Development of Efficient Screening Methods for Resistant Cucumber Plants to Meloidogyne incognita (오이 뿌리혹선충병에 대한 효율적인 저항성 검정법 확립)

  • Root-knot nematodes represent a significant problem in cucumber, causing reduction in yield and quality. To develop screening methods for resistance of cucumber to root-knot nematode Meloidogyne incognita, development of root-knot nematode of four cucumber cultivars ('Dragonsamchuk', 'Asiastrike', 'Nebakja' and 'Hanelbakdadaki') according to several conditions such as inoculum concentration, plant growth stage and transplanting period was investigated by the number of galls and egg masses produced in each seedling 45 days after inoculation. There was no difference in galls and egg masses according to the tested condition except for inoculum concentration. Reproduction of the nematode on all the tested cultivars according to inoculum concentration increased in a dose-dependent manner. On the basis of the result, the optimum conditions for root-knot development on the cultivars is to transplant period of 1 week, inoculum concentration of 5,000 eggs/plant and plant growth stage of 3-week-old in a greenhouse ($25{\pm}5^{\circ}C$). In addition, under optimum conditions, resistance of 45 commercial cucumber cultivars was evaluated. One rootstock cultivar, Union was moderately resistant to the root-knot nematode. However, no significant difference was in the resistance of the others cultivar. According to the result, we suggest an efficient screening method for new resistant cucumber to the root-knot nematode, M. incognita.

  • This study evaluated the in vitro nematicidal activity of Glycyrrhiza uralensis root extracts (GuRE) on Meloidogyne incognita eggs and juveniles. The results showed that treatment of M. incognita eggs with 2.0 mg/mL GuRE for 5 and 10 d resulted in 64.0 and 68.1% hatch inhibition, respectively. Furthermore, the relative mortality of J2 was 96.2% after treatment with 2.0 mg/mL GuRE for 48 h. Changes in the shape of the eggs and juveniles were determined after incubation with 2.0 mg/mL GuRE for 5 d and 48 h, respectively. These preliminary results suggest that GuRE can be used as an environment-friendly bio-nematicide to control root-knot nematodes. In the future, in vivo assays should be conducted using GuRE to ascertain its potential for widespread application as a nematicide.

  • Over 98% of Dendranthema x grandiflorum plants growing under a glasshouse in Gumi, Korea were infested with Meloidogyne hapla. Dendranthema x grandiflorum cv, 'Ardilo' had fewer eggmass (3.3/g root), followed by cv. 'Euro' (6.5), and cv. 'Monarisa' had the most eggmasses(17). The coefficient correlation between plant growth and nematode reproduction(number of eggmass per plant) indicated that nematode reduced chlorophyll content in leaf (r = -0.418, p = 0.059; r = -0.463, p = 0.040). Number of flowers, number of leaves and stem height to flower were positively correlated with flower weight(p < 0.05).

  • 참외는 대부분 플라스틱하우스내에 소형터널을 설치하여 보온하는 형태로 재배되고 있으며, 연작하므로써 뿌리혹선충을 비롯한 각종 토양전염성 병해충의 피해가 많아지고 염류가 집적되는 등 재배상 많은 문제점이 발생되고 있다. 특히 참외를 장기재배하는 성주 등 경북지역에서는 뿌리혹선충으로 인해 생육이 불량하고(박 등, 1995a; 황 등 1999). 과실의 착과율이 떨어지는 등의 피해가 많이 발생되고 있다. (중략)

Survey on Plant-Parasitic Nematodes in Peach Orchards in Gyeongbuk Province (경북지역 복숭아과원에 분포하는 식물기생성 선충의 조사)

  • Plant-parasitic nematodes were surveyed at peach orchards in Gyeongbuk province for three years from 2000 to 2002. Species, density, frequency, Simpson's index, Shannon and Wiener index, and prominence value were analyzed. Plant-parasitic nematodes were isolated from 97 orchards out of 124 orchards. More important plant-parasitic nematode genera on peach were Tylenchorhynchus nudus, Criconemoides informis, Paratylenchus elachistus, Pratylenchus vulnus, and Helicotylenchus digonichus. Aphelenchus sp., Tylenchus sp., Heterodera sp., Scutellonema sp. were found, but were not important. Root-knot nematodes were not found even though they were serious in foreign countried.

