Isolation and Culture Medium Optimization for Thermostable Extracellular α-Amylase Production by Thermophilic Alicyclobacillus acidocaldarius (세포외 고온성 α-아밀라제를 생산하는 Alicyclobacillus acidocaldarius 의 분리 및 효소생산용 최적 배양 조건)

  • A thermophilic $Alicyclobacillus$ $acidocaldarius$, which produces thermostable ${\alpha}$-amylase, was isolated from the hot water effluent of a boiled rice mill near Tirupati, Andhra Pradesh, India. The effect of different culture conditions on the growth and production of extracellular ${\alpha}$-amylase by thermophilic $A.$ $acidocaldarius$ was investigated in laboratory scale. The results showed that the optimum conditions for the production of ${\alpha}$-amylase are a temperature of $60^{\circ}C$, pH of 6.0, and medium starch concentration of 1.0%, and yeast extract and tryptone of 0.2%. Surfactants, like Tween-20 and SDS, up to 0.02%, were found to increase the bacterial growth and enzymes. Further increase in their concentration resulted in significantly decreased enzyme production.

Two-stage Bioprocesses Combining Dark H2 Fermentation: Organic Waste Treatment and Bioenergy Production (혐기성 수소발효를 결합한 생물학적 2단공정의 유기성폐자원 처리 및 바이오에너지 생산)

  • This study was performed to investigate the application of dark $H_2$ fermentation to two-stage bioprocesses for organic waste treatment and energy production. We reviewed information about the two-stage bioprocesses combining dark $H_2$ fermentation with $CH_4$ fermentation, photo $H_2$ fermentation, microbial fuel cells (MFCs), or microbial electrolysis cells (MECs) by using academic information databases and university libraries. Dark fermentative bacteria use organic waste as the sole source of electrons and energy, converting it into $H_2$. The reactions related to dark $H_2$ fermentation are rapid and do not require sunlight, making them useful for treating organic waste. However, the degradation is not complete and organic acids remain. Thus, dark $H_2$ fermentation should be combined with a post-treatment process, such as $CH_4$ fermentation, photo $H_2$ fermentation, MFCs, or MECs. So far, dark $H_2$ fermentation followed by $CH_4$ fermentation is a promising two-stage bioprocess among them. However, if the problems of manufacturing expenses, operational cost, scale-up, and practical applications will be solved, the two-stage bioprocesses combining dark $H_2$ fermentation with photo $H_2$ fermentation, MFCs, or MECs have also infinite potential in organic waste treatment and energy production. This paper demonstrated the feasibility of two-stage bioprocesses combining dark $H_2$ fermentation as a novel system for organic waste treatment and energy production.

Comparisons of Physical Properties of Bacterial Celluloses Produced in Different Culture Conditions Using Saccharified Food Wastes

  • The saccharogenic liquid (SFW) obtained by the enzymatic saccharification of food wastes was used as a medium for production of bacterial cellulose (BC). The enzymatic saccharification of food wastes was carried out by the cultivation supernatant of Trichoderma harziaum FJ1 culture. Acetobacter xylinum KJ1 was employed for the BC production culture. The physical properties, such as polymerization, crystallinity, Young's modulus, and tensile strength, of BCs produced by three culture methods: the static cultures using HS (Hestrin-Schramm) as a reference medium (A) or the SFW medium (B), the shaking culture (C) or the air circulation culture (D) using the SFW medium, were investigated. The degrees of polymerization of BCs produced under the different culture conditions (A-D) showed 11000, 9500, 8500, and 9200, respectively. Young's modulus was 4.15, 5.0, 4.0, and 4.6 GPa, respectively. Tensile strength was 124, 200, 80, and 184 MPa, respectively. All of the BC had a form of cellulose I representing pure cellulose. In the case of the shaking culture, the degree of crystallinity was 51.2%, the lowest degree. Under the other culturing conditions, the trend should remain in the range of 89.7-84%. Overall, the physical properties of BC produced from SFW were similar to those of BC from HS medium, a commercial complex medium, and BC production by the air circulation culture mode brought more favorable results in terms of the physical properties and its ease of scale-up. Therefore, it is expected that a new BC production method, like air circulation culture using SFW, would contribute greatly to BC-related manufacturing.

