  • Free radicals have long been considered damaging to various tissues. An excessive amount of reactive oxygen species (ROS) is known to have detrimental effects on the body and to be linked to numerous pathological conditions, such as cardiovascular disease, cancer, diabetes, and skeletal muscle atrophy. On the other hand, recent findings suggest that ROS is important for maintenance and development of cellular activity. Cells respond to increased oxidative stress by adaptive changes in the expression of a variety of proteins involved in the maintenance of cellular integrity. ROS is also essential for skeletal muscle function and metabolism. It is well known that physical exercise has many health benefits. Paradoxically, physical exercise also stimulates the production of ROS, which result in oxidative stress. Based on evidence amassed in the past decade, exercise itself may be considered an antioxidant because training increases the expression of antioxidant enzymes. In this review, we discuss the processes underlying the generation of ROS and its role in exercise-induced adaptation based on recent evidence. Furthermore, we discuss the possible role of NADPH oxidase in exercise-induced activation of insulin signaling and its effect on longevity.

Pervaporation of Fluoroethano1 and Methacrylic Acid Aqueous Solution Through New Acid-resistant Poly(vinyl alcohol) Membranes (새로운 내산성 PVA가교막을 이용한 불화에탄을 수용액과 메타크릴산 수용액의 투과증발분리)

  • Acid-resistant poly(vinyl alcohol) (PVA) membranes connected with ethylene and ether groups were prepared via a thermal crosslinking reaction by varying the ratio of PVA to ethylene glycol diglycidyl ether (EGDE). The crosslinked membranes were characterized using FT-IR and swelling tests, respectively. Pervaporation behaviors with the PVA membranes were investigated for aqueous TFEA (2,2,2-trifluoroethanol) as a function of EGDE content and operating temperature. The pervaporation properties far MA (methacrylic acid)/water mixture were also carried out with the optimized PVA membrane. The PVA membranes prepared with EGDE showed more excellent acid-resistance than those crosslinked with gluaraldehyde. The membranes showed high permeation fluxes of 0.1 and $0.3\;km^2h$ and high separation factors of 100 and 900 in the $96\;wt\%$ TFEA and MA aqueous fred mixtures at high temperature above $80^{\circ}C$, respectively. These confirmed theses membranes could be used in esterification membrane reactor process for the production of 2,2,2-trifluoroethylmetacrylate (TFEMA).

A Study on the Protective Effect and Its Mechanism of Zinc against Immuno-cytotoxicity of Methylmercury (유기수은의 세포면역독성과 이에 대한 아연의 방어효과 및 기전)

  • This study was carried out to elucidate the protective effect of zinc chloride(ZnCl$_2$) and its mechanism against the immuno-cytotoxicity of methylmercury chloide($CH_3$HgCl). This study was observed in the culture of EMT-6 cells which are originated from mammary adenocarcinoma of Balb/c mouse. Cytotoxicity of metals was measured by cell viability and NO$_2$$^{[-10]}$ , and mitochondrial function was evaluated by adenosine triphosohate (ATP) production. $CH_3$HgCl significantly decreased the sythesis of nitric oxide(NO), ATP and glutathione(GSH) in a dose-dependent manner. ZnCl$_2$ significantly increased the synthesis of GSH in a dose-dependent manner, but synthesis of NO and ATP were not changed. The immuno-cytotoxicity of $CH_3$HgCl was not fully protected when combined addition of ZnCl$_2$, whereas ZnCl$_2$ prior to addition of $CH_3$HgCl completly protected the Hg-induced immuno-cytotoxicity. Similarly, intracellular accumulation of mercury significantly decreased by ZnCl$_2$. Degree of diminution of intracellular mercury was larger in ZnCl$_2$ prior to addition of $CH_3$HgCl than in combined addition of ZnCl$_2$ and $CH_3$HgCl.. Dithiothreitol(DTT) or buthionine sulfoximine(BSO) addition at 50$\mu$M or less, which was not toxic to the cells, did not affect synthesis of NO and ATP. DTT increased intracellular GSH level and DTT pretreatment protected toxicity induced by $CH_3$HgCl as shown complete recover in the NO and ATP values. BSO decreased intracellular GSH level and BSO pretreatment exaggerated toxicity induced by $CH_3$HgCl as shown synergistic reduction in the NO and ATP values. These results indicated that the protective effects of zinc against immuno-cytotoxicity of methylmercury associated with increasing cellular level of GSH. Increased intracellular GSH transports methylmercury to out of cells. In accordance with intracellular level of mercury decreased, immuno-cytotoxicity of methylmercury decreased. These result also suggest that the protective mechanism of zinc against the mercury toxicity would be exerted in the immune system in vivo.

