Bioassays of Polycyclic Aromatic Hydrocarbons Using cyp1a1-Luciferase Reporter Gene Expression System in Mouse Liver Hepa 1 Cells

  • Recent industrial society has human widely exposed to PAHs (polynuclear aromatic hydrocarbons) that are comming from the incomplete combustion of organic material as wider spread environmental contaminants. Biological activities of PAHs are not known although PAHs are considered as carcinogens. Our laboratory have been studied the effect of PAHs in the mouse liver hepa 1 cells. In this study, we examined the mouse liver hepa-l cells as a new bioassay system to evaluate bioactivity of PAHs. We have selected 13 PAHs to examine bioassay using cyp1a1-luciferase reporter gene expression system where cyp1a1 1.6 Kb 5flanking region DNA was cloned in front of luciferase reporter gene and this plasmid was transfected into hepa 1 cells transiently. This cells then used for the study to observe the effect of PAHs. We demonstrated that PAHs induced the CYP1A1 promoter and 7-ethoxyresolufin O-deethylase (EROD) activities in a concentration-dependant manner. Some of PAHs showed stronger stimulatory effect on CYP1 gene expression than TCDD. Acenaphthene, anthracene, fluorine, naphthalene, pyrene, phenanthrene, carbazole were weak responders to cyp1a1 promoter activity stimulation and EROD induction in hepa 1 cells and these chemicals seemed to respond less to EROD than cyp1a1 promoter activity. Benz(a)anthracene, benzo(b)fluoranthene, benzo(k)fluoranthene, chrysene, and dibenzo(a,h)anthracene showed strong response to cyp1a1 promoter activity stimulation and also EROD induction in hepa 1cells. Results of dose response study suggested that four strong responding PAHs, such as benzo(a)anthracene benzo(k)fluoranthene, chrysene, and dibenzo(a, h)anthracene might be mediated through arylhydrocarbon receptor system in hepa1 cells.

Characteristics of Chemical Composition in Carbonaceous Aerosol of PM2.5 Collected at Smoke from Coal Combustion (석탄 연소 시 발생되는 PM2.5 내 탄소 에어로졸의 화학 조성 연구)

  • The $PM_{2.5}$ samples were obtained from coal combustion with the four different combustion temperatures (550, 700, 900 and $1100^{\circ}C$) to understand chemical composition in carbonaceous aerosol. OC concentration was the highest when the combustion temperature was $550^{\circ}C$, while, the highest concentration for EC was shown at $700^{\circ}C$ of the coal combustion temperature. However, OC concentrations were very low and EC was not detected when the temperature was over $900^{\circ}C$. It indicates complete combustion was achieved when the combustion temperature was over $900^{\circ}C$. For six groups of organic compounds, n-alkanes and n-alkanoic acids were predominant at all of the combustion temperature in smoke of coal combustion, while, PAHs was only detected at $550^{\circ}C$. The diagnostic ratios of PAHs calculated in this study were 0.59 for Fluoranthene/(Fluoranthene+Pyrene), reflecting the characteristics of coal combustion. The Carbon number Preference Index (CPI) values of n-alkanes which ranged from 0.9 to 1.3 also showed the characteristics of coal combustion.

  • It is well known that diesel exhaust particulate matter contains mutagenic PAHs, such as benzo[${\alpha}$]pyrene, benz[${\alpha}$]anthracene, chrysene, etc. Therefore it is suspected that these chemicals act on estrogen receptor and reveal endocrine-disrupting effects. Recent attention has focused on causative chemicals of endocrine-disrupting effects. We examined the estrogenic activity of respirable diesel exhaust particulate matter derived from diesel powered vehicle. PM2.5 diesel exhaust of vehicle was collected using a high volume sampler equipped with a cascade impactor. Diesel exhaust samples were fractionated according to EPA methods. The presence of estrogenic and antiestrogenic chemicals in PM 2.5 diesel exhaust was determined using E-screen assay. To quantitatively assess the estrogenic and antiestrogenic activities in diesel exhaust particulate matter, estradiol equivalent concentration (bio-EEQ) was calculated by comparing the concentration response curve of the sample with those of the estrogen calibration curve. Weak estrogenic activities and strong antiestrogenic activities were detected in the crude extract and moderately polar fractions. Higher antiestrogenic potency was observed with higher EROD activities in aliphatic and aromatic compounds fraction. In conclusion, estrogenic/antiestrogenic-like activities were present in diesel exhaust particulate matter. However, the health consequences of this observation was unknown, the presence of these activities may contribute to and exacerbate adverse health effect evoked by diesel exhaust particulate matter.

