HISTOPATHOLOGY AND PERCUTANEOUS ABSORPTION OF TOPICAL FORMULATION CONTAINING NEW CAPSAICIN ANALOG.

  • A new capsaicin analog modified with 4-hydroxyl and alkyl chain of capsaicin was a very potent antiinflammatory analgesic drug and may be clinically useful for those who have rheumatoid arthritis, diabetic neuropathy and cancer. The purpose of this study was to investigate histopathology after short and long term application of poloxamer-based gels, and percutaneous absorption of various topical formulations. Poloxamer-based gel was prepared by cold method using poloxamer 407. The poloxamer gels was applied to dorsal sites of hairless mouse skin during one week or one month for the evaluation of skin irritation. The applied site was then sectioned for histopathologic examination. The topical formulations were also prepared using CMC, HPMC, MC, carbopol and glycerylmono stearate. Skin variation of poloxamer gels was studied using excised hairless mouse, rat, hamster and human penis skin. Franz-type diffusion cells were used far skin penetration of drug against receptor phase filled with about 10$m\ell$ of 0.9% saline solution kept at 32$^{\circ}C$. The concentration of drug was determined by the reverse phased C18, Symmetry HPLC with fluorometeric detector. No skin erythema was observed after dorsal application of poloxamer-based gels for one week or one month. No histopathologic changes was also examined, suggesting no skin toxicity of poloxamer-based gels. The order of flux rate was HPMC > MC ( CMC > poloxamer >> glycerylmono stearate ( carbopol. There was a skin variation of poloxamer gels. The flux rate of poloxamer gels was highest in case of hairless mouse followed by rat, human and hamster skin. 코지산을 함유한 폴록사머 겔 제제의 약물방출 및 피부자극성 (Drug Release and Skin Irritancy of Poloxamer Gel Containing Kojic Acid)

  • Low toxicity, reverse thermal gelation and high drug loading capabilities suggest that poloxamer 407 gels have great potential as a topical drug delivery system. Kojic acid (KA) is an antimelanogenic agent which has been widely used in cosmetics to whiten the skin color. However, it has the drawbacks of skin irritancy due to its acidic pH. Poloxamer gels of different polymer contents were formulated to overcome the problem and compared to the cream type formulations of either w/o/w multiple emulsion cream or o/w type emulsion cream. Using Franz diffusion cells mounted with a synthetic cellulose membrane (MWCO 12,000), drug release characteristics of the formulations were evaluated by the HPLC assay of KA concentration in the receptor compartment of pH 7.4 phosphate buffered saline solutions. Drug release from w/o/w multiple emulsion cream was controlled by oil membrane, showing the apparent zero order release kinetics. The KA release from the poloxamer gels was also controlled by the gel matrix, showing that drug release increased linearly as KA contents increase, but decreased exponentially as the polymer contents increase. In the skin irritancy test, the primary irritancy index(PII) of poloxamer gel base was lower than those of multiple emulsion cream base and o/w cream. Depending on KA contents or polymer contents in the gel. PH values in poloxamer gels were ranged from 1.3 to 2.0, which are interpreted as low or negligible irritation on skin. There was a good correlation between the log value of flux in drug release and PII value in skin irritation. It was possible to conclude that the poloxamer gels containing KA might be a good candidate for an antimelanogenic topical delivery system by virtue of the controlled release of the drug and the reduced skin irritancy.

비가압식 막 공정을 통한 정삼투막 성능 평가 (Evaluation of Forward Osmosis (FO) Membrane Performances in a Non-Pressurized Membrane System)

  • The objective of this study is to develop a novel method for evaluating forward osmosis (FO) membrane performances using a non-pressurized FO system. Basic membrane performance parameters including water (A) and solute (B) permeability coefficients and unique parameter for FO membrane such as the support layer structural parameter (S) were determined in two FO modes (i.e., active layer faces feed solution (AL-FS) and active layer faces draw solution (AL-DS)). Futhermore, these parameters were compared with those determined in a pressurized reverse osmosis (RO) system. Theoretical water flux was calculated by employing these parameters to a model that accounts for the effects of both internal and external concentration polarization. Water flux from FO experiment was compared to theoretical water fluxes for assessing the reliability of those parameters determined in three different operation modes (i.e., AL-FS FO, AL-DS FO, and RO modes). It is demonstrated that FO membrane performance parameters can be accurately measured in non-pressurized FO mode. Specifically, membrane performance parameters determined in AL-DS FO mode most accurately predict FO water flux. This implies that the evaluation of FO membrane performances should be performed in non-pressurized FO mode, which can prevent membrane compaction and/or defect and more precisely reflect FO operation conditions.

