The effect of intracellular trafficking of CD1d on the formation of TCR repertoire of NKT cells

  • CD1 molecules belong to non-polymorphic MHC class I-like proteins and present lipid antigens to T cells. Five different CD1 genes (CD1a-e) have been identified and classified into two groups. Group 1 include CD1a-c and present pathogenic lipid antigens to ${\alpha}{\beta}$ T cells reminiscence of peptide antigen presentation by MHC-I molecules. CD1d is the only member of Group 2 and presents foreign and self lipid antigens to a specialized subset of ${\alpha}{\beta}$ T cells, NKT cells. NKT cells are involved in diverse immune responses through prompt and massive production of cytokines. CD1d-dependent NKT cells are categorized upon the usage of their T cell receptors. A major subtype of NKT cells (type I) is invariant NKT cells which utilize invariant $V{\alpha}14-J{\alpha}18$ TCR alpha chain in mouse. The remaining NKT cells (type II) utilize diverse TCR alpha chains. Engineered CD1d molecules with modified intracellular trafficking produce either type I or type II NKT cell-defects suggesting the lipid antigens for each subtypes of NKT cells are processed/generated in different intracellular compartments. Since the usage of TCR by a T cell is the result of antigen-driven selection, the intracellular metabolic pathways of lipid antigen are a key in forming the functional NKT cell repertoire.

  • Natural killer (NK) cells play a crucial role in innate immune system and tumor surveillance. NK cells are derived from $CD34^+$hematopoietic stem cells and undergo differentiation via precursor NK cells in bone marrow (BM) through sequential acquisition of functional surface receptors. During differentiation of NK cells, many factors are involved including cytokines, membrane factors and transcription factors as well as microenvironment of BM. NK cells express their own repertoire of receptors including activating and inhibitory receptors that bind to major histocompatibility complex (MHC) class I or class I-related molecules. The balance between activating and inhibitory receptors determines the function of NK cells to kill targets. Binding of NK cell inhibitory receptors to their MHC class I-ligand renders the target cells to be protected from NK cell-mediated cytotoxicity. Thus, NK cells are able to discriminate self from non-self through MHC class I-binding inhibitory receptor. Using intrinsic properties of NK cells, NK cells are emerging to apply as therapeutic agents against many types of cancers. Recently, NK cell alloactivity has also been exploited in killer cell immunoglobulin-like receptor mismatched haploidentical stem cell transplantation to reduce the rate of relapse and graft versus host disease. In this review, we discuss the basic mechanisms of NK cell differentiation, diversity of NK cell receptors, and clinical applications of NK cells for anti-cancer immunotherapy.

Construction of a Large Synthetic Human scFv Library with Six Diversified CDRs and High Functional Diversity

  • Antibody phage display provides a powerful and efficient tool for the discovery and development of monoclonal antibodies for therapeutic and other applications. Antibody clones from synthetic libraries with optimized design features have several distinct advantages that include high stability, high levels of expression, and ease of downstream optimization and engineering. In this study, a fully synthetic human scFv library with six diversified CDRs was constructed by polymerase chain reaction assembly of overlapping oligonucleotides. In order to maximize the functional diversity of the library, a ${\beta}$-lactamase selection strategy was employed in which the assembled scFv gene repertoire was fused to the 5'-end of the ${\beta}$-lactamase gene, and in-frame scFv clones were enriched by carbenicillin selection. A final library with an estimated total diversity of $7.6{\times}10^9$, greater than 70% functional diversity, and diversification of all six CDRs was obtained after insertion of fully randomized CDR-H3 sequences into this proofread repertoire. The performance of the library was validated using a number of target antigens, against which multiple unique scFv sequences with dissociation constants in the nanomolar range were isolated.

  • Cat vocalizes to communicate with another and express their internal states. The vocal repertoire of the cat is wide and up to 21 different vocalizations have been described in the literatures. But it is more than probable that the repertoire contains more types of vocalizations. An ethogram was created in this paper describing the actual known vocalisations of the domestic cat based on an auditory classification. However, the audiogram allows also a visual classification which can increase the accuracy of vocalization differentiation. The classification can be risky as it is sometimes unclear if different types of vocalizations are produced in different environments or if a unique type of vocalization is used with variation in the acoustic parameters. As an example, isolation calls produced by kittens differ depending on the context. The environment has an important impact on the vocal behaviour and thus feral cats and pet cats vocalize differently. Pet cats are thus able to create an efficient communication with humans thanks to the flexibility of vocalisation behaviours. This review allowed us to create a simple model of the cat vocal repertory.

재즈 보컬의 레퍼토리 편곡특성에 관한 연구 -발라드를 중심으로- (A Study on the Repertoire Arrangement Characteristic of Jazz Vocal -Focused on the Ballads-)

  • 재즈 연주자들이 음반에 많이 녹음한 재즈 스탠더드 곡을 대상으로 레퍼토리의 장르를 분류하여 선정한 곡을 중심으로 다른 버전의 편곡 특성을 연구한다. 레퍼토리 곡은 재즈 스탠더드 원곡에서 블루스, 스윙, 발라드, 비밥, 라틴의 5가지 장르로 분류되며, 대부분 발라드와 스윙이다. 레퍼토리의 곡 분류를 통해 같은 곡이 다른 버전에서 어떻게 다르게 연주되었는지 보기위해 여성 재즈보컬들에 의해 녹음된 음반을 중심으로 각 버전의 특징을 분석한다. 이 연구를 수행하여 각 여성 재즈 보컬들의 녹음된 곡에서 다른 버전의 편곡 특성을 도출한다.

