Antioxidant Activity of Solubilized Tetraselmis suecica and Chlorella ellipsoidea by Enzymatic Digests

  • In this study, we focused on natural water-soluble antioxidants from Tetraselmis suecica (T. suecica) and Chlorella ellipsoidea (C. ellipsoidea). They were prepared by enzymatic digestion using five carbohydrases (Viscozyme, Celluclast, AMG, Termamyl and Ultraflo) and five proteases (Protamex, Alcalase, Flavourzyme, Neutrase, and Kojizyme), and the potential antioxidant activity of each was assessed. Most enzymatic digests from T. suecica had a higher radical scavenging activity than those from C. ellipsoidea. Among the enzymatic digests, Kojizyme digest from T. suecica exhibited the highest effect on DPPH radical scavenging. Viscozyme (30.2%) and Neutrase (34.6%) digests from T. suecica exhibited higher hydroxyl radical scavenging activity. Kojizyme digest from T. suecica (81.5%) had strong alkyl radical scavenging activity. Neutrase (61.9%) and Kojizyme (61.5%) digest from T. suecica possessed the highest effects on hydrogen peroxide scavenging. Among the tested samples, Neutrase (TN) and Kojizyme (TK) digests from T. suecica showed the highest antioxidant activity (DPPH, alkyl radical, hydrogen peroxide). Therefore, TN and TK digests were selected for use in the further experiments. Those digests showed enhanced cell viability against $H_2O_2$-induced oxidative damage, and relatively good hydrogen peroxide scavenging activity in an African green monkey kidney (Vero) cell line. These results suggested that an enzymatic digestion will be an effective way for the production of a potential water-soluble antioxidant from a microalgae, T. suecica.

Changes of Antioxidant Capacity, Total Phenolics, and Vitamin C Contents During Rubus coreanus Fruit Ripening

  • Changes in antioxidant activity of Rubus coreanus fruit of 3 clones (S13, S114, and S16), which were selected from different sites, were studied at different ripening stages. Antioxidant activities (tree radical scavenging activity and reducing power) were determined and their relationships to total phenolic contents and ascorbic acid were analyzed. The highest tree radical scavenging activities of 3 clones (S13, S14, and S16) were 79.39, 75.80, and 81.16% at $125\;{\mu}g/mL$, respectively. In general, the antioxidant activity and the related parameters, including total phenolic content and vitamin C content decreased during fruit ripening. Total phenolic contents of the R. coreanus fruits (S13, S14, and S16) were correlated with tree radical scavenging activity ($R^2=0.8114$, 0.9186, and 0.9714). These results improve knowledge of the effect of ripening on the antioxidant activity and related compounds contents that could help to establish the optimum R. coreanus fruit harvest data for various usages.

  • We isolated gallotannin, 2,6-digalloyl-1,5-anhydroglucitol, known as acertannin (1), from the barks and xylems of Acer ginnala Maxim. It is a genus of Acer species of shrubs in the family Aceraceae. A. ginnala grows in Korea, Japan and Mongolia. We accomplished the structure elucidation by confirming that the result of $^1H$,$^{13}C-NMR$,MS spectrum data was similar to previous references. We measured DPPH and ABTS radical scavenging activity in vitro to evaluate anti-oxidative activities on acertannin isolated from A. ginnala. Acertannin from A. ginnala exhibited potent DPPH and ABTS radical scavenging activities. We examined the antioxidant and apoptosis modulative effects. This examination shows that A. ginnala has not only 1,1-diphenyl-2-picryhydrazyl(DPPH) radical scavenging activity and ABTS radical scavenging activity, but also human hair dermal papilla cell protection effects. These results indicate that the barks and xylems of A. ginnala might be developed as a potent anti-oxidant, hair growth agent, and ingredient for related new functional cosmetic materials.

골질 보호 작용이 있는 수종 생약 추출액의 래디칼 소거능 및 DNA 보호 효과 (Radical Scavenging Activities and Protective Effects against Oxidative Damage to DNA of Extracts from Medicinal Plants with known Osteoprotective Effects)

  • Among aging diseases, the most basic problem is a decrease in bone mineral density. Patients with osteoporosis are steadily increasing in the world. This study was investigated to show the relationship between osteoporotic effects and oxidative damage. Water extracts of 15 medicinal plants and ethanol extracts of 14 medicinal plants with known anti-osteoporotic effects, were tested for their radical scavenging activity using DPPH, ABTS, SRSA and FRAP assay. Water extract of Cornus officinalis, Rubus coreanus and ethanol extract of Rubus coreanus, Viscium album var. coloratum, Cimicifuga heracleifolia showed about 15-20 mg/g of total phenolic contents. Water extract of Cornus officinalis, Rubus coreanus and Epimedium koreanum showed high radical scavenging activity. Ethanol extract of Drynaria fortunei, Cornus officinalis, Rubus coreanus, Gentiana scabra and Astragalus membranaceus showed high radical scavenging activity. Water extract of Drynaria fortunei, Cornus officinalis, Nelumbo nucifera, Epimedium koreanum, and Gentiana scabra showed very strong protective effect against oxidative damage to DNA. These results suggest the correlation between anti-osteoporotic effects and antioxidative effects.

