We investigate the astronomical phenomenon records of Sukjong's Chunbang-Ilgi made by Sigangwon (Royal Educational Office of the Crown Prince) at which King Sukjong was the crown prince (i.e., 1667 January 24-1674 September 22). From the daily records of 2,799 days, we extract the astronomical records of 1,443 days and classify them into 14 categories. Then, we group the records of each category into five phenomena (Atmosphere, Eclipse, Daylight Appearance, Apparition, and Appulse) and compare them with the results of modern astronomical computations wherever possible. Except for Atmosphere group comprising records of meteorological events, such as solar halo, lunar halo, and unusual clouds, the significant findings in every other group are as follows: In Eclipse group, the solar eclipse that occurred on 1673 August 12 was unobservable in Korea, which is in contrast to the record of Joseonwangjo-Sillok (Annals of the Joseon Dynasty), which states that the sun was in eclipse around sunset time, as observed at Nam mountain. From the lunar eclipse records, we verify that the Joseon court did not change the date of the events observed after midnight. In Daylight Appearance group, we confirm that this phenomenon was observed during the daytime and not during twilight. We further suggest that if observation conditions are met, a celestial body brighter than -2.3 mag could be seen during the daytime with the naked-eye. In Apparition group, we find the possibilities that the Orionid meteor shower had influence on the meteor records and the seasonality on the aurora records. We also find that the Korean records in which the coma of comet C/1668 E1 was located below the horizon were overlooked in previous studies. Finally, we find that the records of Appulse group generally agree with the results of modern calculations. The records of Beom (trespass in literal) and Sik (eating in literal) events show average angular separations of 1.2° and 1.0°, respectively. In conclusion, we believe this work helps study the astronomical records of other logs of Sigangwon, such as Sukjong's Chunbang-Ilgi.
The article discusses the critical role of chromatography in the analysis and purification of proteins in biopharmaceuticals, emphasizing the importance of comprehensive characterization for ensuring their safety and efficacy. It highlights the use of Avantor® ACE® HPLC columns for the separation and purification of proteins, focusing on the analysis of intact proteins using reversed-phase liquid chromatography (RPLC) with fully porous particles. This article also details the application of different mobile phase additives, such as TFA and formic acid, and emphasizes the advantages of using type B ultra-pure silica-based columns for efficiency and peak shape in biomolecule analysis. Additionally, it addresses the challenges of analyzing intact proteins due to slow molecular diffusion and introduces the concept of solid-core (or superficially porous) particles, emphasizing their benefits over traditional porous particles for the analysis of therapeutic proteins. Furthermore, it discusses the development of Avantor® ACE® UltraCore BIO columns, specifically designed for the high-efficiency separation of large biomolecules, such as proteins, and demonstrates their effectiveness in achieving high-resolution separations, even for higher molecular weight proteins like monoclonal antibodies (mAbs). In addition, it underscores the complexity of analyzing and characterizing intact protein biopharmaceuticals, requiring a range of analytical techniques and the use of wide-pore stationary phases, operated at elevated temperatures and with relatively shallow gradients. It highlights the comprehensive range of options offered by Avantor® ACE® wide pore columns, including both fully porous and solid-core particles, bonded with a variety of complementary stationary phase chemistries to optimize selectivity during method development. The use of ultrapure and highly inert base silica is emphasized for enabling the use of lower concentrations of mobile phase modifiers without compromising analyte peak shape, particularly beneficial for LC-MS applications. Then the article concludes by emphasizing the significance of reversed-phase liquid chromatography and its compatibility with mass spectrometry as a valuable tool for the separation and analysis of intact proteins and their closely related variants in biopharmaceuticals.
