국민공통기본교육과정 과학과 생명영역 물질대사 관련 탐구활동 분석 (Analysis of Inquiry Activities Related to Metabolism Presented in Life Science Textbooks Developed According to the National Common Basic Curriculum)

  • The purpose of this study was to examine the connection among inquiry activities related to 'Metabolism' presented in life science textbooks developed according to the national common basic curriculum. To perform this, the type, process and context of inquiry activities of life science textbooks were analyzed. The number of inquiry activities is gradually increased from elementary to secondary school. As result of analysis, types, processes and skills, and context of inquiry were not balanced according to textbooks and school levels. It is suggested that the framework of evaluating inquiry activities based on curriculum objectives be provided when developing science textbooks. It also implies that teachers need to reconstruct inquiry contents and activities suitable to school situations when they teach in the real fields of education.

  • Water, in a living being, is as essential as the essence derived from food is in maintaining one's life. The concepts are expressed in forms of "food and drink" and "drink-food" in the ${\mathbb{\ulcorner}}Hwangjenakyoung{\mathbb{\lrcorner}}$ and most of the other oriental medicine related references. Following the steps of the human body's metabolism, the water source builds up characteristic formations, such as bodily fluids (blood/perspiration/urine/essence), in each transforming phase according to the nature of the Ki that propels the transformation. Furthermore, each characteristic formations has its' own suitable duties, distinctive features and its field of activation. The vital energy of life is identified as a positive property due its fluidity and its formless nature. In order for this vital energy to come into its own, it needs to weld into one with the material-natured body of the negative property which will embrace the positive property and transform it into body fluid. Water taken into a body will undergo the first activation of Ki, dissolving the Wigi and the Wongi and transforming into the primary body fluid. The delicates among the dissolved Ki will once again go through a transformation in the Jungcho. It will turn into red blood, with influence of the vital function. When the vital energy completes its duties in all parts of the body, it combines with water again and transforms into the secondary bodily fluid. This is when the Takgi gets filtered and the new enriched essence is created.

티로신 키나아제 저해제의 간독성에 대한 고찰 (Reviews on the Hepatotoxicity of Tyrosine Kinase Inhibitors)

  • Background: Small-molecule tyrosine kinase inhibitors (TKIs) have had major impacts on anticancer therapy by targeting the catalytic activities of dysregulated tyrosine kinases. TKIs have not presented traditional toxicities; however, some serious adverse effects, including hepatotoxicity, have been documented in clinical trials and post-marketing surveillance. Although TKI-induced hepatotoxicity can cause severe clinical complications in patients, the underlying mechanism is still unclear. Methods: Studies on TKI-induced hepatotoxicity were identified by Pubmed search, and relevant articles were reviewed. Results: Immunoallergic reaction, cytochrome P (CYP) 450 polymorphisms, and formation of reactive metabolites are under consideration as mechanisms of TKI-induced hepatotoxicity. Host protein-drug metabolite conjugates are recognized as antigens by class II major histocompatibility complexes and are believed to cause liver injuries. Polymorphisms in CYP, which influences TKI metabolism, can slow TKI metabolism and may induce development of hepatotoxicity. The formation of reactive metabolites during drug metabolism can induce hepatotoxicity by directly causing cytotoxicity, leading to cell dysfunction, and indirect toxicity by mediating secondary immune reactions. Concurrent use of various medications with TKI can also cause hepatotoxicity by affecting drug transporter or enzyme activities. Conclusion: Periodic monitoring of patients taking TKIs and risk/benefit reassessments though post marketing surveillance are necessary to prevent hepatotoxicity.

Expressed Sequence Tags of the Wheat-rye Translocation Line Possessing 2BS/2RL

  • Hamlet (PI549276) possessing 2RL was obtained by cross between a wheat cultivar ND7532 (Froid/Centurk) and a rye cultivar Chaupon. Chaupon was known to have resistant gene to biotype L of Hessian fly [Mayetiola destructor (Say)] larvae. The wheat-rye translocation line (Coker797*4/Hamlet) was also known to be resistant to biotype L of Hessian fly larvae. We analysed a set of 96 ESTs from the wheat-rye translocation line (2BS/2RL). ESTs were classified by various physiological processings, such as primary metabolism, secondary metabolism, transcription, translation, transport, signal transduction, defense, transposable element, and others. Three sequences encoding thioredoxin peroxidase, 26S rRNA, and rubisco small subunits were homologous to registered genes in rye. Although limited number of clones were used to develop ESTs, these clones and their sequence information may be useful for researchers studying general physiology and molecular biology on the translocation line.

