Analysis of Heme Biosynthetic Pathways in a Recombinant Escherichia coli

  • Bacterial heme was produced from a genetic-engineered Escherichia coli via the porphyrin pathway and it was useful as an iron resource for animal feed. The amount of the E. coli-synthesized heme, however, was only few milligrams in a culture broth and it was not enough for industrial applications. To analyze heme biosynthetic pathways, an engineered E. coli artificially overexpressing ALA synthase (hemA from Rhodobacter sphaeroides) and pantothenate kinase (coaA gene from self geneome) was constructed as a bacterial heme-producing strain, and both the transcription levels of pathway genes and the intermediates concentrations were determined from batch and continuous cultures. Transcription levels of the pathway genes were not significantly changed among the tested conditions. Intracellular intermediate concentrations indicated that aminolevulinic acid (ALA) and coenzyme A (CoA) were enhanced by the hemA-coaA co-expression. Intracellular coproporphyrinogen I and protoporphyrin IX accumulation suggested that the bottleneck steps in the heme biosynthetic pathway could be the spontaneous conversion of HMB to coproporphyrinogen I and the limited conversion of protoporphyrin IX to heme, respectively. A strategy to increase the conversion of ALA to heme is discussed based on the results.

  • Today, it is not difficult to use information technology (IT), especially, Internet based ones. Many people can not only access IT without learning how to use it but also find and develop new techniques and usages which couldn't be expected by system engineers or designers. This is owing to social interactions among users as well as advancement of IT. Theoretically, such social interactions in using IT can be well explained by adaptive structuration theory (AST) which has been considered as one of trying to capture the change of using IT due to social interactions between users and system. This study is to analyze the relationship between social interactions and motivation in using IT which can determine attitude and intention of using IT. For this purpose we provide a research model, in which two AST related variables, faithfulness of appropriation and consensus on appropriation, are independent variables and three beliefs for using IT, usefulness, ease of use and playfulness, are dependent ones. Additionally, for reflection of changing uses, usefulness is formed as second order factor by two first order factors-usefulness of self-expression and communication. An empirical test of our model for blog users which is analyzed by Partial Least Square method shows supporting most of hypotheses except one, consensus-ease of use.

Dieticians' intentions to recommend functional foods: The mediating role of consumption frequency of functional foods

  • This study explored the conceptual framework of dieticians' intentions to recommend functional food and the mediating role of consumption frequency. A web-based survey was designed using a self-administered questionnaire. A sample of Korean dieticians (N=233) responded to the questionnaire that included response efficacy, risk perception, consumption frequency, and recommendation intention for functional foods. A structural equation model was constructed to analyze the data. We found that response efficacy was positively related to frequency of consumption of functional foods and to recommendation intention. Consumption frequency also positively influenced recommendation intention. Risk perception had no direct influence on recommendation intention; however, the relationship was mediated completely by consumption frequency. Dieticians' consumption frequency and response efficacy were the crucial factors in recommending functional foods. Dieticians may perceive risks arising from the use of functional foods in general, but the perceived risks do not affect ratings describing dieticians' intentions to recommend them. The results also indicated that when dieticians more frequently consume functional foods, the expression of an intention to recommend functional foods may be controlled by the salience of past behaviors rather than by attitudes.

  • The purpose of this study is to analyze the background and characteristics of fashion in wanna-be phenomenon. The primary source of data has been a collection of recent books, news repots, many articles from various kinds of mass media and fashion internet cite. The results of this study can be summarized as follow. First, The wanna-be phenomenon can be divided the background into three parts : change to entertainment society, increase of mass consumption, increase mutual communication with star and fan. Second, the function of fashion in wanna-be phenomenon can be divided with the following: the function of self-expression, guide book of trendy lifestyle, and play for pleasure. Third, fashion style in wanna-be phenomenon can be divided with the following : chic & gorgeous style, sexy casual & chav style, bohemian mix & match style.

Tumor-derived CD4+CD25+ Tregs Inhibit the Maturation and Antigen-Presenting Function of Dendritic Cells

  • CD4+CD25+regulatory T cells (Tregs) play a key role in regulation of immnue response and maintenance of self-tolerance. Studies have found Tregs could suppress tumor-specific T cell-mediated immune response and promote cancer progression. Depletion of Tregs can enhance antitumor immunity. Dendritic cells (DCs) are professional antigen-presenting cells and capable of activating antigen-specific immune responses, which make them ideal candidate for cancer immunotherapy. Now various DC vaccines are considered as effective treatment for cancers. The aim of this study was to evaluate variation of Tregs in BALB/C mice with hepatocellular carcinoma and investigate the interaction between tumor-derived Tregs, effector T cells (Teff) and splenic DCs. We found the percentages of Tregs/CD4+ in the peripheral blood of tumor-bearing mice were higher than in normal mice. Tumor-derived Tregs diminished the up-regulation of costimulatory molecule expression on splenic DCs, even in the presence of Teff cells and simultaneously inhibited IL-12 and $TNF-{\alpha}$ secretion by DCs.

