Antioxidant Activity of Hawthorn Fruit in vitro

  • The antioxidant activity of hawthorn fruit (Crataegus pinnatifida Bunge var. typica Schneider) extracts was investigated by several in vitro antioxidants properties, including DPPH free radical scavenging activity, total phenolic and flavonoid contents, hydroxyl radical scavenging activity, reducing power activity, iron-chelating capacity and nitrite scavenging activity. Among the extracts in this study, the 70% EtOH extract showed higher antioxidant activity than the others. The $IC_{50}$ value of DPPH free radical scavenging activity was $99.26\;{\mu}g/mL$. Furthermore, the 70% EtOH extract also showed significantly high total phenolic and flavonoids contents and reducing power activity. However, the MeOH extract exhibited stronger effects on hydroxyl radical scavenging activity, iron-chelating capacity and nitrite scavenging activity. All the results implicated that, the hawthorn fruit may has the available potential to be utilize as a potential source of natural antioxidant.

Optimization and production of protein hydrolysate containing antioxidant activity from tuna cooking juice concentrate by response surface methodology

  • To optimize the hydrolysis conditions in the production of antioxidant hydrolysates from tuna cooking juice concentrate (TC) to maximize the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, TC containing 48.91% protein was hydrolyzed with Alcalase 2.4 L, and response surface methodology (RSM) was applied. The optimum hydrolysis conditions included a 2.2% (w/v) Alcalase concentration and 281 min hydrolysis time, resulting in the highest DPPH radical scavenging activity of 66.49% (0.98 µmol Trolox/mg protein). The analysis of variance for RSM showed that hydrolysis time was an important factor that significantly affected the process (p < 0.05). The effects of different drying methods (freeze drying, hot air drying, and vacuum drying) on the DPPH radical scavenging activity and amino acid (AA) profiles of TC hydrolysate (TCH) were evaluated. Vacuum-dried TCH (VD) exhibited an increase in DPPH radical scavenging activity of 81.28% (1.20 µmol Trolox/mg protein). The VD samples were further fractionated by ultrafiltration. The AA profiles and antioxidant activities in terms of the DPPH radical scavenging activity, 2,2'-azino-bis(3-ethylbenzthiazoline)-6-sulfonic acid (ABTS) radical scavenging activity, ferric reducing antioxidant power, and ferrous ion chelating activity were investigated. Glutamic acid, glycine, arginine, and cysteine were the major AAs found in the TCH fractions. The highest DPPH radical scavenging activity was found in the VD-1 fraction (< 5 kDa). The VD-3 fraction (> 10 kDa) exhibited the highest ABTS radical scavenging activity and ferric reducing antioxidant power. The ferrous ion chelating activity was the highest in VD-1 and VD-2 (5 to 10 kDa). In conclusion, this study provided the optimal conditions to obtain high antioxidant activities through TCH production, and these conditions could provide a basis for the future application of TCH as a functional food ingredient.

Antioxidant activity of ethanol extracts from the root and bark of Ulmus davidiana

  • Antioxidant activities of Ulmus davidiana root and bark extracts were evaluated by various antioxidant tests, including DPPH radical-scavenging, nitric oxide-scavenging, superoxide anion radical-scavenging, and ABTS radical-scavenging assays, and superoxide dismutase (SOD)-like activity and reducing power analysis, along with the determination of total phenolic and flavonoid contents. Both the extracts showed strong antioxidant activities by these testing methods. Ulmus davidiana root extract possessed strong reducing power and nitric oxide-scavenging activity, and high scavenging activities against free radicals including the superoxide anion, and the ABTS and DPPH radicals, but a weaker scavenging activity of SOD. In contrast, the Ulmus davidiana bark extract exhibited a strong SOD-like activity, but all the other activities were weak. It was observed that the total phenolic and flavonoid contents of the Ulmus davidiana root extract were higher than those of the Ulmus davidiana bark extract.

산초(Zanthoxylum Schinifolium) 뿌리, 줄기 및 잎 추출물의 항산화, DDPH Radical 소거 작용 및 항혈전 효과 (Effects of Antioxidative, DPPH Radical Scavenging Activity and Antithrombogenic by the Extract of Sancho (Zanthoxylum Schinilolium))

