스코폴라민의 흰쥐 피부투과에 대한 투과촉진제들의 영향 (Effect of Various Enhancers on Permeation of Scopolamine through Excised Rat Skin)

  • The transdermal therapeutic system (TTS) of scopolamine has various advantages over its oral dosage forms. The ideal scopolamine TTS requires high skin permeation rate in short time after it is applied on the skin. In order to increase the initial skin permeation rate of scopolamine from TTS, various permeation enhancers were employed. Enhancers employed were fatty acids (oleic and linolenic acids), cyclic monoterpenes (menthol, camphor, cineole and limonene) and others (isopropyl myristate, sodium lauryl sulfate and glyceryl monostearate). The concentration of enhancers in the base were fixed to 5% (w/w). While fatty acids had little enhancing effect on the skin permeation of scopolamine, cyclic monoterpenes, isopropyl myristate and sodium lauryl sulfate resulted in $1.5{\sim}2.6-fold$ higher skin permeation rate of the drug compared to the control. However, lag time was not affected by enhancers studied.

Ethanol Extract of Soybean Ameliorates Scopolamine-Induced Memory Impairment in Mice

  • Soy (Glycine max, family Leguminosae) contains isoflavones and saponins as main constituents. In our preliminary study, soybean ethanol extract (SE) ameliorated scopolamine-induced memory impairment in mice in the passive avoidance task. Therefore, to confirm its ameliorating effect for memory impairments, we measured its effect in scopolamine-induced memory-impaired mice in Morris water maze task. SE significantly prevented scopolamine-induced memory impairment in the Morris water maze task. SE also increased the swimming time within quadrant section of the platform on the day after the final training session test. SE protected the reduction of brain-derived neurotrophic factor (BDNF) expression and cAMP response element-binding protein (CREB) phosphorylation in the hippocampi of scopolamine-treated mice. However, SE did not inhibit acetylcholinesterase. To understand the possible role of soysaponins in memory impairments, we prepared soyasaponins-rich (butanol) fraction of soybean (SRF) and investigated its protective effect against in the passive avoidance and Morris water maze tasks. SRF ameliorated scopolamine-induced memory impairment in mice. The memory impairment-ameliorating effect of SRF was more effective than that of SE. Based on these findings, soybean may improve memory impairment by regulating CREB phosphorylation and BDNF expression.

비타민 E와 비타민 C 첨가가 Scopolamine에 의한 인지능력 감퇴에 미치는 영향 (Effects of Vitamin E and Vitamin C Supplementation on the Decrease in Cognitive Function Induced by Scopolamin)

  • With the increasing elderly population in Korea, dementia has become a mojor health problem in Korea. Several studies have been conducted on the association between dementia and dietary intake, especially vitamin E and vitamin C. The purpose of this study was to investigate the effect of vitamin E and vitamin C supplementation on the decrease in cognitive function induced by scopolamine(300mg/kg) in rats. Rats were divided into three groups : control, vitamin C, and vitamin E supplementation(2.4g Vit C or Vit E /100g diet) and fed the diets for 6 weeks. There were significant decreases in active avoidance response and brain acetylcholinesterase activity in the control group, but no significant differences were observed in the vitamin E and C groups after scopolamine treatment. Brain dopamine concentration of vitamin E and C groups was significantly higher than those of control group after scopolamine injection. The concentrations of brain norepinephrine also showed similar tendence, even though it was not statistically significant. These results indicate that vitamin E and vitamin C may protect against the cognitive function decrease induced by scopolamine. However, it is still unclear how vitamin E and C influence brain neurotransmitters and improve cognitive function. Further study is need to elucidate the role of vitamin E and C supplementation in the prevention of dementia.

술후 통증 치료를 위한 몰핀 경막외 주입시 경피적 Scopolamine에 의한 구역 예방 효과 (Prophylactic Effects of Transdermal Scopolamine Patches on Nausea in Postoperative Patients Receiving Epidural Morphine)

  • Background: To evaluate the effect of prophylactic transdermal scopolamine in reducing nausea associated with postoperative epidural morphine. Methods: 30 healthy patients scheduled for cesarean section were given local anesthetics epidurally for surgical anesthesia. After delivery of the baby, transdermal scopolamine patch($Kimite^{(R)}$ Myung Moon Pharm. Co., Seoul, Korea) was applied to the study group(n=15) and placebo patch to the control group(n=15). Postoperative analgesia was provided soley with epidural morphine. Nausea was treated with metoclopramide. Results: During 24 hours postoperatively, the mean nausea score was significantly lower in the study group than in the control group. But the nausea incidence was not significantly different between the two groups. The mean number of times antiemetic drugs which were administered to patients were lower in the study group than in the control group($1.5{\pm}0.5$ vs $3.3{\pm}1.3$, p<0.05). Conclusion: Transdermal scopolamine patch provides antiemetic effect simply, continuously and safely, preventing nausea which could occur during administration of epidural morphine after cesarean section.