당뇨병성 신부전증 환자의 선단 궤양에 의한 하지절단(1예 보고) (Lower Leg Amputation by Calciphylaxis in Diabetic Nephropathy Patient (A Case Report))

  • Calciphylaxis is a rare disease that appear in patients with secondary hyper-parathyroidism or chronic renal failure or that show defect in calcium phosphate metabolism which is characterized by fibrin deposit or calcification of medial wall of vessels causing gradual ischemic skin necrosis. Calciphylaxis is a disease with poor prognosis as skin necrosis can progress rapidly. If left untreated, calciphylaxis will progress to sepsis with high mortality. The treatment is controversial but kidney transplantation or parathyroidectomy is suggested to recover calcium-phosphate metabolism. The authors have experienced calciphylaxis in a patient with chronic renal failure caused by DM nephropathy with characteristic skin lesion and rapid skin necrosis. We describe this case with documentary reviews.

기능획득 돌연변이 인삼 모상근의 대량생산 (Mass Production of Gain-of-Function Mutants of Hair Roots in Ginseng)

  • 본 연구는 아그로박테리움 공동배양법을 이용한 기능획득 인삼 모상근의 대량생산을 위한 조건 확립에 대한 것이다. 일반적으로, 인삼과 같이 형질전환을 통한종자의 확보가 어려운 식물에서는 loss-of-function을 이용한 기능유전체 연구에 한계가 있다. 한편, 유전자의 기능을 활성화시키는 방법 (gain-of-function)인 activation tagging 기술은 이러한 문제점을 극복할 수 있는 대안이 될 수 있으며, Agrobacterium rhizogenes를 이용한 모상근 생산 시스템은 대량의 돌연변이체를 안정적으로 용이하게 얻을 수 있다는 점에서 최적의 시스템이라고 할 수 있다. 본 연구에서는 activation-tagging된 효율적인 형질전환 모상근 생산에 있어서의 최적의 아그로박테리움 균주 및 인삼조직, 배지조성 등에 대한 조건을 확립하였으며, 다양한 배지에서의 형질전환 모상근의 생장률 및 분지율, 표현형 등을 조사하였다. 엽병 절편을 activation-tagging vector pKH01을 가지고 있는 A. rhizogenes R1000와 공동배양하였을 때 배양 4주후 85.9%의 빈도로 모상근이 생산되었다. 모상근의 최대 생장과 분지도를 나타내는 배양조건을 조사한 바 엽병절편을 1/2 SH 배지에서 4주 배양하였을 때 왕성하게 생장하였으며 2.6의 분지도를 보여주었다. 최종적으로 1,989개체의 독립적인 형질전환 모상근 line을 생산하였으며, 이들 모상근 line은 인삼 진세노사이드 생합성 관련 유전자의 발굴 및 기능해석에 유용하게 쓰일 것이다.

Phanerochaete chrysosporium의 액체 배양 및 Lignin Peroxidase 생산 (Submerged Culture of Phanerochaete chrysosporium and Lignin Peroxidase Production)

  • This study characterizes the growth of white rot fungi Phanerochaete chrysosporium IFO 31249) and lignin peroxidase(LiP) activity in different submerged culture media. P. chrysosporium was grown in the form of pellet of various sizes from a spore inoculum under shaking liquid culture condition. While the growth of mycelia was higher under the nitrogen-sufficient culture than under the nitrogen-limited culture, ligninase activity was relatively lower. The lignin peroxidase appeared in nitrogen-limited culture and was suppressed by excess nitrogen. High level(40U/l) of lignin peroxidase activity was obtained in the growth medium containing 1.5mM veratryl alcohol, a secondary metabolite of P. chrysosporium. Lignin peroxidase production was not observed under conditions of nitrogen sufficiency or in balanced media, suggesting that control parameters could increase the activity by manipulating the secondary metabolism.

Proposed Mechanisms of Photobiomodulation (PBM) Mediated via the Stimulation of Mitochondrial Activity in Peripheral Nerve Injuries

  • Evidence shows that nerve injury triggers mitochondrial dysfunction during axonal degeneration. Mitochondria play a pivotal role in axonal regeneration. Therefore, normalizing mitochondrial energy metabolism may represent an elective therapeutic strategy contributing to nerve recovery after damage. Photobiomodulation (PBM) induces a photobiological effect by stimulating mitochondrial activity. An increasing body of evidence demonstrates that PBM improves ATP generation and modulates many of the secondary mediators [reactive oxygen species (ROS), nitric oxide (NO), cyclic adenosine monophosphate (cAMP), and calcium ions (Ca2+)], which in turn activate multiple pathways involved in axonal regeneration.
