Purpose: High intake of cholesterol leads to cardiovascular disruption. Estimating the actual intake of cholesterol can be beneficial for nutrition intervention. This research aimed to develop Semi-Quantitative Food Frequency Questionnaire (SQ-FFQ) to estimate cholesterol intake and analyze its reliability and validity. Methods: SQ-FFQ was developed by sorting high cholesterol food items in Indonesian food database and food items' availability. A total of 30 older adults were randomly chosen from Public Health Center in Jagir District, Surabaya, Indonesia to test its validity. Reliability test was done by measuring the same developed SQ-FFQ in one-month period, while validity test was done by comparing SQ-FFQ results with 6-days food record. Statistical analysis used for reliability test was paired t-test, the Intra-class Correlation Coefficient (ICC), and Cronbach's α to measure the internal consistency. Meanwhile, validity of developed SQ-FFQ was analyzed using paired t-test and Bland-Altman. Results: Reliability of 2 administered SQ-FFQs showed a good agreement based on paired t-test analysis (p = 0.200), ICC (0.609), and Cronbach's α (0.757). Strong agreement was found in most of food items, but agreements for egg yolk and fried duck were poor. Significant difference was found between those food items (p = 0.001 vs. p < 0.001, respectively) with mean difference were -25.3 mg and 46.2 mg. Validity of developed SQ-FFQ2 compared to 6-days food diary records also found a strong agreement based on paired t-test and the Bland-Altman analysis. Conclusion: This baseline research provides a reasonably valid and repeatable measure of cholesterol intake estimation that can be widely used in nutrition and public health study, especially in Indonesia. No study has been conducted in Indonesia on the development of tools to estimate the cholesterol intake.

Response surface analysis was used to study dietary ratios of n-3/n-6 fatty acid and P/S to minimize plasma triglycerides, total cholesterol and LDL ${\cdot}$ VLDL-C levels and maximize plasma HDL ${\cdot}$ C levels of rats. Because the dietary components were not statistically independent, they were studied in combinations of two variables. The two-variable combinations were the most useful in locating the desired maximum or minimum plasma triglycerides, total cholesterol and LDL ${\cdot}$ VLDL-C response in terms of the proportions of the dietary components. Response surface contours and three dimensional plots were developed for each plasma lipid response. The contours and three dimensional plots were used to help determine those combinations of the dietary fatty acid ratios that would produce the desired minimum or maximum lpid responses. The statistical analyses indicated that the minimized plasma cholesterol response levels could be attained with a diet consisting of 2.26 n-3/n-6 fatty acid and 2.15 P/S ratios.
Lipid consumption in early life has been implicated with arteriosclerosis in later life. It has been also claimed that lipid contents of serum in early life would reflect coronary atherosclerosis in later life. Present study has been undertaken to look for serum lipid contents of mother and newborn simultaneously to determine the difference of various lipid classes. Lipids were extracted by Folch method. Total lipid was determined gravimetrically; cholesterol by Zak's method, phospholipids by Fiske-Subba-Row's method, and triglycerides by Sugiura's method. Fatty acids were analysed by gas liguid chromatography. Values of the total lipids of mother was significantly higher than those of newborn. HDI-cholesterol/total cholesterol ratio of newborn's was higher than mother's. The amount of saturated fatty acids in the serum cholesteryl ester of newborn was significantly higher than mother's.