Phytotoxic Effect, DPPH Radical Scavenging Activity and Chlorogenic Acid Level of Methanol Extracts from Aerial Parts of Several Korean Salad Plants

  • A series of aqueous or methanol extracts from four different Korean salad plants were assayed to determine their allelopathy and antioxidant activity. The extracts applied on filter paper in a Petri-dish bioassay significantly inhibited root growth of against alfalfa (Medicago sativa) seedlings. Leaf extracts from 40 g dry tissue $L^{-1}$ of Aster yomena was most phytotoxic to alfalfa root growth, and followed by that of Cirsium japonicum, Taraxacum officinale, and Ixeris dentate. Methanol extracts of plants dose-dependently increased DPPH free radical scavenging activity in vitro. Antioxidant activity of methanol extracts from the same plant species was investigated, and the result showed high DPPH free radical scavenging activity in Cirsium japonicum, Aster yomena, and Ixeris dentate, however, in Taraxacum officinale was least activity. By means of HPLC analysis, chlorogenic acid, p-coumaric acid, and total phenolics with 7.68, 17.47 and 18.64 mg, $100g^{-1}$, respectively, showed the highest amounts in methanol extracts from Cirsium japonicum leaves. These results suggest that Compositae salad plants contain water-soluble substances with allelopathic potential as well as antioxidant activity.

Taguchi 법에 의한 흑목이버섯의 항산화활성 증진을 위한 열수추출 최적화 (Optimization of Hot Water Extraction for Enhancing Antioxidative Activity from Auricularia auricula Using Taguchi Approach)

  • Optimization study of the hot water extraction for enhancing antioxidative activity from Auricularia auricula was performed by Taguchi approach using orthogonal matrix $L_9(3^4)$ method. The correlation between DPPH radical scavenging activity and the components of samples extracted from different extraction conditions were also analyzed. The correlation coefficient between DPPH radical scavenging activity and melanin content of A. auricula were 0.93, indicating 'good correlation'. The optimum extraction conditions was obtained at the extraction time of 1 hr. temperature of $85^{\circ}C$, solid: water ratio of 1: 40(w/v) and frequency of 2 times. Under these conditions, values of maximum DPPH free radical scavenging activity and melanin contents of A. auricula were $67.21{\pm}2.17$ and $52.94{\pm}2.10 mg/g$, respectively. Melanin content of 1.6 times and DPPH free radical scavenging effect of 130% were enhanced by optimization.

가공용 감귤 신품종 하밀감 과피 추출물의 항산화 활성 (Antioxidative Activities of New Citrus Hybrid 'Hamilgam' Peel Extracts)

  • The major objective of this study was to investigate the usability as cultivar for processing of new Citrus hybrid 'Hamilgam'. We investigated various antioxidant activities, such as DPPH radical, ABTS radical, Hydroxyl radical and Superoxide anion radical effect, along with total polyphenol content and total flavonoid content of Citrus hybrid 'Hamilgam', C. unshiu, C. natsudaidai. The total polyphenol content and total flavonoid content were the highest in the Hamilgam. The major flavonoids of Hamilgam and Natsudaidai were naringin and neohesperidin, and those of Unshu were narirutin and hesperidin, as determined by HPLC. Especially, the neohesperidin content of Hamilgam showed much higher (100 times over) than that of Unshu. However, the aglycone form was not found. DPPH and ABTS radical scavenging activities were the highest in Hamilgam peels. Hydroxyl radical scavenging activity was high in the order of Unshu, Hamilgam and Natsudaidai. Superoxide anion radical scavenging of 3 cultivars peels displayed low activities compared to DPPH and ABTS radical scavenging. Based on these results, Hamilgam peel extracts possess antioxidant activities and may thus serve as potential sources of functional food, cosmetic products, etc.