Nitrous Oxide를 이용한 흡입진정 (Inhalation Sedation with Nitrous Oxide in Dental Treatment)

  • The usage of nitrous oxide is increased for the anxious patient to dental treatment. There are two methods to induce the sedation during dental treatment. One is sedation with drugs the other no need of drugs. We discussed here about sedation with drugs. The methods of drug administration are oral, intramuscular, intravenous, inhalation. The method of oral administration of drugs are convenient to patient and doctor but poor controllability. Intramuscular method is a parenteral technique that maintains several advantages over the enteral technique. However its pales in comparison to other parenteral technique. Intravenous method represents most effective method of ensuring predictable and adequate sedation in all patients. But it has inability to reverse the action of drugs after they have been injected except some drugs (e.g., narcotics and benzodiazepine). A variety of gaseous agents may be administered by inhalation to produce sedation. In dental practice, the inhalation administration of gas means use of nitrous oxide. There are many advantages of nitrous oxide administration. First, very short latent period and rapid onset of drug action which lead to possible titration of drug concentration. With nitrous oxide, clinical effects may become noticeable as quickly as 15 to 30 seconds after inhalation. Recovery from inhalation sedation is also quite rapid. In out patient dental practice rapid recovery is very important because it permit to discharge the patient without escort and the patient return to their ordinary life without limit. To success the conscious sedation with nitrous oxide, the administrator should be keep the mind that always titration of nitrous oxide concentration during induction and treatment. Careful observation need during treatment to prevent oversedation because the adequate nitrous oxide concentration to patients changed by environmental stress. Always begins with 100% oxygen and ends with 100% oxygen to prevent diffusion hypoxia which rare in clinical practice.

기중 포름알데히드 측정을 위한 확산포집기의 개발 (The Development of New Diffusive Sampler for Formaldehye in Air)

  • To utilize diffuse sampling of formaldehyde in air, a new sampler was designed. A glass fiber filter was impegnated with 2,4-dinitrophenylhydrazine(DNP) and phosphoric acid and mounted 37mm cassette. The formaldehyde vapor was sampled in the dynamic chamber and measured by high performance liquid chromatograph and compared with solid sorbent tube. The results were as follows ; 1. The desorption efficiencies of diffusive sampler between 97.0% and 100%. 2. the sampling rate is constant as in $58{\sim}61.8m{\ell}/min$ when sampling times are between 120 and 360 min. 3. There was a significant relationship between concentrations of diffusive samples and active samples with the coefficient of determination(R2) of 0.92. 4. Desorbed amount of formaldehyde diffusive sampler was increased by high relative humidity. 5. Wheth diffusive samplers were stored at room temperature or at refrigerator there was no statistically significant difference in the accuracy of result. 6. When the diffusive samplers, which collected formaldehyde vapor, were exposed to clean air for three hours, there was no significant loss of formaldehyde due to reverse diffusion. In conclusion, this study suggest that developed diffusive samplers will be a reasonable substitute for the solid sorbent tube for sampling formaldehyde and practical comparative study of developed diffusive sampler should be performed at workplaces of manufacturing industry.

황해저층냉수의 형성에 관한 수치실험 (Numerical Experiment for the formation of the Yellow Sea cold Water mass)

  • 하계에 나타나는 황해저층내수에 간단한 3차원의 육면체 모델을 적용하여, 조석의 혼합작용과 쿠로시오수괴가 황해저층냉수의 형성에 주는 영향을 연구하였다. 본연구에 서는 수치모델의 기술적인 문제 때문에 조석의 혼합작용을 연직혼합계수로 매계변수화 하였다. 또한 연안을 따라 나타나는 수온전선은 조석의 혼합작용에 의해서 형성되도록 하였으며, 황해저층냉수의 남부에 나타나는 수온전선의 형성에는 고온의 쿠로시오수괴 를 고려하였다. 수치실험의 결과, 모델의 상층과 하층에서 수온분포의 영향이 반대로 나타났다. 상층에서는 모델의 중앙부에서, 하층에서는 연안해역에서 각각 상대적으로 고온의 수온분포가 나타났다. 해수의 순환형태 역시 상층과 하층 사이에 반대로서, 상 층에는 시계방향이, 하층에는 반시계방향의 순환이 나타났다.

