홍삼 추출액 파우치 제품으로부터 분리동정된 변패세균에 관한 연구 (Studies on Saprogenic Bacteria Isolated from Korean Red Ginseng Extrart Product)

  • 홍삼 추출액 제품의 변패원인균을 분리동정하고 생화학적 특성 및 병원성 유무를 판단하였다. 분리된 5가지의 균주는 각각 Bacillus subtilis, Bacillus sp., Paenibacillus sp., Micrococcus sp., Pseudomonas sp.로 동정되었으며, 이 중 Bacillus subtilis와 Paenibacillus sp., Micrococcus sp.는 홍삼 추출액의 원재료로부터 기인한 내생포자를 형성하는 내열성 균주로 추정되었다. 분리된 군주 대부분은 낮은 염도와 중성 pH 조건에서 잘 자라며, 영양요구성이 단순한 호중온성 세균들이었다. 또한 용혈성과 설사유발성을 나타내지 않았으며, 흰쥐 (rat)를 이용한 피부혈관 투과성 항진작용 실험에서도 음성을 나타내어, 수행한 실험 결과 내에서는 분리된 5균주 모두 병원성이 없는 것으로 판단되었다.

Comparative Study of White, Red, and Black Ginseng Extract on Improves the Learning and Memory Impairments by Increases of Synaptic Protein Expression in Scopolamine-induced Dementia Rats

  • To compare and analyze the improvement effects of white ginseng extract, red ginseng extract, and black ginseng extract on cognitive dysfunction and memory impairment caused by scopolamine in rats. In the cognitive behavioral test, the tendency of the SCOP+B group to overcome the escape time delay induced by scopolamine administration was observed, unlike the SCOP group. The frequency on plat form was significantly increased in the group treated with ginseng extracts compared to the SCOP group. As a result of measuring the duration time on goal quadrant, the time spent in the quadrant was significantly increased in the SCOP+B group compared to the SCOP group. In the hippocampus, the SCOP-treated group significantly decreased the activity of AChE compared to the normal group, but the ginseng extract-treated groups significantly increased it compared to the SCOP group. After sacrificing the rats after the behavioral test, the expression of PSD95 protein in the excised brain was significantly decreased in the SCOP group compared to normal, but it was observed that the SCOP+R and SCOP+B groups were significantly increased compared to the SCOP group. CREB1 protein expression was significantly increased in the SCOP+R group, and the expression of Cdk5 was significantly increased in the SCOP+B group. Ginseng extracts significantly restored the memory damaged by scopolamine suggesting that red ginseng increased the expression of CREB1 and PSD95 proteins, and black ginseng increased the protein expression of Cdk5 and PSD95 to induce memory recovery.

홍삼첨가 방법에 따른 홍삼청국장의 품질특성 (Quality Characteristics of Red Ginseng Cheonggukjang According to Addition Methods of Red Ginseng)

  • 본 연구에서는 홍삼과 청국장을 융합하여 기호도 및 기능적 특성이 강화된 홍삼청국장 개발을 위하여 홍삼엑기스와 홍삼분말 첨가량 및 첨가 방법에 따른 품질특성을 조사하였다. 홍삼엑기스를 첨가하여 청국장을 제조한 결과, 설정된 조건에서는 엑기스의 첨가량이 많을수록 점질물을 생성이 많아지고 환원당 함량과 혈전용해능도 높게 나타났다. 홍삼 제품 규격 사포닌 함량은 홍삼엑기스 4%에서 적합하게 나타났다. 또한 홍삼분말 첨가 청국장은 점질물 생성, 환원당 함량 및 혈전용해능이 엑기스 첨가 청국장과 비슷한 경향으로 나타났고 아미노태질소와 암모니아태질소 함량은 분말 첨가량이 높을수록 점점 감소하는 경향으로 나타났으며 조사포닌 함량은 분말 14% 첨가구가 홍삼성분함유 제품 제조 기준에 적합한 것으로 나타났다. 홍삼첨가 시기를 달리하여 청국장을 제조한 결과, 점질물, 아미노태질소, 암모니아태질소, 환원당 함량 및 혈전용해능은 첨가시기에 따른 차이가 크지 않았으며, 조사포닌 함량은 발효전에 첨가한 구간이 발효후에 첨가한 것보다 더 높은 함량을 나타내었다. 진세노사이드 함량은 첨가시기에 따른 차이는 크지 않았으며 $Rg_{1}$, Re, Rf, $Rh_{1}$, $Rg_{2}$, $Rb_{1}$, Rc, $Rb_{2}$, Rd 및 $Rg_{3}$가 확인되었다. 이상의 결과로 홍삼엑기스 4%를 발효전에 첨가하는 방법이 홍삼융합청국장의 품질특성이 우수하여 최적 조건으로 설정할 수 있었다.

