Effect of Cortisone on Serum Protein of Gamma-Irradiated Mice ($\gamma$線에 照射된 마우스의 血淸蛋白質에 미치는 Cortisone의 影響)

  • Male mice of strain SM were given 128 rads of single whole-body gamma-irradiation of $^{60}Co$, 14 to 16 minutes following a subcutaneous injection of physiological saline or cortisone acetate (1mg/day, for 4 days preirradiation). The serum protein patterns and the level of the total serum proteins were determined at various time intervals after exposure. Total serum protein was determined by Biuret method and serum protein fractions and A/G ratio were determined by paper electrophoresis using Whatman No.1 filter paper and barbital buffer (pH 8.6, ionic strength 0.06). 1. Total body gamma-irradiation caused a rise in the level of the total serum protein at 1 day and in the level of the serum albumin-globulin ratio at 5 days in both cortisone acetate-treated and control groups. 2. Cortisone acetate delayed the total serum protein rise at 5, 10, and 20 days after exposure. 3. Cortisone acetate delayed the A/G ratio rise at 1, 5, and 10 days after exposure. 4. It may be inferred that cortisone greatly reduces the sensitivity of mice to gamma-irradiation on the blood protein, probided that cortisone is given before the exposure.

  • This study was conducted to compare the effect of dietary sucrose and starch on the lipid metabolism of Rat fed by low protein diet for 3 and 6 weeks periods. Forty male weanling Spargue-Dawley rats weighing 52.7 grams each, after being adopted for 2 days with standard diet, were blocked into 8 groups and fed experimental diet as designed. Experimental diet were composed of two different levels of proteins, 6% designed as low protein diet and 15% designed as standard protein diet. In each group, the content of serum lipid, serum cholesterol and liver lipid were measured. The results of this experiment were summerized as follows. 1. liver lipid content was tended to be high in the sucrose group of low protein fed animal. 2. the content of total serum lipid was tended to be high in the sucrose group, and this tendency was showed statistical significance in the animals fed by low and standard protein after 6 weeks of experimental period. 3. The difference in the total serum cholesterol content between the sucrose and starch group was not significant

  • Casein or soybean protein was subjected to there action with caffeic acidtyrosinase system at 30-35$\circ$C, pH 6.8 with aeration for 5hr. The resulting brown proteins were washed with acetone until the washings were on longer colored. However, modified protein still retained a light brown. The effects of the modified proteins and brown compounds on male Wistar strain rats were studied by pair-feeding of a cholesterol-free diet for 14days. Significant decrease in protein digestibility for the rats fed with the modified proteins were observed. Weight gain and protein digestibility were not influenced by feeding brown compounds, but the feeding of brown compound from casein caused an enlargement of caecum. The concentrations of serum cholesterol and triglyceride in the rats fed with modified proteins and brown compounds were mostly unchanged against the rats fed with untreated proteins. These results suggest that the decrease in protein digestibility induced by enzymic browning-reaction did not cause the decrease in concentration of serum cholesterol.

Detection and Prognostic Analysis of Serum Protein Expression in Esophageal Squamous Cell Cancer

  • Objective: To assess differences in serum proteins in esophageal squamous cell carcinoma patients. Methods: 144 esophageal squamous cell carcinoma patients and 50 healthy volunteers were included in this study, with surface-enhanced laser desorption-ionization time-of-flight mass spectrometry and weak cation exchange magnetic beads. Follow-up allowed the relations between serum proteins and prognosis to be analyzed. Results: A total of 93 protein peaks were detected (molecular weight range: 1500-30000), 10 demonstrating statistically significant differences. There were no differences in protein peaks between 92 patients with a survival more than 2 years and 52 patients with survival less than 2 years. There were two significantly different protein peaks between 45 stage II patients with a survival more than 2 years and 14 stage II patients with survival less than 2 years. There was one significantly different protein peak between 22 stage III patients with a survival more than 2 years and 29 stage III patients with survival less than 2 years. Conclusion: Differences of serum proteins in esophageal squamous cell carcinoma are related to prognosis of patients. The protein fingerprint can be helpful for clinical diagnosis and treatment.

