Dipenyleneiodonium Induces Growth Inhibition of Toxoplasma gondii through ROS Induction in ARPE-19 Cells

  • Based on the reactive oxygen species (ROS) regulatory properties of diphenyleneiodonium (DPI), we investigated the effects of DPI on host-infected T. gondii proliferation and determined specific concentration that inhibit the intracellular parasite growth but without severe toxic effect on human retinal pigment epithelial (ARPE-19) cells. As a result, it is observed that host superoxide, mitochondria superoxide and $H_2O_2$ levels can be increased by DPI, significantly, followed by suppression of T. gondii infection and proliferation. The involvement of ROS in anti-parasitic effect of DPI was confirmed by finding that DPI effect on T. gondii can be reversed by ROS scavengers, N-acetyl-L-cysteine and ascorbic acid. These results suggest that, in ARPE-19 cell, DPI can enhance host ROS generation to prevent T. gondii growth. Our study showed DPI is capable of suppressing T. gondii growth in host cells while minimizing the un-favorite side-effect to host cell. These results imply that DPI as a promising candidate material for novel drug development that can ameliorate toxoplasmosis based on ROS regulation.

Effects of Dietary Xanthophylls and See Weed By-Products on Growth Performance, Color and Antioxidant Properties in Broiler Chicks (Xanthophylls과 해조 부산물 첨가 급여가 육계의 사양성적, 육색 및 항산화 특성에 미치는 영향)

  • This experiment was conducted to investigate the effects of dietary pigment sources on the performance, color and antioxidant properties in broiler chick. Experimental diet was formulated to have isocalories and isonitrogen during the experiment period. Total xanthophylls content in the experimental diet was formulated to have 30ppm. Experimental trials were done for five weeks with six treatment groups; T1 (Control), T2 (Olo Glo, natural yellow pigment), T3 (Kern Glo, natural red pigment), T4 (canthaxanthin, synthetic red pigment), T5 (asthaxanthine, natural red pigment), and T6 (seaweed by-products). Body weight gain and feed intake were significantly lower (p<0.05) in T6 group than in other treatments. Mortality was lower in T2, T3 and T4 than in control, but higher (p<0.05) in T5 and T6. The sources of pigments did not have any effects on the dressed carcass and abdominal fat pad (p>0.05). The gizzard weight was significantly lower in T6 (p<0.05) than in others. Pigmentation of leg skin was significantly lower (p<0.05) in control and T6. Effects of dietary pigments was greater with red pigments than with yellow pigments, and those were also greater with natural pigments than with synthetic ones. The peroxide value (POV), thiobarbituric acid reactive substances (TBARS) and pH values of chicken meat were increased (p<0.05) in all treatments at 12 day storage, and was higher (p<0.05) in pigments supplementation group. No differences of CIE L$\^$*/(lightness) and b$\^$*/(yellowness) were not found by storage days and xanthophylls sources. The a$\^$*/(redness) after 12 day storage was significantly (p<0.05) decreased in all treatments, but those of T4 and T5 were higher than those of others. These results showed that feeding of xanthophylls sources to chick could improve color intensity and inhibit lipid oxidation of leg meat.

  • This study was performed to establish the precise and rapid method to distinguish croakers through the pigment analysis of colored imported white croakers for adultration. We surveyed the coloring behaviors, extraction test by water and organic solvent and using pigments such as targeting, curcumine, and azo dye products. The pigment of yellow croaker is not stained on wet cloth or tissue which is rubbed on epidermis of yellow croaker and was not eluted in water extraction test, while adulterated pigments were easily extracted by water and acetone, but edible diluted yellow, Yellow No. 4 and Yellow No. 5 were not extracted. Reactive pigment was detected easily by extraction with water and dispersed pigment was also detected by extraction test. As a result of discoloring characteristics of carotene having similar structure to yellow croaker and azo dye by oxidation and reduction, azo dyes were not discolored by oxidation with sodium percarbonate or peracetic acid but that were discolored by oxidation with Fenton reagent after 1hr and by hypochlorite promptly. On the other hand, carotenes were not discolored by sodium precarbonate and Fenton reagent but discolored by sodium hypochlorite after 2 hr and by peracetic acid promptly. Azo dyes were discolored by reduction with sodium hydrosulfite and sodium carbonate but carotenes were not discolored by these reagents. This discoloring test was applicable to detect adulterated pigments and other marine product.