  • The Plastic material had been developed in the middle of the 19th century as an alternative material. Along with the development of the electrical engineering industry, it cropped up into center stage as an effective material and has increasingly expanded its use. As such, the plastic material has bound deep-seated ties with the design activities of industrial products, coming into the main material for a variety of industrial designs. Despite its dose affinity to design function in terms of its materialistic property, we have rarely seen examples of intensive study on the realtionship between plastics and designs. This study aims to find the importance that the substance affects industrial products in designs along with the development of plastic materials. With the objective in mind, we made a review of the streamline stylishness that had flourished in the twenties and thirties of the 19th century. Through this study, we understand that the plastic material has a close realtionshop with design activities in three different aspects. First, its amorphous state of nature makes it possible to change into any shape one desires in plastic surgery, which feature in turn influences the moulding of any design forms. Second, the plastic material is best suited to mass-manufacture, which induces to reduce the cost of production. Hence, the expansion of design industry. Third, the plastic material allows the multiple variety of colors, sensitivity, gloss and patterns and infinitely large possibility ranging from natural senses to human senses with the result that numberless diversity of designs cdould come into being.

Study of an ER bound p80 Homologous to Nucleolar B23 (핵소체 단백 B23과 세포질 단백 p80의 유사성에 관한 연구)

  • Protein B23 is one of the major nucleolar phosphoproteins associated with pre-ribosomal particles, and is localized in the granular region of the nucleolus. Recent studies suggest that protein B23 shuttles between nucleus and cytoplasm and also interacts with HIV Rev. These findings indicate that protein B23 is important in nucleocytoplasmic relationship and viral replication. However, the exact function of protein B23 is not clear yet. In acute nucleolar hypertrophy of rat liver, treated with thioacetamide, there was observed an increase of not only protein B23 but also B23-like protein p45 when anti-B23 monoclonal antibody (MAb) was used for identification. On the basis of the large B23 specific epitope structure composed of 68 amino acids, a hypothesis was formulated to examine that p45 is the pre-B23 resulting from excessive production of B23. In an attempt to investigate the precursor of B23, we analyzed the subcellular fractions and microsomal subfractions. Subsequently, we analyzed the finger printings of B23-like proteins using the tryptic peptide mapping. The results are summarized: 1) Using B23 MAb, we observed the presence of B23-like proteins in nucleolar fraction, nucleoplasmic fraction and microsomal fraction. 2) In the further microsomal subfractionation, we could partially purify B23-like protein in 2M layer of sucrose gradient. 3) When ion exchange chromatography was employed, there were protein species 80kDa(p80), 65kDa(p65) and 60kDa(p60). 4) Based on the tryptic map analysis of $^{125}I$ labeled proteins, the similarity between B23 and p80 was found only in 9 out of 14(B23) and 21(p80) peptides, and difference was found in the remaining peptides. p80 and p60 had 18 common peptides, and all the peptides of p60 were similar to those of p80. From these results, it is proposed that p45 is an abnormal metabolite resulting from carcinogenesis by thioacetamide, and it is not the precursor of B23. In addition, we suggest that p80 may be a precursor of p45.

Dispersion Characteristics of Hydrogen Gas by the Effect of Leakage Hole Size in Enclosure Space (누출공 크기에 따른 밀폐공간 내 수소 가스의 확산 특성)

  • As a potential clean energy resource, the production and consumption of hydrogen gas are expected to gradually increase, so that hydrogen related studies are also increasing. The thermal and chemical properties of hydrogen result in its high flammability; in particular, there is a high risk if leaks occur within an enclosed space. In this study, we applied the computational fluid dynamics method to conduct a numerical study on the leakage behavior of hydrogen gas and compared these numerical study results with an experimental study. The leakage hole diameter was selected as an important parameter and the hydrogen gas dispersion behavior in an enclosed space was investigated through various analytical methods. Moreover, the flammable regions were investigated as a function of the leakage time and leakage hole size. We found that the growth rate of the flammable region increases rapidly with increasing leakage hole size. We also investigated the relation between the mass flow rate and the critical time when the hydrogen gas reaches the ceiling. The analysis of the monitoring points showed that the hydrogen gas dispersion behavior is isotropic and independent of the geometry. We found that the concentration of gas in an enclosed space is affected by both the leakage flow rate and amount of gas accumulated in the enclosure.

  • This study was designed to investigated the effects of cadmium(Cd) feeding on the humoral immune responses such as PFC-responses and production of immunoglobulins in BALB/c mice. The results obtained were summarized as follows; 1. Total PFCs of direct IgM antibody response were significantly decreased in all Cd-fed goups, whereas total PFCs of IgG antibody response were slightly increased. 2. In secondary immunization, total HA titers were increased in all Cd groups as compared with control, especially in 100ppmm group and also IgG titers were slightly increased except for 50ppm group. 3. The levels of $IgG_1$ were increased to 5.5% 18.7%, 17.4% and 7.2% in 25, 50, 100 and 200ppm groups as compared with control, respectively. And also the levels of IgE were increased to 5.7%, 7.3%, 8.7% and 0.4% in those of Cd groups, in order. Conclusively, concentrations of $IgG_1$, and IgE were increased in all Cd groups. Based on the results of this study and previous report, it was shown that Cd might affect humoral immune responses by modifying the distribution and function of cells playing in the cellular immune response.

