Inactive but Dimeric Form of Lipoprotein Lipase in Human Plasma

  • Active lipoprotein lipase (LPL) is known as a noncovalent homodimer of identical subunits, and dissociation of the dimer to a monomeric form renders the lipase inactive. In this study, the oligomerization status of LPL in human and rat plasma was investigated. The LPL activity was barely detectable in the control rat and human plasma. After the injection of heparin, the total lipolytic activity of plasma was rapidly increased, and reached its maximum in 30 min. Changes of the LPL protein correlated well with those of lipolytic activity. The LPL protein that is released by heparin into both human and rat plasma was active and dimeric in the sucrose density gradient ultracentrifugation. In control rat plasma, LPL was inactive, and a great fraction was present as an aggregate. However, the inactive LPL protein in the control human plasma retained the dimeric state, indicating that dimerization can be an entity independent of the catalytic activity of LPL. The released LPL is transported as a complex with lipoproteins in plasma. Lipoprotein profiles, determined by NaBr ultracentrifugation, exhibited typical LDL- and HDL-mammal patterns in humans and rats, respectively, with a smaller amount of the LDL fraction observed in rats. The difference in the lipoprotein profiles might influence the fate of the released LPL in plasma.

The Importance of Nitrogen Release and Denitrification in Sediment to the Nitrogen Budget in Hiroshima Bay

  • The main purpose of this study was to estimate the role of dissolved inorganic nitrogen (DIN) released from sediment and denitrification process in sediment on the nitrogen budget of Hiroshima Bay by means of collecting data on distributions and budgets of nitrogen and phosphorus in the bay, DIN fluxes across sediment-water interface and denitrification rates in the sediments of the same area. The TN : TP and DIN:DIP atomic ratios of the discharged freshwater were about 26 and 21, respectively. The standing stocks in the seawater of the TN : TP atomic ratio varied from 8 to 14 with an annual mean value of 11, while the DIN : DIP atomic ratio varied from 10 to 15 with an annual mean value of 12 in the bay. The residence time of nitrogen and phosphorus were estimated to be about 109 days and 200 days in the bay, respectively. The proportion of DIN released from sediment and denitrification rate to the loading of total nitrogen into Hiroshima Bay were $45\%\;(37\~82\%)\;and\;13\%(0.0\~37\%)$, respectively, and the amount of nitrogen through denitrification process was 6.5 times larger than the outflow of nitrogen from the bay. The results show that DIN released from sediment and denitrification process in sediment play important roles on the nitrogen budget in Hiroshima Bay.

Detection and characterization of excretory/secretory proteins from Toxoplasma gondii by monoclonal antibodies

  • Excretory/secretory proteins (ESP) from Toxoplasma gondii were analyzed to define the function in the penetration process into host cells. Whole ESP obtained at 37$^{\circ}C$ were composed of 15 bands with molecular mass of 110, 97, 86, 80, 70, 60, 54, 42, 40, 36. 30, 28, 26, 22, and 19 kDa. Five ESP of 86, 80, 42, 36, and 28 kDa were reacted with monoclonal antibodies (mAb). named as Tg386 (microheme), Tg485 (surface membrane), Tg786 (rhoptry), Tg378, and Tg556 (both dense granules), respectively. The ESP was released by a temperature-dependent/-independent manner and all at once whenever ready to pour out except Tg786. Each ESP was not exhausted within the parasite but the amount was limited. Tg786 was released continuously with increment, whereas Tg378 and Tg556 were ceased to release after 3 and 4 hr, Dense granular Tg378 and Tg556 were released spontaneously and constitutively before the entry into host cells also. The entry of T. gondii was inhibited by all the mAbs differentially. And the parasite deprived of ESP was inhibited to enter exponentially up to 90.1%. It is suggested that ESP play an essential function to provide appropriate environment for the entry of the parasite into host cells.

