Condition of Exo-polysacchride Production from Submerged Mycelial Culture of Ganoderma lucidum by Using Air-lift Fermenter System (Air-lift Fermenter System을 이용한 Ganoderma lucidum 균사체의 심부배양에 의한 세포외 다당류의 생산 조건)

  • For the efficient production of a new exo-polysaccharide from Ganoderma lucidum ASI 7004, the optimum conditions and methods in submerged cultivation were investigated with an airlift fermenter system. The optimum aeration rate was 2.5 Wm at the initial pH 5.0 and 28$^{\circ}C$. The increase of dissolved oxygen concentration by pure oxygen supply during cultivation did not improved the exo-polysaccaride production and the mycelial growth. The maximum exo-polysaccharide production and the mycelial growth under the optimum culture condition were obtained in media of glucose 60g/L, yeast extract 6g/L, (NH4)2HPO4 1g/L and KH2PO4 0.5g/L. Under these optimum medium and culture conditions, about 7.15g/L of exo-polysaccharide and 13.9g/L of mycelial growth were producted, respectively.

  • The objective of this work was to accomplish the isolation, molecular identification and characterizing the physiology of the causal agent of the algal spot in mango trees. For this purpose, the pathogen growth was assessed in different culture media, with subsequent observation and measurements of the filamentous cells. The molecular identification was made using mycelium obtained from leaf lesions and pure algae colonies grown in culture medium. Descriptions based on DNA sequencing indicated that the algae is Cephaleuros virescens. The algae must be isolated primarily in liquid medium for further pricking into agar medium. The highest mycelial growth average in Petri dishes occurred when the algae were grown in Trebouxia and BBM. Trebouxia enabled larger cells in the filamentous cells when compared to other culture media.

Deciphering Functions of Uncultured Microorganisms (난배양성 미생물의 기능 분석 방법)

  • Microbes within complex communities show quite different physiology from pure cultured microbes. However, historically the study of microbes has focused on single species in pure culture and most of microbes are unculturable in our labs, so understanding of complex communities lags behind understanding of pure cultured cells. Methodologies including stable isotope probing (SIP), a combination of fluorescence in situ hybridization (FISH) and microautoradiography (MAR), isotope micrarray, and metagenomics have given insights into the uncultivated majority to link phylogenetic and functional information. Here, we review some of the most recent literatures, with an emphasis on methodological improvements to the sensitivity and utilities of these methods to link phylogeny and function in complex microbial communities.

A Simple Method for the Preparation of Highly Pure Conjugated Linoleic Acid(CLA) Synthesized from Safflower Seed Oil

  • A simple and rapid method was developed to prepare a large quantity of highly pure conjugated linoleic acid (CLA) chemically-synthesized from safflower seed oil (SSO). CLA-SSO(74.9% in purity) was synthesized from fresh SSO(79.9% of linoleic acid) by alkaline isomerization at 18$0^{\circ}C$. Urea(50g) and CLA-SSO (25g) were completely dissolved in ethanol (750ml) using a water bath(5$0^{\circ}C$) and followed by refluxing for 60 min. The resultant was cooled to room temperature and stored in a cold room (4$^{\circ}C$) for 24hrs. After removing the urea adduct by filtration, the filtrate was rotoevaporated under 4$0^{\circ}C$ and the residue was dissolved in hexane (200ml). The hexane extract was washed with distilled water (100ml$\times$3) and dried over sodium sulfate anhydrous. This urea treatment procedure was repeated three times. The purity of CLA recovered from the hexane extract was 95.0%. This method can be applied to prepare a large quantity of highly pure chemically-synthesized CLA (>0.5kg/a batch) from any plant oils containing high percentages (>70%) of linoleic acid.

Isolation of Chondromyces crocatus in Pure Culture (Chondromyces crocatus의 순수 분리)

  • We have isolated Chondromyces crocatus KYC2823 in pure culture and five other strains in mixed culture with companion bacteria from Korean soil samples. The strain KYC2823, which was isolated from the soil sample collected in Cheongdo-gun, Gyeongsangbuk-do, showed typical characteristics of C. crocatus, including the shape of fruiting bodies and production of a peculiar odor. In addition, the 16S rDNA sequence was 99.8% identical to that of the strain Cm c5, the proposed neotype strain of C. crocatus. Cloning and sequence analysis of the polyketide biosynthetic genes from KYC2823 by performing PCR have revealed that this strain has biosynthetic gene clusters for ajudazols (inhibitors of electron transport systems) and chondramides (substances affecting the function of the actin cytoskeleton), and biosynthetic genes for other polyketide compounds that have not been cloned yet.

Reduction of Hydrogen Sulphide in Chicken Manure by Immobilized Sulphur Oxidising Bacteria Isolated from Hot Spring

  • The rapid development of the poultry industry has led to the production of large amounts of manure, which produce substances like hydrogen sulfide ($H_2S$) that contribute to odor pollution. $H_2S$ is a highly undesirable gas component and its removal from the environment is therefore necessary. Sulfur-oxidizing bacteria (SOB) are widely known to remove contaminating $H_2S$ due to their ability to oxidize reduced sulfur compounds. In this study, three potential SOB (designated AH18, AH25, and AH28) that were previously isolated from a hot spring in Malaysia were identified by 16S rRNA gene analysis. Laboratory-scale biological deodorization experiments were conducted to test the performance of the three isolates-in the form of pure or mixed cultures, with the cells immobilized onto alginate as a carrier-in reducing the $H_2S$ from chicken manure. On the basis of 16S rRNA phylogenetic analysis, isolate AH18 was identified as Pseudomonas sp., whereas isolates AH25 and AH28 were identified as Achromobacter sp. The most active deodorizing isolate was AH18, with an $H_2S$ reduction rate of 74.7% (p < 0.05). Meanwhile, the reduction rates for isolates AH25 and AH28 were 54.2% and 60.8% (p > 0.05), respectively. However, the $H_2S$ removal performance was enhanced in the mixed culture, with a reduction rate of 81.9% (p < 0.05). In conclusion, the three potential SOB isolates were capable of reducing the $H_2S$ from chicken manure in the form of a pure culture immobilized on alginate, and the reduction performance was enhanced in the mixed culture.

