미국흰불나방(Hyphantria cunea D.)의 난황단백질-3의 합성 및 이용 (Synthesis and Fate of Yolk Protein-3 in Hyphantria cunea D.)

  • Yolk protein-3 (YP3) was purified from the ovary of Hvpharatria cunea D. and the synthesis and fate during embryogenesis of WP3 were investisated by electrophoresis and fluorography. YP3 purified through gel slice and electrophoretic elution'was determined to have M. W. of 18 Kd and consist of one subunit. Haemolymph and fat body of male and female %were electrophoresed during vifellogenic stages to indentifv the vitellosenin in female. The result showed that there was no distinct difference in electrophoretic patterns betweerl male and female. However, tissue culture of fat body and maturing ovary indicated that YP3 was svuthesized by fat body. Aiso, vP3 in iaid eggs was maintained constant untii naut s artier oviposition and then decreased, indicating that YP3 was drastically used during late embryosenesis. However, a part of YP3 was present even in newly hatched first instar larvae.

  • This propose a simple DC voltage sensor less single phase PFC(Power Factor Correction Circuit) converter by detecting a AC current sensors are not required to construct the control system. The DC voltage is directly controlled by the command input signal Kd($V_o/V_a$)for the boost chopper circuit. The DC voltage regulation is small because of the feed forward control for the AC line voltage VS and no dependence of the circuit parameters. The sinusoidal current waveform in phase with the AC input voltage can be obtained. These characteristics are confirmed by some experiment results.

The Effect of One Leg Deadlift Exercise on the Muscle Activity of Lower Extremities according to the Weight Characteristics

  • Purpose: This study examined the effects of a kettlebell and aqua bag on the gluteus maximus, gluteus medius, and vastus medialis lower extremity muscle strength during a one-leg deadlift, a top-down exercise. Methods: Twenty healthy adults were enrolled in the one-leg deadlift exercise under two conditions. The muscle activity of the gluteus maximus, gluteus medius, and vastus medialis was measured by surface electromyography (S-EMG). A paired t-test was used for statistical analysis, and the statistical significance criteria were set to p<0.05. Results: A comparison of the muscle activity according to the degree of contraction of the gluteus maximus revealed low muscle activity in AD exercise compared to KD exercise (p<0.05), and there was no significant difference in the gluteus medius and vastus medialis (p<0.05). Conclusion: These results suggest that when the one-leg deadlift exercise is performed according to the weight characteristics, the gluteus maximus is suitable for intervention using a kettlebell, and the gluteus medius and vastus medialis are more suitable for intervention by providing instability using an aqua bag.

Antibiofilm Activity and Binding Specificity of Polyclonal DNA Aptamers on Staphylococcus aureus and Escherichia coli

  • Aptamers are short, chemically synthesized, single-stranded DNA or RNA oligonucleotides that fold into unique three-dimensional structures. In this study, we aim to determine the antibiofilm activity and binding specificity of the six polyclonal DNA aptamers (S15K3, S15K4, S15K6, S15K13, S15K15, and S15K20) on Staphylococcus aureus BPA-12 and Escherichia coli EPEC 4. Aptamer S15K6 showed the highest percentage of antibiofilm activity against S. aureus BPA-12 (37.4%) as shown by the lowest OD570 value of 0.313. Aptamer S15K20 showed the highest percentage of antibiofilm activity against E. coli EPEC 4 (15.4%) as shown by the lowest OD570 value of 0.515. Aptamers S15K13 and S15K20 showed antibiofilm activities against both S. aureus BPA-12 and E. coli EPEC4, and thus potentially have broad reactivity. Furthermore, based on the binding capacity and Kd values from our previous study, the binding specificity assay of selected polyclonal DNA aptamers (S15K3 and S15K15) against S. aureus BPA-12, E. coli EPEC 4, S. aureus BPA-6, S. agalactiae, E. coli MHA-6, and Listeria monocytogenes were performed using qPCR. Aptamers S15K3 and S15K15 showed specific binding to S. aureus BPA-12, E. coli EPEC 4, S. aureus BPA-6, and S. agalactiae, but could not bind to E. coli MHA-6 and L. monocytogenes. Therefore, this study showed that the polyclonal DNA aptamers have antibiofilm activity and were able to bind to S. aureus BPA-12 and E. coli EPEC 4 bacteria.