  • In the rapidly changed 21st century when the flow speed of information is so fast and the quantity of information is too much increasing, personal appearance and characteristic image-formation is highly evaluated. Nowadays personal appearance is very essential to competition and may be considered the non-linguistic expression of self-image. The beauty action of the youth is shown to follow the object which youth want to rely on or long for and it is affected by the influence of the sociocultural elements. The purpose of this study is to identify the corelation between appearance and beauty action according to the sociocultural elements of the juveniles concerning appearance and to make an inquiry into how they pursue beauty in the restrictive bound compared with the adults. As their restrictive beauty action, the practical reflection percentage of hair-style, management of face, hand and nail control will be investigated and analyzed. Furthermore this basic material of this survey was used for the development of beauty article, cosmetics and education program. With the findings of the beauty articles in which the growing generation have a deep interest, this study provided the basic materials to the customer analysis and also the reference materials to the beauty marketing.

The Social Aspects and Costumes of the 1980's Expressed in the Movie 'American Psycho' (영화 '아메리칸 사이코'에 나타난 1980년대의 사회상과 복식에 관한 연구)

  • A movie's fashion style delivers the overall atmosphere of the scene including the characters's class, personality, spiritual world and inner thinking and even their conflicts in the story. The movie 'American Psycho' directed by Mary Harron and based on from Bret Easton Ellis's original novel ridicules the American yuppie culture of the 1980's through the behavior of the hero Patrick Bateman. The life style of the yuppie sees itself as the high-class embodiment of a particular culture, but the various subcultures such as Glam and Punk show that it is merely a two-faced culture suffering from hypocrisy and mammonism. An analysis of the costumes found in the movie indicated an exhibition of the 1980's Haute Couture fashion, which was mainly occupied by the mainstream social class and of the social phenomenon of post-modernism. The anti-fashion presented in the movie as the resistance culture formed by the subculture was in extreme contrast with the expression of self-actualization.

The Symbolic Meaning and Values Portrayed In Models' Characteristics in Fashion Advertisements

  • Various current events provide evidence that society is undergoing changes in perceptions of social relationships. Specifically, visual media in the form of advertisements can convey images which reflect society's values and concepts about role relationships. The purpose of this research was to examine ads in fashion magazines to determine what types of model roles and role relationships typically appear in fashion advertising which can mirror society's values. A content analysis was conducted of ads obtained from US Vogue and US GQ for the year 2002. Six kinds of roles/relationships were found: (1) Narcissism (representing self absorption), (2) sexually enticing opposite-sex relationships, (3) close/romantic same-sex relationships, (4) friend relationships, (5) family relationships, and (6) independent relationships. Of these, narcissism predominated, however, a small number of sexually provocative ads appeared as well as same-sex romantic relationships. Because sole (single) models were more typical, they also were examined to determine ways in which they relate to the audience. Characteristics examined included body presentation & pose, eye gaze, and facial expression. Direct eye gaze was the typical way to engage the audience. Gender differences were apparent: smiling was more typical of women, indifference for men. The symbolic meaning and values investigated from this research are the blurring of gender identity portrayed in homosexual imagery, family values, and the value of youth. The consistency of models' race in ads does not portray the diversity reflected in the demographic census.

  • Notch signaling plays a pivotal role in cell fate determination, cellular development, cellular self-renewal, tumor progression, and has been linked to developmental disorders and carcinogenesis. Notch1 is activated through interactions with the ligands of neighboring cells, and acts as a transcriptional activator in the nucleus. The Notch1 intracellular domain (Notch1-IC) regulates the expression of target genes related to tumor development and progression. The Notch1 protein undergoes modification after translation by posttranslational modification enzymes. Phosphorylation modification is critical for enzymatic activation, complex formation, degradation, and subcellular localization. According to the nuclear cycle, Notch1-IC is degraded by E3 ligase, FBW7 in the nucleus via phosphorylation-dependent degradation. Here, we summarize the Notch signaling pathway, and resolve to understand the role of phosphorylation in the regulation of Notch signaling as well as to understand its relation to cancer.   • The purpose of this study is to analyze the tech fatale fashion image in female models of mobile advertisements and to find the fashion characteristics. The tech fatale emerging as new culture code is a compound word of the femme fatale and technology. It is characteristics of the femme fatale, the post digital culture and the female leadership. The findings of the study were as follows : The tech fatale types were the independence, the transformation and the tradition. The independence was a self expression, appealed to visual image, was showed the coating fabric, denim, space look and street fashion and reflected the creativeness and digital generation. The transformation appealed to sexuality and was showed luster fabric, exposure, body-conscious, glam look. The voluptuous beauty represented the pride of the post digital generation. The tradition appealed to emotion and was showed pale color, simple line, soft texture fabric and a feminine Image. The meanings of tech fatale were the imagination, the public, the duality, the game, the purity and the recurrence. The formative characteristics reflect the mind of post digital generation who is against authority and pursues the human being worth such as the identity establishment and the pure emotion.