Increase in Insulin Secretion Induced by Panax ginseng Berry Extracts Contributes to the Amelioration of Hyperglycemia in Streptozotocin-induced Diabetic Mice

  • Panax ginseng has long been used as a traditional herbal medicine. More recently, it has received attention for its anti-diabetic and anti-obesity effects in humans and in animal models of type 2 diabetes. In the present study, we tested the hypoglycemic effects of ginseng berry extract in beta-cell-deficient mice and investigated the mechanisms involved. Red (ripe) and green (unripe) berry extracts were prepared and administered orally (100 or 200 mg/kg body weight) to streptozotocin-induced diabetic mice daily for 10 wk. The body weight was measured daily, and the nonfasting blood glucose levels were measured after 5 and 10 wk after administration. Glucose tolerance tests were performed, and the serum insulin levels were measured. The proliferation of beta-cells was measured in vitro. The administration of red or green ginseng berry extract significantly reduced the blood glucose levels and improved the glucose tolerance in beta-cell deficient mice, with the higher doses resulting in better effects. Glucose-stimulated insulin secretion was significantly increased in berry extract-treated mice compared with streptozotocin-induced diabetic control mice. Treatment with ginseng berry extract increased beta-cell proliferation in vitro. Both red berry and green berry extracts improved glycemic control in streptozotocin-induced diabetic mice and increased insulin secretion, possibly due to increased beta-cell proliferation. These results suggest that ginseng berry extracts might have beneficial effects on beta-cell regeneration.

흰 쥐의 턱관절 염증성 통증모델에서 홍삼 및 흑삼추출물의 효과 (Effects of Red or Black Ginseng Extract in a Rat Model of Inflammatory Temporomandibular Joint Pain)

  • 실험동물의 TMJ 내에 포르말린으로 유도한 염증성 통증 모델에서 홍삼 및 흑삼 추출물이 통증 발생과 조절에 미치는 영향을 평가하고자 하였다. 홍삼 혹은 흑삼 추출물을 TMJ 내에 투여한 후 통증 행위반응의 변화를 관찰하였고, 또한 실험동물의 연수를 적출하여 Nrf2 경로의 활성 변화를 단백정량분석법을 활용해서 분석한 결과 먼저, 포르말린을 TMJ에 주입한 결과 안면부 통증행위반응이 유의하게 증가되었다. 이것을 토대로 홍삼 및 흑삼 추출물의 경구 투여한 결과 포르말린에 의해 유도된 실험동물에서 통증행위 반응이 효과적으로 감소되었고, 단회투여보다 반복투여가 포르말린 2차 통증행위반응 경감에 더 효과적으로 나타났다. 또한 홍삼 및 흑삼 추출물의 반복 투여 시 포르말린에 주입에 의해 증가된 연수에서의 Nrf2 단백 발현량을 감소시켰고, 간과 신장의 독성검사에서도 영향을 미치지 않는 것으로 나타났다. 따라서, 본 연구에서 홍삼 및 흑삼 추출물은 간과 신장을 부작용을 나타내지 않으면서 포르말린으로 유도된 안면부 통증과 Nrf2의 발현의 조절에 효과적인 것으로 나타났다.

Rg3-enriched red ginseng extract promotes lung cancer cell apoptosis and mitophagy by ROS production

  • Background: Red Ginseng has been used for many years to treat diseases. Ginsenoside Rg3 has documented therapeutic effects, including anticancer and anti-inflammatory activities. However, the anticancer effect of Rg3-enriched red ginseng extract (Rg3-RGE) and its underlying mechanisms have not been fully explored. We investigated whether Rg3-RGE plays an anti-tumor role in lung cancer cells. Methods: To examine the effect of Rg3-RGE on lung cancer cells, we performed cell viability assays, flow cytometry, western blotting analysis, and immunofluorescence to monitor specific markers. Results: Rg3-RGE significantly inhibited cell proliferation and induced mitochondria-dependent apoptosis. Furthermore, Rg3-RGE also increased expression of mitophagy-related proteins such as PINK1 and Parkin. In addition, treatment with Rg3-RGE and mitophagy inhibitors stimulated cell death by inducing mitochondria dysfunction. Conclusions: Rg3-RGE could be used as a therapeutic agent against lung cancer.

