A Linear Beacon System Featuring an Internal Deoxyguanine Quencher Allows Highly Selective Detection of Single Base Mismatches

  • The fluorescence intensity of a single-stranded oligonucleotide containing a fluorene-labeled deoxyuridine $(U^{Fl})$ unit increases by only 1.5-fold upon formation of its perfectly matched duplex. To increase the fluorescence signal during hybridization, we positioned a quencher strand containing a deoxyguanine (dG) nucleobase, functioning as an internal quencher, opposite to the $U^{Fl}$ unit to reduce the intrinsic fluorescence upon hybridization with a probe. From an investigation of the optimal length of the quencher strand and the effect of the neighboring base sequence, we found that a short strand (five-nucleotide) containing all natural nucleotides and dG as an internal quencher was effective at reducing the intrinsic fluorescence of a linear beacon; it also exhibited high total discrimination factors for the formation of perfectly matched and single base-mismatched duplexes. Such assays that function based on clear changes in fluorescence in response to single-base nucleotide mutations would be useful tools for accelerating diagnoses related to various diseases.

  • The fine structure of the hemocytes in Wolf spider, Pardosa astrigera, is discribed and compared with that of similar cells in other spider species and insects. Five hemocyte types are identified in the hemolymph: prohemocyte, plasmatocyte, granulocyte, spherulocyte and adipohemocyte. Prohemocytes are small with a relatively undifferentiated cytoplasm. The nucleus is comparatively large and has a perinuclear space. Plasmatocytes and granulocytes are pinocytotic function in the hemolymph of the body. The plasmatocytes have some coated pits on the plasma membrane and well developed Golgi complex, The granulocytes appear sequence of events in the formation of coated vesicle from a coated pit on its plasma membrane. Golgi complex become well expressed and give rise to small secretory vesicles which fuse to large bodies. The spherulocytes are larger in cell size than other hemocytes. Their cytoplasm is filled with spherules. The spherules contain the floccurent materials and the helical structured materials, which are 220nm in length and 80nm in width. The adipohemocytes are oval shaped and have a number of lipid droplets.

  • Blind identification and equalization of communication channel is important because it does not need training sequence, nor does it require a priori channel information. So, we can increase the bandwidth efficiency. The linear prediction error method is perhaps the most attractive in practice due to the insensitive to blind channel estimator and equalizer length mismatch as well as for its simple adaptive algorithms. In this paper, we propose method for fractionally spaced blind equalizer with arbitrary delay using one-step forward prediction error filter from second-order statistics of the received signals for SIMO channel. Our algorithm utilizes the forward prediction error as training sequences for data estimation and desired signal for channel estimation.

  • The pressing roller was designed and manufactured with high modulus carbon fiber composite material to exploit the high specific stiffness of the composite material. the optimal stacking sequence for the pressing roller was obtained from the FE analysis and the shape of the rubber coating layer was determined based on the calculated deflection for the uniform pressure on the film along the axial length of the pressing roller. Then the static deflection of the manufactured composite pressing roller was experimentally evaluated in comparison with analysis result and dynamic characteristics were measured through vibrational test.

  • When driving on uniformly tining concrete pavement road, the whine noise which has high spectrum at the specific frequency related to the uniform tining space exist. In this paper, a randomization method of tining space for concrete pavements is proposed to reduce the whine noise. The proposed method is developed based on the LCG(linear congruential generator) algorithm. The design parameters used in the method are number of different tining space, ratio of each tining space, total linings of each size. sequence of actual tining under given constant drag length and texture. Monte Carlo simulations are used to validate the proposed method.

  • Chrysanthemum stunt viroid (CSVd) ws identified in chrysanthemum cv. Chunkwang showing symptoms of stunt with leaf distortion (K1) and stunt with chlorosis of leaves (K2) collected from the main cultivation area of Masan, Kyongnam province in Korea. The specific RNAs related with the diseased chrysanthemums were detected. Full-length 354 bp CSVd cDNAs were amplified from infected tissue by reverse transcription and polymerase chain reaction using a pair of primers specific for CSVd sequence. The amplified cDNA products were analyzed by agarose gel electrophoresis and the specific cDNAs were cloned. Nucleotide sequences of the two CSVd isolates K1 and K2 varied. Phylogenetic analysis of the nucleotide sequences of CSVd isolates indicated that K1 was closely related with J2 and Am 2 isolates. K1 and K2 were transmitted by grafting to Dendranthema grandiflorum cv. Mistletoe, Gynura aurantiaca, and Lycopersicon esculentum cv. Rutgers. This is the first report of CSVd in D. grandiflorum in Korea.

Analysis of ITS Nucleotide Sequences in Ribosomal DNA of Morus Species (뽕나무속 식물의 ITS 영역 염기서열 분석)

  • Nucleotide sequence in internal transcribed spacer (ITS) regions of ribosomal DNA among mulberry varieties (Morus species) were analyzed in order to identify the possibility of classification for the species. The variations in the ITS regions were compared among 9 mulberry varieties and one variety of Cudrania species as an outgroup. ITS 1 region of the varieties ranging from 219 to 220 bp in length was 49-50 bp shorter than ITS 2 region. Of 510 sites in the ITS 1 and 2 regions, 148 sites were potentially variable, of which 52% and 48% sites were distributed in ITS 1 and ITS 2 regions, respectively. By pairwise comparisons on the nucleotide sequences in the ITS 1 and 2 regions among 9 mulberry varieties, they were classified into 5 groups. Divergence values of the sequences, however, were considerably low ranging from 0 to 1.3%. Especially, there was no divergence among Backasipmunja, Chungilppong and Milsungpong and Jungyasang, Ssarigol II and Yulbon, respectively.

  • In this paper, a three-dimensional eco-epidemiological model with delay is considered. The stability of the two equilibria, the existence of Hopf bifurcation and the permanence are investigated. It is found that Hopf bifurcation occurs when the delay ${\tau}$ passes though a sequence of critical values. The estimation of the length of delay to preserve stability has also been calculated. Numerical simulation with a hypothetical set of data has been done to support the analytical findings.

  • GF(2)위에서 최대길이를 갖는 n-셀 90/150 셀룰라 오토마타의 각 셀은 길이가 $2^n-1$인 수열을 생성한다. 이러한 셀룰라 오토마타의 임의의 셀에 대한 출력수열은 다른 셀에 대한 출력수열의 위치를 이동함으로써 얻을 수 있다. 본 논문에서는 주어진 셀룰라 오토마타의 상태전이행렬의 특성다항식에 의한 동차 선형점화식을 만족하는 충격반응수열을 이용하여 셀들의 위상이동차를 계산하는 알고리즘을 제안한다.

Preform Design of the Bevel Gear for the Warm Forging using Artificial Neural Network (신경망을 이용한 정밀 베벨기어의 온간단조 예비성형체 설계)

  • In this paper, the warm forging process sequence has been determined to manufacture the warm forged product for the precision bevel gear used as the differential gear unit of a commercial automobile. The preform shape of bevel gear influences the dimensional accuracy and stiffness of final product. So, the design parameters related preform shape such as aspect ratio and chamfer length having an influence the formability of forged product are analyzed. Then the optimal conditions of design parameters have been selected by artificial neural network (ANN). Finally, to verify the optimal preform shape, the experiments of the warm forging of the bevel gear have been executed. The proposed method can give more systematic and economically feasible means for designing preform shape in metal forming process.
