Current Status of Vegetable Grafted Seedling Shipping Export to Japan and Analysis of Transportation Environment (채소 접목묘 일본 선박 수출 현황 및 수송 환경 분석)

  • The purpose of this study was to provide the basis for the vegetable seedlings export research and policy establishment by analyzing vegetable seedlings export status from the agricultural quarantine statistics in Korea and the changes of temperature and relative humidity during the vegetable seedlings shipping export transportation to Japan. From 2007 to 2016, various vegetable seedlings such as cabbage, tomatoes, cucumber, onion and etc., have been exported to more than 20 countries around the world. The main exporting country of vegetable seedlings for commercial purposes is Japan, and the major exported seedlings to Japan in 2016 were fruit vegetable grafted seedlings such as eggplant, tomato, cucumber, watermelon and pepper. Total export amount of the fruit vegetable seedlings to Japan in 2016 were 2,575,446 seedlings and it is approximately 0.7 to 1.6 million dollars. The grafted seedlings exported to Japan were consumed for urban agriculture and farm use. Shipping transportation took about 24 hours in the process of receiving the package ${\rightarrow}$ shipment ${\rightarrow}$ quarantine (Busan port) ${\rightarrow}$ Quarantine (Japan Shimonoseki Port). The growing demand for vegetable seedlings due to the development of urban agriculture in Japan and the growing interest and demand for vegetable grafting seedlings in neighboring countries such as Russia will be an opportunity to expand the export size of Korean vegetable grafting seedlings. In order to expand the export of vegetable seedlings in Korea, it is necessary to ensure further active government policy and research on the production of export seedlings, seedlings storage and transportation technology and analysis of exporting countries' market information.

  • In 2004, average temperature was higher, and rainfall was less than those of previous year, The diseases of rice, barley, pepper, chinese melon, apple and oriental pear were surveyed. Seedling diseases, leaf blast, sheath blight and bacterial blight of rice, phytophthova blight, virus diseases and anthracnose of pepper, and sudden wilt syndrome and powdery mildew of chinese melon grown in plastic greenhouse were severe. Especially, sheath blight and bacterial blight of rice occurred two times higher than those of previous year, Panicle blight of rice decreased about 4 times, compared with the previous year, presumed that the lower rainy days, rainfall and RH suppressed spread of the fungal pathogens to panicles of rice. Lower rainfall during mid- and late Aug caused three-times less occurrence of phytophtora blight of red-pepper than that of the previous year, Another diseases surveyed occurred similar or less than those of the previous year.

Control Efficacy of Streptomyces sp. A501 against Ginseng Damping-off and Its Antifungal Substance

  • Ginseng damping-off, caused by the fungal pathogens Rhizoctonia solani and Pythium sp., is a critical disease in ginseng seedling. In a continuing effort to find microorganisms with the potential of acting as a biocontrol agent against Rhizoctonia damping-off, we found that a Streptomyces sp. A501 showed significant antifungal activity against Rhizoctonia solani. In field experiment to test the efficacy of Streptomyces sp. A501 in controlling ginseng damping-off, the incidence of damping-off disease was meaningfully reduced when ginseng seeds were soaked in the culture broth of Streptomyces sp. A501 before sowing. To perform characterization of the antifungal compound, we isolated it from the culture broth of strain A501 through Diaion HP-20 and silica gel column chromatographies and preparative high-performance liquid chromatography. The structure of the antifungal compound was assigned as fungichromin by spectroscopic methods, mainly nuclear magnetic resonance and electrospray ionization-mass analysis.

  • This study was conducted to investigate the optimal conditions for spore germination, prothallus propagation, sporophyte formation, and seedling growth in Polystichum braunii (Spenn.) $F{\acute{e}}e$. The rate of spore germination and early prothalium development was high in Knop (41.2%), which had low mineral content. The optimal medium for prothallus propagation and sexual organ formation was 2MS medium (2% sucrose). Among the various mixtures of cultivation soil (bedding soil, peat moss, perlite, and decomposed granite), a mixture of bedding soil and decomposed granite at a ratio of 2:1 (v:v) had a positive effect on sporophyte formation (276.3 ea/$7.5m^2$). The most efficient conditions for promoting the growth of sporophytes were pots filled with only bedding soil.

