Anti-inflammatory and Wrinkle Improvement Effects of Peptides from Ginseng Berry Amino Acidic Complex (진생베리 아미노산 복합체로부터 분리한 펩타이드의 항염, 주름개선 효과)

  • Ginseng berry (GB) contains Ginsenoside Re and has anti-inflammatory and anti-wrinkle properties. In this study, TLC fractions 1, 2, and 4 of the ginseng berry amino acid complex were identified and analyzed by HPLC. And identified a peptide (AP-1) by LC/MASS analysis of fraction 1. The anti-inflammatory activity was confirmed by investigating the inhibitory effect of AP-1 on NO production. In addition, collagen synthesis using procollagen type I C-peptide (PIP) ELISA kit was 50% higher effective than that of the control group. From these results, the peptide isolated from ginseng berry amino acid complex is considered to have anti-inflammatory and anti-wrinkle effect, and may be useful as an anti-inflammatory and anti-aging cosmetic raw material.

  • Type I (collagen) and procollagen are reduced in aged human skin. This reduction could result from increased degration by metalloproteinases and from reduced procollagen synthesis and skin collagenase is required for initiation of the degration of type I collagen. In the present study, we study on assay the collagen and collagenase in natural plants using the fibroblast human skin cell. We select the 15 kind of plants used to herbal and 4 kind of fraction(by methylene chloride, ethyl acetate, n-butanol, water). Among these extract, the ethyl acetate fraction from benincasa hispida/prunus persica, trichosanthes kiriowii, trogopterus xanthipes and methylene chloride fractions from benincasa hispida/prunus persica, torilis japonica and n-butanol fraction from cnidium officinale, chrysanthemum sibiricum were selected for further experiments as they exhibited distinctive amount of collagen compared to other natural extracts. These extracts were again subjected to collagenase assay test. Benincasa hispida/prunus persica extract was shown to have exellent collagen synthesis activity from result of the collagen assay test and the other extract was shown to have over 130% of collagen synthesis activity. But, in the study of collagenase assay test just only trogopterus xanthipes extract was shown to have collagenase inhibition.

Protective Effects of Portulaca oleracea L. Extract against Matrix Metalloproteinase Production and Reactive Oxygen Species Generation Induced by Ultraviolet B Radiation in Human Keratinocytes (쇠비름 추출물의 UVB 자외선 조사에 의한 인간각질형성세포 손상에 대한 보호 효과)

  • Portulaca oleracea L. is an edible plant widely consumed in daily diet throughout Europe, Asia and America. In this study, protective effects of P. oleracea L. extracts against oxidative stress and matrix metalloproteinase (MMP) activity induced by ultraviolet B (UVB) radiation were investigated using HaCaT immortal human keratinocytes. In this context, the mRNA and protein productions of MMPs (MMP-1, -2, and -9) and type I procollagen, which are major markers of photoaging induced by UVB radiation in HaCaT keratinocytes, were evaluated. Furthermore, UVB-induced reactive oxygen species (ROS) generation and mRNA and protein expression levels of superoxide dismutase-1 (SOD-1), oxygenase-1 (OH-1), and nuclear factor-erythroid 2-related factor-2 (Nrf-2), all of which are associated with the antioxidant balance, were investigated. As shown by the results, UVB radiation induced ROS formation and led to increased production of MMPs and decreased collagen production in human keratinocytes, which resulted in skin photoaging or photodamage. The treatment with P. oleracea L. extracts downregulated MMP (MMP-1, -2, and -9) production and upregulated type I procollagen expression in UVB-induced HaCaT cells. Furthermore, treatment with the extracts decreased UVB-induced ROS generation and increased the expression of antioxidant enzymes, such as SOD-1 and OH-1, through the Nrf-2 pathway. Taken together, these results suggest that P. oleracea L. extracts could be a potential cosmeceutical agent for the prevention of skin photoaging or photodamage.

Skin Anti-aging Effect of Forsythia viridissima L. Extract (연교추출물의 피부 항노화 효과)

  • Skin anti-aging effect of Forsythia viridissima L. extract was evaluated by using antioxidant assay, expression of type I procollagen, and UVA-induced matrix metalloproteinase-1 in human dermal fibroblasts. Matairesinol-rich Forsythia viridissima L. extract was showed the scavenging activity of radicals and reactive oxygen species with the $IC_{50}$ values of $4.50\;{\mu}m/ml$ against 1,1-diphenyl-2-picrylhydrazly radical and $542.43\;{mu}m/ml$ against superoxide radicals in the xanthine/xanthine oxidase system, respectively. The type I procollagen was increased 33.76% by treatment with matairesinol-rich Forsythia viridissima L. extract, and UVA-induced MMP-1 was reduced 35.78% in a dose dependent manner. In the human skin irritation test, 2% matairesinol-rich Forsythia viridissima L. extract did not show any adverse effect. Also, the clinical study indicated that a cream group treated with 0.2% matairesinol-rich Forsythia viridissima L. extract significantly reduced skin wrinkles, as compared with a non-treated cream group (p < 0.05). These results suggest that Forsythia viridissima L. extract may be useful as a potential source of functional anti-aging cosmetics.

