Sensing Characteristics of Tyrosinase Immobilized and Tyrosinase, Laccase Co-immobilized Platinum Electrodes

  • Tyrosinase was covalently immobilized on platinum electrode according to the method we developed for laccase (Bull. Korean Chem. Soc. 2002, 23(7), 385) and p-chlorophenol, p-cresol, and phenol could be detected with sensitivities of 334, 139 and 122 nA/ ${\mu}M$ and the detection limits of 1.0, 2.0, and 2.5 ${\mu}M$, respectively. The response time ($t_{90\%}$) is 3 seconds for p-chlorophenol, and 5 seconds for p-cresol and phenol. The optimal pHs of the sensor are in the range of 5.0- 6.0. This sensor can tolerate at least 500 times repeated injections of p-chlorophenol with retaining 80% of initial activity. In case of tyrosinase and laccase co immobilized platinum electrode, the sensitivities are 560 nA/ ${\mu}M$ for p-phenylenediamine (PPD) and 195 nA/ ${\mu}M$ for p-chlorophenol, respectively. The sensitivity of the bi-enzyme sensor for PPD increases 70% compared to that of only laccase immobilized one, but the sensitivity for p-chlorophenol decreases 40% compared to that of only tyrosinase immobilized one. The sensitivity increase for the bi-enzyme sensor for PPD can be ascribed to the additional catalytic function of the co-immobilized tyrosinase. The sensitivity decrease for p-chlorophenol can be explained by the “blocking effect” of the co-immobilized laccase, which hinders the mass transport through the immobilized layer. If PPD was detected with the electrode that had been used for p-chlorophenol, the sensitivity decreased 20% compared to that of the electrode that had been used only for PPD. Similarly, if p-chlorophenol was detected with PPD detected electrode, the sensitivity also decreased 20%. The substrate-induced conformation changes of the enzymes in a confined layer may be responsible for the phenomena.

  • Ultra Super Early Strength Cement is a material that satisfies these requirements. early hydration heat however, is significant over regular concrete, thus discretion is advised for thermal cracks in accordance with heat generation when constructing a large-scale structures. In addition, the negative point that it is difficult to achieve required strength in a short period of time following rubbing process while retaining workability, the cement is being used conditionally for engineering material and Ultra Super Early Strength Cement for maintenance material for construction doesn't exist. Magnesia Phosphate Cement, which is currently under studies in overseas uses no extra admixture and has strong points of Ultra Super Early Strength as well as favorable construction-ability and adhesive stability to the prototype concrete. These factors stem recognition that it could be used as maintenance material for construction of diverse applicability. In order to provide necessary data to increase practicality of the magnesia phosphate cement for Ultra Super Early Strength Mortar, the study carried out simulate experiment on member of framework to review field applicability.

Amperometric Detection of Some Catechol Derivatives and o-aminophenol Derivative with Laccase Immobilized Electrode: Effect of Substrate Structure

  • [ $DeniLite^{TM}$ ] laccase immobilized Pt electrode was used for amperometric detection of some catechol derivatives and o-aminophenol (OAP) derivative by means of substrate recycling. In case of catechol derivatives, the obtained sensitivities are 85, 79 and $57 nA/{\mu}M$ with linear ranges of $0.6\~30,\;0.6\~30\;and\; 1\~25 {\mu}M$ and detection limits (S/N=3) of 0.2, 0.2 and $0.3{\mu}M$ for 3,4-dihydroxycinnaminic acid (3,4-DHCA), 3,4-dihydroxybenzoic acid (3,4-DHBA) and 3,4-dihydroxyphenylacetic acid (3,4-DHPAA), respectively. In case of OAP derivative, the obtained sensitivity is $237 nA/{\mu}M$ with linear range of $0.2\~15{\mu}M$ and detection limit of 70 nM for 2-amino-4-chlorophenol (2-A-4-CP). The response time $(t_{90\%})$ is about 2 seconds for each substrate and the long-term stability is around 40-50days for catechol derivatives and 30 days for 2-A-4-CP with retaining $80\%$ of initial activity. The optimal pHs of the sensor for these substrates are in the range of 4.5-5.0, which indicates that stability of the enzymatically oxidized product plays a very important role in substrate recycling. The different sensitivity of the sensor for each substrate can be explained by the electronic effect of the sugstituent on the enzymatically oxidized form.

