The Study on Bleaching of Kenaf Fibers (Part I) -Effect of Bleaching- (케냐프 섬유의 표백에 대한 연구 (제1보) -표백효과를 중심으로-)

  • The study was based on a three-stage, non-repetitive factorial experiment in which chemical-rotted kenaf fibers were treated separately with hydrogen peroxide concentrations of $0.5\%,\;1\%\;and\;2\%$, with pH solutions of 7, 9 and 11, and treatment times of 30, 60 and 90 minutes. Under optimal conditions, the study was conducted to determine the bleaching efficiency by the addition of chelators, penetrants and surfactants. The bleaching effects on the kenaf fibers were high in high hydrogen peroxide concentration, high in alkali solution pH, low in long treatment time. The optimal level of hydrogen peroxide on bleaching effects were at $2\%$ hydrogen peroxide concentration, with pH of 11 and treatment time of 60 minutes. Under the conditions of $2\%$ hydrogen peroxide concentration, pH 11 and treatment time of 60 minutes, the addition of chelator: Sodim Pyrophosphate(SP), Citric Acid(CA) made the bleaching effects of the kenaf fibers high.

A Study on Properties of crystallized Glass in $ZnO-P_2O_5$ System ($ZnO-P_2O_5$계 결정화 유리의 물성에 관한 연구)

  • ZnO-$P_2O_5$ system glasses containing 45 to 60 mol% ZnO have been melted at$1200^{\circ}C$and crystallized through controlled heat treatment. The properties of the base glass and crystallized glass were examined with XRD. FTIR. density. thermal expansion, electric conductivity, hardness. The principal crystalline phase was identified as zinc metaphosphate [$Zn(PO_3)_2$ in crystallized glasses containing 45-55mol% ZnO and zinc pyrophosphate ($Zn_2P_2O_7$) in the sample of 60mol% ZnO with X - ray diffraction analysis. Thermal expansion coefficient and DC electrical conductivity were varied with direction of oriented crystalline in the samples containing 50-60mol% ZnO. This suggests the existance of the oriented crystalline.

An Efficient System for the Expression and Purification of Yeast Geranylgeranyl Protein Transferase Type I

  • To purify the geranylgeranyl protein transferase type I (GGPT-I) efficiently, a gene expression system using the pGEX-4T-1 vector was constructed. The cal1 gene, encoding the ${\beta}$ subunit of GGPT-I, was subcloned into the pGEX-4T-1 vector and co-transformed into E. coli cells harboring the ram2 gene, the ${\alpha}$ subunit gene of GGPT-I. GGPT-I was highly expressed as a fusion protein with glutathione S-transferase (GST) in E. coli, purified to homogeneity by glutathione-agarose affinity chromatography, and the GST moiety was excised by thrombin treatment. The purified yeast GGPT-I showed a dose-dependent increase in the transferase activity, and its apparent $K_m$ value for an undecapeptide fused with GST (GST-PEP) was $0.66\;{\mu}M$ and the apparent value for geranylgeranyl pyrophosphate (GGPP) was $0.071\;{\mu}M$.

  • Magnesium alloys is the lightest by structural metals, but it is not good corrosion resistant because of pit, void. Particularly, AZ31B magnesium alloy sheets that have slag, scratch by rolling process indicate some defects. The objective of this research is to perform uniform plating on AZ31B by studying etching and zincate process. Especially, zincate treatment by zinc salt and pyrophosphate is the most important in the decoration plating. Dissolution of magnesium is reduced by the formation of uniform zinc conversion layer during strick and post process, which decreases defects for plating process.

  • Alkaline powder cleaning agents (APCAs) were prepared by blending of $Na_2CO_3$ tetrasodium pyrophosphate (TSPP). sodium orthosilicate (Na-OSi), Na-dioctyl sulfosuccinate (303C), Demol C, and MJU-100A (100A). The physical properties of APCAs tested with steel specimen showed the following results. The cleaning powers of APCA-6 ($Na_2CO_3$ 250g/TSPP 70g/Na-OSi 40g/303C 60g/Demol C 50g/100A 30g mixture) for press-rust preventing oil was 97% and 98% degreasing at 2wt%, $70^{\circ}C$ and $90^{\circ}C$, respectively; for Quenching oil, the cleaning power of APCA-6 was 95% degreasing at 2wt% and $70^{\circ}C$. From the results, it was ascertained that APCA-6 exhibited a good cleaning power. Also low foaming power tests proved that the APCA-6 maintained good defoaming effect.