Scopolamine유도 치매쥐에서 산수유 추출물의 항치매 효과 (Anti-dementia Effects of Cornus officinalis S. et Z. extract on the Scopolamine Induced Dementia in Mouse)

  • These days, as the average span of population's life increases, the number patients of dementia also increases. But Research on Korean medicine is stilled limited. The research evaluates the effect of the extract from Cornus officinalis S.et Z on cognitive impairment induced by scopolamine in mice. The mice were randomly divided into five groups of ten mice. The normal group was treated with only 0.9% saline. The control group was treated with scopolamine (5 mg/kg, i.p.). The positive control group was treated with tacrin. The C100, 200 group was treated with C. officinalis extracts 100, 200 mg/kg. Memory-related behaviors were evaluated using a morris water maze and a passive avoidance test. Protein levels of BDNF, p-CREB (ser133), immunohistochemistry staining, and cholinergic activities were measured in brain tissue. The effects of C. officinalis extract significantly increased acetylcholine concentration and decreased acetylcholinesterase activity. The C. officinalis extract affected memory formation. Also, to confirm expression of protein BDNF, p-CREB (ser133) in the hippocampus, the researchers observed that immunohistochemistry and western blot increased in C. officinalis extract. These results suggest that C. officinalis provides a significant neuroprotective effect against scopolamine-induced cholinergic system and cognitive impairment.

Effects of Ethylcholine Aziridinium, Scopolamine and Mor-phine on Learning Behaviors in Morris Sater Maze.

  • To assess the behavior of the various chemicals, such as ethylcholine axiridinium (AF54A), scopolamine and morphine, the chemicals were administered into eitheer rat or mice. And water maze tests were performed before and during drug administration. In AF64A-treated groups (3 nmol/each ventricle), the latencies to escape was significantly increased in both of the pretraining-and posttraining groups. In scopolamine-treatment (2mg/kg, sc) to the pretrained group, the latency to escape was significantly shortened after the acute administration of scopolamine. However in subscute treatment group with scopolamine, the latency to escape was significantly increased. In morphine-treated groups (10 mg/kg, ip), the subacute treatmment with morphine, the latency to escape was not ahcnged. The results indicate that each chemical induces the learning impariment. However the chrmical-induced learning impairment may have different characteristics upon the exposed chemical. Also the results suggest that both the motivation and the retrieval of memory might be impaired by AF64A.

기억력 감퇴 모델에서 영신초(靈神草), 원지(遠志), 석창포(石菖蒲) 혼합제제의 기억력 및 인지 기능 개선에 관한 연구 (Nootropic and Anti-amnestic Effect of PPA on scopolamine-induced Cognitive Impairment in Mice)

  • In the present study, we assessed the effect of the PPA (Polygala japonica Houtt., Polygala tenuifolia WILLD. and Acorus gramineus SOLAND. mixed preparation), a traditional herbal prescription, on the learning and memory impairments induced by scopolamine. The cognition-enhancing effect of PPA was investigated using a passive avoidance test, Y-maze test and the Morris water maze test in mice. Drug-induced amnesia was induced by treating animals with scopolamine (1 mg/kg, i.p.). A single PPA (400 and 800 (mg/kg)) administration significantly reversed the scopolamine-induced cognitive impairments in the passive avoidance test (P<0.05). On the Y-maze test, PPA (400 and 800 (mg/kg)) also significantly reversed scopolamine-induced cognitive impairments in mice (P<0.05). PPA also improved escape latencies in training trials and increased swimming times and distances within the target zone of the Morris water maze (P<0.05). These results suggest that PPA attenuates amnesic state induced by scopolamine and that these Effect are mediated by enhancing the cholinergic dysfunction.