Influence of Dry Roasting of Whole Faba Beans (Vicia faba) on Rumen Degradation Characteristics in Dairy Cows, II: Starch

  • Whole faba beans (WFB) were dry roasted at different temperatures of 110, 130 and $150^{\circ}C$ for 15, 30 and 45 minutes (min) to determine the optimal heating conditions to increase bypass starch as glucose source which may be a limiting nutrient in high producing dairy cattle. Ruminant degradation characteristics of starch (St) of WFB were determined using in sacco method in 6 dairy cows fed 60% hay and 40% concentrate. Measured characteristics of St were soluble (washable) fraction (S), potentially degradation fraction (D) and the rate of degradation (Kd) of the insoluble but degradable St fraction. Based on measurement of these characteristics, percentage bypass starch (%BSt) and bypass starch (BSt) were calculated. Degradability of starch in the rumen was reduced by dry roasting at temperature of 130 and $150^{\circ}C$ and increased at $110^{\circ}C$. S varied from 50.0% in the raw whole faba beans (RWFB) and 53.7% in $110^{\circ}C$/15 min to 18.2% in $150^{\circ}C$/45 min. D varied from 49.9% in RWFB and 46.3% in $110^{\circ}C$/15 min to 81.8 % in $150^{\circ}C$/45 min. Kd varied from 9.8% in RWFB and 11.0% in the $110^{\circ}C$/30 min to 4.2 in $150^{\circ}C$/45 min. All these effects resulted in increasing %BSt from 22.1% in the $110^{\circ}C$/45 min and 23.9% in RWFB to 49.9% in the $150^{\circ}C$/45 min. Therefore BSt increased from 91.4 g/kg and 98.4 g/kg to 199.9 g/kg respectively. Dry roasting at $110^{\circ}C$ increased the starch rumen degradation. Treatment at higher temperature (130 and $150^{\circ}C$) decreased rumen degradation of starch and seemed to be linear up to highest values tested. No optimal dry roasting conditions of treatment could be determined at this stage. It may be concluded that dry roasting at temperatures of 130 and $150^{\circ}C$ was effective in shifting starch degradation from rumen to intestine to increase bypass starch.

Theileria sergenti merozoite 수용성 항원의 항원성과 면역성 II. 자연 조건하에서의 감염에 대한 면역시험 (Immunogenicity and Protective Efficacy of Solubilized Merozoite-enriched Theileria sergenti Immunogens. II. Protection against Natural Exposure under Field Conditions)

  • Theileria sergenti merozoite의 면역원성 polypeptide중 29, 34, 35 그리고 105KD를 함유한, 수용성 항원을 흘스타인 송아지 (1개월령)에게 접종한 후, 제주도의 야외 목장에 방목시킴으로써 진드기로부터 병원성 충주의 도전에 따른 항원의 효력, 안전성 그리고 방어력을 관찰하였다. 즉 송아지 20두에 수용성 항원(100 mg/dose)을 Freund's adjuvant와 함께 1차 접종, 4주 후에 추가접종하였으며, 다른 20두의 송아지를 대조군으로 사용하였다. 추가접종 5주 후에 이들 송아지를 야외 목장에 방목, 매월 혈액학적 그리고 생화학적 변화를 관찰하였다 예방접종군에 있어서 hematocrit와 총적혈구수는 진드기 도전 전과 비교하여 약간의 변화를 인정할 수 있었다. 일반적인 혈액학적 소견은 예방접종군과 비교하여 유의한 차이를 인정할 수 있었다(p<0.05). 7월에 있어서 적혈구내 기생률은 예방접종군에 있어서는 0.4%이었으나, 대조군에 있어서는 약 3.6%이었다. 체중 증가율, albumin, aspartate alninotransferase, total protein, 그리고 bilirubin에 있어서도 유의한 차이 (p<0.05)를 인정할 수 있었다. 예방접종군의 혈액학적 그리고 생화학적 측정치는 대조군 보다는 정상치에 접근하였다. 실험종료 직후에 있어서 예방접종군의 30% 송아지와 대조군의 모든 송아지의 치료와 대조군 중 25%는 수혈이 요구되었다. T. sergenti merozoite의 수용성 항원에 대한 이 같은 연구 결과는 앞으로 유전공학적 백신제조를 위한 연구자료로 활용될 수 있을 것이다.

