Quality Characteristics of 4 Year-old Ginseng by Enzymatic Hydrolysis Conditions (4년근 인삼의 효소적 가수분해 조건에 따른 품질특성)

  • This study investigated quality characteristics of 4 year-old ginseng by enzymatic hydrolysis conditions to increase utilization. Ginseng was ground after steaming and was each treated with hydrolase A, B, C and D. When quality characteristics by the enzymes were examined, no significant difference was observed with pH of 5.5~5.6 and the sugar content of 4.0~4.33. The crude saponin content was the highest in ginseng treated with D, followed by B, C and A. The crude saponin, the reducing sugar and the total sugar contents increased until 0.3% (w/w) concentration in enzyme D with no significant difference by its concentration. Although active ingredients increased with time passage of hydrolysis, no significant change was found after three hours and the crude saponin content was the highest when ginseng was treated at $60^{\circ}C$. From these results, optimum conditions for 4 year-old ginseng were $60^{\circ}C$ for 3 hours with 0.3% (w/w) enzyme D, and under these conditions the reducing sugar, the total sugar and the crude saponin contents recorded 18.11, 36.21 and 4.23 mg/g, respectively. Therefore, enzymatic hydrolysis was found to be effective in increasing active ingredients of 4 year-old ginseng with various usages expected.

Evaluation of mango saponin in broilers: effects on growth performance, carcass characteristics, meat quality and plasma biochemical indices

  • Objective: The objective of the present study was to determine whether mango saponin (MS) could be used as a feed additive in broiler chicks by evaluating growth performance, carcass characteristics, meat quality, and plasma biochemical indices. Methods: A total of 216 1-d-old Arbor Acres male broiler chicks were randomly assigned into three dietary treatments supplemented with 0 (control), 0.14% (MS 0.14%), or 0.28% (MS 0.28%) MS. Each treatment had six replicates (cages) with 12 chicks each. The feeding trial lasted for six weeks. Results: Compared with the control, dietary supplemented with 0.14% or 0.28% MS increased average daily weight gain of chicks in the grower (22 to 42 d) and the whole (1 to 42 d) phases, and the final body weight of chicks on d 42 was higher in MS supplemented groups (p<0.05). Lower $L_{45min}{^{\star}}$ (lightness) and $L_{24h}{^{\star}}$ values, lower $b_{24h}{^{\star}}$ (yellowness) value, and higher $a_{45min}{^{\star}}$ (redness) and $a_{24h}{^{\star}}$ values of the breast muscle were observed in chicks fed with 0.28% MS on d 42 (p<0.05). The total antioxidant capacity in plasma increased in MS 0.14% group on d 21 (p<0.001). Lower contents of plasma total cholesterol and triglyceride were observed in chicks fed with 0.28% MS on d 21 and d 42, whereas the group supplemented with 0.14% MS only decreased plasma triglyceride content on d 21 (p<0.05). The glucose content in plasma decreased in MS 0.28% group on d 42 (p<0.001). Conclusion: Overall, MS could be used as a feed additive in broiler chicks, and the supplemental level of 0.28% MS in diet could improve growth performance, meat quality, and plasma lipid metabolism in broiler chicks.

Panaxcerol D from Panax ginseng ameliorates the memory impairment induced by cholinergic blockade or Aβ25-35 peptide in mice

  • Background: Alzheimer's disease (AD) has memory impairment associated with aggregation of amyloid plaques and neurofibrillary tangles in the brain. Although anti-amyloid β (Aβ) protein antibody and chemical drugs can be prescribed in the clinic, they show adverse effects or low effectiveness. Therefore, the development of a new drug is necessarily needed. We focused on the cognitive function of Panax ginseng and tried to find active ingredient(s). We isolated panaxcerol D, a kind of glycosyl glyceride, from the non-saponin fraction of P. ginseng extract. Methods: We explored effects of acute or sub-chronic administration of panaxcerol D on cognitive function in scopolamine- or Aβ25-35 peptide-treated mice measured by several behavioral tests. After behavioral tests, we tried to unveil the underlying mechanism of panaxcerol D on its cognitive function by Western blotting. Results: We found that pananxcerol D reversed short-term, long-term and object recognition memory impairments. The decreased extracellular signal-regulated kinases (ERK) or Ca2+/calmodulin-dependent protein kinase II (CaMKII) in scopolamine-treated mice was normalized by acute administration of panaxcerol D. Glial fibrillary acidic protein (GFAP), caspase 3, NF-kB p65, synaptophysin and brainderived neurotrophic factor (BDNF) expression levels in Aβ25-35 peptide-treated mice were modulated by sub-chronic administration of panaxcerol D. Conclusion: Pananxcerol D could improve memory impairments caused by cholinergic blockade or Aβ accumulation through increased phosphorylation level of ERK or its anti-inflammatory effect. Thus, panaxcerol D as one of non-saponin compounds could be used as an active ingredient of P. ginseng for improving cognitive function.

