the serum concentration-time curve from time 0 to 24 hr) was calculated by the linear trapezoidal rule method. $C_{max}$ (the maximum serum drug concentration) and $T_{max}$ (the time to reach $C_{max}$) were compiled from the serum concentration-time data. Analysis of variance was carried out using logarithmically transformed $AUC_t$ and $C_{max}$. No significant sequence effect was found for all of the bioavailability parameters, indicating that the crossover design was properly performed. The 90% confidence intervals of the $AUC_t$ ratio and the $C_{max}$ ratio for Samchundang Lercanidipine/Zanidip were log 0.9505-log 1.2258 and log 0.9987-log 1.2013, respectively. These values were within the acceptable bioequivalence intervals of log 0.80-log 1.25. Thus, the criteria of the KFDA guidelines for the bioequivalence was satisfied, indicating Samchundang Lercanidipine tablet 10 mg and Zanidip tablet are bioequivalent.

헛개열매추출액발효물이 흰쥐의 에탄올 경구투여에 의한 간손상 지표와 체중 감량 완화에 미치는 영향 (Effect of fermented Hovenia dulcis Thunb fruit water extract on biomarker for liver injury and body weight changes in rats given oral administration of ethanol)

  • 발효에 의한 헛개열매의 기능성 상승 정도를 검토하고자, 헛개열매열수추출물을 발효시킨 후 급성 및 만성 알코올 투여 간손상 동물모델을 통하여 체중감량 억제, 알코올 분해 및 간기능 개선 효능을 검증하였다. 급성 알코올 투여 동물모델에서 헛개열매발효군(ET-FHWE)은 알코올대조군(ET)에 비하여 혈청 알코올 농도가 유의적으로 감소되었고, 특히 알코올 투여 3시간 후의 알코올 농도는 헛개열매추출액발효물에 의해 46.1%, 헛개열매열수추출물에 의해 19.1% 감소된 것으로 나타났다. 또한 알코올 투여에 의해 증가된 혈청 아세트알데하이드 농도는 헛개열매추출액발효물에 의해 알코올 투여 3시간 후에는 48.7%, 5시간 후에는 39%로 알코올대조군(ET)보다 유의적으로 감소하였고, 이는 헛개열매열수추출물은 발효에 의해 알코올 및 아세트알데하이드 분해능이 증가하는 것으로 사료되었다. 만성 알코올 투여 간손상 동물모델 실험에서 알코올 투여에 의해 상승된 혈청 알코올 농도는 헛개열매열수추출물과 헛개열매추출액발효물 투여에 의해 각각 31%, 41% 유의적으로 감소하였다. 혈청 아세트알데하이드 농도와 ${\gamma}$-GTP 활성도는 헛개열매열수추출물과 헛개열매추출액발효물 투여에 의해 알코올대조군(ET)보다 유의적으로 감소되었으며, 장기간 알코올 투여에 의한 체중 감소 억제 및 간조직 지질수준의 유의적 감소를 나타내었다. 또한 헛개열매추출액발효물은 장기간의 알코올 투여로 인해 감소된 혈당을 유의적으로 증가시키는 것으로 나타났다. 본 연구 결과, 급성 알코올 투여 동물모델에서 헛개열매열수추출물은 발효에 의해 알코올 및 아세트알데하이드 분해능이 증진되었고, 만성 알코올 투여 모델을 통한 실험에서는 발효에 의해 헛개열매의 간기능 개선효능이 유지됨과 동시에 일부 효능(혈청 지질 및 혈당 수준 개선능)은 증가하는 것으로 나타났다. 따라서 헛개열매추출액발효물은 급성 및 만성 알코올성 간손상 억제에 있어서 헛개열매열수추출물보다 더욱 강력한 기능성 물질로 활용될 수 있을 것으로 기대된다.

폐경 전 . 후 중년 여성들의 골밀도에 영향을 미치는 요인 (Factors Affecting Bone Mineral Density in Korean Women by Menopause)

  • Bone mineral density (BMD) focus one's attention on prevention effects of osteoporosis. This study was conducted to investigate BMD (lumbar spin : L2- L4, femur neck : m, femur trochanter TR, femur ward's triangle : WT, wrist) and look into the factors that affect BMD by menopause in 89 nonsmoking healthy Korean women (40-60 yr). Anthropometric index and body composition, nutrient intakes, osteocalcin and total protein, albumin, total cholesterol, triacylglycerol and calcium in serum were determined. Body fat mass, diastolic blood pressure and serum cholesterol concentration of postmenopausal women were significantly higher than those of premenopausal women. FN, TR, WT, wrist BMD of postmenopausal women were not different from those of premenopausal women. However L2- L4 BMD of postmenopausal women was lower than that of premenopausal women. Significant positive correlations were found between L2-L4, FN, TR, WT and weight (r = 0.44, r = 0.64, r = 0.58, r = 0.57) and significant positive correlations were found between FN, TR, WT and BMI (r =: 0.54, r = 0.41, r = 0.54) of premenopausal women. Whereas significant positive correlation was only found between TR BMD and weight. BMI (r = 0.38, r = 0.29) of postmenopausal women. FN BMD and WC (waist circumstance) of premenopausal women were found significant positive correlation (r = 0.35) whereas L2-L4 BMD and WC of postmenopausal women was found significant negative correlation (r = -0.31) . In premenopausal women. differences of bone BMD were not shown by exercise and alcohol drinking, but in postmenopausal women, significant difference of FN BMD was shown by exercise and alcohol drinking. These results suggested that by menopause, effects of weight, BMI, WC, exercise and alcohol drinking on bone BMD were different. Therefore, by menopause, we should consider the different ways to increase the bone BMD according to different factors.

