Experimental Hybridization between Some Marine Coenocytic Green Algae Using Protoplasms Extruded in vitro

  • Some marine coenocytic green algae could form protoplasts from the extruded protoplasm in seawater. The dissociated cell components of the coenocytic protoplasm could be reunited into live cells and, hence, the formation of new species by mixing protoplasms from different coenocytic cells has been predicted. Our results showed that an incompatibility barrier was present during protoplast formation in coenocytic algae to exclude foreign inorganic particles or alien cell components. No inorganic particles or alien cell components were incorporated into protoplast formed spontaneously in seawater. Even when the inorganic particles or alien cell and/or cell component were incorporated into protoplast in some experimental condition, they were expelled from the protoplast or degenerated within several days. A species-specific cytotoxicity was observed during protoplast hybridization between the protoplasms of Bryopsis spp. and Microdictyon umbilicatum. The cell sap of M. umbilicatum could destroy the cell components of Bryopsis spp., but had no effect on Chaetomorpha moniligera. Species C. moniligera and Bryopsis did not affect protoplast generation of either species. The wound-induced protoplast formation in vitro might have evolved in some coenocytic algae as a dispersal method, and the incompatibility barrier to alien particles or cell and/or cell component could serve as a protective mechanism for successful propagation.

방사선 조사선량에 따른 생쥐 난소의 미세구조적 연구 (Ultrastructural study of mouse ovary under X-ray irradiation)

  • 본 연구는 6 MV LINAC에서 발생한 X-선을 생쥐 생체에 조사한 후, 방사선 조사선량에 따른 난소조직의 미세구조적 변화를 고배율의 전자현미경을 이용하여 관찰하였다. 방사선 조사에 따른 난소조직의 미세구조 변화를 관찰하기 위하여 고배율의 전자현미경으로 관찰한 결과, 방사선량의 증가에 따라 성장난포의 과립층세포에서 핵과 세포질의 미세구조 변형이 급격히 증가하였으며, 난포동에서는 세포사의 부산물인 세포 잔류체들과 백혈구 및 대식세포 등이 관찰되었다. 과립층세포의 미세구조적 변형은 주로 핵의 응축에 의한 전자밀도의 증가와 핵의 분절화, 그리고 세포질의 위축 등, 전형적인 세포예정사의 특성을 나타내고 있었다. 세포의 괴사도 일부 확인되었으나 그다지 현저하지 않았으며, apoptotic body와 함께 대식세포가 산재되어 있었다.

  • The author thought that the results of cytologic tests could be changed according to the fixative methods. So the author has studied several methods of fixing exfolaiative cells collected from normal adult cheek mucosa with a solution of equal parts of ether and 95% ethylalcohol, 95 ethylalcohol and Isoprophyl alcohol, and compared the results with each other. The results were as follows : The effects of fixation, and staining efficiency of nucleus and protoplasm are similar in each method, but the staining efficiency of cytoplasm and the conversation of cytoplasmic membrane were best in 95% ethylalcohol.

Cytoskeletal changes during nuclear and cell division in the freshwater alga Zygnema cruciatum (Chlorophyta, Zygnematales)

  • Cytoskeletal changes were observed during cell division of the green alga Zygnema cruciatum using flourescein isothiocynate (FITC)-conjugated phallacidin for F-actin staining and FITC-anti-$\alpha$-tubulin for microtubule staining. Z. cruciatum was uninucleate with two star-shaped chloroplasts. Nuclear division and cell plate formation occurred prior to chloroplast division. Actin filaments appeared on the chromosome and nuclear surface during prophase, and the F-actin ring appeared as the cleavage furrow developed. FITC-phallacidin revealed that actin filaments were attached to the chromosomes during metaphase. The F-actin ring disappeared at late metaphase. At telophase, FITC-phallacidin staining of actin filaments disappeared. FITC-anti-$\alpha$-tubulin staining revealed that microtubules were arranged beneath the protoplasm during interphase and then localized on the nuclear region at prophase, and that the mitotic spindle was formed during metaphase. The microtubules appeared between dividing chloroplasts. The results indicate that a coordination of actin filaments and microtubules might be necessary for nuclear division and chromosome movement in Z. cruciatum.

  • We find ourselves living in a post-human era when technology and images coexist with humans. A cyborg, a combination of an organic body and a machine could be called a human fused with a machine. It could be understood as a new species where humans and machines have combined rather than simple protoplasm or an organic body. The purpose of this study is to demonstrate that a concept of a body in post humanism era is finding expression in fashion illustration through cyborg images. The concepts of post humanism and cyborg are analysed by means of research into literature and characteristics in which cyborg images as post humans are expressed through art, and images of a cyborg body in fashion illustration and their meanings are explored with the foregoing as a framework of reference. Cyborg image reflects new images and concept of a human in post humanism. The cyborg body images in fashion illustration are expressed through the addition of mechanical images, digital virtual body, monster image by means of hybrid combination, distorted physical transformation and fragmented body and body absence. And cyborg image in fashion illustration shows the extension of body concepts through the technology and uncertain and ambiguous identity.