  • Due to the oil spill incident in December 2007, every facility of oyster culture was removed in western coast especially in Taean and Seosan. To restore oyster resources in Taean and Seosan, we brought oyster seeds from southern area and monitored their growth. In addition we monitored the culture environment throughout the year, and observed the number of oyster larvae and attached spats on collectors during summer. The factors of water environment were appropriate for oyster culture in both study area. The growth of shell height was larger in Uihang-ri, Taean than Jungwang-ri, Seosan. Spawning was more intensive in a short time in Jungwang-ri, Seosan than Uihang-ri, Taean. The number of oyster larvae and spats of collectors were much more in Jungwang-ri, Seosan than Uihang-ri, Taean. This study showed that transplantation of healthy oyster seeds from southern area can be a way of restoration of oyster resources in western coast. In addition, systematic approaches are necessary by building a better understanding of regional characteristics to restore and enlarge the oyster culture farms in western coast. In summary Uihang-ri, Taean will be appropriate for cultivation farms and Jungwang-ri, Seosan for seedling grounds to increase oyster culture productivity.

Effect of Water Condition and Air Circulation on Fruit Fermentation of Oriental Melon (Cucumis melo L. var. makuwa Makino) (수분조건 및 송풍처리가 참외의 발효과 발생에 미치는 영향)

  • This study was conducted to assess effects of soil moisture condition and ventilation on development of fermented fruit of oriental melon. In higher soil moisture condition (-10 kPa), roots absorbed more water and transpiration was decreased under low temperature and high humidity conditions. Development of fermented fruit may be come from absorbed water concentrated placenta with higher soluble solid. Fermented fruit was developed with higher level at Shintozoa and Elite, was developed with lower level at Hongtozoa, and was not developed at self-rooting of oriental melon. Ventilation on fruit did not affect development of fermented fruit at Shintozoa, Elite, Hongtozoa and self-rooting seedling. Ca contents of flesh and placenta of fruit under ventilation treatment were higher than control but contents of Acetaldehyde, Ethanol and Ethylacetate were lower.

Bacillus spp. as Biocontrol Agents of Root Rot and Phytophthora Blight on Ginseng

  • Ginseng (Panax ginseng) is one of the most widely cultivated medicinal herbs in Korea. However, yield losses reached up to 30-60% due to various diseases during 3 or 5 years of ginseng cultivation in the country. Therefore, successful production of ginseng roots depends primarily on the control of diseases. The objective of this study was to select potential biocontrol agents from rhizobacteria isolated from various plant internal root tissues for the control of multiple ginseng diseases as an alternative to fungicides. Among 106 Bacillus strains, two promising biocontrol agents, Bacillus pumilus strain B1141 and Paenibacillus lentimobus strain B1146, were selected by screening against root rot of ginseng caused by Cylindrocarpon destructans in a greenhouse. Pre-inoculation of selected isolates to seed or l-year-old root of ginseng resulted in stimulation of shoot and/or root growth of seedlings, and successfully controlled root rot caused by C. destructans (P<0.05). Furthermore, drenching of cell suspension of the selected isolates on seedling-growing pots reduced the incidence of Phytophthora blight after the seedlings were challenged with zoospores of Phytophthora cactorum (P<0.05). P. lentimorbus strain B1146 showed antifungal activity against various soil-borne pathogens in vitro, while B. pumilus strain B1141 did not show any. Results of this study suggest that some rhizobacteria can induce resistance against various plant diseases on ginseng.

