Clinical Study for the One Case of Premature Ovarian Failure by Serum Hormone Assay (호르몬 검사를 통하여 관찰한 조기난소부전의 치험 1례)

  • Objectives: The purpose of this study is to asses the effects of oriental medicine on premature ovarian failure. Methods: The patient in this case was 30-year-old female. The chief complain is oligomenorrhea. By serum hormone assay and history interview, we diagnosed premature ovarian failure. She was treated by oriental treatments, such as herbal medicine and acupuncture. And then we estimated the results of treatment by follow-up measurements of serum hormone level. Results: After treatments, symptoms of premature ovarian failure was improved, level of serum gonadotropin decreased and level of serum $E_2$ increased. We confirmed the improvement of case by follow-up measurements of serum hormone level. Conclusions: This results of serum hormone assay showed how medically effective oriental treatments of premature ovarian failure.

Effect of veterinary acupuncture (VA) on the eggs-laying rate (침자극(鍼刺戟)이 생식능력(生殖能力) 저하(低下) 닭에 미치는 영향(影響))

  • Objectives : To investigate the increasing effect of veterinary acupuncture (VA) on the eggs-laying rate, we carried out the experiment in old hens. Methods : Twenty four old hens (ISA Brown; 1,200 - 1,400 g) over 75 weeks old whose eggs-laying rates were under the 50% for recent 15 days were used. In experimental group, VA treatments were performed seven times during 10 days. Unang point at the end of bilateral wing and Migun point at the tail bone were stimulated about 30 seconds by the stainless steel needle (0.3 mm o.d.). In control group, similar manual stress were performed at same frequency. The numbers of laid eggs were observed for 26 days after the beginning of treatment. Bloods were sampled two times from wing veins during and after the treatment period and serum concentrations of luteinizing hormone (LH) and Estradiol were detected. Results : Eggs-laying rates of experimental group $(34.29{\pm}2.79%)$ were significantly increased than that of control group $(27.56{\pm}2.00%,\;p<0.05)$ with the significant increasing of LH serum concentration in experimental group $(from\;0.77{\pm}0.13\;IU/I\;to\; 1.42{\pm}0.22\;IU/I,\;p<0.05)$. Conclusions : Increasing effect of VA treatment on the eggs-laying rate in old hens were proved in this study, and it may be the first prompt on the experimental investigation and development of VA.

  • From the 80% ethanol extraction of onion flesh, fraction I,II and III were separated by Sephadex LH-20 column chromatography. Fraction II and III had a high inhibitory effect on ACE and xanthine oxidase. The ACE was about 80% at 50 ppm of fraction IIand III. The xanthine oxidase was about 82.5% at 100 ppm of fraction In. This research was studied to investigate the effects of the fraction In isolated from onion on the reduction of fat accumulation in rats fed high fat diet for 2 months. The metabolism was examined by analyzing the fat in serum and liver. It was shown that cholesterol in serum was remarkably reduced in onion fraction In as compared with those of control.

Protective effects of saffron against zearalenone-induced alterations in reproductive hormones in female mice (Mus musculus)

  • Objective: Zearalenone (ZEA) is a mycotoxin with potent estrogenic effects. Saffron is an herbal product that has antioxidant activities. The objective of this study was to investigate the protective role of saffron against reproductive toxicity induced by ZEA in female mice. Methods: Ninety 8-week-old female mice were randomly allocated into three treatment groups. The first group received an intraperitoneal injection of ZEA (2.5 mg/kg) on alternate days. The second group received ZEA (2.5 mg/kg) on alternate days plus oral saffron daily (50 mg/kg). The third group was treated with a vehicle of 1% dimethyl sulfoxide (DMSO) on alternate days, as a control. Ten mice were euthanized from each group at 30, 60, and 90 days of treatment. Serum levels of luteinizing hormone (LH), follicle-stimulating hormone (FSH), estradiol ($E_2$), and progesterone (P) were assessed. The uterus and ovaries were examined for changes in size or morphology. Results: Serum levels of LH, FSH, $E_2$, and P in the female mice treated with ZEA plus saffron were significantly higher than in those treated with ZEA alone, and were not significantly different from those treated with 1% DMSO. The female mice treated with ZEA alone showed a reduction in size of the uterus and abnormal architecture of the ovaries. Conclusion: The administration of saffron to female mice resulted in a significant reduction in ZEA-induced alterations in reproductive hormone levels, the size of the uterus, and the morphology of the ovaries.