Ruminal Behavior of Protein and Starch Free Organic Matter of Lupinus Albus and Vicia Faba in Dairy Cows

  • Faba beans (vicia faba) (FB) and lupin seeds (Lupinus Albus) (LS) were dry roasted at three temperatures (110, 130, $150^{\circ}C$) for 15, 30 or 45 min to determine the effects of dry roasting on rumen degradation of crude protein and starch free organic matter ($^{PSF}OM$). Rumen degradation characteristics of $^{PSF}OM$ were determined by the nylon bag incubation technique in dairy cows fed 60% hay and 40% concentrate. Measured characteristics of $^{PSF}OM$ were undegradable fraction (U), degradable fraction (D), soluble fraction (S), lag time (T0), and the rate of degradation (Kd). Based on the measured characteristics, rumen availability ($RA^{PSF}OM$) and bypass $^{PSF}OM$ ($B^{PSF}OM$) were calculated. Dry roasting did not have a greater impact on rumen degradation characteristics of $^{PSF}OM$ (p>0.05). S varied from 32.1 (raw) to 30.0, 27.8, 30.8% (LS) and 15.4 (raw) to 14.4, 20.8, 20.9% (FB); D varied from 65.4 (raw) to 66.3, 66.9, 55.9% (LS) and 54.9 (raw) to 55.0, 51.0, 64.7% (FB); U varied from 2.6 (raw) to 7.3, 7.0, 7.7% (LS) and 29.7 (raw) to 30.6, 28.2, 14.4% (FB); Kd varied from 6.0 (raw) to 7.3, 7.0, 7.7% (LS) and 22.4 (raw) to 24.4, 21.1, 7.9% (FB); $B^{PSF}OM$ varied from 35.5 (raw) to 33.8, 36.6, 38.2% (LS) and 41.3 (raw) to 41.5, 39.7, 47.6% (FB) at 110, 130 and $150^{\circ}C$, respectively. Therefore dry roasting did not significantly affect $RA^{PSF}OM$, which were 353.7, 367.9, 349.6, 336.9 (g/kg DM) (LS) and 12.82, 127.0, 133.7, 117.1 (g/kg DM) (FB) at 110, 130 and $150^{\circ}C$, respectively. These results alone with our previously published reports indicate dry roasting had the differently affected pattern of rumen degradation characteristics of various components in LS and FB. It strongly increased bypass crude protein (BCP) and moderately increased starch (BST) with increasing temperature and time but least affected $^{PSF}OM$. Such desirable degradation patterns in dry roasted LS and FB might be beneficial to the high yielding cows which could use more dry roasted $^{PSF}OM$ as an energy source for microbial protein synthesized in the rumen and absorb more amino acids and glucose in the small intestine.

한천 분해효소를 생산하는 해양 미생물 SH-1의 분리 및 특성 분석 (Isolation and Characterization of Marine Bacterial Strain SH-1 Producing Agar-Degrading Enzymes)