  • 스마트 미디어의 등장은 미디어에 대한 정의와 범위 뿐 아니라 이용의 개념까지 새로운 격변에 몰아넣고 있다. 미디어 이용이 다중화 되는지 아니면 하나의 미디어로 수렴되고 있는지, 또는 이런 미디어 이용 변화가 일상생활에는 어떤 영향을 미치고 있는지 등에 대한 논의가 그것이다. 다중 미디어 이용 현상에 대한 기존 연구와는 달리, 이 연구는 '미디어 레퍼토리'를 이론적 근거로 삼는다. 하지만 많은 기존 연구들은 미디어 레퍼토리에 따른 이용 현실을 결코 완전하게 설명하지 못한다. 즉 레퍼토리 유형을 설정하고 유형별 이용 시간 등이 어떤 차이가 있는가에 대해 언급함으로써 끝을 맺는다. 이 이론의 현실 설명력을 높이기 위해서는 이용시간 등과 같은 정태적인 분석뿐만 아니라 미디어 레퍼토리 유형별로 일상생활이 어떻게 달리 구성되는지 등과 같은 동태적인 분석이 필요하다. 이 연구는 그 동안의 미디어 레퍼토리 관련 연구의 한계를 지적하고, 이에 근거해 미디어 레퍼토리와 일상생활 간의 관계를 검증함으로써 관련 연구의 지평을 넓히고 있다.

  • 본 연구는 채널수의 급격한 증가로 인해 이용 가능성이 증대하고 있는 TV 선호채널 설정기능 이용에 대한 태도 및 의도에 영향을 미치는 변인들을 찾고자 하였다. 이론적인 분석틀로 개혁확산이론, 채널레퍼토리, 채널전환행위, 사회적영향 등을 활용하여 연구가설 및 연구문제를 설정하였다. 일반 시청자 250명을 대상으로 온라인 설문조사를 실시한 뒤 다중회귀분석을 통해 실증적인 분석을 실시하였다. 분석결과, TV 선호채널 설정기능에 대한 태도에는 내재적 개혁성, 상대적장점, 사회적 임계점 등이 유의미한 영향을 미쳤으며, TV 선호채널 설정기능에 대한 이용의도에는 합치성, 관찰가능성, 사회적 임계점 등이 유의미한 영향을 미쳤다.

Impaired Memory in OT-II Transgenic Mice Is Associated with Decreased Adult Hippocampal Neurogenesis Possibly Induced by Alteration in Th2 Cytokine Levels

  • Recently, an increasing number of studies have focused on the effects of CD4+ T cell on cognitive function. However, the changes of Th2 cytokines in restricted CD4+ T cell receptor (TCR) repertoire model and their effects on the adult hippocampal neurogenesis and memory are not fully understood. Here, we investigated whether and how the mice with restricted CD4+ repertoire TCR exhibit learning and memory impairment by using OT-II mice. OT-II mice showed decreased adult neurogenesis in hippocampus and short- and long- term memory impairment. Moreover, Th2 cytokines in OT-II mice are significantly increased in peripheral organs and IL-4 is significantly increased in brain. Finally, IL-4 treatment significantly inhibited the proliferation of cultured adult rat hippocampal neural stem cells. Taken together, abnormal level of Th2 cytokines can lead memory dysfunction via impaired adult neurogenesis in OT-II transgenic.

Immunosignature: Serum Antibody Profiling for Cancer Diagnostics

  • Biomarkers for preclinical diagnosis of cancer are valuable tools for detection of malignant tumors at early stages in groups at risk and screening healthy people, as well as monitoring disease recurrence after treatment of cancer. However the complexity of the body's response to the pathological processes makes it virtually impossible to evaluate this response to the development of the disease using a single biomarker that is present in the serum at low concentrations. An alternative approach to standard biomarker analysis is called immunosignature. Instead of going after biomarkers themselves this approach rely on the analysis of the humoral immune response to molecular changes associated with the development of pathological processes. It is known that antibodies are produced in response to proteins expressed during cancer development. Accordingly, the changes in antibody repertoire associated with tumor growth can serve as biomarkers of cancer. Immunosignature is a highly sensitive method for antibody repertoire analysis utilizing high density peptide microarrays. In the present review we discuss modern methods for antibody detection, as well as describe the principles and applications of immunosignature in research and clinical practice.

  • 번식시기동안 상호 이소종인 노랑턱멧새와 붉은뺨멧새의 song을 충청북도 청원군 강내면에서 녹음하여 분석하였다. 두종의 수컷들을 대상으로 playback실험을 실시하였다. 노랑턱멧새와 붉은뺨멧새가 각각 자연적 그리고 인공적인 song repertoires로 playback을 하였을 때 이웃-낯선개체를 인식하는지의 여부를 조사하였다. 또한 붉은뺨멧새에 대해 전반부의 song과 후반부의 song을 playback하여 이웃-낯선개체의 인식 여부도 조사하였다. 노랑턱멧새는 자연적인 song repertoires로 이웃-낯선개체를 인식하였으나, 인공적인 song에 대해서는 반응하지 않았다. 붉은뺨멧새는 자연적, 인공적, 그리고 song의 전반부를 playback하였을 때 이웃-낯선개체를 구별하였다. 반면에 song의 후반부를 playback하였을 때는 반응하지 않았다.