Morinda citrifolia Inhibits Both Cytosolic $Ca^{2+}$-dependent Phospholipase $A_2$ and Secretory $Ca^{2+}$-dependent Phospholipase $A_2$

  • This study investigated the effects of the methanol extracts of Morinda citrifolia containing numerous anthraquinone and iridoid on phospholipase $A_2$ ($PLA_2$) isozyme. $PLA_2$ activity was measured using various $PLA_2$ substrates, including 10-pyrene phosphatidylcholine, 1-palmitoyl-2-[$^{14}C$]arachidonyl phosphatidylcholine ([$^{14}C$]AA-PC), and [$^3H$]arachidonic acid (AA). The methanol extracts suppressed melittin-induced [$_3H$]AA release in a concentration-dependent manner in RAW 264.7 cells, and inhibited $cPLA_2/sPLA_2$-induced hydrolysis of [$^{14}C$]AA-PC in a concentration- and time-dependent manner. A Dixon plot showed that the inhibition by methanol extracts on $cPLA_2$ and $sPLA_2$ appeared to be competitive with inhibition constants ($K_i$) of $3.7{\mu}g/ml$ and $12.6{\mu}g/ml$, respectively. These data suggest that methanol extracts of Morinda citrifolia inhibits both $Ca^{2+}$-dependent $PLA_2$ such as, $cPLA_2$ and $sPLA_2$. Therefore, Morinda citrifolia may possess anti-inflammatory activity secondary to $Ca^{2+}$-dependent $PLA_2$ inhibition.

Characteristics of Polycyclic Aromatic Hydrocarbons Degradation by Stenotrophomonas maltophilia (Stenotrophomonas maltophilia에 의한 방향족 화합물의 분해특성)

  • In this study, Isolation was attempted to acquire a phenol utilizing bacterium for PAH degradation and to investigate the characteristics of PAH degradation. The isolate was identified by BIOLOG test as Stenotrophomonas maltophilia. Lower first order reaction constant was detected in the presence of lower phenol concentration. The yield coefficient of phenol was 0.1447mg cell/mg phenol. In the presence of naphthalene and phenol, phenol degradation was favorable. The isolate was capable of utilize naphthalene and phenanthrene as growth substrate but PAH, containing over 4-ring structure such as pyrene, was not degradable. The possible phenanthrene degradation pathway would be the addition of two hydroxy group on C-1 and C-2 position, followed by ortho cleavage, and then decarboxylation.

Effect of Chemical Carcinogens on the Replication, Cytolyticity, DNA Synthesis, and Protein Expression of Herpes Simplex Virus in Viral Infected Cells (발암성 화학물질들이 Herpes Simplex Virus의 복제, 세포융해, DNA 합성 및 단백질 합성에 미치는 효과)

  • We investigated effects of several chemical carcinogens, i.e., $benzo({\alpha})pyrene$ (BP),7,12-dimethylbenz(a)anthracene (DMBA), nitrosomethyl urea (NMU), and nicotine on the replication, cytolyticity, DNA synthesis, and protein synthesis of type 1 herpes simplex virus (HSV-1) in viral infected Vero cell monolayers. We observed that the BP and DMBA did not show such activity. All chemical carcinogens did not inhibit the synthesis of viral DNA, but the expression of gamma viral proteins that are expressed from the newly synthesized progeny viral DNA was somewhat notably inhibited by BP and DMBA. However, the synthesis of alpha and beta viral proteins was not altered by the chemical carcinogens. These data indicate that the gamma viral proteins expressed from the newly synthesized DNA in the presence of chemical carcinogens in the culture medium may be defective. This is further supported by the fact that the virus fail to replicate in the presence of these chemical carcinogens, in spite of viral DNA and proteins are somewhat normally synthesized.

Source Profile of Road Dust for Statistical Apportionment Modeling in Seoul (통계 수용모델을 위한 서울시 도로변 화학성분 원인 프로파일)

  • Sources related to road dust is one of the biggest sources, which is responsible for a large portion of emission. In particular, PM2.5 is a potential cause for respiratory diseases, thus it should be managed and a mitigation plan using results of statistical apportionment modeling such as chemical mass balance needs to be established. Recently, identifying sources of PM2.5 and analyzing the contribution of the road dust through a contribution assessment is required. Therefore, this study provides the chemical source profiles of PM2.5 using IC, GC/MS, OCEC, and XRF for both paved sidewalk and paved roadway collected at seven different sampling sites. As a result, for paved sidewalk, $NH{_4}^+$ (70%), $NO{_3}^-$ (12%), $PO{_4}^-$ (9%), and $SO{_4}^{2-}$ (9%) have been analyzed in PM2.5 mass. Major molecular marker such as Si has been indicated as $12.0{\pm}3.4%$ and $13.6{\pm}6.9%$ for paved sidewalk and roadway, respectively. PAHs such as Fluoranthene, Pyrene, Chrysene, and 1,3,5-Triphenylbenzene are suggested as molecular markers for road dust.