  • Effects of root, stem and leaf extract of sancho (Zanthoxylum schinifolium) on the inhibition of lipid peroxidation in the hepatic microsome of rat, DPPH radical scavenging activity and activated partial thromboplastin times (APTT) were examined in vitro. The highest inhibition of hepatic microsomal lipid peroxidation was observed by ethyl acetate fraction than that of methylene chloride fraction of the root and stem extracts. The high inhibition of lipid peroxidation was determined in the leaf, the root and the stem in order. The DPPH radical scavenging activity of ethyl acetate fraction was higher than that of n-butanol fraction and it was similar to the root and the steam extract. It was similar to the inhibition of hepatic microsomal lipid peroxidation. The DPPH radical scavenging activity was the highest in 2.500mg/mL of ethyl acetate fraction and it was 4.4 fold higher than that of $\alpha-tocopherol$, as an antioxidant standard. The DPPH radical scavenging activity was dependent on the extract concentration in the range of 0.125-5.000 mg/mL. The throm-boplastin times were higher than that of n-butanol fraction and it was similar to the root and the steam extracts. The leaf extract showed the highest antithrombogenic effect followed by the stem and then the root extract. The activated partial thromboplastin times were ependent on the extract concentration in the range of 0.100-2.000 mg/mL. Consequently, the effects of antioxidative, DPPH radical scavenging activity and antithrombogenic of Z. schinifolium was observed due to the inhibition of lipid peroxidation and the DPPH radical scavenging activity by methylene chloride, n-butanol and ethyl acetate fraction of the leaf extract. 김치유래 Lactobacillus plantarum K-21의 DPPH 라디칼 제거활성 증진 및 다양한 항산화 효과 (Enhanced DPPH Radical Scavenging Activity of Lactobacillus plantarum K-21 Isolated from Kimchi and its Various Antioxidant Effects)

  • Lactic Acid Bacteria (LAB) are among the representative probiotics that have been used for a long time in fermented food. Although there are many studies on detecting the radical scavenging activity of LAB, few studies have been conducted on the environmental factors that improve scavenging activity. This study investigated the environmental factors affecting the DPPH radical scavenging and various antioxidant activities of Kimchi-derived Lactobacillus plantarum K-21 with antihypertensive and radical scavenging activities. The optimal conditions for scavenging DPPH radicals were glucose 2%, bactopeptone 0.5%, Tween 80 0.05%, L-cysteine 0.05%, and an initial pH 6.5 at 35℃. Under optimal conditions, the DPPH radical scavenging activity was 94.8±2.2%, which was 1.5 times higher than that of the basic medium. In addition, L. plantarum K-21 had other antioxidant activities; ABTS radical scavenging (93.6±1.5%), hydroxyl radical scavenging (8.5±0.9%), metal chelating (65.9±0.5%), NO scavenging (53.1±19%), SOD-like (25.1±1.5%), and reducing power (11.7±1.4%) activities were detected. Therefore, L. plantarum K-21 may act not only as a starter for lactic acid-fermented foods with improved functionality but also as a drug for various diseases caused by oxidative stress.

한국산 감자의 기관별, 품종별, 중량별, 분포별 및 조리 방법에 따른 총 페놀 함량과 DPPH 라디칼 소거능에 관한 연구 (The Total Phenolic Contents and DPPH Radical Scavenging Activities of Korean Potatoes according to Physical Characteristics and Cooking Methods)

  • This study was conducted to measure the total polyphenol contents and DPPH radical scavenging activities of different potato plants parts, varieties, and grades, and by distribution and different cooking conditions. The results were as follows. For the plant parts, total phenolic content and DPPH radical scavenging activity were highest in the flowers followed by leaves and stems, respectively. Among 5 potato varieties, 'Jasim' had the highest DPPH radical scavenging activity and the activity of its pulp was lower than that of its cortex(peel). Regardless of potato grade, the cortexes(peel) of samples had two-fold higher DPPH radical scavenging activity than pulp, and the Grade SS potato had the highest phenolic content. It was also found that the bud ends and stem ends had comparably larger amounts of phenolic compounds in horizontally cut potatoes. Finally, the descending order for DPPH radical scavenging activity, according to different cooking conditions, was as follows: gas oven range-baking, frying, microwave-heating, sauteing, boiling in 1% saline solution, steaming, and boiling in 3% saline solution.

Evaluation of Physiological Activities of the Citron (Citrus junos Sieb. ex TANAKA) Seed Extracts

  • Citron seed extracts (CSEs) were made using distilled water (CSEW), ethanol (CSEE), and n-hexane (CSEH), to measure the total polyphenol contents, DPPH and ABTS radical scavenging activities, and anti-complementary activity. The total polyphenol content was observed the highest in CSEE (188.71 ${\mu}g/mL$), and occurred in the following order: CSEE>CSEW (141.11 ${\mu}g/mL$)>CSEH (26.19 ${\mu}g/mL$) at 10 mg/mL. CSEE (63.56%) and CSEW (56.61%) showed significantly higher DPPH radical scavenging activities when compared with CSEH (28.57%). ABTS radical scavenging activities of CSEE (45.53%) and CSEW (40.02%) were also observed to be higher, whereas CSEH did not show ABTS radical scavenging activity. Anti-complementary activity of CSEE (26.85%) showed a greater activity than that of CSEW (7.84%) at 1,000 ${\mu}g/mL$. Limonin and nomilin contents had the highest values (1.882% and 2.089%) in CSEE, and with 0.327% and 0.139% in CSEW; however, CSEH showed relatively very low values at 0.061% and 0.026%, respectively. Among the CSEs tested, CSEE as a by-product from citron may provide an important source of dietary antioxidant compounds with rich polyphenol and limonoid contents, and immunopotentiating activity, including the complement activation factor.