A Survey of Nutrition and Blood Contents of Rural Women in Korea (일부 농촌지역 부인의 영양실태 및 혈액성상에 관한 연구)

  • The nutritional survey and blood sampling was conducted from May 23 to 27, 1971, in order to investigate the state of the nutritional intake in relation to blood contents of rural women. This survey covered a total of 47 women residing in Youngam-gun, Cholla-namdo, Korea. The daily diet of these subjects was observed by 24-hour recall method and weighing method for 3 days and hemoglobin (Hb), hematocrit(Hct), mean corpuscular hemoglobin concentration (MCHC), serum protein, serum albumin, A/G ratio, serum iron, total iron binding capacity (TIBC), transferrin saturation (TS), serum cholesterol and blood specific gravity (SG) were measured. The results were summarized as follows ; The rate of intake of vegetable foods was high, especially cereals, and that of animal foods was low, especially eggs and milk and milk products. The daily intake of niacin was sufficient, and the intakes of energy and iron were also tend to be adequate. On the contrary, the intakes of vitamin A, $B_2$ and C, calcium and protein were insufficient. The mean red blood cell number was $410.8{\pm}26.4{\times}10^{4}/mm^{3},$ the mean values of Hb, Hct and MCHC were $12.5{\pm}0.9gm%$, $37.5{\pm}2.4%$ and $33.2{\pm}0.5%$ thus the anemic prevalences were 21.3%, 12.8% and 93.6% comparing with the anemic criteria of WHO. The levels of serum protein and serum albumin were $7.1{\pm}0.4gm\;%$ and $4.3{\pm}0.3gm\;%$so the A/G ratio was indicated $1.6{\pm}0.2.$. The levels of serum iron and TIBC were $91.9{\pm}27.5{\mu}g/100ml and $297.5{\pm}31.4{\mu}g/100ml,$ so TS was calculated $31.2{\pm}9.7\;%$. The mean value of SG was $1.044{\pm}0.002,$ and the level of serum cholesterol was $164.3{\pm}25.4mg/dl$. In the relation between each of bloood content, Hb showed positive correlation not only with Hct but also protein at the level of significance of 1 per cent and 5 per cent, and protein also showed a positive correlation with serum cholesterol at the 1 per cent level of significance. In the relation between the nutrient intake and blood contents, vitamin C intake showed a positive correlation with Hb, serum protein and serum iron at the level of significance of 1 per cent, iron intake also showed a positive correlation with serum iron and serum cholesterol at the same significant level, and protein intake showed a positive correlation with serum iron at the at the level of significance of 5 per cent.

  • The effects of the serum total protein and immunoglobulin levels of the colostrumconferred Korean native calves at 2 days of age on the disease incidence during the suckling period were studied. The results obtained were summerized as follows: 1. Serum total protein, total immunoglobulin, IgG, and IgM levels of 6 Korean native claves died from septicemia were lower than those of calves survived (p<0.05). 2. Korean native calves having low serum total protein and immunoglobulins were affected with diarrhea earlier in life, when compared with calves having high serum levels (p<0.05). 3. Korea native calves having low serum total protein were affected with bronchopneumonia earlier in life, when compared with calves having high serum levels (p<0.05). 4. Korean native calves having low serum neutralizing antibody titers to bovine viral diarrhea virus had high incidence rate of BVD, when compared with calves having high serum neutralizing antibody titers (p<0.05).

Effects of Dietary Chromium Picolinate on Performance, Egg, Quality, Serum Traits and Mortality Rate of Brown Layers