A Review of Clinical Studies about Acupuncture Therapy for Whiplash Associated Disorder (교통사고로 인한 편타 손상의 침 치료에 대한 임상연구의 국내외 동향)

  • Objectives This study aimed to research current trends of acupuncture treatment of whiplash associated disorder (WAD). Methods Clinical studies about acupuncture therapy on WAD were searched in 7 databases. The included studies were classified according to their publication dates, methods, interventions, outcome measures. Interventions were analyzed by accompanied intervention, acupoint, acupuncture retaining time and treatment period. Results 14 articles were included. The number of studies on acupuncture for WAD shows increasing tendency since 1990. The included studies consist of 6 randomized controlled trials (RCTs), 2 non-randomized controlled trials (nRCTs), 2 cohort studies, 1 before and after study, 1 case study and 2 case reports. Most studies used visual analogue scale (VAS) as primary outcome. Conclusions This review demonstrates that further clinical studies need to be carried out under the formal clinical practice guidelines of acupuncture therapy for WAD in order to support clinical treatment objectively.

The Scope of Regenerative Endodontics on Open-Apices in Young Permanent Teeth (영구치의 개방성 치근첨에 대한 재생 근관치료적 의견)

  • Numerous challenges in treating permanent dentition are encountered, especially in treating immature teeth with pulp necrosis. Historically, calcium hydroxide application during long periods of time was used to induce the formation of a calcific barrier across the open apex. In 2004, a new treatment modality for the management of the open apex was introduced. This treatment was named as 'revascularization' and gained acceptance among dentists. The protocol was different from the traditional apexification techniques in that the canal was irrigated and disinfected with a combination of three antibiotics (ciprofloxacin, metronidazole and minocycline). At the next appointment, bleeding was induced and the canal was sealed with MTA. Successful regenerative endodontic treatment of necrotic immature permanent teeth can provide continued root development, increased thickness in the dentinal walls and apical closure. These developments of a functional pulp-dentin complex have a promising impact on retaining the natural teeth, the goal of the dental health care.

Design of a 10-bit SAR ADC with Enhancement of Linearity On C-DAC Array (C-DAC Array내 선형성을 향상시킨 10비트 CMOS SAR ADC 설계)

  • In this paper, CMOS SAR A/D converter 1.8V supply for the design of an A/D converter having an middle speed for the biological signal processing was designed. This paper proposes design of a 10-bit SAR Analog to Digital Converter improving linearity driven by MSB node of C-DAC array divided into 4 equal parts. It enhances linearity property, by retaining the analog input signal charging time at MSB node. Because MSB node samples analog input, it enhances resolution through getting initial input signal precisely. By using split capacitor on C-DAC array, it reduced chip size and power dissipation. The Proposed SAR A/D Converter is fabricated in 0.18um CMOS and measured 7.5 bits of ENOB at sampling frequency 4MS/s and power supply of 1.8V. It occupies a core area of $850{\times}650um^2$ and consumes 123.105uW. Therefore it results in 170.016fJ/step of FOM(Figure of Merit).