Saponin Composition and Physico-Chemical Properties of Korean Red Ginseng Extract as Affected by Extracting Conditions (추출조건에 따른 홍삼추출액의 사포닌 조성 및 이화학적 특성)

  • In this study, the composition of saponin and physico-chemical properties of Korean red ginseng extract was analyzed based on various extracting conditions. The total saponin and individual ginsenoside concentration of the red ginseng extract showed a decreasing trend as the extracting temperature and time increased; also, the extracting condition at $75^{\circ}C$ for 24 hours showed the highest concentration. In contrast, the concentration of $Rg_3$ increased as the extracting temperature and time increased within the particular range. It was suggested that a certain part of ginsenosides changes to $Rg_3$ according to extracting conditions; thus, the concentration of $Rg_3$ increased. Physico-chemical properties of Korean red ginseng extract based on the extracting conditions were different compared to those for saponin; so, as the extracting temperature and time increased, brix and color difference increased but pH decreased indicating stabilization of the overall quality of the product. Therefore, the most appropriate extracting condition for both the product quality of Korean red ginseng extract and stable extraction of saponin was $80^{\circ}C$ within 48 hours, minimizing the loss of ginsenosides.

Effect of Total Ginseng Saponin on the Opioid Receptor Binding in Mouse Brain (Mouse뇌에서 Opioid 수용체 결합력에 미치는 인삼의 영향)

  • The modulatory effects of total ginseng saponin (TGS) on the 1, 6, and opioid receptor binding in morphine tolerance and dependence were examined in this study. The specific [$^{3}H$]DAGO ([D-$Ala^2$, N-$Mephe^4$, $Glyco^4$] enkephalin) binding was significantly increased in chronic morphine (10 mg/kg, i.p.) treated mouse striatum. The specific [$^{3}H$]DPDPE ([D-$Pen^2$, D-$Pen^5$] enkephalin) binding was ignificantly increased following morphine treatment in the mouse striatum and cortex. Also, an apparent decrease in the affinity of [$^{3}H$]DPN (diprenorphine) was observed after chronic morphine treatment in mouse striatum and cortex. 7GS produced a sleight increase of specific [$^{3}H$]DAGO, [$^{3}H$]DPDPE binding and a significant increase of specific [$^{3}H$]DPN binding in the mouse brain striatum. In cortex, TGS produced an inhibition of specific [$^{3}H$]DAGO and [$^{3}H$]DPDPE binding and increase of the specific [$^{3}H$]DPN binding. The prolonged administration of TGS (25, 50, 100, and 150 mg/kg, i.p., 3 wks) produced an inhibition of increased [$^{3}H$]DAGO specific binding following morphine without significant changes in the agonist binding to and receptors in mouse striatum and cortex. These contracted alterations in $\mu$, $\gamma$ and $\kappa$ opiate receptor binding were dependent in TGS dogs and brain sites.

Effects of the Saponin of Polygalae Radix on the Renal Function and CNS-Depression (원지(遠志) Saponin의 이뇨효과(利尿效果) 및 중추억제작용(中樞抑制作用)에 관한 연구(硏究))

  • Polygalae Radix was used as diuretics, analgesics and expertorants in oriental medicine. The root of Polygala tenuifolia Willd. (Polygalaceae) is comprised saponin (Onjisaponin A,B,C,D,E,F and G) polygalitol, onsitin and sugars. The pharmacological action of crude Polygala-saponin (PS) obtained from the roots are studied. The following results were obtained; 1) The median lethal dose $(LD_{50})$ of PS in mice is presented 71.1mg/kg s.c. and 694. 5mg/kg p.o.. 2) PS demonstrated diuretic action of relatively long acting duration in mice. 3) The diuretic mechanism of PS was found due to inhibitory effect of renal tubular reabsorption of electrolytes and glomerular vascular dilatation. 4) The group, administered simultaneously PS and cefadroxil monobydrate was significantly increased with PS alone group on diuretic action. Synergistic effect cefadroxil monohydrate on the diuretic action of PS seems due to competitive inhibition of plasma protein binding with PS. 5) PS demonstrated analgesic action by the acetic acid stimulating method and Randall-Selitto test in mice. 6) PS presented antipyretic action against febrile treated with the typhoid vaccine. 7) PS was significantly prolonged against the hypnotic duration of pentobarbital in mice. 8) Onset time convulsion and death induced by picrotoxin and strychnine in mice were not delayed. According to the above results, the PS was identified as a pharmacological active component obtained from roots of Polygala tenuifolia Willd.

