Cotylodon Development and Seeding Growth of Lotus corniculantus , Lotus tenuis and Their Reciprocal Hybrids (Lotus corniculantus , Lotus tenuis 및 이들 상호 교잡종의 자엽과 유식물의 생장발육)

  • Seedling development among reciprocal FI hybrids and their parents, Lotus comiculatus and L renuis, were studies using a simple method of measuring cotyledon area. Cotyledon area attached to the plant could be measured with simple rapid method. Cotyledons expanded very rapidly during the first week, then slightly expanded until maximum size was reached around 3 weeks after emergence. Cotyledon area upon emergence was correlated with seed size (P=0.05), but after emergence there was a divergence in cotyledon expansion rate. Rapidity of photosynthetic area development was determinant factor for seedling growth of trefoil species. 2 $\times$ m2 and 1 $\times$ m2 among the hybrids promised the possibility increasing seedling vigor of birdsfoot trefoil.

Simple sequence repeat marker development from Codonopsis lanceolata and genetic relation analysis

  • In this study, we developed 15 novel polymorphic simple sequence repeat (SSR) markers by SSR-enriched genomic library construction from Codonopsis lanceolata. We obtained a total of 226 non-redundant contig sequences from the assembly process and designed primer sets. These markers were applied to 53 accessions representing the cultivated C. lanceolata in South Korea. Fifteen markers were sufficiently polymorphic, and were used to analyze the genetic relationships between the cultivated C. lanceolata. One hundred three alleles of the 15 SSR markers ranged from 3 to 19 alleles at each locus, with an average of 6.87. By cluster analysis, we detected clear genetic differences in most of the accessions, with genetic distance varying from 0.73 to 0.93. Phylogenic analysis indicated that the accessions that were collected from the same area were distributed evenly in the phylogenetic tree. These results indicate that there is no correlative genetic relationship between geographic areas. These markers will be useful in differentiating C. lanceolata genetic resources and in selecting suitable lines for a systemic breeding program.

Effect of Cell Size on Growth and Development of Plug Seedlings of Three Indigenous Medicinal Plants (플러그 셀 크기가 세 가지 자생 약용식물 묘 생육에 미치는 영향)

  • There have not been many studies conducted on the seedling production, especially in plug trays, of traditional medicinal plant species. In an effort to establish guide lines for seedling production, this study investigated the effect of plug cell size on the growth and development of plug seedling of three medicinal plant species. Seeds were sown in either 128, 200, or 288-cell plug trays, containing a commercial medium. Growth and development of individual seedling was generally promoted with increasing size of a plug cell in all of the three species. The greatest biomass of the seedlings gained in a plug tray was obtained in the 288-cell trays in Perilla frutescens var. acuta Kudo and Sophora tonkinensis, and the 200-cell trays in Angelica gigas Nakai. Overall growth and development of the shoot and root of a single seedling of Perilla frutescens var. acuta Kudo, except total chlorophyll and anthocyanin contents, was the greatest in the 128-cell tray. However, length of the longest root, length, width and area of the leaf, internode length, root fresh weight, and root ball formation in the 200- and 288-cell trays were not significantly different each other. In Sophora tonkinensis, although length of the longest root, stem diameter, leaf width, leaf area, shoot fresh weight, and root ball formation were not significantly different among the treatments, length of the longest root and root ball formation of a single seedling were the greatest in the 128-cell tray. Overall shoot and root growth, except total chlorophyll content, of a single seedling of Angelica gigas Nakai was the greatest in the 128-cell tray. Based on the total biomass, it is concluded that 288-cell trays are recommended for production of plug seedlings of medicinal plant species P. frutescens var. acuta Kudo and S. tonkinensis. In A. gigas Nakai, it would be more economical to use the 200-cell trays than 128-cell trays due to total biomass.

Effect of Several Herbicides on Weed Control and Seedling Growth in Rice Nursery (수도(水稻)못자리에 있어서 몇가지 제초제처리(除草劑處理)가 잡초방제(雜草防除) 및 묘생육(苗生育)에 미치는 영향(影響))

  • This experiment was carried out to clarify the effect of herbicide chlormethoxynil 7G, CNP 9G and butachlor 3.5/pyrazolate 6G on weed control and seedling growth in rice seedbed. Different quantities of the herbicides were applied as soil incorporation or surface broadcasting. At earlier sowing annual grasses such as Echinochlva spp. and Alopecurus spp. were dominated, but Cyperus spp. were dominated at later sowing. Weed suppression by herbicides was greater in soil incorporation than in broadcasting and chlormethoxynil 7G and CNP 9G application (30kg/ha) showed about 70% weed control, while butachlor 3.5/pyrazolate 6G (10-20kg/ha) showed 90 to 100% weed control. At lower rate of butachlor 3.5/pyrazolate 6G (10kg/ha) emergence ratio, healthy seedling ratio and relative growth rate of rice were higher, and phytotoxicity was lower than in the others.