발아 특수미의 항산화 활성 (Antioxidative Activity of Germinated Specialty Rices)

  • 발아에 의한 거대배아미와 유색미의 기능성 변화를 항산화 활성에 중심을 두고 조사하였다. 전반적으로 환원력은 일반현미보다 거대배아미와 유색미가 높았고, 발아처리는 환원력을 증가시키는 경향이 있었다. 지질과산화에 대한 억제효과를 조사한 결과, in vitro의 linoleic acid peroxidation system 및 ex vivo의 토끼 적혈구 막지질 과산화 system 모두에서 억제효과도 유색미가 가장 높았고, 그 다음이 거대배아미, 일반현미의 순서로 나타났다. Superoxide radical에 대한 소거활성은 유색미>거대배아미>일반현미의 순서로 낮아졌으며, 무발아 조건과 비교할 때 발아처리는 라디칼에 대한 소거활성을 증진시켰다. Hydroxyl radical 소거활성도 유색미가 가장 높았으며, 그 다음이 거대배아미, 일반현미의 순서였다. 발아에 의하여 일반현미의 hydroxyl radical 소거활성을 급격히 증가되었지만, 발아미간에 소거활성의 순위는 변하지 않았다. In vitro에서 조사된 ROS 소거활성과 같은 경향이 HL-60 세포에서 TPA로 자극으로 발생된 ROS에 대한 소거활성에서도 나타났다. 실험 결과, 유색미와 거대배아미에서는 발아와 연관되어 세포독성의 증가가 발견되지 않는 차별적인 ROS 소거활성의 증가 현상이 발견되었다.

청혈단(淸血丹)의 6-hydroxydopamine에 의해 유발된 독성에 대한 신경세포보호효과 (The Protective Effect of Chunghyul-dan(Qingxuedan) Against 6-hydroxydopamine Induced Neurotoxicity.)

  • Objectives : This Study was performed to assess the antioxidant and neuroprotective effect of Chunghyul-dan(Qingxuedan) in PC12 cells and primary rat mesencephalic dopaminergic neurons. Methods : The anioxidant effect was investigated using the DPPH radical and ABTS cation scavenging assays and total polyphenol amout of Chunghyul-dan(Qingxuedan). The neuroprotective effect of Chunghyul-dan(Qingxuedan) in PC12 cells was evaluated using MTT assay. The scavenging activity of Chunghyul-dan(Qingxuedan) on ROS production induced by 6-OHDA(6-hydroxydopamine) in PC12 cells was evaluated, as well as the attenuating effect on GSH reduction. Finally, we examined the neuroprotective effect of Chunghyul-dan(Qingxuedan) against 6-0HDA-induced toxicity in the primary culture of rat mesencephalic doperminergic neurons. Results : Chunghyul-dan(Qingxuedan) showed concentration-dependent scavenging activities in DPPH radical and ABTS cation scavenging assays and it was not cytotoxic to PC12 cells. In postand co-treatment, Chunghyul-dan(Qingxuedan) protected PC12 cells from the 6-OHDA induced toxicity at 50 and 100 ${\mu}$g/mL significantly. And Chunghyu!-dan(Qingxuedan) decreased the 6-OHDA induced ROS production at a dose dependent manner, while increaing the 6-OHDA induced GSH reduction at 50 and 100 ${\mu}$g/mL significantly. Finally, Chunghyul-dan(Qingxuedan) showed signicant protection of rat mescencephalic dopaminergic neurons from 6-OHDA at 1 ${\mu}$g/mL. Conclusions : These results demonstrate that Chunghyul-dan(Qingxuedan) has the antioxidant and neuroprotective effect against 6-0HDA induced cytotoxicity through decreasing ROS production and increasing GSH reduction.

Betula Platyphylla var. Japonica Extract Prevent Ultraviolet C Light-induced Cell Damage in Chinese Hamster Fibroblast (V79-4) Cells

  • The present study reports the protective properties of a total methanol extract of B. platyphylla var. japonica against ultraviolet (UV)-C irradiation. Pretreatment of Chinese hamster fibroblast (V79-4) cells with a total methanol extract significantly increased cell survival following $300\;J/m^2$ of UV-C irradiation. The total methanol extract was further fractionated into 5 fractions: n-hexane, dichloromethane, ethylacetate, n-butanol and water fractions. Among these fractions, B. platyphylla var. japonica ethylacetate, butanol and water fractions showed significant protective effects against the cellular damage induced by UV-C irradiation. In order to elucidate the mechanism underlying this protective effect, DPPH (Editor note: abbreviations should be spelled out at first use.) radical scavenging and lipid peroxidation inhibitory activity were measured. Significant radical scavenging and lipid peroxidation inhibitory activities were observed for the ethylacetate fraction. In summary, the present data demonstrate that an extract of B. platyphylla var. japonica has a significant protective effect against UV-C irradiation. The underlying mechanism of this protective effect may involve radical scavenging and inhibition of lipid peroxidation by the B. platyphylla var. japonica extract.