  • This experiment was conducted with total 960 brown layers, consisted of 8 treatment to investigate the effects of dietary chromium as chromium picolinate on egg production, egg quality, nutrient utilizability, serum traits and mortality in brown layers. Layers were fed diets with two levels of dietary protein (14% and 16%) and supplemented with 0, 200, 400, 800 ppb/kg of chromium as chromium picolinate, respectively. The highest egg production, egg weight and egg mass were found in 800 ppb chromium picolinate supplementation group with high protein level (16%) (p < 0.05). Although there was no significant difference, layers receiving 400 ppb of chromium picolinate with high protein (16%) represented the lowest broken eggs. The utilization of energy, dry matter and crude protein of 400 ppb chromium picolinate group with low protein level (14%) were significantly higher than those of control or other chromium picolinate group (p < 0.05). 400 ppb chromium picolinate with low protein level (14%) showed the lowest serum glucose concentration. But serum glucose concentrations in all treatments showed no significant differences. Present date revealed that the lowest serum cholesterol concentration of layers was found at 400 ppb chromium picolinate group with high protein level (16%) (p < 0.05). Crude protein content in yolk was significantly higher in eggs of layers received 800 ppb chromium picolinate and the lowest in eggs from layers received 400 ppb chromium picolinate among chromium picolinate levels (p < 0.05). Mortality was remarkably decreased by chromium picolinate supplementation and the lowest mortality value was found in layres receiving 800 ppb chromium picolinate with high protein level.

The Effect of Serum Chemical Values in Recipients on Pregnancy Rate following Embryo Transfer (수란우의 혈청화학치의 농도가 수정란이식 후 수태율에 미치는 영향)

  • This study was done to examine the relationship between serum chemical values (urea nitrogen, glucose, total protein and cholesterol) of recipients and pregnancy rate following embryo transfer. Blood samples were taken from 184 Holstein heifers or cows on Day 6 or 7 (Day 0=day of estrus) to analysis for serum urea nitrogen, glucose, total protein and cholesterol concentrations. After selection of recipients, frozen Holstein embryos were thawed and directly transferred to recipients non-surgically. The average serum concentrations of urea nitrogen, glucose, total protein and cholesterol were 13.8 mg/dl, 56.5 mg/dl, 7.2 mg/dl, 124.8 mg/dl, respectively. The average concentrations of serum urea nitrogen and cholesterol were lower (P<0.05) in pregnant recipients (10.7 mg/dl, 99.2 mg/dl) than in non-pregnant recipients (13.0 mg/dl, 122.2 mg/dl), respectively, although the concentrations of glucose and total protein were not different. These results show serum urea nitrogen and cholesterol concentrations can be used important factors for selection of recipients in Holstein.

Effects of Soy Isoflavones on Lipid Profiles and Hepatic LDL Receptor mRNA Level in Growing Female Rats

  • The present study examined the effect of soy isoflavones on lipid metabolism in growing female rats. Rats were randomly assigned to three different groups and provided experimental diets for 9 weeks. The experimental groups were classified into 1) a control group, 2) a soy protein isolate group: soy (+)) group and 3) a soy protein concentration group: soy (-)) group. Diets contained either casein or one of two soy proteins with (soy (+)) or without isoflavones (soy (-)). Serum triglyceride concentration showed no significant differences among the experimental groups. Serum total cholesterol concentration was significantly lower in both the soy (+) and soy (-) groups than in the control group and LDL-cholesterol concentration was significantly lower in the soy (+). Serum HDL-cholesterol concentration was significantly higher in the control group than in the soy protein groups but the HDL-cholesterol share rate in total cholesterol tended to be lower in the control group than in the soy protein groups, insignificant as it was. Hepatic IDL receptor mRNA level was significantly increased in the soy (+) group when compared to the other two groups to be 20% higher than the control group. In conclusion, soy protein isolate, soy protein rich with isoflavones reduced serum total cholesterol and LDL-cholesterol concentration and increased hepatic IDL receptor mRNA expression in growing female rats. Therefore, it is considered that the intake of soy isoflvones during puberty can be advantageous in terms of the long-tenn control of serum lipid.

  • The effect of hepatic injury produced by CCL, was studied on rats receiving a low protein-high carbohydrate (7% casein), standard protein (20% casein) and a high protein diet (30% casein). The rats fed low protein diet are resistant to CCl$_4$ in its effects on the liver as judged by histology, serum enzymes(guanase, ALT) and the content of hepatic protein. On the other hand, the pretreatment of hydrocortisone before injection of CCl$_4$ to the rats fed a standard diet, slightly decreased both serum ALT and guanase activities. In the pretreatment of actinomycin D, the liver and serum guanase activities were significantly decreased. It indicates that the cause of increasing serum guanase is based on the alteration of membrane permeability and the result of accelerated enzyme synthesis in liver cells of CCl$_4$ intoxicated rats.