  • Effect of eight different treatments to stimulate germination and of seedling ageson the growth and tuber yields of Liriope platyphlla was carried out in vitro(room temperature)and in vivo(field). Treatments, in vitro, of sulfuric acid(2%), $GA_3$(100ppm), and stratification were most stimulated the germination of L.platyphylla. Among them the stratification was also the most effective for emergence in field and was shortened the days to germination in vitro and in vivo. Plant height and the number of leaves and tillers per plant in the sulfuric acid and the vinyl mulch treatments were the poorest, but those in other treatments were highered without any differences. Seeding of L.platyphylla was taken 66 days to emerge and the growth of that was more decreased than that of the seedlings. The younger seedling, the shorter the day of emergence of a new shoot. The olders were bloomed earlier and more increased the growth than the yongers and the divided seedlings, but the tuber yield was most increased in the 3 years seedling.

Comparison of Pepper Grafting Efficiency by Grafting Robot (접목로봇의 고추묘 접목 효율성 비교)

  • Manual grafting is a labor-intensive operation and highly susceptible to human error. Development of grafting robot has been considered as an effective alternative to manual grafting. The study was conducted to investigate the grafting efficiency between the domestically produced automated grafting robot and traditional manual grafting performed in a commercial plug seedling greenhouse. Plug seedlings of pepper (Capsicum annuum L.) 'Buchon' and 'Anseongmatchum' were sown as scions and rootstocks, respectively on March 29, 2013. The grafting was performed by a grafting robot, amateur grafters, and professional grafters on May 31, 2013. After grafting, the seedling efficiencies and seedling characteristics were compared in terms of plant height, the number of success as well as failure of seedling grafts produced in an hour. The results showed that plant heights were most uniform in the grafts performed by the professionals that had a 0.6 cm difference whereas an uneven difference of 2.1 cm difference was observed in plug seedling grafted by an amateur. However, plug seedlings grafted by the grafting robot were found to be uniform with 1 cm plant height difference. Moreover, the maximum number of grafting processed plug seedlings per hour (667 plants) was achieved by the grafting robot on compared with the seedlings grafted (466 plants) by the professionals. Furthermore, the least failure rate of 21.7% was noted upon the usage of grafting robot than the manual grafters. Thus, the results demonstrate that the technology of automated grafting robots is the most appealing and can be employed in commercial plug seedling greenhouses to overcome the drawbacks of manual grafting.

Seed Gravity, Germination, and Optimum Seeding Rate for Machine Transplanting in Colored Rice Varieties (유색벼의 품종별 종자 비중, 발아 특성 및 기계이앙 육묘시 적정 파종량)

  • Seeds of colored rice generally have high anthocyanin and flavonoid content but some cultivars have low seed germination rate. This defect poses unstable seedling establishment in nursery bed. The seed gravity variations of 10 colored rice varieties and its effect on germination, seed soaking duration and their optimum seeding rates for seedling raising in machine transplanting were investigated in the experiment. Based on seed gravity distribution of the 7 black- and 3 red-colored rice varieties at three levels of seed gravity (1.0<, 1.0~1.06, 1.06>), the black colored seeds such as Josaengheugchal and Sinmyungheugchal had high amount of specific gravity of below 1.0 ranging from 86~ 96%, while they had only 3~13% of seeds in specific gravity above 1.06. Sintoheugmi, Heugjinju, Heugnam, Heugkwang had various mass of seeds, showing 29~44% for specific gravity of below 1.0, 24~39% for 1.0~1.06 and 25~45% for above 1.06. On the other hand, the red colored rice such as Hongjinju, Jeogjinju and Geonkanghongmi had high percentage of specific gravity of above 1.06 with 84~86% while they had only 9~12% of specific gravity below 1.0 similar to Ilmibyeo of noncolored rice. The black colored seed generally showed low germination percentage, slow germination speed and long mean germination time, and low water absorption rate as compared with seed of the red colored rice which was similar to those of Ilmibyeo used as a control cultivar. The black colored seeds took 2~4 days longer seed soaking duration than the red colored rice before germination. This was related to high seed amount of specific gravity below 1.0 in black colored rices. The high amount of seed gravity above 1.06 in the colored rice seeds was positively correlated with percentage of germination, germination speed and mean germination time, ripened grain ratio and water absorption. Seed gravity distribution effect were less pronounced between back- and red- colored seeds in seedling emergence. The black colored rice has slightly lower normal seedling emergence rate than the red colored rice and Ilmibyeo in the seedbed soil due to high percentage of ungerminated seed. Normal seedling emergence rate of the black colored rice in the seedbed was 75.2~82.2% for 10-day old seedling and 85.3~ 90.9% for 30-day old seedling which was lower by 4.5~ 8.0% and 0~3.3%, respectively, than the red colored rice. Based on the normal seedling number per tray of Ilmibyeo for seeding rate of 10-day and 30-day old seedlings, the recommended seeding rate of black colored rice is 200~ 220g seeds for the 10-day old seedling and 110~130g for the 30-day old seedling in transplanting rice while the seeding rate of the red colored rice was 220g seeds for the 10-day old seedling and 130g for the 30-day old seedling.