  • Adipose tissue is an important endocrine organ involved in the control of whole body energy homeostasis and insulin sensitivity. Considering the increased incidence of obesity and obesity-related disorders, including diabetes, it is important to understand thoroughly the process of adipocyte differentiation and its control. Therefore, we performed a differential proteome mapping strategy using two-dimensional gel electrophoresis combined with peptide mass fingerprinting to identify intracellular proteins that are differentially expressed during adipose conversion of 3T3-L1 pre-adipocytes in response to an adipogenic cocktail. In the current study, we identified 46 differentially expressed proteins, 6 of which have not been addressed previously in 3T3-L1 cell differentiation. Notably, we found that phosphoribosyl pyrophosphate synthetase (PRPS), a regulator of cell proliferation, was preferentially expressed in pre-adipocytes than in fully differentiated adipocytes. In conclusion, our results provide valuable information for further understanding of the adipogenic process.

Nonstructural Protein 5B of Hepatitis C Virus

  • Since its identification in 1989, hepatitis C virus has been the subject of extensive research. The biology of the virus and the development of antiviral drugs are closely related. The RNA polymerase activity of nonstructural protein 5B was first demonstrated in 1996. NS5B is believed to localize to the perinuclear region, forming a replicase complex with other viral proteins. It has a typical polymerase structure with thumb, palm, and finger domains encircling the active site. A de novo replication initiation mechanism has been suggested. To date, many small molecule inhibitors are known including nucleoside analogues, non-nucleoside analogues, and pyrophosphate mimics. NS5B interacts with other viral proteins such as core, NS3, 4A, 4B, and 5A. The helicase activity of NS3 seems necessary for RNA strand unwinding during replication, with other nonstructural proteins performing modulatory roles. Cellular proteins interacting with NS5B include VAMP-associated proteins, heIF4AII, hPLIC1, nucleolin, PRK2, ${\alpha}$-actinin, and p68 helicase. The interactions of NS5B with these proteins might play roles in cellular trafficking, signal transduction, and RNA polymerization, as well as the regulation of replication/translation processes.

Radiolabelled Monoclonal Antibodies (McAb): An Alternate Approach to the Conventional Methods for the Assessment of Cardiomyocyte Damage in an Experimental Brain-Death Pig Model

  • The present study was carried out to determine the possible use of cTn-I in the cardiac myofibrillar architecture, as a potential target for in vivo radioimmunodetection of cardiac damage in a brain death pig model. Radioiodiantion of the anti-cTn-I 5F4 McAb was carried out by lactoperoxidase method. the percentage iodine incorporation achieved was 70-75%. The radioiodinated McAbs were purified on Sephadex G-25 column and characterised by Paper chromatography, Phast Gel electrophoresis and electroimmunoblotting. Radioiodinated anticTn-I 5F4 McAbs were employed alongside Pyrophosphate($Tc_{99m}$-PPi$) and $Thallium^{201}$ chloride($TI^{201}$) in 24 landrace pigs (brain-dead=18 & sham-operated=6). The percentage cardiac uptake of the radiolabelled antibody injected dose was significantly higher in the brain dead animals(0.196%) as compared to that of sham-operated animals (0.11%). Specific in vivo localization of radiolabelled McAbs in the infarcted cardiac tissue was confirmed by computer-aided reconstruction of 3-D images of the isolated heart. The preliminary results of the study revealed preferential uptake of radiolabelled antibody at the site of myocyte damage resulting from artificially induced brain death.

  • Mutated forms of the ras oncogenes are associated with about 30% of human tumors. The ras genes encode 21KDa proteins, called p21 or Ras, that are associated with the plasma membrane. FPTase is a dimeric enzyme that catalyses the transfer of the farnesyl group from farnesyl pyrophosphate onto cysteine 186 at the C-terminus of the Ras protein. This is mandatory process for triggering ras oncogene toward tumor formation. Therefore, selective inhibitors of FPTase have the potential to be used as antitumorgenic agents.

  • RF sputtering process was applied to produce thin hydroxyapatite(HAp) films on Ti-6Al-4V alloy substrates. The effects of different heat treatment conditions on the chemical composites between HAp thin films and Ti-6Al-4V alloy substrates were studied. After deposition, the HAp thin films were heat treated for 1h at $400^{\circ}C,\;600^{\circ}C\;and\;800^{\circ}C$ under the atmosphere, and analyzed O/M, FESEM-EDX and ESCA, respectively. Experimental results represented that interface of HAp thin films and Ti-6Al-4V alloy substrates was composed Ti-OH, TiO, TiN, $Al_2O_3,\;V_2O_3,\;VO_2$. pyrophosphate and decreased carbide followed by the increase of heat treatment temperature.