합환피의 인지 및 기억력 개선 효과에 대한 연구 (Effect of Albizziae Cortex Water Extract on Cognition and Memory Impairments)

  • The aim of this study was to investigate the effects of the water extract of Albizziae Cortex (AC) on the learning and memory impairments. AC was administered to normal mouse and scopolamine-injected amnesia mouse model. Passive avoidance test, Y-maze test, and Morris water maze test were conducted to confirm the cognitive-enhancing activities of AC. Acetylcholinesterase (AChE) activity and acetylcholine (Ach) content were measured after oral administration of AC. On the passive avoidance test, AC (200 mg/kg) significantly increased latency time and recovered scopolamine-impaired learning and memory in mice. In addition, AC (200 mg/kg) reduced Exploration time in target quadrant and reversed the scopolamine-induced cognitive impairments in the Y-maze test. Moreover, AC (200 mg/kg) increased exploration time in target quadrant and improved scopolamine-reduced escape latencies in the Morris water maze test. These effects were presented by regulatory effects of AC on AChE activity and Ach content. Taken together, AC increases cognitive-enhancing activities and ameliorates scopolamine-induced learning and memory impairment. AC might be a potential agent for prevention and treatment of amnesia and dementia.

기억력 감퇴모델에서 산사의 기억력 개선 효과에 관한 연구 (The Fruits of Crataegus pinnatifida Bunge ameliorates Learning and Memory Impairments Induced by Scopolamine)

Methods : The cognition-enhancing effect of C. pinnatifida was investigated using a passive avoidance test, the Morris water maze test and Y-maze test in mice. Drug-induced amnesia was induced by treating animals with scopolamine (1 mg/kg, i.p.). Results : The ethanolic extract of C. pinnatifida (100, and 200 mg/kg) significantly reversed the scopolamine-induced cognitive impairments in the passive avoidance test (p < 0.05). Moreover, C. pinnatifida (200 mg/kg) also improved escape latencies in training trials and increased swimming times and distances within the target zone of the Morris water maze (p < 0.05). On the Y-maze test, C. pinnatifida (100, and 200 mg/kg) also significantly reversed scopolamine- induced cognitive impairments in mice (p < 0.05). Conclusions : The ethanolic extract of Crataegus pinnatifida dramatically possesses the anti-amnestic and cognitive-enhancing activities related to the memory processes, and these activities were parallel to treatment duration and dependent on the learning models.

녹용 추출물이 치매 동물모델의 기억력 개선과 관련효소 활성에 미치는 효과 (Effects of the Deer Antler Extract on Scopolamine-induced Memory Impairment and Its Related Enzyme Activities)

  • 본 연구는 녹용 추출물이 scopolamine으로 기억손상을 유발한 실험동물의 기억력 개선과 AChE, MAO-B의 활성억제, 신경전달 물질인 ACh 함량을 치매 치료제인 tacrine과 비교하여 분석하였다. 공간인지력을 평가하는 Morris water maze test에서 scopolamine(2 mg/kg, i.p.)으로 기억손상을 유발한 경우 4일간의 인지훈련에도 불구하고 platform을 찾아가는 escape latency 시간이 증가되었으며, tacrine과 녹용추출물 투여군은 훈련 2일째부터 유의적으로 escape latency 시간이 감소하였다. 실험 5일째 platform을 제거한 probe test에서도 녹용 추출물 처리군은 scopolamine 처리군보다도 escape latency 시간이 유의적으로 감소하여 장기기억 개선효과가 있음을 보여주었다. 뇌의 신경전달물질인 ACh 생성은 대조군 대비 scopolamine 투여군은 84.7% 감소하였으나, 녹용 추출물 투여군(97.5%)은 치매 치료제인 tacrine(97.8%)과 비슷하게 정상적인 수치로 ACh 생성량을 증가시켰다. Tacrine 처리군은 scopolamine 투여로 상승한 뇌조직과 혈청의 AChE 활성을 유의적으로 감소시켰고, 녹용 추출물 처리군은 뇌조직의 AChE 활성을 유의적으로 감소시켰다. 뇌조직의 MAO-B 활성은 그룹 간 유의적인 차이는 없었으나, tacrine과 녹용 추출물 처리군이 scopolamine 투여군보다 MAO-B 활성을 감소시키는 경향을 보였다. 따라서 녹용 추출물은 scopolamine으로 기억손상을 유발한 치매 동물모델에서 신경전달물질을 분해하는 효소의 활성을 저해함으로써 장기기억 활성을 촉진하고 콜린성 신경계를 자극하여 기억 및 학습 증진에 효과적으로 작용하는 천연물 유래 기능성 물질로 사료된다.