오계란 단백질 가수 분해물 제조 및 한외여과 분획물의 in vitro 항산화 활성 특성 (In vitro Antioxidant Activity of Ogae (Korean Native Black Fowl) Egg White Protein Hydrolysates Fractionated by Ultrafiltration)

  • 식물 및 동물성 단백질 유래 펩타이드 형태의 단백질 가수 분해물들은 항산화, 고혈압 완화, 면역조절, 진통완화 및 항균작용 등 생리활성이 있는 것으로 알려져 왔다. 본 연구는 연산오계란 단백질 가수 분해물을 Ultrafiltration를 이용하여 HDS(분획되지 않은 가수 분해물), 1 kDa, 5 kDa, 10 kDa, 50 kDa로 분획된 기능성 펩타이드의 DPPH radical scavenging activity, superoxide anion radical scavenging activity, hydroxyl radical scavenging activity 및 $Fe^{2+}$ chelation ability을 평가하였다. 그 결과 DPPH radical scavenging activity 최대값은 1 kDa(70.83 %), hydroxyl radical scavenging activity 최대값은 5 kDa (47.01 %), superoxide anion radical scavenging activity 최대값은 5 kDa(40.57 %), $Fe^{2+}$ chelation ability 최대값은 5 kDa(29.87 %)로 나타났다. Ultrafiltration를 이용하여 fractionation된 단백질 가수 분해물의 항산화 저해 능력 $IC_{50}$ 평가하였다. 그 결과 HDS의 최대값은 superoxide anion radical scavenging activity($IC_{50}$, 5.42 mg/ml)이고, 1 kDa의 최대값은 $Fe^{2+}$ chelation ability($IC_{50}$, 1.67 mg/ml)이고, 5 kDa의 최대값은 $Fe^{2+}$ chelation ability($IC_{50}$, 2.09 mg/ml)이고, 10 kDa의 최대값은 $Fe^{2+}$ chelation ability($IC_{50}$, 2.61 mg/ml)이고, 50 kDa의 최대값은 $Fe^{2+}$ chelation ability($IC_{50}$, 4.53 mg/ml)이다. 그러므로 본 연구 결과를 바탕으로 5 kDa를 이용하여 오계란 단백질에서 분획한 펩타이드는 항산화 기능성 식품소재로서 활용할 가치가 높을 것으로 기대한다.

두리안 추출물의 항산화 활성 (Antioxidative Activity of the Durian (Durio zibethinus) Extract)

  • The purpose of this study was to examined the antioxidant activities by water and 70% ethanol extract from durian (Durio zibethinus.) seed, sarcocarp and peel. Durian extract were studied for reducing sugar content, polyphenol content, superoxide dismutase (SOD) like activity, electron donating ability, nitrite scavenging ability, flavonoid content, hydroxy radical scavenging activity. Reducing sugar content were increased peel > sarcocarp > seed. Total polyphenol, flavonoid content, DPPH radical scavenging ability and SOD like activity were increased seed > peel > sarcocarp. Total polyphenol content was relatively high as $21.90{\pm}0.50mg/g$ in the ethanol extract of the seed. DPPH radical scavenging ability was relatively high as $62.08{\pm}2.63%$ in the water extract of the seed. Nitrite scavenging ability was no significant difference. Hydroxy radical scavenging activity was increased seed > peel > sarcocarp, was relatively high as $58.27{\pm}1.13%$ in the water extract of the seed.

In vitro anti oxidant activity of methanol extract of Clerodendrum infortunatum Linn

  • The antioxidant potency of methanolic extract Clerodendrum infortunatum Linn. (MECI), which are widely used in the Indian indigenous system of medicine for different purposes, was studied. The antioxidant potential was evaluated using different established in vitro antioxidant tests viz. determination of total amount of polyphenolics compounds, DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging, nitric oxide scavenging, superoxide anion radical scavenging, hydroxyl radical scavenging and reductive power assay. It was found that MECI contain a high amount of polyphenolics and possesses significant free radical scavenging activity in all the assay. The higher activity was may be due to presence of richest amount of polyphenolics and flavonoids in it.