Poliovirus Decrease Effect by Activated Sludge Microbes (활성슬러지 구성 미생물에 의한 폴리오바이러스의 감소 효과)

  • The biological wastewater treatment system is known to have an important role in reducing the quantify of enteric virus in water environments. To clarify the roles of activated sludge microbes in decreasing the virus infectivity, the behavior of the virus in bacteria, protozoa, and metazoa was examined by pure or mixed culture system using poliovirus type 1(Lsc, 2ab strain). In the bacterial culture systems, the virus infectivity in the liquid phase decreased by a reversible adsorption of the virus to the bacteria or bacterial flocs. On the other hand, in the protozoa and the metazoa culture systems using T. pyriformis and P. erythrophthalma, respectively, with a variety of bacterial strains as prey, the main virus decrease mechanism of reversible adsorption in early stage was changed to irreversible predation, which was not eluted in this study. The virus decrease was more effective in the P. erythrophthalma culture system, which had high predation and floc forming abilities. However, in the mixed culture system of Z. ramigera and P. erythrophthalma, the more rapid reversible adsorption of virus to Z. ramigera flocs preceded the irreversible predation of P. erythrophthalma.

Observations on Some of the Mycelial Growth and Pigmentation Characteristics of Cordyceps militaris Isolates

  • Characteristic growth patterns of Cordyceps militaris isolates on various media, under varying light conditions and at varying incubation periods were examined. Light was found to be the most critical single factor in determining the density, texture, and pigmentation of the mycelial culture of the fungus. However, under the light condition, the degree of pigmentation and mycelial density were found to be affected by the incubation period and type of medium. Irrespective of the variations in medium type or incubation period, there was no pigmentation of the mycelium under dark condition. Radial growth of the mycelium was faster under dark incubation rather than under light incubation. Abundant mycelial density and darkest pigmentation of C. militaris isolates were produced in nutritionally rich media like SDAY, SMAY and CZYA, suggesting that these media may fulfill all the requirements for vegetative growth of the fungus. Growth characteristics of C. militaris isolates could be easily observed by the simple agar culture method, which would be useful to characterize the phenotypic characteristics of large number of pure cultures of the fungus under given conditions of growth factors such as medium, light and temperature.

A Study on the Biocompatibility of Anodized Titanium (양극산화 티타늄의 생체적합성에 관한 연구)

  • The high biocompatibility of titanium is connected with the high corrosion resistance of the surface oxide, its high dielectric constant, and some other specific biochemical properties of the oxide. The corrosion resistance of titanium can be improved with the formation of passive film by anodic oxidation. In other to characterize the titantium oxlde film formed by anodic oxidation, titanium plates were anodized in 0.5M $H_3SO_4$ electrolyte at voltages between 5V and 100v. The oxide film was examined by an X-Ray Diffractometer(XRD) and a Scanning Electron Microscope(SEM). In addition, the corrosion resistance of oxide film was tested by dipping in physiological NaCl,5% HCI,5% $H_3PO_4$ and its biocompatability was evaluated by the fibroblast-like cell culture. The results obtained are as follows : 1. The thickness of surface oxide and micropore are increased with the increase of electrode potential and formed deeply along the grain boundary. 2. The solubilities of titanium in electrolyte solution shows that the anodized titanium has more corrosion resistance than the untreated pure titanium. 3. The biocomatibility of anodized titanium is superior to untreated pure titanium.

  • The Chosun Dynasty in the $18^{th}$ century was a period of revival for science, art, and culture, bringing about Silhak(practical science), a new trend in the history of Chosun thoughts. In the history of fine arts, realistic landscape paintings and genre paintings were popular as realism became prevalent. From the aspect of food culture, in particular, the luxurious and elegant Korean-styled food culture was completed during this period. Iwanwasuseoksihoedo was painted by Jeong Hwang (1735-1800) in 1789, depicting a banquet on an elder's $60^{th}$ birthday. It is classified as a genre painting in the late Chosun Period but it contains things that comes into our heart, which are white porcelain with blue celadon pictures, white porcelain bowls, busy atmosphere of a banqueting house where food is being carried in a hurry and elders' serene appearance. All these things show the abundance of life, the room and comfort of old ages, and the beauty and relish of life in the well-arranged living ground. Hoehonyedo was painted on an elder's $60^{th}$ marriage anniversary by an unknown artist presumably in the $18^{th}$ century is a painting as realistic as a documentary photograph. The work gives viewers pleasure and comfort because it describes not a mighty clan but the superb later years of an official who had lived right and upright life. In the aspect of food culture, it displays the food culture of the splendid sixtieth marriage anniversary of a Korean official through noble etiquette among family members, seat planning, unique table culture and high quality tableware including white porcelain with celadon pictures and pure white porcelain.
