Phosphate-Induced Rat Vascular Smooth Muscle Cell Calcification and the Implication of Zinc Deficiency in A7r5 Cell Viability

  • The calcification of vascular smooth muscle cells (VSMCs) is considered one of the major contributors for vascular disease. Phosphate is known as the inducer for VSMC calcification. In this study, we assessed whether phosphate affected cell viability and fetuin-A, a calcification inhibitor protein, both which are related to VSMC calcification. Also, VSMC viability by zinc level was assessed. The results showed that phosphate increased Ca and P deposition in VSMCs (A7r5 cell line, rat aorta origin). This phosphate-induced Ca and P deposition was consistent with the decreased A7r5 cell viability (P<0.05), which implies phosphate-induced calcification in A7r5 cells might be due to the decreased VSMC cell viability. As phosphate increased, the protein expression of fetuin-A protein was up-regulated. A7r5 cell viability decreased as the addition of cellular zinc level was decreased (P<0.05). The results suggested that zinc deficiency causes the decreased cell viability and it would be the future study to clarify how zinc does act for VSMC cell viability. The results suggest that the decreased VSMC viability by high P or low Zn in VSMCs may be the risk factor for vascular disease.

Expression and diagnostic application of p12 protein of African swine fever virus by recombinant baculovirus (재조합 baculovirus에 의한 아프리카 돼지콜레라바이러스 p12 단백질의 발현과 진단적 적용)

  • African swine fever (ASF) is an infectious disease of domestic and wild pigs for which there is no vaccine in the world. A proper surveillance of viral activity and a timely response to ASF outbreaks depend upon the rapid diagnosis of ASF viral infection. Internationally prescribed enzyme-linked immunosorbent assay (ELISA) is a fast, sensitive test routinely used in the diagnosis of the ASF. However, inactivated whole ASF virus antigen used in this test is a tedious to prepare and has a risk of outside exposure of infectious virus by laboratory accident during the preparation. An ASF virus noninfectious recombinant antigen is a safe and easily produced alternative antigen for use in diagnostic assay. We have cloned the ORF O61R gene of the ASF virus to generate a recombinant baculovirus producing the p12 protein in insect cells under control of the polyhedrin promoter as non-fusion protein. When used in an indirect ELISA, the p12 antigen showed reactivity with all known ASF positive pig sera but not with negative pig sera. Our results indicated that the p12 protein would be one of alternative antigens for diagnosis of the ASF.

Digital Watermarking for Multi-Level Data Hiding to Color Images (컬러 영상에서 다중-레벨 데이터 은닉을 위한 디지털 워터마킹)

  • Multi-level has advantage to express image in all levels with different images. This paper proposes digital watermarking built-in technique to transform color image to YCbCr color space to guarantee robustness and imperceptibility of the watermark in the various expression of color images, and to hide multi-level data which shows spread spectrum from low resolution to whole resolution for the Y-signal of multi-level. In color signal, Y-signal and low resolution built-in watermark has risk to be visible, but it can guarantee the robustness of watermark in various colors and transformed images. As a result of the experiment, wavelet compression image with built-in watermark showed robustness and imperceptibility of watermark.

Analysis on Statins for The Treatment of Bone Fracture (스타틴계 고지혈증치료제의 골절치료효과에 대한 분석)

  • 3-Hydroxy-3-methylglutaryl coenzyme A reductase inhibitors (Statins) are potent inhibitors of cholesterol biosynthesis. Cholesterol-lowering therapy using statins significantly reduces the risk of coronary heart disease. Various discovery of statins as bone anabolic agents has spurred a great deal of interest among both basic and clinical bone researchers. In-vitro and some animal studies suggest that statins increase the bone mass by enhancing bone morphogenetic protein-2 (BMP-2)-mediated osteoblast expression. Clinical and animal test results of statins focusing on the prevention and treatment of bone fractures was collected. Three independent literature searches were performed by using from January 1, 2002 to September 2008 for clinical and animal test results. Search term included statins, HMG-CoA reductase inhibitors, pleiotropic effects, fracture, osteoporosis and clinical and animal test. No consensus has been reached whether clinical use of statins has beneficial effects on bone health, partly due to lower statin concentrations because of first-pass metabolism by the liver. Experimental use of statins as stimulators of bone formation suggests that they may have widespread applicability in the field of orthopaedics. With their combined effects on osteoblasts and osteoclasts, statins have the potential to enhance resorption of synthetic materials and improve bone ingrowth. In conclusion, The use of statins in the prevention and treatment of bone fractures requires further study. But observational studies suggest that statins for decreasing bone fractures including osteoporosis have to be considered local direct administration like transdermal or subcutaneous type over oral adminstration.

Screening and Characterization of a Novel RNA Aptamer That Specifically Binds to Human Prostatic Acid Phosphatase and Human Prostate Cancer Cells

  • Prostatic acid phosphatase (PAP) expression increases proportionally with prostate cancer progression, making it useful in prognosticating intermediate to high-risk prostate cancers. A novel ligand that can specifically bind to PAP would be very helpful for guiding prostate cancer therapy. RNA aptamers bind to target molecules with high specificity and have key advantages such as low immunogenicity and easy synthesis. Here, human PAP-specific aptamers were screened from a 2'-fluoropyrimidine (FY)-modified RNA library by SELEX. The candidate aptamer families were identified within six rounds followed by analysis of their sequences and PAP-specific binding. A gel shift assay was used to identify PAP binding aptamers and the 6N aptamer specifically bound to PAP with a Kd value of 118 nM. RT-PCR and fluorescence labeling analyses revealed that the 6N aptamer bound to PAP-positive mammalian cells, such as PC-3 and LNCaP. IMR-90 negative control cells did not bind the 6N aptamer. Systematic minimization analyses revealed that 50 nucleotide sequences and their two hairpin structures in the 6N 2'-FY RNA aptamer were equally important for PAP binding. Renewed interest in PAP combined with the versatility of RNA aptamers, including conjugation of anti-cancer drugs and nano-imaging probes, could open up a new route for early theragnosis of prostate cancer.