만응환(萬應丸) 에탄올 추출물의 메티실린 내성 포도상구균에 대한 항균활성 및 내성억제 효과 (Antibacterial Activity and Inhibition of Resistance in Methicillin-resistant Staphylococcus aureus by Maneung-hwan Ethanol Extract)

  • Objectives In this study, we investigated the antimicrobial activity of a 70% ethanol extract of Maneung-hwan (MEH), which is prescribed by practitioners of oriental medicine for use against methicillin-resistant Staphylococcus aureus (MRSA). Methods The antibacterial activity of MEH against MRSA strains was evaluated using the disc diffusion method, broth microdilution method (minimal inhibitory concentration, MIC), checkerboard dilution test, and time-kill test. The mechanism of action of MEH was investigated by bacteriolysis using detergents or ATPase inhibitors Additionally, mRNA and protein expression were investigated by quantitative reverse transcription-polymerase chain reaction and western blot assay, respectively. Results The MIC of MEH was 25~1,600 ㎍/mL against all the tested bacterial strains. We showed that MEH extract exerts strong antibacterial activity. In the checkerboard dilution test, the fractional inhibitory concentration index of MEH in combination with antibiotics indicated synergism or partial synergism against S. aureus. The time-kill study indicated that the growth of the tested bacteria was considerably inhibited after a 24-h treatment with MEH and selected antibiotics. To measure the cell membrane permeability, MEH (3.9 ㎍/mL) was combined with Triton X-100 (TX) at various concentrations N,N-dicyclohexylcarbodimide (DCCD) was also tested as an ATPase inhibitor. TX and DCCD cooperation against S. aureus exhibited synergistic action. Accordingly, the antimicrobial activity of MEH in the context of cell membrane rupture and ATPase inhibition was assessed. Additionally, the expression of genes and proteins associated with resistance was reduced after exposing MRSA to MEH. Conclusions These results suggest that MEH possesses antibacterial activity and acts as a potential natural antibiotic against MRSA.

고속 열 확산에 의한 얕은 접합 형성과 Ti-실리시이드화된 $n^+$ -p 다이오드 특성 분석 (The Formation of the Shallow Junction by RTD and Characteristic Analysis for $n^+$ -p Diode with Ti-silicide)

  • The ultra shallow junction was formed by 2-step RTP. Phosphorus solid source(P$_{2}O_{5}$) was transfered on wafer surface during RTG(Rapid Thermal Glass Transfer) of which process condition was 80$0^{\circ}C$ and 60sec. The process temperature and time of the RTD(Rapid Thermal Diffusion) were 950~105$0^{\circ}C$ during 5~15sec respectively sheet resistances were measured as 175~320$\Omega$/m and junction depth and dopth and dopant surface concentration were measured as 0.075~0.18$\mu$m and 5${\times}10^{19}cm^{4}$ respectively. Ti-silicide was formed by 2-step RTA after 300$\AA$ Titanium was deposited. The 1st RTA (2nd RTA) was carried out at the temperature of $600^{\circ}C$(700~80$0^{\circ}C$) for 30 seconds (10~60 seconds) under N$_2$ ambient. Sheet resistances after 2nd RTA were measured as 46~63$\Omega$/D. Si/Ti component ratio was evaulated as 1.6~1.9 from Auger depth profile. Ti-Silicided n-p junction diode (pattern size : 400$\times$400$\mu$m) was fabricated under the RTD(the process was carried out at the temperature of 100$0^{\circ}C$ for 10seconds) and 2nd RTA(theprocess was carried out at the temperature of 750$^{\circ}C$ for 60 seconds). Leakage current was measured 1.8${\times}10^{7}A/mm^{2}$ at 5V reverse voltage. Whent the RTD process condition is at the temperature of 100$0^{\circ}C$ for 10seconds and the 2nd RTA process condition is at the temperature of 75$0^{\circ}C$ for 60 seconds leakage current was 29.15${\times}10^{9}A$(at 5V).