Biochemical parameters and reproductive traits in female rabbits (oryctolagus cuniculus) exposed to psidium guajava leaf aqueous extract

  • Background: The potential impact of aqueous extracts from Psidium guajava leaves on the reproductive system of female rabbits was evaluated. Methods: Twenty-eight rabbits, aged five to six months were utilized. Rabbits were divided into four groups and were randomly assigned to receive one of the following oral doses of the guava leaf extracts: 0 (control group), 10, 20, or 30 mg/kg of body weight. After a treatment period of 30 days, blood was collected via jugular venipunture and the serum was extracted for the assessment of serum biochemical traits levels. The females were bred and monitored throughout their pregnancy to ascertain reproductive outcomes. Results: The results indicated that the guava leaf extract significantly increased the body weight of the rabbits during both pre- and post-pregnancy compared to the control group (p < 0.05). The litter size at three weeks post-birth, prolificity rate, FSH, LH, and protein levels were notably higher (p < 0.05) at a dose of 20 mg/kg of body weight. The viability rate three weeks post-birth increased with escalating extract doses, and the highest values were observed at doses of 20 and 30 mg/kg of body weight (p < 0.05). Conclusions: This study demonstrated that, the aqueous extract of guava leaves appears to stimulate the production of FSH, LH and enhance body weight, prolificity, and pregnancy outcomes in mammals. As such, it is suggested that a dose of 20 mg/kg body weight could be beneficial in improving the reproductive performance of female.

Effects of Ovarian Function on the Hypophyseal Gonadotropin Secretion in Rats (흰쥐의 난소기능(卵巢機能)이 하수체(下垂體)의 성선(性腺) 날극(剌戟)호르몬 분비(分泌)에 미치는 영향(影響))

  • The study was carried out to elucidate the feedback mechanism on the hypothalamo-hypophyseal system from the functional changes of ovary in female rats. One hundred and forty-four mature female rats were lloted into the three groups; ovariectoimzed group, estradiol treated group and intact control group. The varies of 48 heads of rat were completely removed. Forty eight heads of rat were administered with $200{\mu}g$ of estradiol benzoate every 48 hours. Serum FSH, LH and prolactin levels were determined with radioimmunoassay method at 3,6,12,24 ours, and 5,10, and 15days after treatment. The rats were necropsied to measure the weights of hypophysis and to examin the histological changes in the organs. The results obtained were as follows: The weights of hypophysis were increased after ovariectomy and decreased after estradiol injection. The differences in hypophysis weights were significant between the group from 5 days after treatment. The histological changes in hypophysis were appeared from 5th day after ovariectomy. Proliferation and hypertrophy began to occur in basophilic from 10th day after ovariectomy, chromophobes were slightly hypertrophied and acidophilic cells were atrophied. In estradiol injected rats the histological findings were appeard to be contrary to those of ovariectomized rats. Serum FSH levels significantly changed after ovariectomy and estradiol injection and were higher in both the treated groups than in the intact control group. Within 18 hours after treatment the level was the highest in ovariectomized group, and thereafter the highest level was found in estradiol treated gorup. In ovariectomized rats the levels were rapidly increased 3 hours after treatment and maximum levels were found 18 hours after treatment. In estradiol treated rats the levels started to increase 18 hours after treatment and reached maximum levels 24 hours treatment. 4. Serum LH levels started to increase 3 hours after ovariectomy and estradiol injection and reached maximum levels 12 hours after ovariectomy and 24 hours after estradiol injection. There were significant differences in LH levels between the groups in each observation time. Up to 18 hours after treatment levels were higher in ovariectomized rats than in estradiol treated rats. but thereafter the levels were higher in estradiol treated rats than in ovariectomized rats. The multiple range test showed that a significant difference in LH levels was not found between ovariectomized group and estradiol treated group 18 hours and 5 days after treatment. 5. Serum prolactin levels were significantly changed after ovariectomy and estradiol injection. The levels were lower in ovariectomized rats than in intact control rats.