벌나무 열수 및 에탄올 추출물의 항산화 활성 및 알코올 분해능 (Antioxidant and Alcohol Degradation Activities of Extracts from Acer tegmentosum Maxim.)

  • 벌나무의 에탄올 및 열수 추출물을 각각 제조하여 추출물별 항산화 활성을 비교하고, in vitro 및 in vivo에서의 알코올 분해능을 검토하였다. 벌나무 열수 및 에탄올 추출물의 총 폴리페놀 함량은 각각 $171.93{\pm}2.2{\mu}g/mg$, $152.69{\pm}1.25{\mu}g/mg$, 총 플라보노이드 함량은 각각 $7.51{\pm}1.34{\mu}g/mg$, $5.01{\pm}0.83{\mu}g/mg$이었고, FRAP값은 각각 $1.75{\pm}0.32{\mu}M/{\mu}g$, $1.67{\pm}0.28{\mu}M/{\mu}g$으로 나타났다. 벌나무 열수 및 에탄올 추출물은 DPPH 및 ABTS에 대해 높은 소거활성을 보였고, 지질과산화 억제활성이 높음을 알 수 있었다. 벌나무 열수추출물은 HepG2 세포에서 알코올에 대한 in vitro에서의 간세포 보호 효과가 있었고, 또한 알코올을 투여한 rat에서 벌나무 추출물을 투여하면 알코올 투여 30분이 경과한 후 알코올 투여군보다 혈중 알코올 함량이 유의적으로 저하되었다. 따라서 벌나무 추출물은 높은 항산화 작용을 하며, 알코올 분해 작용 및 알코올에 대한 간 보호 효과가 높은 것으로 생각된다.

Bulnesia sarmienti 를 함유한 숙취해소 음료가 알코올대사 및 항산화 효소활성에 미치는 영향 (Effect of Alcohol Detoxification Beverage that Contained Bulnesia sarmienti on Alcohol-metabolizing Enzymes and Antioxidant Enzyme Activities)

  • 허브의 일종인 B. sarmienti를 이용하여 숙취 해소 음료를 개발하고 그에 따른 알콜 대사 및 항산화 효소에 미치는 영향을 알아보고자 하였다. 알코올을 섭취한 쥐에 B. sarmienti 음료군(BSP)을 투여 한 경우 타사 제품군인 PC 그리고 B. sarmienti만을 제외한 음료군인 NBS보다 더 뛰어난 알코올 분해능 및 ADH, ALDH에서도 뛰어난 활성을 보였다. 또한 생화학적 검사를 통해 안전성을 확인하였으며, ALP, ALT 그리고 AST에서도 PC군에 비해 유의적인 감소 효과를 보였다. 항산화 활성 및 효소계에 있어서도 BSP군에서는 GSH에서는 CO군에 비해 효소의 함량은 높으나, 다른 군에 비해 유의적인 차이는 보이지 않았으며, TBARS에는 다른 군에 비해 함량이 감소됨을 알 수 있었으나 뚜렷한 차이는 보이지 않았다. 그러나 GST, CAT 그리고 SOD 효소에 있어서는 CO군에 비해 유의적인 효소 활성이 감소되었음을 알 수 있었다. 이상과 같은 결과를 토대로 B. sarmienti 추출물이 첨가된 음료는 뛰어난 숙취 해소 및 간세포의 손상을 보호하는 음료로서 이용 가능하리라 기대 된다.