Production of the Rare Ginsenoside Rh2-MIX (20(S)-Rh2, 20(R)-Rh2, Rk2, and Rh3) by Enzymatic Conversion Combined with Acid Treatment and Evaluation of Its Anti-Cancer Activity

  • The ginsenoside Rh2 has strong anti-cancer, anti-inflammatory, and anti-diabetic effects. However, the application of ginsenoside Rh2 is restricted because of the small amounts found in Korean white and red ginsengs. To enhance the production of ginsenoside Rh2-MIX (comprising 20(S)-Rh2, 20(R)-Rh2, Rk2, and Rh3 as a 10-g unit) with high specificity, yield, and purity, a new combination of enzymatic conversion using the commercial enzyme Viscozyme L followed by acid treatment was developed. Viscozyme L treatment at pH 5.0 and $50^{\circ}C$ was used initially to transform the major ginsenosides Rb1, Rb2, Rc, and Rd into ginsenoside F2, followed by acid-heat treatment using citric acid 2% (w/v) at pH 2.0 and $121^{\circ}C$ for 15 min. Scale-up production in a 10-L jar fermenter, using 60 g of the protopanaxadiol-type ginsenoside mixture from ginseng roots, produced 24 g of ginsenoside Rh2-MIX. Using 2 g of Rh2-MIX, 131 mg of 20(S)-Rh2, 58 mg of 20(R)-Rh2, 47 mg of Rk2, and 26 mg of Rh3 were obtained at over 98% chromatographic purity. Then, the anti-cancer effect of the four purified ginsenosides was investigated on B16F10, MDA-MB-231, and HuH-7 cell lines. As a result, these four rare ginsenosides markedly inhibited the growth of the cancer cell lines. These results suggested that rare ginsenoside Rh2-MIX could be exploited to prepare an anti-cancer supplement in the functional food and pharmaceutical industries.

Yield and Production Forecasting of Paddy Rice at a Sub-county Scale Resolution by Using Crop Simulation and Weather Interpolation Techniques (기상자료 공간내삽과 작물 생육모의기법에 의한 전국의 읍면 단위 쌀 생산량 예측)

  • Crop status monitoring and yield prediction at higher spatial resolution is a valuable tool in various decision making processes including agricultural policy making by the national and local governments. A prototype crop forecasting system was developed to project the size of rice crop across geographic areas nationwide, based on daily weather pattern. The system consists of crop models and the input data for 1,455 cultivation zone units (the smallest administrative unit of local government in South Korea called "Myun") making up the coterminous South Korea. CERES-rice, a rice crop growth simulation model, was tuned to have genetic characteristics pertinent to domestic cultivars. Daily maximum/minimum temperature, solar radiation, and precipitation surface on 1km by 1km grid spacing were prepared by a spatial interpolation of 63 point observations from the Korea Meteorological Administration network. Spatial mean weather data were derived for each Myun and transformed to the model input format. Soil characteristics and management information at each Myun were available from the Rural Development Administration. The system was applied to the forecasting of national rice production for the recent 3 years (1997 to 1999). The model was run with the past weather data as of September 15 each year, which is about a month earlier than the actual harvest date. Simulated yields of 1,455 Myuns were grouped into 162 counties by acreage-weighted summation to enable the validation, since the official production statistics from the Ministry of Agriculture and Forestry is on the county basis. Forecast yields were less sensitive to the changes in annual climate than the reported yields and there was a relatively weak correlation between the forecast and the reported yields. However, the projected size of rice crop at each county, which was obtained by multiplication of the mean yield with the acreage, was close to the reported production with the $r^2$ values higher than 0.97 in all three years.

Standard Procedure for the Aerodynamic Design of Small Wind Turbine Blades (소형 풍력 블레이드 공력 설계를 위한 표준 절차 구축)

  • There have been many academic researches on the aerodynamic design of wind turbine based on blade element method (BEM) and momentum theory (MT, or actuating disk theory). However, in the real world, the turbine blade design requires many additional constraints more than theoretical analysis. The standard procedure is studied in the present paper to design new blades for the wind turbine system ranged from the small size from 1 to 10 kW. From the experience of full design of a 10 kW blade, the authors tried to set up a standard procedure for the aerodynamic design based on IEC 61400-2. Wind-turbine scale, rotating speed, and geometrical chord/twist distribution at the segmented span positions are calculated with a suitable BEM/MT code, and the geometrical shape of tip and root should be modified after considering various parameters: wing-tip vortex, aerodynamic noise, turbine efficiency, structural safety, convenience of fabrication, and even economic factor likes price, etc. The evaluated data is passed to the next procedure of structural design, but some of them should still be corresponded with each other: the fluid-structure interaction is one of those problems not yet solved, for example. Consequently, the design procedure of small wind-turbine blades is set up for the mass production of commercial products in this research.