Antioxidant vitamin supplementation focuses one's attention on the prevention of age-related diseases. This study was conducted to investigate the antioxidant status and lipid profiles and to look into the antioxidant vitamin supplementation that affects lipid metabolism in 20 elderly non-smoking Korean women (placebo group: n = 6, vitC suppl: n = 7, vitE suppl: n = 7). Age, height, weight, muscle, percent of fat and WHR were not significantly different among the groups, however $\%$ of fat was above $33\%$ and WHR was above 0.9. And blood pressure of the placebo group was 131.7/81.7 (border line hypertension), that of vitamin C supplement was 141.4/87.1 (hypertension) and that of vitamin E supplement was 151.4/92.9 (hypertension). Although nutrient intakes of all groups were poor, antioxidant status (blood vitamins C, E, A, and beta-carotene) and lipid profile (TG, total-cholesterol, VLDL-cholesterol, LDL-cholesterol, HDL-cholesterol) were normal. For nutritional intervention, the vitamin C supplement group received L-ascorbic acid 1,000 mg, and vitamin E supplement group received d-alpha-tocopherol 400IU for 4 weeks, showing the effects of vitamin E supplementation. Response total cholesterol of HDL-cholesterol (T-Chol/HDL) in vitamin E supplement group was significantly decreased from 4.3 to 3.2. And response LDL-cholesterol of HDL-cholesterol (LDL/HDL) in the vitamin E supplement group was also significantly decreased from 2.6 to 1.7. In addition, after the adjustment for plasma lipids (TG, total cholesterol), plasma vitamin A levels in vitamin E supplement group were significantly increased from 7.89 mg/g to 14.91 mg/g. And systolic blood pressure in vitamin E supplement group was significantly reduced. These results suggested that vitamin E supplementation affects the lipid profiles and blood pressure in elderly non-smoking women. So various nutrition programs must be implemented against age-related diseases and further studies are needed regarding sorts and amounts of antioxidant nutrients and supplementation periods.
Little information on the cholesterol content and the fatty acid composition of avian species other than chicken is available. This study was conducted to compare the yolk cholesterol content and the fatty acid profiles of some wild birds maintained in captivity on commercial grain-based chicken diets. The concentration of cholesterol/g of yolk as well as the total yolk cholesterol per egg varied among species. Yolk cholesterol concentration, expressed as mg/g of yolk, was highest in chukar, followed by pheasant, guinea fowl and quail, while total yolk cholesterol in an egg was highest in guinea fowl, followed by pheasant, chuckar and quail. An inverse relationship between yolk cholesterol concentration and egg weight was observed among species with an exception of quail. Although major fatty acids of egg yolk were oleic acid, palmitic acid, linoleic acid and stearic acid in all birds, the composition varied among species. Chukar and quail showed higher oleic acid content than pheasant and guinea fowl, while showing lower linoleic acid. Fatty acids of chukar and guinea fowl eggs were more saturated than those of pheasant and quail. Chukar and especially quail had higher monounsaturated fatty acids (MUFA) than pheasant and guinea fowl; in quail egg 51.6% of total fatty acids were MUFA. Polyunsaturated fatty acids (PUFA), essential fatty acids (EFA) and the ratio of PUFA to saturated fatty acid (P/S ratio) were higher in pheasant and guinea fowl than in chukar and quail. Differences in fatty acid profile of triglyceride (TG) among birds were largely similar to those of total lipid. In comparison to TG, phosphatidyl choline (PC) was low in MUFA while high in saturated fatty acids (SFA), PUFA, P/S ratio and EFA. PC was most saturated in guinea fowl egg yolk, followed by chukar, quail and pheasant. PUFA, P/S ratio and EFA in PC were highest in pheasant followed by chukar, guinea fowl and quail. PE was distinguished from PC by its high contents of stearic acid, eicosapentenoic acid (EPA) and docosahexenoic acid (DHA) while low in palmitic, oleic and linoleic acids. In egg yolk of all birds MUFA was significantly lower in PE than in PC except in quail. Compared to other species, quail had a considerably higher content of MUFA in PE at the expense of SFA and PUFA.

This study was designed to investigate effects of KH-305 on erectile dysfunction in hyperlipidemic rat. KH-305 has been evaluated for antihyperlipidemic and antierectile dysfunction effects on experimental hyperlipidemic rats induced by high cholesterol-diet. After oral administration of KH-305 water extract (50, 100, 200, 300 mg/kg) to hyperlipidemic rats for 8 weeks, the variables including body weight, total cholesterol, HDL-, and LDL-cholesterol levels in serum, the expression of eNOS and nNOS in penis were measured. Also, erectile function was determined by the measurements of intracavernosal pressure (ICP) and maximal arterial pressure (MAP) after electrical stimulation of the cavernosal nerve. Oral administration of KH-305 extracts significantly inhibited the increase of serum total cholesterol and LDL-cholesterol levels and the decrease of serum HDL-cholesterol level in hyperlipidemic rats induced by high fat diet. The penile expression level of the two enzymes (eNOS, nNOS) were increased significantly after oral administration of the KH-305 extracts. Erectile function after 10 volts stimulation was significantly decreased in the hyperlipidemic rat compared with the normal rat, but increased in KH-305 group compared with hyperlipidemic group. These results suggest that KH-305 is effective for erectile dysfunction in hyperipidemia.

