To select potential inhibitors of platelet aggregation from large numbers of crude plant extracts, the modified thin smear method for the anti-platelet aggregating activity using platelet rich plasma was further modified by direct observation under a light microscope without staining the smear. The activities determined by the method were coincided with those by the electrical impedence method using whole blood, when ADP or collagen was employed as the aggregating agent. Among 130 varieties of edible and herbal plants which collected from markets or experimental farms of agricultural research institutes, those showed the anti-platelet aggregating activities were selected by testing the crude methanol extracts: Aster scaber, Aster tataricus, Ligularia stenocephala, Platycodon glaucum Allium victorialis, Allium oderum, Moros bombycis, Portulaco oleracea, Aamthopanax sessiliflorus and Rosa davurica. However, some of them activated the platelet aggregation under the same assay conditions: Pimpinella brachycarpa, Hosta plantaginea, Capsella bursapastoris, Fagopyrm esculentum, Prunus mume, Rubus coreanus and Limaria japonica. In addition, those revealed the antioxidant activities were selected by measuring the abilities to scavenge superoxide anion radicals: Pteridium aquilinum, Aster scaber, Ligularia fischeri, Chrysanthemum zawadskii, Artemisia capipparis, Cirsium setidens, Commelina communis and Capsella bursapastoris among edible plants.

The purpose of this study was to develop a practical caries activity test by fluorescence using laser, plasma light and halogen light. The subjects of study were 45 children of 7-8 years old Argon laser, plasma light and halogen light were irradiated to buccal or labial surface of all teeth. Fluorescence of initial carious lesion from teeth was observed through barrier filter and the number of teeth showing lesion was counted. Visual examination for the dDfFtT, mutans streptococci screening test and Lactobacilli colony counting were also done. Data analysis was accomplished by Axelsson's method. The result from the present study can be summarized as follows. 1. Laser, plasma light and halogen light could detect the initial carious lesions better than visual examination(p<0.05). 2. There was positive correlation between laser(r=0.42), plasma light(r=0.41), halogen light(r=0.39) and dBfFtT rate(p<0.01). 3. The specificity sensitivity and predictive value was showed highest value in laser, but was showed favorable value in plasma light and halogen light. In regard to above results, laser, plasma light and halogen light all considered to be reliable method for determining individual caries activity. And they were also considered to be practical method because it would be simple, inexpensive, and time saving method.

Purpose: The purpose of this paper was to describe the attributes of nursing with humor based on an integrative review and to present basic information for developing humor intervention programs for children. Methods: Using combinations of the terms 'nursing' and 'humor' as key words, the researchers searched four electronic databases to relevant identify studies. Thirteen studies were selected through full text screening of related research published in academic journals from January 1993 to April 2013. Results: Through in-depth discussion and investigation of the relevant literature, three components emerged; 1) using instruments of coping for stress relief, 2) symptoms relief through pleasant feeling, and 3) job satisfaction and work improvement of nurse using humor. Conclusion: The results of this study suggest that humor is valuable as a nursing intervention for children and the possibility of bringing about positive results in nursing care if nurses utilize the sense of humor as fully as they can. Therefore, it is critical for nurses to develop a diversity of humor interventions for children as well as measurements to test quality and quantity of humor used in clinical pediatric settings.
Subcellular localization of a protein containing nuclear localization signals (NLS) has been well studied in many organisms ranging from invertebrates to vertebrates. However, no systematic analysis of NLS-containing proteins available from Mollusks has been reported. Here, we describe in silico screening of NLS-containing proteins using the mollusks database that contains 22,138 amino acids. To screen putative proteins with NLS-motif, we used both predict NLS and perl script. As a result, we have found 266 proteins containing NLS sequences which are about 1.2% out of the entire proteins. On the basis of KOG (The eukaryotic orthologous groups) analysis, we can't predict the precise functions of the NLS-containing proteins. However, we found out that these proteins belong to several types of proteins such as chromatin structure and dynamics, translation, ribosomal structure, biogenesis, and signal transduction mechanism. In addition, we have analysed these sequences based on the classes of mollusks. We could not find many from the species that are the main subjects of phylogenetic studies. In contrast, we noticed that cephalopods has the highest number of NLS-containing proteins. Thus, we have constructed mollusks NLS database and added these information and data to the mollusks database by constructing web interface. Taken together, these information will be very useful for those who are or will be studying NLS-containing proteins from mollusks.
Extracts from 63 kinds of Pholiota sp. fruiting bodies were prepared using water and methanol, and then their physiological functionalities were investigated. The methanol extracts from Pholiota adiposa PAD030 showed high fibrinolytic activity and those of P. adiposa ASI PAD-022 showed potential inhibitory activity of 76.8% against ${\beta}-hydroxy-{\beta}-methylglutaryl(HMG)-CoA$ reductase. The highest antioxidant and tyrosinase inhibitory activities were found in the water extracts of Pholiota sp. PSP-015 (72.7%) and methanol extracts of P. nameko PNA-024 (69.5%), respectively. However, superoxide dismutase(SOD)-like activity and elastase inhibitory activity were low in almost of the extracts. The HMG-CoA reductase inhibitor from the fruiting body of P. adiposa PAD-022 which showed the highest functionality was extracted maximally when powder of the fruiting body was shaked at $30^{\circ}C$ for 12 h by methanol and its HMG-CoA reductase inhibitory activity was 80.2%.
Extracts from 52 samples of mushrooms were prepared by using water, ethanol and methanol, and then yields and angiotensin I-converting enzyme(ACE) inhibitory activity were investigated. Sample mushrooms contained crude proteins of $7.1{\sim}56.5%$, curde lipids of $0.2{\sim}4.4%$ and carbohydrates of $30.3{\sim}86.6%$. Among 52 samples, the water extract from fruiting body of Pholiota spp. ASI 24027 showed the highest extraction yield of 68%. Water extract of Pholiota spp. ASI 24012 fruiting body had potential ACE inhibitory activity of 66%. The optimal extraction condition of the ACE inhibitor from the fruiting bodyies of Pholiota spp. ASI 24012 was In water at $30^{\circ}C$ for 1 hr and ACE inhibitory activity was 67.6% on the condition with 0.2 mg of $IC_{50}$.