  • In a hot water system using solar energy, solar heat is not simply collected by the heat collecting plate, but by heat exchange between the solar collector (flat or vacuum type) and the hot water storage tank. Therefore, the amount of collected solar energy depends on the hot water usage patterns that determine the temperature of the thermal storage tank. Also, if the temperature of the hot water stored in the storage tank exceeds the dangerous temperature during the summer, the heat must be released for safety. If the temperature of the hot water in the storage tank is low, it is necessary to heat by the auxiliary heat source. In this study, three buildings are defined as hotel, swimming pool, and school facilities. And we calculated the released heat energy, auxiliary heat source, and pure storage heat energy based on different hot water usage patterns and installation angle of the solar collectors.

  • Predicting radioactive fission product (FP) behaviors in the reactor coolant system and the containment of a nuclear power plant (NPP) is one of the major concerns in the field of reactor safety, since the amount of radioactive FP released into the environment during the postulated accident sequences is one of the major regulatory issues. Radioactive FPs circulating in the primary coolant loop and released into the containment are basically in the form of gas or aerosol. In this study, a multi-component and multi-sectional analysis module for aerosol fission products has been developed based on the MAEROS model [1,2], and the aerosol transport model has been developed and verified against an analytic solution. The deposition of aerosol FPs to the surrounding structural surfaces is modeled with recent research achievements. The developed aerosol analysis model has been successfully validated against the STORM SR-11 experimental data [3], which is International Standard Problem No. 40. Future studies include the development of the resuspension, growth, and chemical reaction models of aerosol fission products.

A study on the release of nickel and chromium from simulated orthodontic appliances (교정장치로부터의 니켈과 크롬의 유리에 관한 연구)

  • Nickel and chromium are two major metals used in the alloys of most orthodontic appliances. But these metals are known to cause hypersensitivity, dermatitis, and asthma. In addition, a significant carcinogenic and mutagenic potential has been demonstrated for compounds containing these metals. The purpose of this study was to find out how much nickel and chromium was released from orthodontic appliances, and which factors would influence the release. The simulated orthodontic appliances were constructed for a half of a mandibular arch and incubated in $0.05\%$ NaCl solution at $37^{\circ}C$. Nickel and chromium release was quantified with an Inductively Coupled Plasma (ICP) spectroanalyzer. The results were as follows : 1. From simulated orthodontic appliances, nickel was released $9.83-70.0{\mu}g/day$ but the release of chromium was not detectable in Limit of 10ppb. 2. The amount of nickel release was significantly different between the types of appliances. 3. The galvanic condition increased the amount of nickel release, which was not statistically significant. 4. The sand blasting increased the amount of nickel release, which was also not statistically significant.

Estimation of Ammonia Emission with Compost Application in Plastic House for Leafy Perilla Cultivation (시설잎들깨 재배의 퇴비 시용에 의한 암모니아 배출량)

  • BACKGROUND: Concerns have been raised about the impact of recent high concentrations of fine dust on human health. Ammonia(NH3) reacts with sulfur oxides and nitrogen compounds in the atmosphere to form ultrafine ammonium sulfate and ammonium nitrate (PM2.5). There is a growing need for accurate estimates of the amount of ammonia emitted during agricultural production. Therefore, in this study, ammonia emissions generated from the cultivation of leafy perilla in plastic houses were determined. METHODS AND RESULTS: Cow manure compost, swine manure compost, and poultry manure compost each at 34.6 ton ha-1, the amount commonly used by farmers in the field, was sprayed on the soil surface. Just after spraying cow manure compost, swine manure compost, and poultry manure compost, the ammonia was periodically measured and analyzed to be 22.5 kg ha-1, 22.8 kg ha-1, and 85.2 kg ha-1, respectively. The emission factors were estimated at 70.0 kg-NH3 ton-N, 62.8 kg-NH3 ton-N, and 234.1 kg-NH3 ton-N, respectively. Most ammonia was released in the two weeks after application of the compost and then the amount released gradually decreased. CONCLUSION: Therefore, it is necessary to improve the emission factor through a study on the estimation of ammonia emission by type of livestock manure and major farming types such as rice fields and uplands, and to update data on the production, distribution, and sales of livestock manure.