This study investigated the effects of polymannuronate feeding on cholesterol levels of serum, liter tissue, and feces in rats. After one week of general diet feeding, four week old S.D. rats were fed basal group, cholesterol diet group (1$\%$ cholesterol), and rolymannuronate diet group (1$\%$ cholesterol,5$\%$ polymannuronate). The total cholesterol levels in the serum and liver tissue was significantly decreased in the polymannuronate diet group. The polymannuronate diet group showed increased amounts of feces, total fecal bile acid and dietary fiber contents compared to basal diet group and cholesterol diet groups. Also, the histology of liver showed serious localized fat drops in the cholesterol diet group, but the polymannuronate diet group showed fewer localized fat drops. These results suggest that feeding of polymannuronate improves the lipid metabolism profile of rats by changing the serum and liver tissue cholesterol levels with increased level of the total bile acid, total dietary fiber and total cholesterol level in the feces.
This study investigated whether dietary factors are more influential factor than other health behavior such as drinking, smoking and exercise on abnormal serum cholesterol level inspite of Korean dietary pattern differences compared to Europeans and Americans. A double case control study model has been used for the study design. One model consisted of high blood cholesterol cases and control. the other model consisted of low blood cholesterol cased and controls. 5.398 sedentary male workers who had taken medical examinations at a university hospital were used as the study subjects. Out of the study subjects, 36individuals with high blood cholesterol cases and 30 individuals with low blood cholesterol cases were selected. For the 66 individual control selection, the individual control selection, the individuals matching method was adopted. The food frequency method was used to collect the data for assessment of the dietary factors. A standardized questionnaire was used to investigate other health behavior. logistic regression analysis was employed to measure the relative importance between the factors considered. There were no statistically significant differences observed in nutrients consumption or other health behavior among the low, normal and high blood cholesterol groups, An overmatching effect had been suspected as the cause of those findings. However, the results of logistic regression analysis to identify the factors influencing high serum cholesterol showed that odd ratios of dietary factors such as tocopherol(3.0) and saturated fatty acid(1.6) were higher than I. I of smoking and 1.2 of drinking. Similar results were also observed incases of low serum cholesterol. The above findings imply that although the dietary pattern is quite different from that of Europeans and America, the dietary factor is still a significant factor for abnormal blood cholesterol in Koreans. Therefore, the dietary risk factor identified in high fat consumption populations are still relevant for the relatively healthy Korean as guideline for preventive health practices. Steroids are a class of compounds with cyclopentane polyhydrophenanthrene as the parent nucleus, and they usually have unique biological and pharmacological activities. Most of the biosynthesis of steroids is completed by a series of enzymatic reactions starting from cholesterol. Synthetic biology can be used to synthesize cholesterol in engineered microorganisms, but the production of cholesterol is too low to further produce other high-value steroids from cholesterol as the raw material and precursor. In this work, combinational strategies were established to increase the production of cholesterol in engineered Saccharomyces cerevisiae RH6829. The basic medium for high cholesterol production was selected by screening 8 kinds of culture media. Single-factor optimization of the carbon and nitrogen sources of the culture medium, and the addition of calcium ions, zinc ions and citric acid, further increased the cholesterol production to 192.53 mg/l. In the 5-L bioreactor, through the establishment of strategies for glucose and citric acid feeding and dissolved oxygen regulation, the cholesterol production was further increased to 339.87 mg/l, which was 734% higher than that in the original medium. This is the highest titer of cholesterol produced by microorganisms currently reported. The fermentation program has also been conducted in a 50-L bioreactor to prove its stability and feasibility.