  • The study was conducted to investigate the infestation and distribution of plant-parasitic nematodes on Chinese yam (Dioscorea batatas) in Andong, Korea. Root-knot nematodes, Meloidogyne incognita was the most prevalent nematode species which is found from 43 yam fields (81.1%) with high population densities (average of 450 juveniles/$300cm^3$). Other nematodes, root-lesion nematode (Pratylenchus spp.) and pin nematode (Paratylenchus spp.) were less problematic. Density of juveniles (J2) of M. incognita was $10\;J2/300cm^3$ by August, then increased to 274 in September and 624 in October. The highest J2 density was found at the soil depth between 40 and 50 cm ($1,840\;J2/300cm^3$). M. incognita was able to infest yam tuber down to a depth of 70 cm and developed galls outside and brown spots inside. The highest number of females were found at 40-50 cm (79 females/10 cm piece) tuber from the top.

Effect of Entomopathogenic Nematodes on Egg Mass Formation by the Northern Root-knot Nematode, Meloidogyne hapia (곤충병원성 선충이 당근뿌리혹선충의 난낭 형성에 미치는 영향)

  • The entomopathogenic nematodes, Steinernema carpocapsae All strain (ScA), S.glaseri NC strain (SgN) and H. bacteriophora NC 1 strain (HbN), were evaluated for the effects on egg mass formation by the northern root-knot nematode, Meloidogyne hapla in pot experiment using tomato. In the first experiment, 2.5$\times$10$^{5}$ infective juveniles (Ijs) of entomopathogenic nematodes were inoculated to 100 g of the soil infected with ca. 450 Ijs of M. hapla/100 ㎤ in 150 $mell$ container. The number of egg mass was significantly decreased to 9.4-36.5 in ScA, to 5.7-24.7 in SgN and to 11.2-16.0 in HbN treatments compared with 62.5 in M.hapla alone. In the second experiment, ScA and S.carpocapsae Pocheon strain (ScP) and SgN and S.glaseri Dongrae strain (SgD) were treated to 350 g of the soil infected with 100, 200 M.hapla larvae/100 ㎤ in 450 $mell$ container The entomopathogenic nematodes were inoculated at the rate of 2,020 Ijs and 1.6$\times$105 Ijs in 350 g soil. The number of egg mass of M.hapla were significantly decreased in the entomopathogenic nematode treatments compared with M.hapla alone although no differences were observed among Steinernema species, strains, or infection concentrations. Treatments of entomopathogenic nematodes 3 days before M.hapla inoculation were more effective on reduction of egg mass formation than those of 3 days after M.hapla treatments. Growth of tomato was not affected by entomopathogenic nematode treatments.

  • Egg masses of the root- knot nematodes ( Meloidogyne incognita, M. arenaria and M. haply) were exposed to two different regimes of soil moisture (459 and 2459) and temperature ( -2$^{\circ}C$ and 33$^{\circ}C$), quite extreme condition in their natural environment, and their survival rate was compared. Three species did not show any difference in the rate when exposed to either soil moisture for 25 days, with the rate in the range of 8.6% to 10.4%. In response to temperature treatment, however, they differed : the best survival rate was obtained from M. incognita at high temperature ( 33"C) and from M, hapla at low temperature (-2$^{\circ}C$) plot. The third species (M. arenaria) was intermediate in both temperature regimes.imes.

Cell Wall Localization of Two DUF642 Proteins, BIIDXI and TEEBE, during Meloidogyne incognita Early Inoculation

  • The root-knot nematode Meloidogyne incognita infects a variety of plants, including Arabidopsis thaliana. During migration, root-knot nematodes secrete different proteins to modify cell walls, which include pectolytic enzymes. However, the contribution of host cell wall proteins has not been described during this process. The function of two DUF642 cell wall proteins, BIIDXI (BDX, At4g32460) and TEEBE (TEB, At2g41800), in plant development could be related to the regulation of pectin methyl esterification status in the cell walls of different tissues. Accordingly, the expression of these two genes is up-regulated by auxin. BDX and TEB were highly induced during early M. incognita inoculation. Moreover, cell wall localization of the proteins was also induced. The cell wall localization of BDX and TEB DUF642 proteins during M. incognita early inoculation suggested that these two proteins could be involved in the regulation of the degree of pectin methylation during cell separation.
