Efficient Production of Ginger (Zingiber officinale Roscoe) Rhizome by Shoot-Tip Culture

  • High productivity of ginger (Zingiber officinale Roscoe) was obtained from the rhizome produced by shoot-tip culture with Korean native variety, Seosanjong. Seed rhizomes induced by shoot-tip culture were successfully established in the field. The rhizomes induced by both plant or rhizome were higher in emergence rate and faster in days to emergence than those of home seed production. The seed rhizome production induced by shoot-tip culture was two times heavier than that of home seed production. These results suggest that shoot-tip culture might be one of mass propagation methods in seed rhizome of ginger plant.

Habitat Environment and Massive Propagation Method of Rare Species Miscanthus changii Disappeared in Seoul Area (서울지역에서 사라져 가는 장억새(Miscanthus changii)의 서식환경조사 및 대량번식 방안 연구)

  • This study was conducted to investigate the draft data for the habitat environment and massive propagation method of Miscanthus changii, which is classified as rare species. The in-site analysis of the native soil give a indication of pH 5.24~5.48, electric conductivity (EC)0.03~0.02 $dS{\cdot}m^{-1}$, organic matter (OM) 1.13~1.15%, Ca 3.4 $mg{\cdot}kg^{-1}$, K 0.25 $mg{\cdot}kg^{-1}$, Mg 0.53 $mg{\cdot}kg^{-1}$, N 0.23%, P 2.15 $mg{\cdot}kg^{-1}$ showed low and 0.33~9.12 $cmol{\cdot}kg^{-1}$ CEC. Growth and tiller number in rhizome propagation of Miscanthus changii showed that horticulture media (perlite 1+vermiculate 1+peat moss 1) is the most suitable soil to habitat. As for seed propagation, germination percentage is 0% (0/20) at first seeding, 2% (1/50) at second seeding and 2.1% (6/280) at third seeding. The average germination percentage was below 2%. This is the reason why Miscanthus changi disappear at habitat of poor soil environment and low germination. We gained 61 plants that 60 rhizome propagation and 1 by seed propagation, renatural to habitat.

  • Ligusticum chuanxiong has been an important oriental crude drug and cultivated for a long time in Korea. Ligusticum chuanxiong has been vegetatively propagating crop without seed due to its sterility and would be an allodiploid derived from the hybridization of two allied species. Generally, vegetative propagation is more costly per unit propagule than sexual(seedling) propagation. This study was conducted to find out the techniques of mass propagation by stem cutting for reduced seedling cost. When cutted on August 30 with 1st to 2nd internode, its highest rooting percentage was 81 percent. Rhizome yield of plants cutted in 50 percent on September 1 decreased to 9 percent less than control (rhizome propagation). Rooting percentage of rooted cutting plants after field planting on October 25 was the highest being 87 percent. Rhizome yield with planting appeared 127 and 120kg/10a on September 25 and October 25, respectively.

Effects of Different Mulching Materials on Rhizome Yield and Crude Saponin Contents in Anemarrhena asphodeloides Bunge (피복비닐 종류에 따른 지모의 근경수량 및 조사포닌 함량)

  • In this study effect of different mulching materials on yield and crude saponin contents in Anemarrhena asphodeloides Bunge. The results of this study are summarized as follows: The rhizome yield of seed propagation type in transparent PE mulching cultivation was of which increased 16.7% more than those of non-mulching cultivation. The rhizome yield of young-plant propagation type in green PE mulching and transparent PE mulching cultivations were of which increased 29.4% and 26.5%, respectively more than those of non-mulching cultivation. The contents of methanal extract of seed propagation type in transparent PE mulching and black PE mulching cultivations were of increased each 31.0%, 15.4% more than those of non-mulching cultivation. Crude saponin contents of seed propagation type in transparent PE mulching cultivation was increased up to 30.3% compared to those of non-mulching cultivation. However, the crude saponin contents of seed propagation type in green PE mulching and black & white PE mulching cultivations were not affected by the non-mulching cultivation. Crude saponin contents of young-plant propagation type in transparent PE mulching cultivation was increased 24.0% and 15.4% more than those of green PE mulching and black PE mulching cultivations.

On the occurrence and control of the rhizome rot of the common ginger caused by fusarium oxysporum f. zingiberi (생강마름썩음병의 발병분포 및 방제에 관하여)

  • The rhizome rot of ginger, caused by fusarium oxysporum f.zingiberi, gives a great deal of damages to the ginger farming in Korea. Main symptoms to the disease include the yellowing and blighting of the stems as the rotting of rhizomes. The infection rate was 20 percent in Bongdong, Jeonbug, whereas 10 percent in Susan, Chungnam. The rate was higher at the farm where gingers had been cultivated for many years. To control the disease, soil fungicides Dowfum MC-2 and Tolene C-17 were turned out to be more effective than Di-Trapex nad Daconil. Treatment of Benlate to the rhizome pieces, which were cut for propagation, also showed a better result as compared with that of Othocide. Dual treatment of both Dowfum MC-2 or Tolene C-17 to the farm soil and Benlate to the rhizome pieces was recommended for the best chemical control.