Meat Quality of Lambs Fed on Palm Kernel Meal, a By-product of Biodiesel Production

  • This study aimed to establish the optimum level of palm kernel meal in the diet of Santa Ines lambs based on the sensorial characteristics and fatty acid profile of the meat. We used 32 lambs with a starting age of 4 to 6 months and mean weight of $22{\pm}2.75kg$, kept in individual stalls. The animals were fed with Tifton-85 hay and a concentrate mixed with 0.0, 6.5, 13.0 or 19.5% of palm kernel meal based on the dry mass of the complete diet. These levels formed the treatments. Confinement lasted 80 days and on the last day the animals were fasted and slaughtered. After slaughter, carcasses were weighed and sectioned longitudinally, along the median line, into two antimeres. Half-carcasses were then sliced between the 12th and 13th ribs to collect the loin (longissimus dorsi), which was used to determine the sensorial characteristics and fatty acid profile of the meat. For sensorial evaluation, samples of meat were given to 54 judges who evaluated the tenderness, juiciness, appearance, aroma and flavor of the meat using a hedonic scale. Fatty acids were determined by gas chromatography. The addition of palm kernel meal to the diet had no effect on the sensorial characteristics of meat juiciness, appearance, aroma or flavor. However, tenderness showed a quadratic relationship with the addition of the meal to the diet. The concentration of fatty acids C12:0, C14:0 and C16:0 increased with the addition of palm kernel meal, as did the sum of medium-chain fatty acids and the atherogenicity index. Up to of 19.5% of the diet of Santa Ines lambs can be made up of palm kernel meal without causing significant changes in sensorial characteristics. However, the fatty acid profile of the meat was altered.

Expression and Characterization of a Single-Chain Variable Fragment against Human LOX-1 in Escherichia coli and Brevibacillus choshinensis

  • The single-chain variable fragment (scFv) against lectin-like oxidized low-density lipoprotein receptor-1 (LOX-1) is a promising molecule for its potential use in the diagnosis and immunotherapy of atherosclerosis. Producing this scFv in several milligram amounts could be the starting point for further engineering and application of the scFv. In this study, the abundant expression of the anti-LOX-1 scFv was attempted using Escherichia coli (E. coli) and Brevibacillus choshinensis (B. choshinensis). The scFv had limited soluble yield in E. coli, but it was efficiently secreted by B. choshinensis. The optimized fermentation was determined using the Plackett-Burman screening design and response surface methodology, under which the yield reached up to 1.5 g/l in a 5-L fermentor. Moreover, the properties of the scFvs obtained from the two expression systems were different. The antigen affinity, transition temperature, and particle diameter size were 1.01E-07 M, $55.2{\pm}0.3^{\circ}C$, and 9.388 nm for the scFv expressed by B. choshinensis, and 4.53E-07 M, $52.5{\pm}0.3^{\circ}C$, and 13.54 nm for the scFv expressed by E. coli. This study established an efficient scale-up production methodology for the anti-LOX-1 scFv, which will boost its use in LOX-1-based therapy.

Deproteinized Mulberry Leaf Juice - A New Media for Growth of Microorganisms

  • Mulberry being a foliage crop is grown extensively for feeding of silkworms and are also used for cattle feeding. These loaves are highly nutritious, which contain various mineral elements and bio-molecules such as carbohydrates, proteins, lipids and other essential amino acids, etc. In the present study, deproteinized mulberry leaf juice was used for preparation of the medium for cultivation of various types of microbes. Results revealed that deproteinized mulberry leaf juice medium is best for isolation of fungi, bacteria and actinomycetes and this medium can be substituted with synthetic media, which are haying the costly ingredients for isolation and identification of bacteria, fungi and actinomycetes. Further, this deproteinized mulberry juice medium can also be used in mass multiplication of useful/beneficial microbes to enhance soil microflora to improve soil fertility and to avoid root diseases. Perspective enterprises can take up the mass multiplication/large-scale production of useful microbes such as Trichoderma, Rhizobium, Pseudomonas and Bacillus to use in mulberry and in other agricultural crops using deproteinized mulberry leaf juice.

Fluidized Bed Drying Effect on the Aerogel Powder Synthesis

  • A fluidized bed drying approach was utilized to the synthesis of water glass based silica aerogel powders. The effects of the fluidized bed drying conditions such as the superficial velocity and temperature of hot air and bead size as well as bead/wet-gel ratio, on the physical properties such as tapping density and productivity of the aerogel powders were systematically investigated. The experimental results showed that the amount of beads mixed with wet-gels in the fluidized bed column has the most profound impact on the fluidization efficiency, greatly enhancing the yield of the aerogel powders up to 98% with a proper bead/wet-gel weight ratio as compared to 72% without using beads. No significant change was observed in the tapping density over a wide range of the fluidized drying condition. Consequently the fluidized bed drying approach shows a good promise as an alternative route for the large-scale production of the aerogel powders.