Preparation and Release Characteristics of Dextromethorphan HBr Ion-exchange Albumin Microcapsule (브롬화수소산 덱스트로메토르판의 이온교환 알부민 마이크로캅셀의 제조 및 용출특성)

  • Dextromethorphan HBr (DMP HBr) ion exchange albumin microcapsules were prepared by the interfacial polymerization method. The incorporation of drugs in empty albumin microcapsules was more increased in case of glutaraldehyde (GA) and formaldehyde (FA) than terephthaloyl chloride (TC) as a cross linking agent. The amount of DMP HBr incorporated into empty albumin micorcapsules was augemented with increasing DMP HBr concentration and the amount of empty microcapsules in the incorporation medium. Increasing the salt concentration in the release medium, the release rate and the DMP HBr amount released from microcapsules were increased. The release rates of DMP HBr from microcapsules retarded considerably compared with DMP HBr powder.

A STUDY CONCERNING THE CHARACTERISTICS OF KOREAN NI-TI ALLOY ORTHODONTIC WIRE (국산 Ni-Ti합금 교정용 선재의 특성에 관한 연구)

  • To estimate the characteristics of Korean Ni-Ti alloy orthodontic wire, this study investigated compositions, tensile properties, bending properties, heat treatment effects, and ion releasing degrees, and compared these characteristics to those of the imported Ni-Ti alloy wire. The results obtained are as follows ; 1. Ti and -Ni elements in ORTHOLLOY were in a range showing superelasticity, and there was a little difference in the Ni and Ti contents of ORTHOLLOY as compared with those of SENTALLOY. 2. The results of the tensile test concerning ORTHOLLOY exhibited a superelastic effect, indicating an area of a definite amount of stress in spite of the changes in the range from $2\%\;to\;8\%$ in the strain rate. 3. ORTHOLLOY presented higher load values than SENTALLOY in the same deflection values when the wire was tested in three-point bending. A load range displaying a superelastic effect was 80-l00g, 140-l80g, and 130-200g respectively, in wire diameters of 0.014', 0.016', and 0.018' 4. By heat treatments at $400^{\circ}C$ and at $500^{\circ}C$, a load range showing the effect of superelasticitly was lessened by the duration of the heat treatment time. The superelastic effect was destroyed as a result of the 10 minutes heat treatment at $600^{\circ}C$. 5. The quantity of the Ni ion released from ORTHOLLOY, tended to be greater than the amount of released Ni ion in SENTALLOY. The Co ion released was very little(<0.01ppm) in SENTALLOY and ORTHOLLOY irrespective of the lapse of time. Released Ni ions on the 1st day were at the maximum, and the releasing rate showed plateaus after three days. 6. The surface morphology of SENTALLOY was relatively regular irrespective of the lapse of rime, and the corrosion tendency was not observed. However, the surface morphology of ORTHOLLOY was rather irregular and shelved fitting corrosion after immersion.

  • All liquids contain a small amount of gaseous components and the amount of gases dissolved in a liquid is in accordance with Henry's Law. In a multi-stage thermal-type seawater desalination plant, as the supplied seawater undergoes variations in temperature and pressure in each evaporator, the gases dissolved in the seawater are discharged from the liquid. The discharged gases are carbon dioxide, nitrogen, oxygen, and argon, and these emitted gases are non-condensable. From the viewpoint of convective heat transfer, the evaluation of non-condensable gas released during a vacuum evaporation process is a very important design factor because the non-condensable gases degrade the performance of the cooler. Furthermore, in a thermal-type seawater desalination plant, most evaporators operate under vacuum, which maintained through vacuum system such as a steam ejector or a vacuum pump. Therefore, for the proper design of a vacuum system, estimating the non-condensable gases released from seawater is highly crucial. In the study, non-condensable gases released in a thermal-type seawater desalination plant were calculated quantitatively. The calculation results showed that the NCG releasing rate decreased as the stage comes getting a downstream and it was proportional to the freshwater production rate.
