증손오적환(增損五積丸) 비적방(脾積方)이 생쥐의 복강암에 미치는 영향 (Effects of Jeungson-Ojeok-Hwan-Bijukbang on Solid Tumor in Mice)

  • Objective : Jeungson-Ojeok-Hwan Bijukbang(JOH) has been used to treat patients with tumor etc. In the theory of Korean medicine, JOH can treat Juk-Chui. JOH is known to have treat Juk-Chui. Juk-Chui is analogous to tumor. Methods : For these reasons, the present study was designed to investigate the effects of JOH on solid tumor in mice in terms of immune-potentiating and direct cytotoxic action of JOH in vitro and vivo study. The present author investigated thymocyte and splenocyte proliferation to confirm immune-potentiating activity of JOH and also investigated tumor/body weight ratio and survival rates in tumor bearing mice. Results : In this study, administration of JOH decreased tumor/body weight ratio significantly, and prolonged survival duration compared to non-treated control. In addition, treatment with JOH suppressed proliferation rate of Sarcoma 180 (S-180) cells significantly, and elevated proliferation rates of thymocytes isolated from normal mice. These results were co-related with in vivo study. Conclusion : these results suggest that JOH is useful to treat patient with solid tumor, because JOH has direct toxic action for tumor cell and immune-potentiating action for T cells. Further study will need to elucidate exact mechanisms related in anti-cancer action of JOH.

벼메뚜기(Oxya japonica japonica Thunberg) 물 추출물 LCT-CT의 항암면역 활성에 관한 유세포 분석학적 연구 (A Flow Cytometrical Analysis of the Antitumor and Immunostimulatory Effects of LCT-CT, a Cold-water Extract Prepared from Rice Grasshopper Oxya japonica japonica Thunberg)

  • Water extracts of rice grasshopper (Oxya japonica japonica Thurnberg) were prepared and their antitumor and immunostimulatory activities were investigated using a flow cytometer. When LCT-CT was ip injected into ICR mice at the dose of 33.3 mg/kg before and after the implantation of $4{\times}10^5$ cells/mouse of sarcoma 180 tumor cells, it inhibited the growth of the tumor cells by 96.6%, showed lymphoblstogenic activities on the splenic lymphocytes and increased the expression of CD25 molecules on the splenic T lymphocytes. When co-cultured with the splenic lymphocytes of a BALB/c mouse, LCT-CT showed strong immunostimulatory activities at the concentration of $25{\sim}100{\mu}g/ml$ by significantly increasing lymphoblasts ratio and CD25 expression.

  • 목단피의 mouse 복수암에 대한 항암성을 다른 생약제의 항암성과 비교하여 연구하였다. 천연생약제(목단피, 주목, 울금, 인경쓱, 여정실, 맥문동) methanol 추출물을 hexane, chloroform ($CHCl_3$), ethylacetate (EtOAc), butanol (BuOH)로 fractionation하여 mouse leukemia L1210 cell과 Sarcoma 180 (S-180) cell에 강한 독성을 나타낸 fraction에 대해 mouse 복수암 억제실험을 실시하였다. 복수형 종양세포 S-180을 ICR mouse (male, $6{\sim}7$주령, $23g{\pm}3g$, 처리당 7마리)의 복부에 주사 ($1{\times}10^{6}$ cells/0.1 ml PBS)한 1일 후 부터 10% DMSO에 용해한 시료 ($30{\mu}g/g$ body weight)를 매일 0.1ml씩 10일간 주사하고 수명연장 효과와 몸무게의 변화를 조사하였다. 처리 fraction 중에서 목단피의 EtOAc fraction이 가장 강한 항암성을 보였는데, 수명연장에서는 대조구의 17.2일 (100%)에 비하여 28.7일로서 167%로 연장되었으며, 체중 증가율도 대조구보다 낮았다 (p<0.05). 목단피의 농도별 (5, 10, 30, $60{\mu}g/g$ body weight) 항암효과는 $30{\mu}g$에서 가장 높았고, $60{\mu}g$처리구 에서는 독성이 나타났다. 목단피의 EtOAc fraction으로부터 GC-MS에 의해 잠정적으로 동정된 2-methoxyphenol, 1-(4-hydroxy-3- methoxyphenyl)-ethanone, 8-methyl-2,4(1H,3H)-pteridinedione, 2,5-furandicarboxylic dimethyl ester 가 mouse 복수암 억제에 관련이 있는 주요 화합물로 추정된다.

