Synthesis and Characterization of a Hydroxylated Dendrimeric Gene Delivery Carrier

  • Arginine conjugated PAMAM dendrimer, PAMAM-R was modified with propylene oxide via hydroxylation of primary amines of arginine residues. About 49 amines were detected to be converted to amino alcohols by 1H NMR. The newly synthesized polymer, PAMAM-R-PO was able to completely retard pDNA from a charge ratio of 2. The average diameter of PAMAM-R-PO polyplex was found to be 242 nm at a charge ratio of 30. The Zeta-potential value of PAMAM-R-PO polyplex was able to reach 20-30 mV over a charge ratio of 10. PAMAM-R-PO indicated higher cell viability than unmodified PAMAM-R on HeLa and 293 cells because of its hydroxylated amines. Transfection experiments on 293 cells showed that the transfection efficiency of PAMAM-R-PO was found to be 1.5-1.9 times higher than that of PEI25kDa at a charge ratio of 30. The polymer eventually displayed about 2 times greater transfection efficiency than PAMAM-R at the same charge ratio in the absence of serum. Therefore, we concluded that the modification of primary amines of PAMAMR to amino alcohols gives positive effects such as reduced cytotoxicity and enhanced transfection efficiency on 293 cells for gene delivery potency of PAMAM-R.

Molecular Area and Interfacial Tension Behavior of High Efficiency Cosurfactants (보조계면활성제의 계면에서의 분자면적과 계면장력 거동)

  • Gibbs' adsorption isotherms are studied to assay the structural effects of ethylene oxide (EO) and propylene oxide (PO) moieties on the molecular area and the interfacial tension behavior of molecules at the interface. Several industrial alcohols and isomerically pure alcohols, which have a general stucture of C4H9O(EO)m(PO)nH, are examined for their cosurfactant properties. They are high molecular weight alcohols and more surface active than the cosurfactants usually used. Results show that the number and the sequence of EO and PO units significantly affect the molecular areas and the interfacial tension (IFT) behavior of these molecules at the water/oil interface. The following conclusions are drawn from the result: 1) PO is more efficient in lowering the IFT and less effective in adsorption than EO. 2) For molecules having the same molecular weight but different structures, smaller molecules are more efficient in lowering the IFT. 3) When more EO, PO, or both units are added to the same hydrophobe, the molecule become bigger and more efficient in lowering the IFT.

  • Electrochemical characteristics and physical properties of polymer electrolyte which immobilized lithium salts such as $LiClO_4$ and $LiCF_3SO_3$ and plasticizers such as ethylene carbonate(EC) and propylene carbonate(PC) in high molecular weight poly(ethylene oxide)[PEO] polymer was investigated. PEO-Li based polymer electrolyte with plasticizers showed ionic conductivity of $10^{-4}S/cm$ at room temperature and high electrochemical stability up to 4.5 V(vs. $Li^+/Li$), so it can be applied to lithium secondary battery. The crystallinity of PEO decreased with the addition of lithium salts and plasticizers, especially $LiClO_4$ and PC showed more effective than and $LiCF_3SO_3$ and EC. Glass transition temperature($T_g$) of polymer electrolyte increased with increasing lithium salt concentration whereas melting temperature ($T_m$) decreased. Polymer electrolyte with plasticizers crystallized at $6^{\circ}C$.

The Cycling Performance of Graphite Electrode Coated with Tin Oxide for Lithium Ion Battery (리튬이온전지용 주석산화물이 도포된 흑연전극의 싸이클 성능)

  • Tin oxide was coated on graphite particle by sol-gel method and an electrode with this material having microcrystalline structure for lithium ion battery was obtained by heat treatment in the range $400-600^{\circ}C$. The content of tin oxide was controlled within the range of $2.25wt\%\~11.1wt\%$. The discharge capacity increased with the content of tin oxide and also initial irreversible capacity increased. The discharge capacity of tin oxide electrode showed more than 350 mAh/g at the initial cycle and 300 mAh/g after the 30th cycle in propylene carbonate(PC) based electrolyte whereas graphite electrode without surface modification showed 140 mAh/g. When the charge and discharge rate was changed from C/5 to C/2, The discharge capacity of tin oxide and graphite electrode showed $92\%\;and\;77\%$ of initial capacity, respectively. It has been considered that such an enhancement of electrode characteristics was caused because lithium $oxide(Li_2O)$ passive film formed from the reaction between tin oxide and lithium ion prevented the exfoliation of graphite electrode and also reduced tin enhanced the electrical conduction between graphite particles to improve the current distribution of electrode.