The quantitative separations of a mixture containing equal amounts of each cation such as Mn(Ⅱ), Cr(Ⅲ), V(Ⅴ), Cu(Ⅱ), Ni(Ⅱ), Co(Ⅱ), and Fe(Ⅲ) are carried out by the elution through $35cm{\times}3.14cm^2$ column of cation exchange resin, $Dowex 50w{\times}12$. The eluents are a mixture of 0.6 M sodium chloride and 0.1 M sodium tartrate (pH = 2.00 and 4.50) for Fe(Ⅲ), V(Ⅴ), Cu(Ⅱ), Ni(Ⅱ) and Co(Ⅱ), and a mixture of 3 M sodium chloride and 0.1 M sodium tartrate (pH = 4.50) or a mixture of 0.7 M sodium chloride and 0.5 M sodium oxalate (pH = 4.50 and 5.00) for Mn(Ⅱ) and Cr(Ⅲ). The subsidiary cations in a standard iron mixture such as V(Ⅴ), Cu(Ⅱ), Ni(Ⅱ), Mn(Ⅱ) and Cr(Ⅲ) are separated together from the large amount of Fe(Ⅲ) through $15cm{\times}3.14cm^2$ column of the resin, $Dowex 1{\times}8$, by elution with the eluent of 4.0 M hydrochloric acid. A small amount of Fe(Ⅲ), however, is eluted together with Cu(Ⅱ). V(Ⅴ), Ni(Ⅱ), Mn(Ⅱ) and Cr(Ⅲ) eluted together are separated quantitatively through $10cm{\times}3.14cm^2$ column of the resin,$Dowex 50w{\times}12$. Cu (Ⅱ) and a small amount of Fe(Ⅲ) are separated quantitatively through $10cm{\times}3.14cm^2$ column of the resin, $Dowex 50w{\times}12$, by the elution with a mixture of 0.6 M sodium chloride and 0.1 M sodium tartrate (pH = 2.00 and 4.50) as an eluent. By the conditions obtained in the separations of the standard iron mixture, Fe(Ⅲ) and all of the subsidiary cations in steel are quantitatively separated.
The physico-chemical properties of nine selected thallium borosilicate glasses and other 21 supplementary compositions were investigated. Their composition-property curves are found to be in many respects analogous to those of other borosilicate glasses containing lithia, soda, and lead oxide. It is indicated that certain minima found in the composition-property curves of thallium borosilicate glasses might be caused by a change in boron coordination as has been observed to occur in the $Na_2O-B_2O_3-SiO_2$ glasses. Typical effects of thallium ions on the borosilicate glass are summarized as follows: 1) Addition of thallium ions increased density, refractive index, water solubility, linear coefficient of thermal expansion, and dielectric constant. 2) Increased concentration of thallium decreased the softening point of the glasses, caused fluorescence under ultraviolet radiation and smeared out the absorption edges up to $15{\mu}$ in the infrared region. An extensive liquid immiscibility was found by replication electron microscope technique in the $Tl_2O-B_2O_3-SiO_2$ system. The immiscibility covers a composition range roughly from 55 wt. % Tl2O to the binary system $B_2O_3-SiO_2.$ By acid treatment, it was found that the immiscible glass consists of separate silica-rich and boron-rich phases.
The crystal structure of bis(N-methylphenazinium) bis(oxalato)palladate(II) has been determined by X-ray crystallography. Crystal data: ((C_{13}H_{11}N_2)_2[Pd(C_2O_4)_2]) $M_w$ = 672.93, Triclinic, Space Group P1 (No = 2), a = 7.616(8), b = 9.842(3), c = $20.335(7)\AA$, $\alpha$ = 103.53(3), $\beta$ = 90.00(5), $\gamma$ = $112.38(5)^{\circ}$, Z = 2, $V = 1363(2){\AA}^3\;D_c = 1.639\;gcm^{-3},\;{\mu} = 7.3\;cm^{-1},\;F(000) = 680.0$. The intensity data were collected with $Mo-K\alpha$ radiation (${\lambda}$= 0.7107\;\AA)$ on an automatic four-circle diffractometer with a graphite monochromater. The structure was solved by Patterson method and refined by full matrix least-square methods using Killean & Lawrence weights. The final R and S values were $R = 0.069,\;R_w = 0.050,\;R_{all} = 0.069$ and S = 5.45 for 3120 observed reflections. Both cation and anion complexes are essentially planar and have dihedral angles of 6.3(6) and $57.06(6)^{\circ}$ between their planes. The planar complex anions are sandwiched between slightly bent cations. The interplanar separations of two triads are 3.328 and 3.463 $\AA$, respectively. The triads are stacked along b-axis, but their orientations are different based on dihedral angle $59.08(9)^{\circ}$ of two complex anions.
A rapid analytical method based on capillary electrophoresis is described for the determination of trace anions in high-purity chemicals which is used to prevent corrosion demage in nuclear power plants. Separations are carried out at 20kV using trimethylsilane-coated fused-silica capillary ($70cm{\times}50$ or $75{\mu}m$ i.d.) with the electrolyte of 5mM Chromate(pH=8). Detection was achieved using on-column indirect photometry at 254nm. The simultaneous analysis of inorganic anions, chloride, nitrate, sulfate, azide and phosphate was performed using methods of hydrodynamic(>1ppm) and / or electrokinetic(<1ppm) injection. The results of studies on the coexisting anions on analyte ions shows that peak responses of analyte in hydrodynamic injection is constant without effect of coexisting anions, but those of analysis in electrokinetic injection is strongly dependant upon the kind of coexisting anions and its ionic mobility. The analyte enrichment rate, hence peak response, is positive relationship with the resistance of the sample solution. Thus, appropriate measures, such as standard addition or internal standard technique, must be used to account for differences in conductance of standard and sample solutions.
