Effects of baicalein on hydrogen peroxide productions in RAW 264.7 mouse macrophages stimulated by poly-IC and lipoteichoic acid (바이칼레인(baicalein)이 poly-IC와 lipoteichoic acid로 자극된 마우스 대식세포 RAW 264.7의 hydrogen peroxide 생성에 미치는 영향)

  • Objectives : The aim of this study was to investigate the effect of baicalein (BA) on the production of hydrogen peroxide and nitric oxide (NO) in RAW 264.7 mouse macrophages stimulated with polyinosinic-polycytidylic acid (poly-IC) and lipoteichoic acid. Methods : RAW 264.7 co-stimulated with poly-IC and lipoteichoic acid were incubated with baicalein at concentrations of 25 and 50 μM. Incubation time is 16 h, 18 h, 20 h, 22 h, and 24 h. After incubation, The production of hydrogen peroxide in RAW 264.7 was measured with dihydrorhodamine 123 assay. Chrysin was used as a comparative material. NO production was evaluated by griess assay. Results : For 16 h, 18 h, 20 h, 22 h, and 24 h incubation, BA at the concentration of 25 and 50 μM significantly inhibited the production of hydrogen peroxide in RAW 264.7 stimulated by poly-IC and lipoteichoic acid (p <0.001). In details, production of hydrogen peroxide in 'poly-IC and lipoteichoic acid'-stimulated RAW 264.7 treated for 16 h with BA at concentrations of 25 and 50 μM was 82.36% and 77.24% of the control group treated with poly-IC and lipoteichoic acid only, respectively; the production of hydrogen peroxide for 18 h was 83.15% and 77.91%, respectively;production of hydrogen peroxide for 20 h was 82.88% and 77.82%, respectively; production of hydrogen peroxide for 22 h was 83.27% and 78.17%, respectively; production of hydrogen peroxide for 24 h was 83.54% and 78.35%, respectively. Additionally, BA at the concentration of 50 and 100 μM significantly inhibited NO production in lipoteichoic acid-induced RAW 264.7 (p <0.001). Conclusions : BA might have anti-oxidative activity related to its inhibition of hydrogen peroxide production in 'poly-IC and lipoteichoic acid'-stimulated RAW 264.7 macrophages.

Study on Biological Effect of Multi-Herbal Drug KOCO-Pl on Mouse Macrophage Raw 264.7 Cells (마우스 대식세포(Raw 264.7)에 대한 한약조성물 KOCO-P1의 세포활성 연구)

  • Objectives : The purpose of this study is to investigate the biological Effect of multi-herbal drug 'KOCO-P1' on mouse macrophage Raw 264.7 cells. Methods : Multi-herbal drug 'KOCO-P1' was composed of Ginseng Radix, Astragali Radix, Polygonati Rhizoma, Liriopis Tuber, and Scrophulariae Radix. Cytotoxicity and cytoprotective activity of K0C0-P1 was verificated by MTT assay. And antioxidative effect of K0C0-P1 against EtOH, Nicotine was inspected by Hydroperoxide assay. Results : K0C0-P1 showed no cytotoxicity on RAW 264.7 cells for 24, 48, 72 hours. KOCO-P1 at 200, 100, and 50 ug/mL reduced the production of H202 in Raw 264.7 cells by EtOH. KOCO-P1 at 50 ug/mL reduced the production of H202 in Raw 264.7 cells by Nicotine. Conclusions : KOCO-P1 could be supposed to have antioxidative effect on macrophage with no cytotoxicity.

Microbial Risk Assessment of Processed Foods in Korea (우리나라의 가공식품에 대한 미생물학적 위험도 평가)

  • This study was undertaken to evaluate microbial risk degree of some processed foods in Korea. In this study the data on the outbreak of foodborne diseases during recent 18 years (1976-1989, 1993-1996. 8) were analyzed. The most frequently isolated pathogens were Salmonella (36.9%); followed vibrio (22.0%), Staphylococcus (15.7%) and Escherichia coli (13.3%). Outbreak rate of Staphylococcus, Vibrio, E. coli and Salmonella, was 33.0%, 23.5%, 17.5% and 17.1%, respectively. Overall risk degree of pathogens by fatality rate, outbreak rate and pathogen amount for foodborne outbreak was Clostridium, 5, Staphylococcus and Vibrio, 4, Salmonella and E. coli, 3. Based on foodborne pathogens, the risk degree of raw seafoods, raw eggs and processed seafoods were 4, and those of raw meats, Doshiraks and milk products were 3. Also, based on processing characteristics of foods, the risk degree of surimi-based imitation crab was 3. Foods of the highest actual risk degree were raw seafoods and raw eggs (16); followed raw meats (15), surimi-based imitation crab (12), Doshirak (9) and milk products (6).

  • These studies were brought about to know how much the elctronic methods of raw silk testing is correct, effective and convenient in the testing process. These effectiveness and convenience for the testing process under the emergency of the automatic system in raw silk inspecting process, the convenience of inspection in crease in it's capability decreasing the necessary working members, but as certain defect, the corresponding time span needs more than the former case in kind. Judging from the above mentioned carefully, it is necessary or even immidiate for participants to deeply study and search in earnest as well, before adopting the so called International Method of Raw Silk Test and Classification which was proposed by S.S.C in 1990.

