Determination of Nitrite-scavenging Activity of Seaweed (해조류에 의한 아질산염 소거활성 분석-연구노트-)

  • Nitrite plays an important role in the formation of carcinogenic nitrosamine. In order to effectively inhibit the formation of carcinogenic nitrosamine in food and biological systems, scavenging of residual nitrite is necessary. In screening test of nitrite-scavenging effect of seaweed, The methanol extract from Phaeophyta was more effective than those from Rhodophyta and Chlorophyta. Among the brown algae family, Laminariaceae, A. cribrosum (90.4$\%$) and E. cava (80.1$\%$), E. stolonifera (79.0$\%$) that belong to genus Ecklonia showed marked nitrite-scavenging effect at pH 1.2. Furthermore, these algae showed potent reducing power and radical scavenging activity. The nitrite-scavenging effect was correlated with level of reducing power.

DPPH Radical Scavenging Effect and Antimicrobial Activities of Plantain (Plantago asiatica L.) Extracts (질경이 추출물의 DPPH 라디칼 소거효과 및 항균활성)

  • To develop the functional food, methanol extracts of dried plantain (Plantago asiatica L.) was fractionated in hexane, chloroform, ethyl acetate, butanol and aqueous fraction. DPPH radical scavenging effect, reducing power, nitrite scavenging effect and antimicrobial activities of these fractions were investigated. Ethyl acetate fraction among these fractions showed the highest effect of all experiments. Ethyl acetate fraction showed about 60% scavenging effect on the DPPH radicals at 400 $\mu$g/5 mL. Nitrite scavenging effect of ethyl acetate and butanol fraction was more than 70~80% at concentration above of 500 $\mu$g. Among various solvent fractions from methanol extracts of plantain, ethyl acetate fraction showed the highest inhibitory effect on the microorganism such as Staphylococcus aureus, Bacillus cereus and Vibrio parahaemolyticus at 5 mg/disc.

Antioxidative Activities and Nitrite-scavenging Abilities of Some Phenolic Compounds (일부 페놀성 화합물의 항산화효과 및 아질산염 소거능)

  • This study investigated the antioxidant and synergistic effects and nitrite scavenging ability of some phenolic compounds(catechin, rutin, quercetin and naringin), The electron donating abilities of naringin, quercetin, rutin and catechin were 6.7%, 92.8%, 87.6% and 92.21%, respectively, The antioxidant activities in O/W emulsion substrates were in order of rutin > quercetin > catechin > naringin. The antioxidant effect of rutin was stranger than that of BHT or ${\alpha}$-tocopherol. ${\alpha}$-tocopherol showed synergistic effect with catechin and quercetin, but ascorbic acid not showed effect. The nitrite scavenging abilities of catechin, quercetin, rutin and naringin were 99.9%, 98.6%, 25.5% and 0.2%, respectively. The nitrite scavenging abilities of quercetin and actechin were very potent as compared with those of BHT and ascorbic acid.

Radical Scavenging Effect of Methanol Extracts from Seaweeds and Their Active Compounds (해조류 추출물 및 활성성분의 라디칼 소거능)

  • The radical scavenging activity of methanol extracts of seaweeds and their active compounds, alginic acid, fucoidan and phloroglucinol, were investigated under in vitro. Among methanol extracts of seaweeds (sea mustard, sea tangle, seaweed papulosa, fusiforme, sea lettuce, purple laver and chlorella), seaweed papulosa and sea tangle showed strong scavenging activities of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and hydroxyl radical (${\cdot}OH$). In addition, under in vitro, the scavenging activities on DPPH radical of alginic acid and fucoidan, which are active compounds of brown algae, and phloroglucinol, the active compound from Ecklonia species, were evaluated and compared. Fucoidan and phloroglucinol showed strong DPPH scavenging effect, in particular, phloroglucinol had strongest activity among the active compounds. On the other hand, alginic acid did not exert DPPH scavenging activity. From the present study, we could confirm the antioxidative activity of seaweeds and its active compounds.

Nitrite Scavenging Effect by Flavonoids and Its Structure-Effect Relationship

  • Nineteen flavonoids, five phenolics, two coumarins, maltol and L-ascorbic acid were tested as scavenger of nitrite which is believed to participate in the formation of N-nitroso compounds. Many were found to be potent scavenger and the five most potent ones were (+)-catechin, (-)-epicatechin, phloroglucinol, caffeic acid and L-ascorbic acid. The nitrite scavenging effect was higher at pH 1.2 than pH 3.0 and increased when the incubation time was longer. The possible relationship of structures to scavenging effect of the flavonoids tested was discussed.