  • Effects of ethylene-propylene diene monomer (EPDM)/polypropylene (PP), zinc oxide, stearic acid, and clay on the combustive properties based on EDPM/PP were investigated. The EDPM/PP/clay nanocomposites was compounded to prepare specimen for combustive analysis by cone calorimeter (ISO 5660-1). It was found that the specific mass loss rate (SMLR) in the nanocomposites decreased due to the fire resistance compared with unfilled EDPM/PP, while the nanocomposites showed the higher total heat release (THR), higher CO production release, and higher specific extinction area (SEA) than those of virgin EPDM/PP. The stearic acid for softening ruber increased the THR and amount of smoke by itself, combustible.

  • An almost pure phase of $Ba_3Si_6O_{12}N_2$ doped with $Eu^{2+}$ was successfully synthesized through the ammonia nitridation of an oxide precursor prepared through an aqueous-solution method, using propylene- glycol-modified silane. The emission peak intensity of the obtained $Ba_3Si_6O_{12}N_2:Eu^{2+}$ was -2.2 times higher than that of the sample prepared through a solid-state reaction method.

  • In this study, three different metal-containing ionic liquid catalysts were prepared by metal insertion and characterized by various physicochemical analytic methods. The catalytic performance of the metal containing ionic liquids in the cycloaddition of $CO_2$ with propylene oxide (PO) to produce propylene carbonate (PC) was investigated under the solvent free condition. The order of approximate rate constants ($K_{app}$) for the metal containing ionic liquid catalysts was $(MeIm)_2ZnCl_2$, > $(MeIm)_2FeCl_2$ > $(MeIm)_2CuCl_2$. These results are in accord with the experimentally obtained activity order of the different metal containing ionic liquid catalysts.

Silymarin Modulates Cisplatin-Induced Oxidative Stress and Hepatotoxicity in Rats

  • Cisplatin (CDDP) is a widely used anticancer drug, but at high dose, it can produce undesirable side effects such as hepatotoxicity. Because silymrin has been used to treat liver disorders, the protective effect of silymarin on CDDP -induced hepatotoxicity was evaluated in rats. Hepatotoxicity was determined by changes in serum alanine aminotransferase [ALT] and aspartate aminotransferase [AST], nitric oxide [NO] levels, albumin and calcium levels, and superoxide dismutase [SOD], glutathione peroxidase [GSHPx] activities, glutathione content, malondialdehyde [MDA] and nitric oxide [NO] levels in liver tissue of rats. Male albino rats were divided into four groups, 10 rats in each. In the control group, rats were injected i.p. with 0.2 ml of propylene glycol in saline 75/25 (v/v) for 5 consecutive days [Silymarin was dissolved in 0.2 ml of propylene glycol in saline 75/25 v/v]. The second group were injected with CDDP (7.5 mg /kg, I.P.), whereas animals in the third group were i.p. injected with silymarin at a dose of 100 mg/kg/day for 5 consecutive days. The Fourth group received a daily i.p. injection of silymarin (100 mg/kg/day for 5 days) 1 hr before a single i.p. injection of CDDP (7.5 mg/kg). CDDP hepatotoxicity was manifested biochemically by an increase in serum ALT and AST, elevation of MDA and NO in liver tissues as well as a decrease in GSH and the activities of antioxidant enzymes, including SOD, GSHPx in liver tissues. In addition, marked decrease in serum NO, albumin and calcium levels were observed. Serum ALT, AST, liver NO level, MDA was found to decreased in the combination group in comparison with the CDDP group. The activities of SOD, GSHPx, GSH and serum NO were lower in CDDP group than both the control and CDDP pretreated with silymarin groups. The results obtained suggested that silymarin significantly attenuated the hepatotoxicity as an indirect target of CDDP in an animal model of CDDP-induced nephrotoxicity.

Conductivity properties of ion conducting polymer electrolyte based on poly(ethylene oxide) (이온전도성 poly(ethylene oxide)고분자전해질의 전도특성)

  • The purpose of this study is to research and develop solid polymer electrolyte(SPE) for Li secondary battery. We investigated the effects of lithium salts, plasticizer addition, temperature dependence of conductivity and electrochemical stability window of polyethylene oxide(PEO) electrolytes. PEO electrolyte completed with LiCIO$\_$4/ shows the better conductivity than the others. PEO-LiCIO$\_$4/ electrolyte, when EO/Li$\^$+/ ratio is 8, showed adequate conductivity around room temperature. By adding propylene carbonate and ethylene carbonate to PEO-LiCIO$\_$4/ electrolyte, its conductivity was higher than that of PEO-LiCIO$\_$4/ without those. Also PEO$\_$8/LiCIO$\_$4/ electrolyte remains stable up to 4.5V vs. Li/Li.