일부 농촌 지역 흡연 여고생의 영양 섭취 및 혈액 성상 조사 (A Study on the Dietary Nutrient Intakes and Blood Profiles of Smoking Teenage Girls Living in a Rural Community in Korea)

  • Cigarette smoking has a negative effect for nutrient intakes and nutritional status of antioxidant nutrients including vitamin C. This study has been performed to compare the dietary nutrient intakes, serum vitamin C concentration and serum cotinine concentration of smoking teenage girls(n=104) with those of non-smoking teenage girls(n=791). The subjects were 895 high school girls(15-19 years of age) living in a rural community in Korea. Of the subjects, 11.6% stated that they were currently smoking. The frequency of taking alcohol, soda and coffee was higher in smokers than in non-smokers, respectively. The usage rate of vitamin/mineral supplements was higher in smokers than in non-smokers. The smokers tended to skip breakfast and lunch and to take snack more often than did the non-smokers. The dietary intakes of calorie, protein, vitamin A, vitamin B, vitamin C, calcium and iron were lower in smokers than in non-smokers. Daily mean intakes of these nutrients were less than the 2/3 of the Korean RDAs in smokers while these intakes were similar to or more than the RDAs in non-smokers. The ratio of under weight or lean tended to be higher in smokers. Serum vitamin C concentration was lower in smokers than in non-smokers. These findings show that smokers have unsound eating habits and poor nutirtional status. In particular, nutritional status of vitamin C is significantly poor in smokers, and this tendency can be found more notably in heavy smokers. Therefore, oxidative stress may be present in young teenage girls with short smoking history as evidence by above results. (Korean J Nutrition 34(3) : 338∼347, 2001)

Evaluations of Poly{vinyl alcohol)/Alginate Hydrogels Cross-linked by r-ray Irradiation Technique

  • In this work, we prepared hydrogels for wound dressing from a mixture of poly(vinyl alcohol) (PVA) and alginate using the $\^$60/Co ${\gamma}$-ray irradiation technique. We examined the physical properties of these hydrogels, including gelation, water absorptivity, and gel strength, to evaluate the applicability of these hydrogels for wound dressings. The biocompatibility of these hydrogels was also evaluated in vitro, in cultures of mouse fibroblasts, and in vivo, by subcutaneous implantation studies in rats. The gel content and strength increased upon increasing the radiation dose and upon decreasing the concentration of alginate. The degree of swelling was inversely proportional to the gel content and strength. The degree of cytotoxicity of the ${\gamma}$-ray-treated hydrogels was ca. 60% compared to the (-) control (serum) after 1 day of incubation. When the incubations were prolonged up to 2 days, the toxicity of all the samples decreased remarkably and reached that of the control. Subcutaneous implantation studies in rats indicated that foreign body reactions occurring around the implanted hydrogels were moderate and became minimal upon increasing the implantation time.

밭미나리 발효액의 항산화 활성과 흰쥐에서 알코올성 간 손상 보호효과 (Antioxidant Activity of Dropwort (Oenanthe javanica DC) Fermented Extract and its Hepatoprotective Effect against Alcohol in Rats)

  • Antioxidant activity of dropwort fermented extract (DFE) was measured according to fermentation period, and liver protective effects were examined using Sprague-Dawley rats. Total polyphenol and flavonoid contents as well as DPPH and ABTS radical scavenging activities increased up to 60~80 days and then decreased slightly. Proper fermentation time for DFE was more than 60 days and less than 80 days. Administration of alcohol to rats for 10 days at 10 mL/kg/day raised serum AST, ALT, total cholesterol, and triglyceride (TG) levels, which were then lowered by DFE and sugar liquid with the same soluble solids. While sugar liquid increased the blood lipid profile, especially TG levels, DFE had no effect due to its antioxidant activity. When TBARS content of the DFE group in liver tissue significantly decreased in a concentration-dependent manner compared to that of the ALH group (p<0.05). Liver damage was recovered by DFE treatment and was confirmed by hamatoxylin-eosin staining. These results suggest that DFE has a protective effect against alcohol-induced hepatotoxicity in SD rats.

인디언구스베리와 님잎 추출물을 함유한 복합 처방의 항산화 및 숙취해소 효과 (Antioxidant and Hangover Cure Effects of Compound Prescription Containing Phyllanthus emblica and Azadirachta Indica Leaf Extract)

  • The purpose of this study was to investigate the antioxidant and hangover cure effects of compound prescription containing Phyllanthus emblica and Azadirachta Indica leaf extract (CP). In vitro experiments, HepG2 cells were induced oxidative stress by hydrogen peroxide (H2O2) and treated with CP at 50, 100, 200 ㎍/㎖ concentration. Antioxidant enzyme (superoxide dismutase (SOD), catalse (CAT), glutathione peroxidase (GPx), glutathione reductase (GR) activity and glutathione (GSH) content were decreased by hydrogen peroxide-induced oxidative stress, but CP was increased that. In vivo experiments, experiment rats were orally administered alcohol 3 g/kg and, after 30 min administered CP 200 mg/kg. After 1 and 3 h of alcohol administration, blood was collected from the tail vein, while after 5 h, blood was collected from the heart. CP modulates alcohol dehydrogenase (ADH) and acetaldehyde level, thereby decreased alcohol level in serum. Also, CP decreased the levels of aspartate aminotransferase (AST) and alkaline phosphatase (ALP). These results suggest that CP has antioxidant effects and alleviates alcohol hangover symptoms.
