PETTIS CONDITIONAL EXPECTATION OF CLOSED CONVEX RANDOM SETS IN A BANACH SPACE WITHOUT RNP

  • In this paper we study the existence of conditional expectation for closed and convex valued Pettis-integrable random sets without assuming the Radon Nikodym property of the Banach space. New version of multivalued dominated convergence theorem of conditional expectation and multivalued $L{\acute{e}}vy^{\prime}s$ martingale convergence theorem for integrable and Pettis integrable random sets are proved.

  • RNA granules such as Stress Granules (SG) and P-Bodies (PB) are aggregates of translationally stalled messenger ribonucleoprotein (mRNP) complexes induced by a wide range of stresses. Over the past decade, extensive studies described key components of RNA granules, their molecular interactions and signaling pathways require for their assembly and disassembly. However, researches defining their exact roles under stress conditions have not been performed so far, although several studies suggested their roles in neurodegenerative diseases recently. In this review, we provide an introduction about their basic properties, key components, and the dynamic nature for their assembly.

CRISPR/Cas9 System을 활용한 배스의 불임 유도에 대한 연구 (A Study on the Induction of Infertility of Largemouth Bass (Micropterus salmoides) by CRISPR/Cas9 System)

  • 배스(Micropterus salmoides)는 수생태계에서 최상위단계에 위치하는 생태계교란 어종으로 심각한 담수생태계의 불균형을 초래하고 있다. 배스의 퇴치 및 관리를 위한 다양한 시도를 하고 있지만 효과적인 방안은 없는 상황이므로 배스의 고유한 특성에 기반한 개체군 감소의 효율성을 극대화할 수 있는 방식을 모색하였다. 본 연구에서는 배스의 Transcriptom 분석으로 Unigene contigs는 182,887개, 그리고 정자-난자 인식 단백질인 IZUMO1과 Zona pellucida sperm-binding protein의 유전자에서 CRISPR/Cas9 system을 적용할 최종 Target sequence는 12종을 산출하였다. 각 Target sequence를 인식할 수 있는 12종의 sgRNA를 합성한 후 후속 연구에 사용할 12종의 Cas9-sgRNA ribonucleoprotein (RNP) complex를 제작하였다. 본 연구에서는 차세대염기서열 분석법으로 정자-난자 인식 단백질을 암호화하는 유전자를 탐색하였고, CRISPR/Cas9 system으로 유전자를 편집하여 번식행동은 하지만 수정란을 형성하지 못하는 생식세포를 생산하는 불임개체를 유도하기 위한 조성물 개발 과정을 확립하였다. 그리고 배스와 동일한 수계에 있는 고유 생물종의 서식에는 영향을 미치지 않는 생태교란종 관리 방안으로서의 유용성을 검증하기 위한 후속 연구의 귀중한 기초 자료를 확보하는데 기여했다고 판단된다.

국제기준에 따른 항공정보업무 관련 규정 개선에 관한 연구 (A Study on the Regulation Improvement relevant to Aeronautical Information Services)

  • 항공정보업무란 항행의 안전, 효율성을 위해 책임질 수 있는 범위 내에서 필요한 항공정보 및 자료를 제공하기 위해 설정된 업무를 말한다. 특히 RNAV의 이행으로 인해 항공정보 및 자료의 역할과 중요성은 나날이 증대되고 있고, 이에 따라 향상된 RNP 및 항행시스템이 절실하게 요구되어지는 상황이다. 국제민간항공기구(ICAO)는 항공안전 확보를 위하여 공동으로 지켜야 할 표준 및 권고사항(SARPs)을 준수하도록 의무를 정하고 있다. 따라서 항행시스템에 의해 항공기 운항에 사용되는 항공정보 및 자료 역시, 각 체약국은 국제적으로 사용되도록 설립된 이러한 규정에 따라 각국의 제 규정을 정비하고 적용하여야 하나, 국가마다 처한 환경의 차이로 인해 서로 다른 규정을 적용하고 있는 것이 현실이기에 항공기 운영자로 하여금 커다란 혼란을 야기하고 있으며, 나아가 항공안전에 중대한 영향을 미치기도 한다. 본 연구는 항공기의 안전운항에 직접적으로 영향을 미칠 수 있는 항공정보업무와 관련된 국제민간항공기구 및 미국연방항공청(FAA)의 관련 규정들을 검토하고 이에 대응한 우리나라의 항공정보업무와 관련된 항공법 및 관련규정 비교하여 그 차이점을 보완하고 국제적 수준으로 향상시키는데 도움이 될 수 있는 방안을 제시하고자 한다.

Surface glycoproteins determine the feature of the 2009 pandemic H1N1 virus

  • After the outbreak of the swine-origin influenza A H1N1 virus in April 2009, World Health Organization declared this novel H1N1 virus as the first pandemic influenza virus (2009 pH1N1) of the $21^{st}$ century. To elucidate the characteristics of 2009 pH1N1, the growth properties of A/Korea/01/09 (K/09) was analyzed in cells. Interestingly, the maximal titer of K/09 was higher than that of a seasonal H1N1 virus isolated in Korea 2008 (S/08) though the RNP complex of K/09 was less competent than that of S/08. In addition, the NS1 protein of K/09 was determined as a weak interferon antagonist as compared to that of S/08. Thus, in order to confine genetic determinants of K/09, activities of two major surface glycoproteins were analyzed. Interestingly, K/09 possesses highly reactive NA proteins and weak HA cell-binding avidity. These findings suggest that the surface glycoproteins might be a key factor in the features of 2009 pH1N1.