Enhancement of Anticancer Activities of Kimchi by Manipulating Ingredients

  • To enhance the antitumor activity of Chinese cabbage kimchi, four kinds of kimchi, which ere differently prepared in kinds and levels of sub-ingredients, were fermented at 15$^{\circ}C$ for 1 day and then at 5$^{\circ}C$ up to pH 4.3. The solid tumor formation, hepatic glutathione S-transferase activity and glutathione contents in the liver, and natural killer (NK) cell activity of spleen were determined from the sarcoma-180 cell injected Balb/c mice that were treated with methanol extracts of the kimchi samples. Kimchi IV, prepared with organically cultivated Chinese cabbage, red pepper powder, garlic, Chinese pepper powder mustard leaf and heat processed salt (Gueun salt), reduced the tumor formation by 39.3% compared to the sarcoma-180 cell treated group, resulting in the smallest tumor weight. Methanol extracts of the kimchi III and kimchi IV recovered the activities of hepatic glutathione S-transferase(GST) that was decreased by the transplantation of the sarcoma-180 cells to th mice. The injections of methanol extracts of kimchi II and kimchi IV increased glutathione contents in sarcoma-180 cells treated mice. The methanol extract of kimchi IV increased the natural killer (NK) cell activity of spleen lymphocytes a more effectively (p<0.05) than those the other kimchi samples. These results suggest that the anticancer activities of kimchi can be increased by changing the kinds and levels of sub-ingredients.

인진쑥 추출물의 세포독성 효과 (Cytotoxic Effect of Artemisia capillaris Extracts on the Cancer Cells on in vitro)

  • Artemisia capillaris THUNB is a perennial herb that belongs to the family Compositae spp and the most common plant among the various herbal folk remedies used in treatment of abdominal pain, hepatitis, chronic liver disease, jaundice and coughing in Korea. This experiment was conducted to investigate cytotoxic effects of Artemisia capillaris extracts on the Hepa-1c1c7 and Sarcoma 180 cancer cells on in vitro experimental tests. On in vitro tests using MTT assay and SRB assay, the extracts showed prominent cytotoxic effects on the two kinds of cancer cell lines, respectively. Antihumor effects appeared in the concentration of over $250{\mu}g/mL$ of both ethanol and ethyl acetate extract, over $500{\mu}g/mL$ of methanol extract, over $5000{\mu}g/mL$ in water extract and over 50% cytotoxicity on the Hepa-1c1c7 and Sarcoma 180. The results suggest that Artemisia capillaris extracts have prominent cyotoxic effects on the cancer cell lines Hepa-1c1c7 and Sarcoma 180.

해암단이 수종의 암세포에 미치는 항암 효과 (Anti-tumor Activities of Haeamdan on Various Cancer Cells)

  • Objective : The objective of this study was to investigate the anti-tumor activity of the complexed herbal formula, Haeamdan (HAD). Methods : Seven Cancer cell lines, LoVo, MCF-7, AGS, Sarcoma 180, HL-60, NCI-H69, LL/2, were prepared and the cytotoxicity was assessed by 3-(4,5-dimethylthiazol-2yl)-2,5-dephenyl tetrazolium bromide (MTT) assay. HAD was applied with various concentrations from 0.1 to 1.0 mg/ml to figure out the appropriate dosage. ICR male mice were intraperitoneally implanted with Sarcoma 180 and divided into 8 species for each group. Control group was treated with normal saline, positive control group was treated with cyclophosphamide 8mg/kg, and experimental group was treated with HAD 1g/kg. Results : Among seven cancer cell lines, HAD exhibited strong cytotoxic activities to followed four cancer cell lines, that is, Sarcoma 180, HL-60, NCI-H69, and LL/2. These cytotoxic activity was expressed under 0.50 mg/ml of IC50 under 0.1~1mg/ml of OBW. When Sarcoma 180 cancer cell was implanted in ICR male mice and treated with the HAD, HAD prolonged the median overall survival for 3.6 days, from 17.5 to 21.1 days. Conclusion : HAD showed strong cytotoxicity to the cancer cells, Sarcoma 180, HL-60, NCI-H69, on in vitro study and it showed anti-tumor activity in vivo with the peritoneal cancer mice by prolonging the median survival for 3.6 days. Further researches would be expected to support the anti-tumor efficacy of HAD.

Sarcoma-180 유발 생쥐에 대한 Cyclophosphamide, Picibanil 및 Tubercin-3의 투여효과 (Anti-Tumor Effect of Cyclophosphamide, TUbercin-3 , and Picibanil on Sarcoma-180 Bearing Mice)

  • This study was carried out to detemrine the efficacy of combined treatment of cyclophosphamide with tubericin-3 and or picibanil. One hundred sixty sarcoma-180 bearing mice were divided eight groups. Each group received saline, tubercin-3, picibanil , and cyclophosphamide along and/or received cyclophosphamide with tubercin-3 , with picibanil or with both tubercin-3 and picinanil, respectively.Average surviving time of each group of animals was as follows ; control was 10.9days, tubercin-3 was 15.1 days. picibanil was 12.6 days, and cylophosphamide was 17.9 days, In combined therapyy that cylophosphamide injected with tubercin-3 , the surviving time was 26.8 days an din the case of other therapy that cyclophosphamide injected with tubercin-3, the surviving time was 26.8 days an din the case of other therapy that cyclophosphamide injected with picibanil, the surviving time was 21.9 day and cyclophosphamide treated with both turbercin03 and picibanil, the surviving time was found to be 18.2 days, conclusively , the therapeutic potentiation seemed to be extended when combined tretment of the chemotherapeutics cyclophosphamide with either one of immunotherapeutics tubericin-3 or picibanil was tried, Combinatin of tubercin-3 and picibanil showed to be atagonistic each other. Yield of ascites fluid were determined 7 days after injectino of sarcoma-180 ascites tumor cells. Adminitration of cyclophosphamide, tubercin-3 , and picibanil alone and their various combinations reduced the yield of ascites fluid except for picibanil group.