The gene for ${\gamma}$-casein, a milk protein, is a member of a family of casein gene which is expressed in mammary glands of the animal during the late gestation and lactation periods binder the influence of various hormones. In order to elucidate tile mechanisms b)'which hormones regulate the coordinate induction of milk protein genes, the mouse ${\gamma}$-casein gene was isolated and characterized. The ${\gamma}$-casein gene was screened from a mouse genomic library constructed in bacteriophage EMBL3 with the ${\gamma}$-casein CDNA used as probe and one clone was obtained. The ${\gamma}$-casein CDNA as probe was partially sequenced and contained ATG start codon and 5'-noncoding region. The cloned genomic DNA was digested with Sal I restriction enzyme, by which the insert DNA can be isolated from EMBL3 vector. Three DNA bands were observed and the size of DNAs was approximately 28kb, 14kb and 9Kb, respectively Accordingly the size of the insert DNA was calculated with approximately 23Kb. The result of Southern blot analysis, however, showed that the cloned genomic DNA was not hybridized with the synthetic oligonucleotides (40 mer) of cDNA 5'-end region, but it was hybridized with the y -casein CDNA. This means that tile cloned y -casein gene may not contain its promoter region. The ${\gamma}$ -casein genomic DNA containing the promoter region has been screening from mouse genomic library with oligonucleotides of CDNA 5'-end region as probe, and twenty-nine clones was obtained.
The structural gene of rabbit hemoglobin was cloned into Pst I site of pBR322 in E. coli. The complementary DNA (cDNA) was synthesized from rabbit globin mRNA with avian myeloblastosis viral reverse transcriptase, and then RNA was destroyed at pH 11. The double stranded cDNA was synthesized with both Klenow fragment of E. coli DNA polymerase I and reverse transcriptase and then the hairpin loop was opened with Sl nuclease. Double stranded cDNA was subsequently tailed with dCTP and annealed to dGMP-tailed vector DNA. After transformation and initial screening of appropriate clones by plasmid size, the cloned colonies were identified by in situ colony hybridization using by plasmid size, the cloned colonies were identified by in situ colony hybridization using $[^32P]$-labeled cDNA probes and characterized the inserts with restriction endonucleases. The expression of cloned globin gene was investigated by standard radioimmunoassay using rat anti-rabbit Hb serum as primary antibody and goat antirat IgG serum as secondary antibody. The result suggested that the chimeric proteins (the part of $\\beta$-lactamase from the vector pBR322 and globin from rabbit) were supposedly produced in E. coli and the product had the antigenic determinant of rabbit hemoglobin.
Two kinds of composite seasoning products (beef broth powder, polk bone extract powder) were used for a detection trial of gamma irradiation treatment up to 10 kGy by analyzing photostimulated luminescence (PSL), electron spin resonance (ESR) and thermoluminescence(TL). PSL results showed that the photon counts of non-irradiated samples were lower than 700, while those of irradiated samples were higher than 5000, which makes it possible to screen irradiated composite seasoning products at 1 kGy or over from the non-irradiated control. ESR signals measured for both irradiated samples were not irradiation-specific, even though they were dose dependent in the signal intensity. Radiation-induced TL glow curves were found in irradiated beef broth powder and furthernmore, TL ratio $(TL_4/TL_2)$ obtained by a re-irradiation step could verify the detection result of TL1 glow curves, showing ratios lower than 0.05 in the non-irradiated sample and higher than 1.00 in irradiated ones.
In a continuing screening of selected medicinal plants native to Mongolia, the antioxidant and ${\alpha}$-glucosidase inhibitory activities of methanol extract of Iris bungei were investigated. After extraction with 80% of methanol, the methanol fraction was further extracted with n-hexane, EtOAc and n-BuOH in order to obtain four different solvent-soluble fractions, namely n-hexane-soluble, EtOAc-soluble, n-BuOH-soluble and $H_2O$ residue. The antioxidant properties were evaluated by radical scavenging assay using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) ($ABTS^+$) radicals. The anti-diabetic efficacy of I. bungei extract was investigated by ${\alpha}$-glucosidase assay. All tested samples showed dose-dependent radical scavenging and ${\alpha}$-glucosidase inhibitory activities. Among the tested extracts, the EtOAc-soluble fractions showed the greatest radical scavenging activity and ${\alpha}$-glucosidase inhibitory properties among other solvent-soluble fractions. This result suggested that there was a significant relationship between the total phenolic content and biological efficacy. Thus, I. bungei extract might be considered as a new potential source of natural antioxidant and as a ${\alpha}$-glucosidase inhibitory source. A more systematic investigation of this biomass sill be performed for further investigation of activity against antioxidative and anti-diabetic effects.