Experimental Studies on the Effects of Palmiwon (팔미원(八味元)이 성기능(性機能)과 항피로(抗疲勞)에 미치는 영향(影響))

  • It has been known for a long time that Palmiwon water extract can be prescribed for the treatment of deficiency of Yang in oriental medicine. But the exact mechanism of it has not been Known yet. In the present experiments, it was attempted to explain the effect of Palmiwon water extract on the serum Hormone level in rabbits and the antifatigue in mice. Palmiwon water extract oral intake showed a increase of serum hormone level in rabbits and the effects of the antifatigue in mice. The results obtained were as follows. 1. The administration of this medicine was observed to cause continuously a significant increase of the testosterone content in the serum from the 7th day to the 21th day. 2. The estradiol centent in the serum, as compared with the control experiment, was seen to increase significantly by the administration of this medicine from the 7th day to the 21th day. 3. The LH and FSH content in the serum, as compared with the control experiment, indicated a tendency toward an increase by the administration of this medicine from the 7th day to the 21 th day, but with no significant difference. 4. In the effect to Palniwon on the antifatigue in mice, swimming time was prolonged in both sample A, B groups than control group, and significance revealed in both sample A, B groups. These data suggest that Palmiwon water extract can increase serum hormone level and that it can have therapeutic effectiveness to the antifatigue.

Effects of Paljinickmohwan on the Ovulation and Ovary in Rats (팔진익모환이 백서의 배란과 난소에 미치는 영향)

  • Paljinickmohwan(八珍益母丸) is used in female infertility. especially due to deficiency of qi and blood or Qihyulyanghe(氣血兩虛). An attempt was made to evaluate the influences of PJIMH on the serum concentrations of FSH, LH, and estradiol(E2), the histological changes of ovary and the immunohistochemical staining for AT2 receptor in ovary of rats. The results of the study were as follows : Blood FSH level significantly increased in experimental group as compared with control group. In blood LH level, experimental group as compared with control group showed no efficacy. Blood E2 level significantly increased in experimental group as compared with control group. In histological observations of ovary, ovulation increased in experimental group as compared control group, which showed no efficacy. In observations of immunohistochemical staining for AT2 receptor in ovary, there is no difference between control group and experimental group. According to these results, it can be concluded that PJIMH influences ovary to increase the ovulation of rats.

Spermatogenic index and hormonal profile in the rats received chromatographic fractions of ethanol extract of Crotalaria juncea L. seeds

  • The ethanol extract of the Crotalaria juncea seeds, which showed promising antispermatogenic and antiandrogenic activities in albino mice, was taken up further for the isolation of the active fractions present in it. Two fractions that were obtained from thin layer chromatography were subjected for testing to know their antispermatogenic and antiandrogenic activities. After preliminary trials the fraction I showed maximum antifertility activity at the dose level of 200 mg/kg body weight when administered orally to the rats for 50 days. The fraction I was found to affect spermatogenesis as well as the endocrine functions of the testis as indicated by gravimetric, histopathological and biochemical changes. Further this fraction has caused degenerative changes in the seminiferous tubules and Leydig cells of the testis. The accessory reproductive organs like epididymis, seminal vesicles, vas deferens, prostrate, Cowper's gland and Levator Ani muscle showed significant malfunction. Cauda epididymal sperm count and sperm motility were reduced significantly. The treatment has also resulted in increase in the cholesterol level and alkaline phosphatase activity, and decrease in protein, glycogen, sialic acid contents and acid phosphatase activity in testis. It is noteworthy that RIA studies have shown significant reduction in serum FSH, LH and testosterone. Scanning electron microscopic observations revealed abnormalities in sperm structure.

Identification of Genes Regulated by PKC${\zeta}$ during Ovulation in the Rat

  • Our previous study demonstrates a rapid activation of atypical PKC${\zeta}$ by the ovulatory dose of LH/hCG. The present study was therefore designed to identify PKC${\zeta}$ regulated-genes in rat ovarian preovulatory granulosa cells. Preovulatory granulosa cells cultured in the presence of myristoylated PKC${\zeta}$ pseudosubstrate peptide were subjected to identify differentially expressed genes by using anneling control primer RT-PCR. As a result, among sixteen genes identified, six genes (testin, glypican-4, retrovirus SC1, connective growth factor, aminolevulinic acid synthase1 and serum- inducible kinase) were rapidly stimulated by hCG. Northern blot analysis demonstrated that all these genes were rapidly stimulated by hCG and declined thereafter. In situ hybridization analysis revealed the expression of these genes in granulosa cells of preovulatory follicles. The present study demonstrates time- and cell-specific expression of PKC${\zeta}$-regulated genes, and may imply that these genes play a specific role(s) during LH-induced ovulation.