Soil Physico-Chemistry and Saponins Content of Platycodon grandiflorum Radix Cultured from Different Sites in Gyeongnam Province (경남지방에서 도라지 재배지역별 토양이화학적 특성과 사포닌 함량)

  • This study was carried out in order to survey the soil physico-chemical, morphology and content of saponins of 5 year-old Platycodon grandiflorum radix from different areas in Gyeongnam. Soil physical properties was showed 1.01-1.29 of bulk density of soil and soil hardness was increased with increasing soil depth, especially below the 30 cm, however it was maintained or small increased below the 40 cm. T-N and OM contents of top soil were highest at Geochang as 0.27% and 56.9 g/kg, respectively, compared to other areas. The root length was longer at Kimhea and Geochang as 33.8 and 33.7 cm, respectively, and fresh weight was heavy at Geochang as 208.5 g. The contents of saponin of P. grandiflorum radix was higher in fine root compare to main root. By region, content of saponins of P. grandiflorum radix were higher at Hapcheon than other area. Mg content was highly negatively correlated at p < 0.01 with platycodin D3, deapioplatycodin D, platycodin D, and deapioplatycodin D ($-0.499^{**}$, $-0.433^{**}$, $-0.421^{**}$, $-0.511^{**}$ respectively). These results suggest that no-fertilized Mg effected on the improvement of saponin contents in P. grandiflorum.

Identification of a novel triterpene saponin from Panax ginseng seeds, pseudoginsenoside RT8, and its antiinflammatory activity

  • Background: Panax ginseng Meyer (Araliaceae) is a highly valued medicinal plant in Asian regions, especially in Korea, China, and Japan. Chemical and biological studies on P. ginseng have focused primarily on its roots, whereas the seeds remain poorly understood. This study explores the phytochemical and biological properties of compounds from P. ginseng seeds. Methods: P. ginseng seeds were extracted with methanol, and 16 compounds were isolated using various chromatographic methods. The chemical structures of the isolates were determined by spectroscopic data. Antiinflammatory activities were evaluated for triterpene and steroidal saponins using lipopolysaccharide-stimulated RAW264.7 macrophages and THP-1 monocyte leukemia cells. Results: Phytochemical investigation of P. ginseng seeds led to the isolation of a novel triterpene saponin, pseudoginsenoside RT8, along with 15 known compounds. Pseudoginsenoside RT8 exhibited more potent antiinflammatory activity than the other saponins, attenuating lipopolysaccharide-mediated induction of proinflammatory genes such as interleukin-1β, interleukin-6, inducible nitric oxide synthase, cyclooxygenase-2, and matrix metalloproteinase-9, and suppressed reactive oxygen species and nitric oxide generation in a dose-dependent manner. Conclusion: These findings indicate that pseudoginsenoside RT8 has a pharmaceutical potential as an antiinflammatory agent and that P. ginseng seeds are a good natural source for discovering novel bioactive molecules.

Effects of Storage Methods and Periods on Root Hardness and Content of Saponin in Platycodon grandiflorum Radix (저장방법과 기간이 도라지 뿌리의 경도와 사포닌함량 미치는 영향)

  • The research was conducted to provide basic information for store temperature which were low and room temperature and packing methods which were vacuum, packing and nitrogen of 3 year-old Platycodon grandiflorum. We investigated hardness and content of saponins, 1) platycodin D3, 2) polygalacin D and 3) deapioplatycodin D, in Platycodon grandiflorum and hardness of Platycodon grandiflorum, which were reduced by increasing storage period and decreased with increasing storing temperature, respectively. The packed storing method was better than others storing methods in low temperature. The high root hardness was significantly related with storing temperature and methods. The content of saponins in Platycodon grandiflorum, i.e., platycodin D3 and polygalacin D were reduced during storing period, however, the content of deapioplatycodin D was increased during storing period.

Effect of Mulching Materials on the Growth and Medicinal Ingredients in Platycodon grandiflorum Radix Organic Cultivation (도라지 유기재배시 피복재료에 따른 생육 및 약용성분 변화)

  • This study was conducted to elucidate the effect of mulching materials on growth characteristics, saponin contents and antioxidant activity of Platycodin grandiflorum Ridix roots for organic farming. Plastic film, sawdust and rice bran were treated by mulching with several different methods after transplanting P. grandiflorum Radix. In root length, when plastic film plot, sawdust plot and rice bran plot were treated, root length was recorded the highest scores (24.0~27.5 cm) in plastic film plot and sawdust plot. Root width was recorded the highest score (30.0 mm) in plastic film plot. Also, the number of fine-roots was the highest in plastic film plot (36.0). Fresh weight, which affects directly yield, was the highest in plastic film plot (130 g/plant). The contents of platycodin D3 and deapioplatycodin D were shown to 111.2 and 48.1 mg/100 g, the highest values were observed in sawdust plot. The total polyphenol and flavonoid contents were recorded the highest scores (11.0 and 8.6 mg/g, respectively) in plastic film plot. In this study we confirmed that there were differences among mulching materials in growth characteristics, saponin contents and antioxident activity of P. grandiflorum roots.