Antiallergic and Antipsoriatic Effects of Korean Red Ginseng

  • Antiallergic and antipsoriatic effects of korean Red Ginseng (KRG, steamed root of panax ginseng C.A. Meyer, Family Araliaceae) were measured. Orally administered KRG water extract potently inhibited passive cutaneous anaphylaxis (PCA). KRG water extract also showed the potent inhibition in oxazolone-induced mouse dermatitis, and suppressed mouse ear swelling by $39\%$ at 16 days at a dose of $0.1\%$. KRG water extract reduced the levels of mRNA of cyclooxygenase (COX)-2, $IL-1\beta$, $TNF-\alpha$ and $INF-\gamma$ increased in oxazolone-applied mouse ears, however, did not inhibit that of IL-4. KRG water extract also inhibited iNOS and COX-2 mRNA expression level of RAW264.7 cell induced by lipopolysaccharide. Based on these findings, we suggest that KRG can improve atopic and contact dermatitis by the regulation of $ IL-1\beta$ and $TNF-\alpha$ produced by macrophage cells and $interferon-\gamma$ produced by Th1 cells.

Antiviral effects of Korean Red Ginseng on human coronavirus OC43

  • Background: Panax ginseng Meyer is a medicinal plant well-known for its antiviral activities against various viruses, but its antiviral effect on coronavirus has not yet been studied thoroughly. The antiviral activity of Korean Red Ginseng (KRG) and ten ginsenosides against Human coronavirus OC43 (HCoV-OC43) was investigated in vitro. Methods: The antiviral response and mechanism of action of KRG extract and ginsenoside Rc, Re, Rf, Rg1, Rg2-20 (R) and -20 (S), Rg3-20 (R) and -20 (S), and Rh2-20 (R) and -20 (S), against the human coronavirus strain OC43 were investigated by using plaque assay, time of addition assay, real-time PCR, and FACS analysis. Results: Virus plaque formation was reduced in KRG extract-treated and HCoV-OC43-infected HCT-8 cells. KRG extract decreased the viral proteins (Nucleocapsid protein and Spike protein) and mRNA (N and M gene) expression, while increased the expression of interferon genes. Conclusion: KRG extract exhibits antiviral activity by enhancing the expression of interferons and can be used in treating infections caused by HCoV-OC43.

백삼(白蔘)과 홍삼(紅蔘)이 포함된 이중탕(理中湯)의 마우스 대식세포 내 hydrogen peroxide 생성에 미치는 영향 (Effects of Red Ginseng-Ejung-tang and White Ginseng-Ejung-tang Water Extract on Hydrogen Peroxide Production in RAW 264.7 Cells)

  • The purpose of this study is to investigate whether the intracellular hydrogen peroxide productions of mouse macrophage RAW 264.7 are modulated by Red Ginseng-Ejung-tang water extract (ER) and White Ginseng-Ejung-tang water extract (EG). Red Ginseng-Ejung-tang were composed of Red Ginseng, Atractylodes rhizome white, Zingiberis Rhizoma Siccus, and Glycyrrhizae Radix. White Ginseng-Ejung-tang were composed of White Ginseng, Atractylodes rhizome white, Zingiberis Rhizoma Siccus, and Glycyrrhizae Radix. The intracellular hydrogen peroxide productions were measured by dihydrorhodamine 123 assay with spectrofluorometer (excitation 485 nm; emission 535 nm). For 4, 20, 24, 44, 48, 68, and 72 h incubation, ER significantly increased hydrogen peroxide productions of RAW 264.7 at the concentration of 25, 50, 100, and $200{\mu}g/mL$ (P <0.05). EG for 4, 20, 24, 44, and 48 h incubation significantly increased hydrogen peroxide productions of RAW 264.7 at the concentration of 25, 50, 100, and $200{\mu}g/mL$ (P <0.05). For 68 and 72 h incubation, EG at the concentration of 50, 100, and $200{\mu}g/mL$ significantly increased hydrogen peroxide productions in RAW 264.7 (P <0.05). These results suggest that ER and EG have the immune-enhancing properties related with their increasing effects on the intracellular hydrogen peroxide production of macrophage.