Callus induction and plant regeneration of Iris dichotoma Pall. in endangered species

  • Iris dichotoma Pall. is an important endangered plant belonging to the family Iridaceae. A method was developed for the rapid micropropagation of I. dichotoma through plant regeneration from leaf, rhizome, and root explant-derived calli. Leaf, rhizome, and root segments were cultured on Murashige and Skoog (MS) medium supplemented with 2,4-dichlorophenoxy acetic acid (2,4-D; $0-3.0mg{\cdot}L^{-1}$) for callus induction. Callus production was highest at $1.0mg{\cdot}L^{-1}$ 2,4-D, where 73.8% and 45.5% of cultured rhizome and root cuttings, respectively, produced calli. The viable calli were maintained at an induced concentration of 2,4-D ($3.0mg{\cdot}L^{-1}$). They were then transferred to MS medium supplemented with various concentrations of 2,4-D ($0-3.0mg{\cdot}L^{-1}$) in combination with 6-benzyladenine (BA: 0, 1.0 and $3.0mg{\cdot}L^{-1}$) for adventitious shoot regeneration. The addition of a low concentration of 2,4-D into BA-containing medium significantly increased the frequency of shoot regeneration in leaf, rhizome, and root-derived calli. The highest number of adventitious shoots (26.4 per callus) formed at $0.5mg{\cdot}L^{-1}$ 2,4-D and 1.0 mg/l BA. For rooting of the shoots, half- strength MS medium supplemented with different concentrations of indole 3-butyric acid (IBA) $0-3.0mg{\cdot}L^{-1}$ was tested. The optimal results were observed using half-strength MS medium supplemented with $1.0mg{\cdot}L^{-1}$ IBA, on which 98% of the regenerated shoots developed roots with an average of 3.5 roots per shoot within 45 days. The plantlets raised in vitro were acclimatized and transferred to soil with 95% success. This in vitro propagation protocol will be useful for conservation and mass propagation of this endangered plant.

Effects of Peduncle Cutting on Yield and Crude Saponin Content in Anemarrhena asphodeloides Bunge (화경제거가 지모의 수량 및 조사포닌함량에 미치는 영향)

  • In this study, effects of wiling peduncle on production yield and crude saponin content in Anemarrhena asphodeloides Bunge were investigation. The peduncle cutting were conduct on May 30, June 15, June 30, non-cutting of seed propagation type and young plant propagation type in Anemarrhena asphodeloides Bunge. The highest rhizome yield was 276.9g/plant on May 30, cutting puduncle in seed propagation type. The rhizome yield at seed propagation type was increased about 40.8%, compared to that at non-cutting. Crude saponin content in rhizome was 7.50% on May 30, cutting puduncle in seed propagation type. The crude saponin content at seed propagation type was increased about 32.2%, compared to that at non-cutting.

Effects of Plant Growth Regulators on in vitro Propagation of Cymbidium kanran and Cymbidium hybrida (한란 및 심비디움의 기내 증식에 미치는 생장조절물질의 효과)

  • This study was carried out to determine the effects of plant growth regulators on organogenesis from Cymbidium kanran and Cymbidium hybrida. Optimal rhizome formation from Cymbidium kanran was obtained on MS medium with 10 ppm kinetin+2 ppm NAA. and optimal protocorm formation from Cymbidium hybrida was obtained on MS medium with 10 ppm kinetin+0.05 ppm NAA. However, in this study the optimal media for the callus induction from both explants was not identified. Optimal shoot induction from rhizome of Cymbidium kanran was obtained on MS medium with 10 ppm BA+2 ppm NAA and 5 ppm BA+2 ppm NAA. Optimal shoot induction from protocorm of Cymbidium hybrida was obtained on MS medium with 10 ppm kinetin+2 ppm NAA.

Propagation Characteristics of Seed and Rhizome of Green kyllinga(Kyllinga brevifolia var. leiolepsis H.) (파대가리(Kyllinga brevifolia var. leiolepsis H.) 종자형성과 근경(根莖)의 번식특성)

  • In this study, a propagation characteristics of green kyllinga(Kyllinga brevifolia var. leiolepsis H.) was investigated. Two to three rhizomes(1st rhizome) arised from the basal part of main shoot at 4th leaf stage and were grown to extend on the soil surface. Then new shoots(lst tillers) emerged from every node of the 1st rhizome. Second rhizomes also arised from the basal part of lst tiller at the time of 4th leaf openning. With such a regularity, 5th rhizomes and about 688 shoots were produced from one plant during one year-cultivation without competition under the natural condition. The degree of vegetative growth and seed formation was 3 times and 2.5 times higher in rhizomeoriginated plant than in that from seed, respectively. The amount of seed formation and the 1000 seeds weight was highest in one planted on June 1 and Aug. 1, respectively. Short-day treatment of less than 14hr appeared to be necessary for the induction of flowering and it was effective as treated not during germination but since at least 2 leaf stage of green kyllinga. Each shoot individually responded to short-day. When plants were exposed to short-day (9hr, day / 15hr, night) treatment at the stage of 2-3 leaves, more than 7 cycles were required for flowering induction. Bolting in main shoot occurred after emergence of 4 leaves under the short-day condition. Exogenous $GA_3$ slightly accelerated the velocity of bolting only in short-day condition.

  • Alstroemeria (Alstroemeriaceae) is one of the most important cut flowers in international market. But, its characteristics such as low multiplication rates, time consuming process and high risk of carrying viral disease, in vitro propagation techniques based on rhizome meristems culture have been developed. In this study, we conducted this experiment to investigate the effect of hormones and gelling agents on the growth for Alstroemeria in vitro tissue culture. The highest number of shoot and root formation were obtained from the growth medium contains BA 1.0 mg/L and NAA 0.1 mg/L. When 0.25% of gelrite was added up to 50% shoots and roots length were observed compared to other gelling agents. Using these results, it is expected to contribute to the establishment of mass production systems in Alstroemeria.