  • The reactive oxygen species (or free radicals) generated by ultraviolet radiation cause damage on cellular components and pigment of skin. The aim of this study was to investigate the skin-whitenig effect of Okyong-san. Inhibitory effects of okyong-san extracts on melanin synthesis were studied. Namely, UV-absorbing ability, free radical scavenging activity and tyrosinase inhibitory activity of okyong-san extracts were investigated. As a result, the extracts of okyong-san were found to inhibit the activity of tyrosinase and they showed an absorbance in the UV-B region and UV-C region. We also observed that extracts of okyong-san had free radical scavenging activity.

A Comparative Study of Recognition Rate of Color QR Code Printed on Tyvek and Cotton Material

  • This purpose of this study to analyze effect material properties have on change in QR code recognition rate according to change of materials by comparing recognition rate of color QR code. QR code applied to textile materials has the advantage of being washable and being applicable to lost child prevention goods or clothes or a person with dementia through record of information relating to the material or input of additional information, differently from QR code printed on the conventional paper. An effective method of entering QR code in textile materials is Digital Textile Printing(DTP), that facilitates printing by rapidly applying diverse information, and small quantity production. It is possible to tailor various QR codes according to use. Regarding samples to use, cotton material used in clothing products and Tyvek material recently applied to clothing and related products were selected. Reactive dyes were used for cotton, pigment was used for Tyvek, and QR code was printed with an inkjet printer by direct printing method. Printing methods and surface textures are different between cotton and Tyvek. It was revealed that consequent print results and results of recognition rate were different. Regarding color to be printed, 2015 S/S - 2017 S/S color presented by Pantone was used. Color combination affected recognition rate of color QR code. Understanding color combination, material properties and print characteristics may be helpful in increasing recognition rate of color QR code, and may contribute to usability of color QR code applied to textile materials in the future.

Biological Characterization of the Chemical Structures of Naturally Occurring Substances with Cytotoxicity

  • Screening for the cytotoxicity from plant origin is the first stage for anti-cancer drug development. A variety of terpenoids with exomethylene, epoxide, allyl, $\alpha,\beta-unsaturated$ carbonyl, acetylenes, and $\alpha-methylene-\gamma-lactone$ induces apoptosis and/or differentiation as well as cytotoxicity through the ROS signal transduction pathways. These are found among monoterpenes, sesquiterpenes, triterpenes, flavonoids, coumarins, diarylheptanoids, and even organosulfuric compounds. The most essential characteristics of natural cytotoxic substances is to possess the strong electrophilicity that is susceptible to nucleophilic biomolecules in the cell. Thiol-reductants and superoxide dismutase can block or delay apoptosis. Thus, ROS and the resulting cellular redox-potential changes can be parts of the signal transduction pathway during apoptosis. Disturbance of the balance of oxireduction by the pigment of natural quinones also caused the induction of the differentiation and apoptosis. Saponins with the cytotoxicity are restricted to their monodesmosides, rather than to bisdesmosides. Those saponins exhibited calcium ion-mediated apoptosis in addition to cytotoxicity whereas they showed also differentiation without extracellular calcium ion. The properties on cytotoxicity, apoptosis, and differentiation were assumed to depend on resultant oxidative stress to the cells. In this review, we describe a spectrum of cytotoxic compounds with various action mechanisms.

Structure of Epithelium and Gland cells in a Korean leech, Whitemenia edentula (한국산 작은 말거머리 (Whitemenia edentula)의 상피조직과 선세포의 구조적 특성)

  • Dorsal and ventral epithelium of leech, Whitemenia edentula, were observed, using both light and electron-microscope. Results are as follows. Epithelia are composed of simple columnar or irregularly-shaped epithelium, the transveral folds were seen in the cross-sectioned dorsal and ventral epithelia, but only longitudinal folds(depth $500{\mu}m$, width $350{\mu}m$) were discovered in the ventral epithelium. As to muscular tissue under the dorsal epithelium of Whitemenia edentula, that of longitudinal muscle layer is thicker and more developed than that of circular muscle layer. Seven gland cells (type-A, B, C, D, E, F and H cells) and one pigment cell (type-G cell) were discovered in the whitemenia edentula. Those were identified as three kinds of the neutral mucopolysaccharide granoles (type-A, B and F cells), two kinds of acidic mucopolysaccharide granules (type-C and D cells), and two kinds of non-reactive granules respectively (type-E and F cells). The glanules of the type-A and type-B gland cells ae electronly high-dense, and surrounded by the muscular tissue. As to the distribution of gland cells, the type-A, type-B, type-C, type-D, type-I and type-H gland cells were discovered only between the connective tissue and the circular muscle layer, while type-F gland cells were discovered only between tile circular muscle layer and the longitudinal muscle layer.