  • Objectives : This study was performed to evaluate the effect of CJ(Cuscuta japonica Chois) on osteoclast differentiation and gene expression. Methods : The osteoclastogenesis and gene expression were determined in RANKL(receptor activator of nuclear factor kappa B ligand)-stimulated RAW 264.7. The results were summarized as followes. Results : CJ decreased the number of TRAP positive cell in RANKL-stimulated RAW264.7 cell. CJ decreased the expression of RANK(receptor activator of nuclear factor kappa B), $TNF{\alpha}$, and IL-6 in RANKL-stimulated RAW264.7 cell. CJ decreased the expression of iNOS and COX-2 in RANKL-stimulated RAW264.7 cell. CJ decreased the expression of Cathepsin K in RANKL-stimulated RAW264.7 cell. Conclusions : It is concluded that CJ might decrease the bone resorption resulted from decrease of osteoclast differentiation and it's related gene expression.

Fundamental Studies on the Food Sanitation for Raw Milk in Korea (국내산 원유에 대한 식품위생학적 연구)

  • Sanitary condition for raw milk in Korea was investigated in this study. It is hoped that the information will be used for reference in future endeavors of study in the field of public health and food sanitation in Korea. The results were summerized as follows: 1) The viable cell counts of bacteria in raw milk were tend to be increased under the various atmospheric temperature, and the correlation coefficient between temperature and total viable cells was r=+0.921(p<0.01). 2) The correlation coefficient between methylene blue reduction time test and viable cell counts of bacteria in raw milk was r=-0.799(p<0.01). 3) The relationship between total solid rate(%) and milk fat rate(%) was highly significant level as r=+0.745(p<0.01). 4) Highly significant correlation coefficient was r= +0.945(p<0.01) between milk fat and protein rate in raw milk.

  • This study was carried out to investigate the effect of sulfamethazine and trimethoprim on TTC test in raw milk. Ten raw milks which does not contain any antibiotics were collected from milk farms and one hundred twenty raw milks which collected from three raw milk receiving stations and those milk samples were mixed up sulfamethazine and trimethoprim for TTC test. The results were as follows ; 1. Positive reactions of TTC test were shown above the concentration of 3, 000ppm for sulfamethazine and 10ppm for trimethoprim. 2. When the TTC test was conducted with trimethoprim and sulfamethazine mixture, a pair of the concentration of trimethoprim and sulfamethazine shown to positive reaction were 0.1-2, 000ppm, 0.5-1, 000ppm, 1.0-250ppm and 2.0-100ppm, respectively. 3. Of 120 raw milk samples tested, 16 samples(13%) were shown quasi-positive reaction when the trimethoprim at the concetration of 2ppm was added in the samples.

  • An effective nesting algorithm has been proposed to allocate the arbitrary shapes on one or several raw sheets by applying the well-known simulated annealing algorithm as the optimization technique. In this approach, both the shapes to be allocated and the raw sheets are represented as the grid-based models. This algorithm can accommodate every possible situations encountered in cutting apparel parts from the raw leather sheets. In other words, the usage of the internal hole of a shape for other small shapes, handling of the irregular boundaries and the interior defects of the raw sheets, and the simultaneous allocation on more than one raw sheets have been tackled on successfully in this study. Several computational experiments are presented to verify the robustness of the proposed algorithm.

Dairy Industry Situation in Hokkaido, Japan (Sanitary Quality of Raw Milk) (일본 북해도의 낙농 ${\cdot}$ 유업현항 (위생적 유질을 중심으로))

  • Hokkaido, as center of Japanese dairying, 3.48 million tons of raw milk which was about 40% of total production in Japan were produced from 11,400 farms in 1996. The average herd size and the annual volume of milking per head in Hokkaido were 78 head, 7,200kg, and the quality of raw milk also is much better than that of other area in Japan. Raw milk having less than 100,000/ml in viable count and less than 300,000/ml of somatic cell count were 99.6%, 93% respectively. In spite of producing large amount of high quality milk, only 26% of total amount was processed as market milk and 76% was used for making dairy products like butter, cheese and milk powder. Therefore, because of big difference in price between the raw milk for market milk and for dairy products. the income of dairy farms are much less comparing to other parts of Japan, where most of the raw milk are consumed as market milk.

Saccharification of Raw Starch in Ethanol Fermentation (에탄올발효에서 전분질무증자당화의 가능성연구)

  • The possibility of the ethanol fermentation from raw cassava starch without cooking was investigated. Saccharification yield in the simultaneous saccharification-fermentation (SSF) system was compared with that in saccharification of raw cassava starch, using glucoamylase of Aspergillus shirousmi. Although the saccharification yield of raw cassava starch with 10 folds of the enzyme was 60% compared to cooked cassava starch, higher saccharification could be obtained by SSF This result is maybe due to the elimination of end product inhibition in saccharification of raw starch by glucoamylase. Final ethanol yield from raw cassava starch was about 88% under the condition of 3$0^{\circ}C$, 120 rpm shaking after 3 days in the SSF system.