Effects of Temperatures and Relative Humidities on the Development of Brown Planthopper, Nilaparvata Zugens (Stal) (온도와 습도가 벼멸구의 생육에 미치는 영향에 관한 연구)

  • The newly hatched nymphs of brown planthopper(BPH) were reared individually for two generations in test tubes, where young rice seedling was planted on agar solution, at $30^{\circ}C,\;25^{\circ}C\;and\;20^{\circ}C\;with\;95\%,\;75\%,\;65\%\;and\;35\%$ R.H. Effects of $30^{\circ}C$ on the development of BPH when compared with those of $25^{\circ}C$ are followings. Egg period, nymphal period, and adult longevity were shortened. Nymphal mortality was increased and the number of oviposited eggs was decreased. Hatchability was zero per cent because the eggs were either unfertilized or died before finishing the development. At the low temperature of $20^{\circ}C$, in comparision with $25^{\circ}C$, the developmental period of nymphs and eggs was considerably lengthened, and adult longevity was shortened, the number of oviposited eggs was decreased. The nymphal mortality was higher at high relative humidity $(above\;75\%\;RH)$ than that at low relative humidity $(under\;65\%\;RH)$. Under the condition of high relative humidities, the adult longevity was shortened, and the number of oviposited eggs was decreased.

Resin Flow, Symptom Development, and Lignin Biosynthesis of Two Pine Species in Response to Wounding and Inoculation with Fusarium circinatum

  • Resin flow, symptom development, and lignin biosynthesis in response to wounding and fungal inoculation were investigated in Pinus rigida and Pinus densiflora. The two-year-old seedling stems were subjected to three types of treatments: (i) wounding without inoculation, (ii) wound-inoculation with a conidial suspension of Fusarium circinatum, and (iii) pre-wounding woundinoculation with the fungus 20 days after the initial wounding. Resin flow from wounding sites was more evident in P. rigida than P. densiflora in all treatments. The wound-inoculation with the fungus induced almost two-fold higher levels of resin flow than the other treatments in both species. The pre-wounding woundinoculation appeared to result in a decrease in pitch canker development in the two pine species. Some reductions in disease severity were observed in the prewounding wound-inoculated P. rigida, showing a mean disease severity of less than 85%, compared with approximately 100% in the wound-inoculated stems. Disease severity was approximately 50% in the woundinoculated P. densiflora, whereas 10% in the pre-wounding wound-inoculated stems. Higher amounts of lignin were found from bark (ca. 40%) than from xylem (ca. 30%). The wound-inoculated bark and the pre-wounding wound-inoculated bark exhibited higher amounts of lignin among the other treatments. These results suggest that the wound-inoculation apparently prompt the increase in resin flow and lignin biosynthesis from the two pine species, and the prior wounding may be involved in decreased disease severity against the further invasion of F. circinatum.

Selection of Self-Fertilization Stevia and Development of Multiple Propagation Method using Hypocotyl Explants (자가수정 스테비아의 선발과 배축절편체를 이용한 대량증식 방법 개발)

  • This study was carried out to assess the potential of hypocotyl explants for multiple-propagation by intensive sweetener variety with high yielding in stevia of self-fertilization. Agronomical character of self-fertilization shown a height range 72.0~120.7 cm, branch's 4.9~56.6 in stevia. Selection of high content of Rebaudioside A (RA) from segregating population in mixing variety have a variety, 'DA1' (55.2%) from HPLC analysis. On a developing of multiple-propagation in stevia, hypocotyl explants used to select for reasonable medium under the influence of each plant growth regulators concentrations. In a Mix treatment of plant growth regulators, as Murashige and Skoog medium with 1.5 mg/L BA and 0.5 mg/L Kinetin showed the most favorable. We got the multiple-propagation of 30 shoots from one inoculated hypocotyl explants as the result of these bases. Rooting induced superbly on MS medium with 0.1 mg/L IBA with inoculated shoots. All of the rooting plants were adapted to a normal seedling. This method of multi-propagation is used effectively in Rebaudioside A productivity elevation.