Strains of Fusarium oxysporum f. sp. lycopersici isolated from Korea, Japan and U.S.A. were used for electrophoretic karyotype (EK) analysis. Chromosome separations on FastLane agarose gels (FMC BioProducts, Rockland, ME), called pulsed field gel electrophoresis (PFGE), were performed by CHEF-DRII apparatus (Bio-Rad Laboratories, Melville, NY) using TAE as a running buffer. To obtain optimal condition for separation of chromosome sized DNAs, variable running conditions such as field strengths, swithching intervals, and running time were applied in CHEF gel electrophoresis. We were able to resolve 9 to 11 chromosome sized DNAs ranging in size from 0.76 to 6.41 Mb in isolates from Korea and estimate that the total genome size was ranging from 35.29 to 38.92 Mb. Distinct differences in length range and genome size exist among isolates from different countries. Isolates from Japan and U.S.A. were resolved 9 to 11 chromosome sized DNAs ranging in size from 1.24 to 6.85 Mb and estimated that the total genome size was ranging from 35.32 to 43.87 Mb. Isolates from variable provinces in Korea had the same or similar chromosomal polymorphism and showed different chromosomal DNA patterns compared to isolates from the other countries.
The seed of cowpea (Vigna sinensis K.) was extracted with 80% aqueous methanol (MeOH). And the concentrated extract was partitioned with ethyl acetate (EtOAc), n-butanol (n-BuOH) and $H_2O$, successively. The repeated silica gel and octadecyl silica gel (ODS) column chromatographic separations for the EtOAc and n-BuOH fractions led to isolation of four sterols. And the chemical structures of the compounds were determined as a mixture of stigmasterol and $\beta$-sitosterol with the ratio of 4 to 3 (1), 7-ketositosterol (2), and stigmasterol 3-O-$\beta$D-glucopyranoside (3) from the interpretation of spectroscopic data including nuclear magnetic resonance (NMR) spectrum metric, mass (MS) spectrum metric and infrared (IR) spectroscope. This study reports the first isolation of $\beta$-sitosterol, 7-ketositosterol, and stigmasterol 3-O-$\beta$-D-glucopyranoside from the seed of Vigna sinensis K. In addition, compound 2, 7-ketositosterol, is rarely occurred in natural source including plant.

Runoff characteristics of the Oedocheon in Jeju island were investigated using the long-term stream stage monitoring data. At the Cheonah valley in the upstream area and Oedocheon downstream, annual runoff occurred 21 and 12 times, respectively, and their average runoff periods were 21 days and 12 days, respectively. Stream stage response time to rainfall was 4 hours, and storm-water transfer from the upstream, Cheonah valley, to the Oedocheon downstream took about 2 hours. The stream discharge measurements had been carried out from Feb. 2004 to Jul. 2005, and showed that normal discharge of the Oedocheon was 0.39 $m^3$/sec in average. Stage-discharge curves were developed to estimate base flow (normal discharge) and (direct) surface runoff. The base flow separations by a numerical filtering technique illustrated that annual surface runoff and base flow accounted respectively for 31.8${\sim}$36.5%, 63.5${\sim}$68.2% of the total stream discharge.

Purpose: To analyze differences in the dose uniformity for the computed breast radiation therapy planning with tangential beam between conventional RT using wedge filter and FiF-IMRT using multileaf collimator based onsizes and volumes of breasts. Materials and Methods: Thirty breast cancer patients were classified according to the sizes and volumes of the breasts using Eclipse treatment planning system ($Varian^{TM}$, USA, V8.0). Conformity Index and Homogeneity Index were computed along with Dose Volume Histogram. Results: No differencein CI (${\pm}1.2%$) was observed. However, lower mean HI (1.67%) in FiF-IMRT was observed compared to that of the conventional RT. Statically significant (P<0.01) correlation was identified between the values of ${\Delta}HI$ (%) and physical parameters such as breast volumes and separations. Conclusion: Increase in breast volume and separation improves the dose uniformities in computed radiation therapy planning for FiF-IMRT. Physical dimension of the breast should be considered to optimize the compured radiation therapy planning.