Proteomic profiles and ultrastructure of regenerating protoplast of Bryopsis plumosa (Chlorophyta)

  • When a multinucleate cell of Bryopsis plumosa was collapsed by a physical wounding, the extruded protoplasm aggregated into numerous protoplasmic masses in sea water. A polysaccharide envelope which initially covered the protoplasmic mass was peeled off when a cell membrane developed on the surface of protoplast in 12 h after the wounding. Transmission electron microscopy showed that the protoplasmic mass began to form a continuous cell membrane at 6 h after the wounding. The newly generated cell membrane repeated collapse and rebuilding process several times until cell wall developed on the surface. Golgi bodies with numerous vesicles accumulated at the peripheral region of the rebuilding cell at 24 h after the wounding when the cell wall began to develop. Several layers of cell wall with distinctive electron density developed within 48-72 h after the wounding. Proteome profile changed dramatically at each stage of cell rebuilding process. Most proteins, which were up-regulated during the early stage of cell rebuilding disappeared or reduced significantly by 24-48 h. About 70-80% of protein spots detected at 48 h after the wounding were newly appeared ones. The expression pattern of 29 representative proteins was analyzed and the internal amino acid sequences were obtained using mass spectrometry. Our results showed that a massive shift of gene expression occurs during the cell-rebuilding process of B. plumosa.

  • Protoplasts were isolated from mesophyll of tobacco(Nicotiana glauca) transformed with kanamycin-resistant gene (NPT II gene) and potato hairy root callus containing Ri plasmid of Agrobacterium rhiEogenes, and protoplasm fusion was made between the isolated protoplasts. The transgenic tobacco leaf tissue could grow on the media containing high concentrations of kanamycin, but not on the phytohormone-free media. On the other hand, the potato hairy root calli could be cultured on the phytohormone-free media but not on media containing more than 40 ㎍/ml kanamycin. In these conditions, the viability of both protoplasts were above 90%, These selection markers were used for the selection of protoplasts fused between the two, i.e. protoplast fusion was detected using selection media containing 100㎍/ml kanamycin and with no phytohormone. The mixture of 1.0% cellulase, 0.3% macerozyme, and 0.7M mannitol was best for the maximum protoplast production for tobacco, and that of 2.0% cellulase, 2.0% macerozyme, 1.0% dricelase, and 0.5M mannitol for potato. Both tobacco mesophyll and potato callus protoplasts were fused by using PEG solution on the selectable medium. Cell walls were regenerated after 5 days in this medium, and colonies were alive until 4 weeks after cultural, but died after 6 weeks.

  • The extracts from Agastache rugosa O. Kuntze, their chloroform and hexane fractions, and estragole identified from hexane fraction were tested to investigate the effects on the growth and metabolic activities of several true fungi. The fungi used were: Aspergillus oryzae KFCC 890, Aspergillus niger KCCM 11240, Saccharomyces cerevisiae IAM 4597, Saccharomyces ellipsoideus PNU 2215. The growth of S. Cerevisiae by treatment of water extract(1%), hexane fraction (0.05%), and estragole (0.05%) were inhibited 93%, 50%, and 33% respectively, and S. ellipsoideus was also inhibited markedly with delaying the alg phase maximum 12 hrs. The growth of A. oryzae was inhibited by treatment of extracts and fractions. The echanol production by S. cerevisiae was increased more than two times in the highest value around 42 hrs incubation by water extract, but chloroform fraction inhibited its production. The glucoamylase actibities by A. niger were strongly inhibited by hexane and chloroform fractions (0.05%). The invertase activity by S. cerevisiae using estragole (0.05%) reached to 57.5% of control group. S. cerevisiae treated with the estragole was damaged the cell wall and cell membrane, leaked the protoplasm, and observed broken pieces of cell.

담배의 바이러스 병엽과 건전엽에 있어서의 유이아미노산에 관한 정량적 연구(예보)

  • A comparative study of free amino acid content in healthy and virus diseased tobacco leaves was carried out by author throughout the gorwing season from June to November of 1963. The methods of qualitative analysis of free amino acids applied in this experiment is followed by Moore and Stein. 1,2 Free amino acids determined in this experiment are shown in Fig. Ⅰ, Ⅱ and Table Ⅰ. As the figure and the table are shown, four more amino acids such as a spartic acid, glutamic acid, tyrosine and phenylalanine are detected in the healthy leaves; these four additional amino acids in the healthy leaves are conspicuous. More quantities of asparagine and alanine are detected in the diseased leaves than the healthy leaves and more quantities of tryptophan is detected in the healthy leaves. It is presumed that such amino acids as tyrosine and phenyllanine are decreased by the incooperation of free amino acid to TMV protein in the process of the process of the leaf protein metabolism which is caused by TMV-RNA trapping action in the diseased leaf protoplasm. It is thought that the decrease of asparagine and the increase of asparic acid in the healthy leaves are the results of in incooperaton of NH2, produced by the protein dissimilation in the diseased leaves, to aspartic acid; it's reaction is caused by the respiration of the diseased leaves accelerated by TMV attack. It is presumed, consequently, that the check of the diseased tobacco leave growth is influenced by the reduction of such amino acids as tryptophane and glutamic acid, which reduction may be due to the abnormal protein metabolism and the action of certain enzyme caused by TMV attack on host protoplast.