Improvement Effect of Non-alcoholic Fatty Liver Disease by Curcuma longa L. Extract (강황 추출물의 비알코올성 지방간 질환 개선 효과)

  • Background: Non-alcoholic fatty liver disease (NAFLD) is a chronic liver disease associated with multiple metabolic disorders. The medicinal plant Curcuma longa L. is widely distributed in Asia and has been used to treat a spectrum diseases in clinical practice. To date, there are inadequate reports of the effects of C. longa 50% EtOH extract (CE) on NAFLD. Therefore, in this study, we evaluate the CE on an NAFLD animal and elucidate the mechanism of action. Methods and Results: C57BL/6J mice fed a methionine-choline deficient diet (MCD) were treated with CE or milk thistle, and changes in inflammation and stetosis were assessed. Experimental animals were divided into six group (n = 10); Normal, MCD, MCD + CE 50 mg/kg/day (CE 50), MCD + CE 100 mg/kg/day (CE 100), MCD + CE 150 mg/kg/day (CE 150), and the Control, MCD + Milk thistle 150 mg/kg/day (MT 150). Body weight, liver weight, liver function, and histological changes were assessed in experimental animals. Quantitative real-time polymerase chain reaction and western blot analyses were performed on samples collected after 4 weeks of treatment. We observed that CE administration improved MCD-diet-induced lipid accumulation, and triglyceride (TG) and total cholesterol (TC) levels in serum. Treatment with CE also decreased hepatic lipogenesis through modulation of the sterol regulatory element binding protein-1 (SREBP-1), CCAAT-enhancer binding protein α (C/EBPα), fatty acid synthase (FAS), and peroxisome proliferator-activated receptor γ (PPARγ) expresion. In addition, the use of CE increased adenosine monophosphate-activated protein kinase (AMPK) phosphorylation and inhibited the up-regulation of toll-like receptor (TLR)-2 and TLR-4 signaling and the production of inflammatory mediators. Conclusions: In this report, we observed that CE regulated lipid accumulation in an MCD dietinduced NAFLD model by decreasing lipogenesis. These data suggeste that CE could effectively protect mice against MCD-induced NAFLD, by inhibiting the TLR-2 and TLR-4 signaling cascades.

Proteome Profiling of Murine Macrophages Treated with the Anthrax Lethal Toxin (탄저 치사독소 처리에 의한 생쥐 대식세포의 단백질체 발현 양상 분석)

  • Intoxication of murine macrophages (RAW 264.7) with the anthrax lethal toxin (LeTx 100 ng/ml) results in profound alterations in the host cell gene expression. The role of LeTx in mediating these effects is unknown, largely due to the difficulty in identifying and assigning function to individual proteins. In this study, we have used two-dimensional polyacrylamide gel electrophoresis to analyze the protein profile of murine macrophages treated with the LeTx, and have coupled this to protein identification using MALDI-TOF mass spectrometry. Interpretation of the peptide mass fingerprint data has relied primarily on the ProFound database. Among the differentially expressed spots, cleaved mitogen-activated protein kinase kinase (Mek1) and glucose-6-phosphate dehydrogenase were increased in the LeTx treated macrophages. Mek1 acts as a negative element in the signal transduction pathway, and G6PD plays the role for the protection of the cells from the hyper-production of active oxygen. Our results suggest that this proteomic approach is a useful tool to study protein expression in intoxicated macrophages and will contribute to the identification of a putative substrate for LeTx.

Zearalenone Affects Immune-Related Parameters in Lymphoid Organs and Serum of Rats Vaccinated with Porcine Parvovirus Vaccine

  • Rats were administered zearalenone (ZEA) via gavage at dosages of 0, 1, 5, and 30 mg/kg for 36 days. On treatment day 8, inactivated porcine parvovirus vaccine (Vac) was injected intraperitoneally. Antibody production against porcine parvovirus was then measured as a function of ZEA treatment. Compared to the vaccine alone, ZEA treatment, with or without Vac, decreased the serum level of IgG. The level of IgM decreased in all ZEA groups at day 22, but the decrease was sustained only in the medium-dose ZEA group at day 36. The level of IgA was unchanged in the Vac only and ZEA groups at day 22, but was decreased in the 5 mg/kg ZEA plus Vac group compared to the Vac only group at day 36. The level of IgE was decreased by all doses of ZEA at day 22, but was unaffected in ZEA plus Vac groups compared to the Vac only group. The levels of IL-1 in the thymus and spleen; INF-${\gamma}$ in serum; IL-2, IL-6, and IL-10 in the thymus; and IL-10 and IFN-${\gamma}$ in the spleen decreased after ZEA administration. Furthermore, the levels of IL-$1{\beta}$ in the spleen and mesenteric lymph node, IL-$1{\beta}$ in the thymus, IL-2 in the thymus and spleen, IL-6 in the thymus, IL-10 and IFN-${\gamma}$ in the spleen, and GM-CSF and TNF-${\alpha}$ in the thymus decreased after vaccination in rats exposed to ZEA. In conclusion, these results suggest that ZEA exposure via drinking water can cause an immunosuppressive effect by decreasing immunoglobulins in serum and cytokines in lymphoid organs.