Pulse Exposure to Ethanol Augments Vascular Contractility Through Stress Response

  • Drinking excessive alcohol has been recognized as a risk factor for hypertension. However, the mechanism by which alcohol intake causes hypertension still remains elusive. We tested the hypothesis that ethanol itself acts as a stress factor on vasculature and indirectly modulates vascular contractility. After end of exposure to 1, 2.5 and 5% ethanol for 45 min, rat aortic strips were subjected to contractile responses, immunoblot for Hsp70 and the measurement of levels of myosin light chain phosphorylation. Exposure to 5% ethanol not only augmented contractions to KCl or phenylephrine, but also increased expression of Hsp70 and the levels of myosin light chain phosphorylation. There were no significant differences in contractions produced by $1\;{\mu}mol/L$ phorbol 12,13-dibutyrate, a protein kinase C activator, whether the tissues were exposed to 5% ethanol or not. This is the first report to show that even short exposure to ethanol has a delayed effect to increase vascular smooth muscle contractility through a modulation of thick filament regulation. It may be a mechanism by which ingestion of alcohol induces hypertension.

Application of Hyperion Hyperspectral Remote Sensing Data for Wildfire Fuel Mapping

  • Fire fuel map is one of the most critical factors for planning and managing the fire hazard and risk. However, fuel mapping is extremely difficult because fuel properties vary at spatial scales, change depending on the seasonal situations and are affected by the surrounding environment. Remote sensing has potential to reduce the uncertainty in mapping fuels and offers the best approach for improving our abilities. Especially, Hyperspectral sensor have a great potential for mapping vegetation properties because of their high spectral resolution. The objective of this paper is to evaluate the potential of mapping fuel properties using Hyperion hyperspectral remote sensing data acquired in April, 2002. Fuel properties are divided into four broad categories: 1) fuel moisture, 2) fuel green live biomass, 3) fuel condition and 4) fuel types. Fuel moisture and fuel green biomass were assessed using canopy moisture, derived from the expression of liquid water in the reflectance spectrum of plants. Fuel condition was assessed using endmember fractions from spectral mixture analysis (SMA). Fuel types were classified by fuel models based on the results of SMA. Although Hyperion imagery included a lot of sensor noise and poor performance in liquid water band, the overall results showed that Hyperion imagery have good potential for wildfire fuel mapping.

  • Prevention is very important in terms of the fact that the percentage of people in their 10s and 20s experience noise-prone hearing is so serious that it can lead to serious hearing later on. This study examines the effect of the hearing loss prevention public advertisement message on the hearing loss prevention intent of the acceptor according to the type of framing(positive/negative) and self-efficacy information provision (provide information/do not provide information). The perceived risk and future time orientation as individual characteristics of individual participants were set as variables that could influence the message effect. In this paper, we propose a message expression method and effective message delivery method in the aspect of message strategy and effect of execution when promoting understanding and interest in hearing loss prevention.

  • Depression is one of the most important psychiatric disorders worldwide. Most depression-related data mining and machine learning studies have been conducted to predict the presence of depression or to derive individual risk factors. However, since depression is caused by a combination of various factors, it is necessary to identify the complex relationship between the factors in order to establish effective anti-depression and management measures. In this study, we propose a methodology for identifying and interpreting patterns of depression expressions using the method of deriving random forest rules, where the random forest rule consists of the condition for the manifestation of the depressive pattern and the prediction result of depression when the condition is met. The analysis was carried out by subdividing into 4 groups in consideration of the different depressive patterns according to gender and age. Depression rules derived by the proposed methodology were validated by comparing them with the results of previous studies. Also, through the AUC comparison test, the depression diagnosis performance of the derived rules was evaluated, and it was not different from the performance of the existing PHQ-9 summing method. The significance of this study can be found in that it enabled the interpretation of the complex relationship between depressive factors beyond the existing studies that focused on prediction and deduction of major factors.

Inhibitory Effect of Fermented Spanish Extract on Inorganic phosphate-induced Vascular Calcification in ex vivo Aortic Rings (발효 시금치 추출물의 무기인산염에 의해 유도된 혈관 석회화 저해 효과)

  • Spinach (Spinacia oleracea L.), a green leafy vegetable, is well known as a functional food due to its biological activities. Vascular calcification is associated with several disease conditions including atherosclerosis, diabetes, and chronic kidney disease (CKD), and is known to raise the risk of cardiovascular diseases related morbidity and mortality. However, there are no previous studies that have investigated the effects of fermented spinach exract (FSE) against aortic and its underlying mechanisms. Therefore, this study investigated the effects and action of possible mechanisms of FSE on inorganic phosphate (PI)-induced vascular calcification in ex vivo mouse aortic rings. PI increased vascular calcification through calcium deposition in ex vivo aortic rings. FSE inhibited calcium accumulation and osteogenic key marker, runt-related transcription factor 2 (Runx2), and bone Morphogenetic Protein 2 (BMP-2) protein expression in ex vivo aortic rings. And, FSE inhibited PI-induced extracellular signal-regulated kinase (ERK) and p38 phosphorylation in ex vivo aortic rings. These results show that FSE can prevent vascular calcification which may be a crucial way for the prevention and treatment of vascular disease association with vascular calcification.