Suppressed CD31 Expression in Sarcoma-180 Tumors after Injection with Toxoplasma gondii Lysate Antigen in BALB/c Mice

  • The anti-tumorigenic effects of Toxoplasma gondii (RH) antigens were studied in a murine sarcoma-180 tumor model. To determine the anti-tumor effects, the reduction in tumor size and expression of CD31 (an angiogenesis marker in the tumor tissue) were examined after injection of BALB/c mice with T. gondii lysate antigen (TLA) or formalin-fixed, proliferation-inhibited, T. gondii tachyzoites. Tumors were successfully produced by an intradermal injection of sarcoma-180 cells with plain Matrigel in the mid-backs of mice. After injection with TLA or formalin-fixed T. gondii tachyzoites, the increase in tumor size and weight nearly stopped while tumor growth continued in control mice that were injected with PBS. CD31 expression in TLA-treated or formalin-fixed T. gondii-injected mice was lower than the control mice. Accordingly, the present study shows that the treatment of mice with formalin-fixed T. gondii or TLA in the murine sarcoma-180 tumor model results in a decrease of both tumor size and CD31 expression.

개불 (Urechis unicinctus)에서 추출한 당단백질의 항암효과 및 면역활성 (Antitumor Effect and Immunological Activity of Glycoprotein from Urechis unicinctus)

  • 개불 당단백질의 sarcom 180 cell 고형암 성장 저지능은 4mg/kg에서 43.63%로 나타났으나, 20mg/kg이상의 농도에서는 고형암 성장 저지능을 보이지 않았다. Sarcom 180 cell에 대한 수명 연장 실험에서도 2mg/kg와 4mg/kg의 저농도에서는 상당한 수명연장 효과를 보였으나, 2Omg/kg 이상의 고농도 투여군에서는 수명연장 효과를 나타내지 않았다. 개불 당단백질의 sarcoma 180 cell에 대한 직접적인 세포 독성 효과는 농도의 증가에 따라 약간의 종양세포 살해효과를 보였으나, 그 효과는 미약하였다. 개불 당단백질의 면역능 증진효과는 체중에 대한 간과 비장의 중량비 증가, 총 복강세포수의 증가 그리고, 당단백질을 고농도 투여시 30.78%~46.30%정도의 백혈구 수 증가 등으로 확인되었다. 이러한 결과로서 개불에서 추출한 당단백질의 항암효과는 면역활성의 증진과 면역자극의 효과라고 볼 수 있었으나, 전반적인 면역활성의 증가에도 불구하고, sarcoma 180 cell에 대한 고형암 성장 저지실험과 수명연장실험에서 고농도로 투여시에는 오히려 활성이 나타나지 않았다. 따라서 개불 당단백질의 최적투여 농도와 고농도 투여시의 부작용에 관한 더 많은 연구가 필요하였다. 이상의 결과에서 개불에서 추출한 당단백질의 면역자극에 의한 항종양 활성을 확인 되므로, 일반적으로 사용되어지는 항암 화학요법제와 더 불어 병행될 수 있는 면역증진제로서의 개발 가능성이 있다고 보여진다.

  • The Artemisia capillaris THUNB is a perennial herb that belongs to the family Compositae spp and probably the most common plant among the various herbal folk remedies being used in the treatment of abdominal pain hepatitis chronic liver disease, jaundice and coughing in Korea. Recently the biological and pharmacological actions of herb have been studied well such as antibacterial, antidiabetic and antitumor activities. This experiment was conducted to investigate antitumor and immunomodulatory effects of Artemisia capillarix extracts against Hepa-1c1c7 and Sarcoma 180 cancer cells on in vivo experimental tests. On in vivo experimental tests using 280 ICR mice the gain of body weight in the control-group mice bearing Sarcoma 180 ascites tumor was 1.5 times more than that of the normal-group mice after 33 days. However, the gain of body weight in all experimental groups administered with Artemisia capillaris extracts was significantly lower than that of the control-group mice (P<0.05). The mean survival times of mice administered with Artemisia capillaris extracts of 25 and 100 mg/kg for 28 days were shown to be 25.39% and 15.39% longer than that of the control-group mice injected with saline (P<0.05). Artemisia capillaris extracts showed the highest tumor inhibition effects, which were 42.4% and 27.2% when intraperitoneally injected with doses of 25 and 100 mg/kg once a day for 28 days in inoculated ICR mice with Sarcoma 180 solid tumor cells (P<0.05). The results suggest that Artemisia capillaris methanol extracts have prominent antitumor effects on the cancer cell lines Hepa-1c1c7 and Sarcoma 180.