공기중 유기용제 농도 측정에 있어서 수동식 시료채취기의 성능평가 및 한국산 수동식 시료채취기의 개발에 관한 연구 제 2 부 : 한국산 수동식 시료채취기의 개발 (Evaluation of Commercially Available Passive Samplers and Development of New Passive Samplers Part 2 : Development of New Passive Samplers)

  • A new type of passive samplers were designed and produced by authors. After evaluating the quality of activated carbon by measuring recovery rate of organic vapors and steadiness of sampling rate, activated carbon with 30 - 35 mesh produced by Company S in Korea was selected. In each passive sampler, an amount of 400 mg of the activated carbon was filled in 25-mm cassette and covered by fixed screen (or wire screen with 100 mesh). In addition to the fixed screen, a wind screen (or wire screen with 300 mesh) was also attached at outer face. The sampling rate of the new Korean passive samplers was estimated Conclusions obtained in the study are as follows. 1. Sampling rates of the newly developed Korean passive samplers were affected by sampling time. For n-hexane, sampling rates of 15- and 60-minute samples were 70.92 and 37.45 ml/min, respectively. Sampling rate of both 200- and 450-minute samples was 25.96 ml/min. It is concluded that, when passive samplers are used for measuring organic vapors, samples be collected longer than 60 minutes. 2. Sampling rate of the passive samplers was also affected by airborne concentration of organic vapors. Lower sampling rates were determined at level of 1/2 threshold limit values (TLVs) recommended by the American Conference of Governmental Industrial Hygienists (ACGIH). It is recommended that sampling rate of the passive samplers be obtained at site by measuring concentrations using both the NIOSH Method and passive samplers simultaneously. 3. When the passive samplers, which collected organic vapors, were exposed to clean air for five hours, there was no significant loss of organic vapors due to reverse diffusion. 4. When samples were stored at room temperature ($21.8{\pm}0.7^{\circ}C$) and refrigerator ($3.8{\pm}0.7^{\circ}C$), there was no significant difference in the accuracy of results. For trichloroethylene and n-hexane, accuracies were within 25 % at both temperatures until seven days. However, poor accuracy exceeding 25 % was indicated in toluene from the first day. It is recommended that samples be stored at freezing temperature below $0^{\circ}C$. 5. Sampling efficiency was significantly affected by direction of the passive samplers. Results of samplers facing wind and down, respectively, were compared. Lower amount of organic vapors were collected when the sampler was oriented down. It is recommended that, when air velocity is low in plants, the passive samplers be oriented to the wind. However, when air velocity is high, the passive samplers be oriented down.

가압형 정삼투의 간헐적 운전이 콜로이드 파울링 및 물리세정 효율에 미치는 영향 (Effect of Intermittent Pressure-Assisted Forward Osmosis (I-PAFO) Operation on Colloidal Membrane Fouling and Physical Cleaning Efficiency)

  • 가압형 정삼투(pressure-assisted forward osmosis, PAFO) 공정은 기존의 정삼투(forward osmosis, FO) 공정의 단점인 낮은 수투과도 및 유도용질의 역확산을 극복하여 전체 공정 효율을 향상시킨다. 하지만 가압에 의한 추가적인 수리학적 압력의 작용은 파울링을 가속화 시킨다는 단점이 있다. 본 연구는 PAFO의 간헐적 운전방법인 간헐적 가압형 정삼투(Intermittent pressure-assisted forward osmosis, I-PAFO)의 파울링 저감 가능성을 평가하기 위해 수행되었다. 비교를 위해 FO 및 PAFO를 동시에 운전하여, 세 가지 운전에서의 파울링 거동을 관찰하였다. 파울링 실험을 위한 오염물질로 콜로이드 실리카 입자를 사용하였고, 분리막 및 입자의 정전기적 상호작용 에너지 변화가 파울링 거동에 미치는 영향도 확인하였다. 실험결과, I-PAFO 운전에서, 용액 pH 변화에 관계없이 가압구간, 압력완화 구간에서 각각 PAFO, FO보다 높은 수투과도를 유지하였다. 파울링 실험 후, PAFO에 비해 I-PAFO운전에서 더 적은 수투과도 감소가 관찰되었고, 이로 인해 물리세정 후 향상된 수투과도 회복률 또한 관찰되었다.