TNF-${\alpha}$ Up-regulated the Expression of HuR, a Prognostic Marker for Ovarian Cancer and Hu Syndrome, in BJAB Cells

  • Background: Hu syndrome, a neurological disorder, is characterized by the remote effect of small cell lung cancer on the neural degeneration. The suspicious effectors for this disease are anti-Hu autoantibodies or Hu-related CD8+ T lymphocytes. Interestingly, the same effectors have been suggested to act against tumor growth and this phenomenon may represent natural tumor immunity. For these diagnostic and therapeutic reasons, the demand for antibodies against Hu protein is rapidly growing. Methods: Polyclonal and monoclonal antibodies were generated using recombinant HuR protein. Western blot analyses were performed to check the specificity of generated antibodies using various recombinant proteins and cell lysates. Extracellular stimuli for HuR expression had been searched and HuR-associated proteins were isolated from polysome lysates and then separated in a 2-dimensional gel. Results: Polyclonal and monoclonal antibodies against HuR protein were generated and these antibodies showed HuR specificity. Antibodies were also useful to detect and immunoprecipitate endogenous HuR protein in Jurkat and BJAB. This report also revealed that TNF-${\alpha}$ treatment in BJAB up-regulated HuR expression. Lastly, protein profile in HuR-associated mRNAprotein complexes was mapped by 2-dimensional gel electrophoresis. Conclusion: This study reported that new antibodies against HuR protein were successfully generated. Currently, project to develop a diagnostic kit is in process. Also, this report showed that TNF-${\alpha}$ up-regulated HuR expression in BJAB and protein profile associated with HuR protein was mapped.

Thermodynamic Analyses of the Constitutive Splicing Pathway for Ovomucoid Pre-mRNA

  • The ovomucoid pre-mRNA has been folded into mini-hairpins adaptable for the RNA recognition motif (RRM) protein binding. The number of mini-hairpins were 372 for pre-mRNA and 83-86 for mature mRNA. The spatial arrangements are, in average, 16 nucleotides per mini-hairpin which includes 7 nt in the stem, 5.6 nt in the loop and 3.7 nt in the inter-hairpin spacer. The constitutive splicing system of ovomucoid-pre-mRNA is characterized by preferred order of intron removal of 5/6 > 7/4 > 2/1 > 3. The 5' splice sites (5'SS), branch point sequences (BPS) and 3' splice sites (3'SS) were identified and free energies involved have been estimated in 7 splice sites. Thermodynamic barriers for splice sites from the least (|lowest| -Kcal) were 5, 4, 7, 6, 2, 1, and 3; i.e., -18.7 Kcal, -20.2 Kcal, -21.0 Kcal, -24.0 Kcal, - 25.4 Kcal, -26.4 Kcal and -28.2 Kcal respectively. These are parallel to the kinetic data of splicing order reported in the literature. As a result, the preferred order of intron removals can be described by a consideration of free energy changes involved in the spliceosomal assembly pathway. This finding is consistent with the validity of hnRNP formation mechanisms in previous reports.

A systematic mRNA control mechanism for germline stem cell homeostasis and cell fate specification

  • Germline stem cells (GSCs) are the best understood adult stem cell types in the nematode Caenorhabditis elegans, and have provided an important model system for studying stem cells and their cell fate in vivo, in mammals. In this review, we propose a mechanism that controls GSCs and their cell fate through selective activation, repression and mobilization of the specific mRNAs. This mechanism is acutely controlled by known signal transduction pathways (e.g., Notch signaling and Ras-ERK MAPK signaling pathways) and P granule (analogous to mammalian germ granule)-associated mRNA regulators (FBF-1, FBF-2, GLD-1, GLD-2, GLD-3, RNP-8 and IFE-1). Importantly, all regulators are highly conserved in many multi-cellular animals. Therefore, GSCs from a simple animal may provide broad insight into vertebrate stem cells (e.g., hematopoietic stem cells) and their cell fate specification.

  • The end of eukaryotic genomic DNA is capped by a specialized structure called as "telomere" which consists of the repetitive array of nucleotide sequence, TTAGGG, in humans and mice, and a variety of binding proteins. Telomerase is a ribonucleoprotein (RNP) complex responsible for the elongation of telomeres to maintain the genomic integrity, and is composed of telomerase reverse transcriptase (TERT), telomerase RNA component (TERC), and their associated factors regulating the catalytic activity of telomerase. Although it is now apparent that telomerase protects cells from apoptosis via the maintenance of genomic integrity by stabilizing telomeres, our understanding for the physiological role of telomerase is yet far from completion, and emerging evidence suggests that telomerase has additional extratelomeric roles in mediating cell survival and anti-apoptotic functions against various cytotoxic stresses. Here we summarize and discuss how telomerase and telomeres are involved in mediating cellular protection against apoptosis.

Identification of Neuregulin-2 as a novel stress granule component

  • Stress Granules (SGs) are microscopically visible, phase dense aggregates of translationally stalled messenger ribonucleoprotein (mRNP) complexes formed in response to distinct stress conditions. It is generally considered that SG formation is induced to protect cells from conditions of stress. The precise constituents of SGs and the mechanism through which SGs are dynamically regulated in response to stress are not completely understood. Hence, it is important to identify proteins which regulate SG assembly and disassembly. In the present study, we report Neuregulin-2 (NRG2) as a novel component of SGs; furthermore, depletion of NRG2 potently inhibits SG formation. We also demonstrate that NRG2 specifically localizes to SGs under various stress conditions. Knockdown of NRG2 has no effect on stress-induced polysome disassembly, suggesting that the component does not influence early step of SG formation. It was also observed that reduced expression of NRG2 led to marginal increase in cell survival under arsenite-induced stress.