A Review on the Atmospheric Concentrations of Polycyclic Aromatic Hydrocarbons (PAHs) in Asia Since 2000 - Part I: Data from Developed Countries

  • Among all hazardous air pollutants, polycyclic aromatic hydrocarbons (PAHs) are more significant owing to their carcinogenic, mutagenic and teratogenic properties. Many researchers worldwide have focused on determining the concentrations of PAHs in ambient air. However, the literature survey reveals that the seriousness of air pollution in Asia in terms of PAHs, compared to the rest of the world. Owing to the importance of PAHs in Asia, this paper reviews the concentrations of PAHs in Asia in recent years. For convenience, this paper describes the concentrations of PAHs in developed Asian countries reported during 2000-2011 as Part-I and in developing Asian countries as Part-II. The first section of this review provides a brief description of the properties, sources of PAHs and the health effects caused by their presence in the atmosphere. The ambient air PAH concentrations in both particle and vapor phases in developed Asian countries are then discussed. This study finds the ambient air concentrations of PAHs in developed Asian countries was higher than the Western countries but was lower than the developing Asian countries. The present review predicts the accurate toxicity due to the presence of PAHs in the atmosphere by calculating the Risk Weighted Concentration (RWC), regardless of the total amount of PAHs. The total data obtained during the literature survey is tabulated and presented as supplementary information to the readers.

Characteristics of Hazardous Air Pollutants in the Steel Industrial City, Pohang ( I ) - Polycyclic Aromatic Hydrocarbons (철강산업도시 포항지역 유해대기오염물질의 오염특성 ( I ) - 다환방향족탄화수소)

  • This study was carried out to characterize the ambient levels of PAH concentrations in Pohang city, where steel industry has been intensively developed. A large amount of PAH emissions from steel production and related industries in Pohang are concerned about the environmental health hazards of nearby residents. TSP samples were seasonally collected during 10 days at four sites in Pohang from April to December 2010. Each sample was collected for 24 hours using high-volume air sampler. All samples were extracted by $soxtec^{TM}$ before concentration of 1.0 mL. The concentrates were analyzed by GC/MS. The prevailing wind direction was southwest and west-southwest in Pohang during the measurement period. Variations of TSP concentrations appeared to be similar between four sites. The highest TSP concentration was found in fall, while the lowest in summer. The highest PAH concentration was found at an industrial site, while the lowest at a residential/commercial site. In the case of BaP, the average concentration at the industrial site was $0.63{\pm}0.66ng/m^3$, which was approximately 2 times of the residential/commercial site. The paired t-test (p<0.05) showed significant differences between industrial and residential area. Particulate PAH concentrations were higher in the winter and lower in the summer. The concentrations of BaP in winter were 2~10 times higher than those in the other three seasons. This study clearly demonstrated the impact of steel industry on the elevated levels of PAH in Pohang city.

Effects of Sediment Elutriates on the Early Reproductive Outputs in the Pacific oyster, Crassostrea gigas

  • This is a subsequent study to our previous finding that Pacific oysters, Crassostrea gigas, gained a so-called upper plateau concentration, around 30,000 ng/g dry weight digestive gland for benzo(a)pyrene, showed reproductive behavior but produced their ensuing reproductive outputs damaged. A serial dilution of sediment elutriates from Jinhae Bay, Korea, where pollution was progressive, were exposed to gametes of the Pacific oyster for 0, 5, 10, 20, 30, and 60 min to detail the pollutant effects on very young specimens. There was an apparent critical dilution over which adverse effects are evident. This was $10\%$ of the present sediment elutriate, corresponding to 0.3 ng/g on the basis of total polycyclic aromatic hydrocarbons (PAHs) for the oyster. Within the dilution the embryonic development was not influenced by the duration of exposure to its gamete stage. At higher dilutions over the critical dilution, occurrence of abnormality increased dependent on the pollutant dilution and the duration of exposure. Similar trends were also found in larval mortality. However, overall, the chemical toxicity was more significant to morphogenesis than to survival, suggesting a potential recruitment of the pollutants-induced abnormal larvae in the wild population to threaten the population integrity.