Scavenging Effect on NO, DPPH and Inhibitory Effect on IL-4 in Astragali Radix Herbal-Acupuncture Solution (황기약침액(黃耆藥鍼液)의 NO, DPPH 소거(消去) 및 IL-4 억제효과(抑制效果))

  • Objective : This study was designed fo find out whether Astragali Radix Herbal-Acupuncture Solution(ARHA) can scavenge NO, DPPH and IL-4 or not. Astragali Radix has been clinically used to reforce the depression of immune reponse etc. in Oriental Medicine. Methods : Free radical metabolism seems to occupy a remarkably common position in the mechanisms of ageing and ageing related disease. Oxidative damage to DNA, lipids, proteins and other molecules may contribute to the development of cancer, cardiovascular disease and possibly neurodegenerative disease. The effects of ARHA on NO, DPPH and IL-4 were measured. Results : The followings are the summary of the result; (1) There is a significant scavenging effect of ARHA on NO in 1, $10mg/m{\ell}$ group in after 24hrs. (2) There is no significant scavenging effect of ARHA on DPPH. (3) There is a significant scavenging effect of ARHA on IL-4 in 1, $10,100{\mu}g/m{\ell}$ group. Conclusions : These results suggest that ARHA has significant scavenging effect on NO, IL-4 and not on DPPH. This study shows that ARHA can be used for ageing related disease and further studies are required to investigate the antioxidative effects of it.

Nitrite Scavenging Effect of Methaol Fraction Obtained from Green Yellow Vegetable Juices (녹즙추출물의 아질산염 소거능에 대한 연구)

  • The objectives of this study were to investigate biological activities such as nitrite scavenging effect of water and methanol fractions obtained from each green juice(Angelica keiskei, kale, carrot) under different pH conditions, and antioxidant vitamin contents in green juice made of green yellow vegetables. The result shows that nitrite scavenging effect of each green juice was 14.0~91.2% in model system. The methanol fraction provided higher effect than the water fraction. The nitrite degradation was the highest at pH 1.2. Nitrite scavenging effect of methanol fraction obtained from kale juice was 91.2% at pH 1.2 which was the highest among tested samples in this experiments. Consequently, extracts of kale juice were shown to provide the greatest functional properties among the vegetables tested. Contents of vitamine E and C in the extracts of kale were higher than the others.

Nitrite Scavenging Effect of Umbelliferaeceae (미나리과 산채의 아질산염 소거능)

  • The nitrite scavenging effect of three kinds of edible wild plants belong to Umbelliferaeceae family was investigated. The contents of crude protein, crude lipid and $\beta$-carotene of the plants were in the range of 22.99∼31.00%, 3.51∼5.08% and 5.40∼19.81 mg%, respectively. The content of ascorbic acid was very low and selenium was not detected in any samples. The contents of linoleic acid and linolenic acid were 66.46∼77.44%. The content of total phenol was 875.77 mg% in Ledebouriella seseloides WOLFF, which was the highest among the samples. The methanol fraction of the plant extract showed higher nitrite scavenging effect than the water fraction. The nitrite scavenging effects of the samples were the highest at pH 1.2 and lowest at pH 6.0, suggesting that it's pH dependent. Nitrite scavenging effect of methnol-soluble fractions obtained from Ledebouriella seseloides WOLFF was 95.3% at pH 1.2, which was the greatest among tested samples. There was a high correlation between the nitrite scavenging effect and the total phenol content.

Scavenging Property of Pyungwi-san Herbal-acupuncture Solution on ROS and RNS (평위산(平胃散) 약침액(藥鍼液)의 활성산소 및 활성질소 소거능)

  • Pyungwi-san(PWS) have been using as a basic prescription of digestive disorder in Korean traditional medicine. This study was performed to examine the in vitro antioxidant activity of the extract using different antioxidant tests including by 1,1-diphenyl-2-picryl-hydrazil (DPPH) radical scavenging, superoxide anion radical scavenging, metal chelating hydrogen peroxide scavenging, lipid peroxydation protective effect and scavenging effect of nitric oxide and peroxynitrite. Herbal-acupuncture solution of PWS(PWS-HS) exhibited a concentration-dependent inhibition of DPPH radical adduct formation and it showed dose-dependent free radical scavenging activity onto superoxide anions. In addition, the result of metal chelating hydrogen peroxide scavenging and ammonium thiocyanate experiments showed that PWS-HS was an active scavenger of hydroxyl radicals. Furthermore, it was also found to be effective in scavenging nitric oxide and peroxynitrite, well-known cytotoxic species that can oxidize several cellular components such as proteins, lipids and DNA.