Motility and Absorptive Capacity of the Ileum in Acute Hemorrhage (급성실혈시의 회장운동과 흡수기능)

  • The influences of the acute hemorrhage on the intestinal functions were studied in the rabbits subjected to acute bleeding, amounting 1.5-2% of the body weight. The motility and the absorptive capacity of the ileum were compared before and after the bleeding. Transfusion of shed blood was also performed in order to see whether the deteriorations were reversible or not. The tension developed in the direction of the longitudinal axis of the ileum was recorded through an appropriate transducer, and the frequency of the rhythmic contraction was counted throughout the procedure. Test solution, 10ml in amount, was placed in the loop of the ileum, and the samples were drawn at zero time and at 20 minutes. Triplicated procedures were repeated on the same loop;namely, before and after bleeding and after transfusion. The test solution was composed of 200 mg% urea, 218 mEq/l of NaCl and 150 mg% of polyethylene glycol (PEG) No. 4,000 in distilled water. The latter substance was used as a marker substance for the volume change of the loop. The results obtained were as follows; 1. The motility of the ileum suffered little effects by acute hemorrhage. However, minor fluctuations were seen in the frequency of the rhythm, showing a slight tendency of decreasing rhythmicity, and it was reversed by transfusion. 2. Diminution of absorptive capacity of urea was noticed in acute hemorrhage and it was interpreted as the consequence of the secondary effect of the retardation of the active transport mechanism governing the sodium transport 3. Absorption rate of the sodium ion was dropped in the hemorrhage, suggesting the indispensable need of the blood supply. 4. Osmolarity of the luminal fluid remained higher in the case of acute hemorrhage. 5. There was a tendency of retaining more fluid in the intestinal lumen in acute hemorrhage, comparing with that observed prior to the bleeding. 6. The deteriorations in the absorptive capacity were restored by transfusion of shed blood.

Changes in Vitamin C and Minerals Content of Perilla Leaves by Different Cooking Methods (조리에 의한 깻잎의 비타민C 및 무기성분의 변화)

  • The purpose of this study was to investigate the differences in vitamin C and mineral contents in perilla leaves as a result of different cooking methods. The results are summarized as follows. In cutting up the perilla leaves, the vitamin C content decreased by 40% with no significant difference made to the mineral content. Steaming was more effective in retaining vitamin C compared to blanching. With increasing treatment times. decreasing amounts of vitamin C remained. There was no significant change in mineral contents except for potassium. In the case of treating with vinegar or lemon juice, more vitamin C remained than when not treated, and there was little influence on the mineral contents. In the solubilization effect of calcium seasoned with acid, rice vinegar was most effective followed by lemon juice, apple juice, brown rice vinegar, then persimmon vinegar. There was a little or almost no vitamin C remaining in kkaennip kimchi. In terms of storing time, raw seasoned kkaennip kimchi had higher mineral contents than blanched or steamed kkaennip kimchis.

Derivation of Flexural Rigidity Formula for Two-row Overlap Pile Wall (2열 겹침주열말뚝의 휨 강성 산정식 유도)

  • Two-row overlap pile wall, currently under development for use in deep excavations, is a novel retaining structure designed to perform itself as a cutoff wall as well as a high-stiffness wall by constructing four overlapping piles arranged in zigzag manner at a time using a tetra-axis auger. This wall has a relatively complex cross-section, compared with other types of pile wall, which would make it difficult to determine design parameters related to cross-section. In this study, a flexural rigidity equation has been derived by analyzing both theoretically and statistically various wall cross-sections with different pile diameters and overlap lengths. The flexural rigidity equation was found to show the maximum error rate of 3%.

Control of MR Haptic Simulator Using Novel S-chain Model (새로운 S-Chain 모델을 이용한 MR 햅틱 시뮬레이터 제어)

  • Due to difficulty in minimally invasive surgery, training simulator is actively researched. A volumetric deformable organ is created by employing a shape-retaining chain-linked (S-chain) model to realize positioning a human organ model in virtual space. Since the main principle of the S-chain algorithm is that the repulsive force is proportional to the number of chain elements, the calculation time can be increased according to the magnitude of deformation. In this work, the advanced S-chain algorithm is used to calculate the repulsive torque according to spin motion. Finally, haptic architecture was constructed using this S-chain model by incorporating the virtual organ with a real master device, which allows the repulsive force and target position to be transferred to each other. The control performance of S-chain algorithm has been evaluated via experiment.