  • 한천을 분해하는 해양미생물을 한천을 유일한 탄소원으로 하는 인공 해수 한천 배지를 이용하여 제부도 개펄에서 분리하였다. SH-1으로 명명한 분리된 균주는 그람음성균이며 한 개의 극성 편모를 가지는 균이었다. 16S rRNA 유전자의 염기서열의 유사성 분석 결과 분리된 균주는 Neiella marina J221 [9]과 가장 높은 상동성을 보였다(96.5%). 분리 균주는 $28-37^{\circ}C$에서 생장하였지만 $42^{\circ}C$에서는 생장하지 못하였고 한천분해효소의 활성은 $37^{\circ}C$보다 $28^{\circ}C$에서 높은 활성을 보였다. 또한 SH-1균주는 1-5% NaCl (w/v)를 포함하는 배양액에서 생장이 가능했으며 3%의 농도에서 가장 좋은 생장을 보였고 한천을 분해하는 효소의 활성도 3% 염분농도의 배양액에서 가장 높았다. 48시간 배양한 세포배양액을 농축하여 조효소액을 준비하여 효소의 적정 pH와 적정 온도를 조사한 결과 pH 7.0에서와 $40^{\circ}C$에서 최적 효소 활성을 보였다. 조효소액을 사용하여 zymogram 분석을 실시한 결과 분자량 15, 35, 52 KD 크기의 3개 이상의 한천 분해효소를 생산하는 것으로 보인다. 박막크로마토그라피(TLC) 분석 결과 아가로스를 분해하여 네오올리고당을 생성하는 ${\beta}$-agarase 를 생산하는 것으로 추정된다.

콩 품종의 생육특성 및 생육단계별 ADP-Glucose Pyrophosphorylase의 활성변화 비교 (Comparison of Agronomic Characteristics and Activity Variation of ADP-Glucose Pyrophosphorylase at Different Growth Stages in Soybean Cultivars)

  • 콩의 생산성을 높이는데 중요한 역할을 하는 효소의 활성변화와 종실 수량과의 관련성을 탐색하기 위해 등숙관련효소인 ADP-glucose pyrophosphorylase(AGP)의 활성변화를 콩 품종별로 등숙기간에 따라 조사한 결과를 요약하면 다음과 같다. 1. 풍산나물콩은 협수 및 잎수가 131개 및 102개로 가장 많았으며, 100립중은 10.4g으로 가장 낮았으나 수량은 275kg/10a으로 가장 높아 물질생산 및 건물축적 효율이 우수한 것으로 나타났는데, 개화시기$(R_1,\;R_2)$에 AGP의 활성도 가장 높은 경향이었다. 2. 품종별 $CO_2$ 동화량은 풍산나물콩이 $20.96{\mu}mol\;m^{-2}s^{-1}$로 가장 많았으며 검정콩1호는 $12.54{\mu}mol\;m^{-2}s^{-1}$로 가장 적었다. 3. 단파흑은 개체당 잎면적이 $3,968cm^2$로 가장 많고 100립중도 30.5g으로 가장 높은 반면 수량은 149kg/10a으로 가장 낮아 건물축적 효율이 가장 낮았으며, 생육단계별 AGP활성도 가장 낮은 수치를 나타냈다. 4. AGP의 small subunit은 60KD의 single band를 나타냈는데 개화기 이후 AGP의 활성변화와 일치하는 경향을 보였다.

Synthetic Peptide-Based Enzyme-Linked Immunosorbent Assay for Human $\alpha$-Fetoprotein

  • $\alpha$-Fetoprotein(AFP) is a good marker for the detection of several diseases such as hepatocellular carcinoma, gonadal germ cell tumor, gastric tumor, and Down's syndrome. In this study, we developed ELISA, using synthetic peptides corresponding to the epitopes of AFP. Five kinds of peptides were synthesized from AFP to produce antibodies in rats that recognize AFP in human plasma as well as amniotic fluid and do not cross-react with serum albumin. All five kinds of antibodies showed good reactivities with their peptide-keyhole limpet hemocyanin conjugates. Anti-synthetic peptide 1 (R-N-E-Y-G-I-A-S-I-L, 4-13) antibody, in particular, reacted well with AEP as well as synthetic peptide 1-KLH but not with human serum albumin. The binding affinity(Kd) was 2.7$\times$10$^{-9}$M for peptide 1 and 6.8$\times$10$^{-8}$M for AEP. The range for measurement of AFP was 10~1,000 ng/ml. The within-assay and between-assay coefficients of variance(CV) were 4.83% and 10.97%, respectively. In a sample of 31 sera and 33 amniotic fluids, there was a good correlation between AFP values determined in this assay and those in a commercial kit. These results indicate that the antibodies against synthetic peptides corresponding to the epitopes of AFP are highly specific to APP and synthetic peptide-based ELISA would be useful for the measurement of human AFP.