Induction of Oxidative Stress by Hexavalent Chromium in Human Bronchial Epithelial Cells (BEAS-2B) (배양 기관지 상피세포(BEAS-2B cells)에서 6가 크롬에 의한 산화적 스트레스)

  • Chromium compounds are widely used in diverse industries including pigment manufacturing, painting, metal plating and leather tanning. With the wide uses of chromium, various adverse effects of the compounds on the environment and human health have been reported. Among them, hexavalent chromium [Cr (VI)], which is a carcinogenic heavy metal, has been widely studies. Epidemiological investigations have shown that respiratory cancers had been found in workers who had been occupationally exposed to Cr (VI). In this study, cell toxicity and induction of reactive oxygen species (ROS) by Cr (VI) (1, 2, 4, $8{\mu}M$) in cultured human bronchial epithelial cells were investigated. Exposure of the cells to Cr (VI) led to cell death, ROS increase, and cytosolic caspase-3 activation. The ROS increase was related with the decreased level of GSH. Chromatin condensation and fragmentation were occurred by Cr (VI) when evaluated by DAPI staining or agarose gel electrophoresis of the extracted DNA. Expression of ROS related genes including glutathione S-transferase, heme oxygenase-1, metallothionein were significantly induced in Cr (VI) treated cells. This result suggests the toxicity in cultured cells by Cr (VI) was expressed through the apoptotic process with ROS induction.

Inhibition of Invasion and Induction of Apoptosis by Curcumin in H-ras-Transformed MCF10A Human Breast Epithelial Cells

  • Curcumin, a dietary pigment in turmeric, posseses anti-carcinogenic and anti-metastatic properties. The present study was conducted to study in vitro chemopreventive effects of curcumin in transformed breast cells. Here, we show that curcumin inhibits H-ras-induced invasive phenotype in MCF10A human breast epithelial cells (H-ras MCF10A) and downregulates matrix metalloproteinase (MMP)-2 dose-dependently. Curcumin exerted cytotoxic effect on H-ras MCF10A cells in a concentration-dependent manner. Curcumin-induced cell death was mainly due to apoptosis in which a prominent downregulation of Bcl-2 and upregulation of Bax were involved. We also suggest a possible involvement of caspase-3 in curcumin-induced apoptosis. Curcumin treatment resulted in the production of reactive oxygen species (ROS) in H-ras MCF10A cells. Apoptotic event by curcumin was significantly inhibited by pretreatment of an antioxidant N-acetyl-$_L$-cysteine (NAC), suggesting redox signaling as a mechanism responsible for curcumin-induced apoptosis in H-ras MCF10A cells. Taken together, our results demonstrate that curcumin inhibits invasion and induces apoptosis, proving the chemopreventive potential of curcumin .

Effects of Submersion Aging in Chilled Water on Lipid Oxidation, Myoglobin Oxidation and Purge Loss of Vacuum-Packed Hanwoo Meat (진공포장 한우육의 냉수 침지 숙성이 지방산화, 육색소 산화 및 포장감량에 미치는 영향)

  • Efficacy of submersion aging in chilled water of vacuum-packed Hanwoo beef was investigated. At 24 hours post-mortem, the somimemebranosus portion of Hanwoo carcass was excised and sliced (2.5cm, thickness). After beef core samples(6$\textrm{cm}^2$${\times}$2cm) were prepared and vacuum packed, the samples were randomly assigned to the four treatments which were conventional refrigerator at 4$^{\circ}C$(control), conventional refrigerator at 1$^{\circ}C$(T1), submersion in chilled water at 4$^{\circ}C$(T2) and submersion in chilled water at 1$^{\circ}C$(T3). Samples were stored for 3, 7, 10 and 14 days to measure meat color(CIE L*, a*, b*), deoxymyoglobin, oxymyoglobin and metmyoglobin percentage, thiobarbituric acid reactive substance (TBARS) value, purge loss(%) and total heam pigment content. CIE a* value of samples was not significantly changed by treatment during the storage. However, metmyoglobin content of T2 samples was significantly (p<0.05) lower than those of control and T1,T3 during storage. Also, TBARS value of T3 samples was significantly (p<0.05) lower than those of control and T2 samples. T3 showed the lowest purge loss%, whereas control remarked the highest purge loss(%). Total heam pigment of all samples were not significantly changed during the storage. These results suggested that submersion aging in chilled water could keep the myoglobin stability and reduce lipid oxidation and purge loss of vacuum packed beef during storage.