Effect of Torilis Fructus on Procollagen Biosynthesis and Activity of Matrix Metalloproteinase-I(MMP-1) in Human Dermal Fibroblast (사상자(Torilis Fructus)가 섬유아세포의 Procollagen 생합성과 Matrix metalloproteinase-I(MMP-1)의 활성에 미치는 영향)

  • Skin wrinkle formations are associated with collagen synthesis and matrix metalloproteinase-I(MMP-1) activity. This study was carried out to find out skin wrinkle reducing components in Torilis Fructus. Torilis Fructus were extracted with 70% ethanol and the ethanol extracts were systematically fractionated with n-hexane, ethylacetate, n-butanol and distilled water. Among them, antiwrinkle component from n-hexane fraction was purified by several column chromatographies and HPLC, which identified as torilin by $^1H-NMR,\;^{13}C-NMR$ and ESI-MS. To determine cell viability, collagen biosynthesis and MMP-1 activity, human dermal fibroblast was treated with 1-5 ppm concentrations of Torilis Fructus extract fraction and torilin. Cell viability was showed 84-102% at all group treated with 1-5 ppm. Collagen synthesis was increased in all group, especially torilin-treated group was highest amount. Active forms of MMP-1 were decreased in all group. From these results, we consider that Torilis Fructus have several antiwrinkle components and torilin may be one of the effective components.

Effects of Vegetable Peptones on Promotion of Cell Proliferation and Collagen Production (Vegetable Peptones의 세포증식 및 콜라겐생성 촉진효과)

  • Skin aging appears to be principally attributed to a decrease in both levels of Type I collagen and regeneration ability of dermal fibroblasts. It is important to introduce an efficient and safe agent for effective management of skin aging. To this end, we performed screening for anti-ageing agents and then found that vegetable peptones (pea and wheat) promoted cell proliferation of adult stem cells. Vegetable peptones may be considered as useful medium additives because it can supply nutrients, peptides, amino acids or growth factor analogues. This study was designed to investigate effects of vegetable peptones on cell proliferation/collagen production and their possible mechanisms in human dermal fibroblasts. In cell proliferation assay, vegetable peptones significantly promoted cell proliferation in a concentration-dependent manner. In addition, human COL1A2 promoter luciferase and type I procollagen synthesis assays showed that vegetable peptones induce type I procollagen production through the activation of COLlA2 promoter. In both TGF-${\beta}1$ luciferase reporter and ELISA assays, vegetable peptones was found to induce TGF-${\beta}1$ production, suggesting that vegetable peptones induce type I procollagen production through the activation of TGF-${\beta}1$. When applied topically in a human skin twice a day for an 4-week period of time, vegetable peptones did not induce any adverse reactions. Theretore, based on these results, we suggest the possibility that vegetable peptones may be considered as an attractive, wrinkle-reducing candidate for topical application.

Anti-oxidant and Anti-aging Activities of Sericinjam Gland Hydrolysate Extract in Human Dermal Fibroblasts (사람 섬유아세포에서 세리신잠 실샘가수분해물(Sericinjam Gland Hydrolysate)의 항산화 및 항노화 효과)

  • We studied the anti-oxidant and anti-aging activities of Sericinjam Gland Hydrolysate (SJGH) in the human dermal fibroblasts. SJGH effectively defended cell death and ROS generation under high H2O2 in human dermal fibroblasts. Moreover SJGH reduced the expression of SA-${\beta}$-Gal and MMP-1 under low concentration of $H_2O_2$ whereas biosynthesis of procollagen-I was increased. This results demonstrate the anti-oxidant and anti-aging activities of SJGH. SJGH could be a good candidate for anti-aging cosmetics ingredient.

The Effects of Polydatin Isolated from Polygonum cuspidatum on Melanogenesis and Wrinkle Formation (호장으로부터 분리한 Polydatin의 미백 및 주름억제 효능에 대한 연구)

  • 5,4'-dihydroxystilbene-3-O-${\beta}$-D-glucopyranoside (Polydatin) is one of the stilbenes found in Polygonum cuspidatum (P. cuspidatum), however, the effects of polydatin on skin biology remain to be elucidated. In this study, we obtained polydatin from P. cuspidatum and investigated the effects of polydatin in skin-derived melanocytes and fibroblasts. In melanocytes, polydatin inhibited not only the tyrosinase activity and melanin production but the expression of melanogenic factors, tyrosinase and microphthalmia-associated transcription factor (MITF). In addition, the level of type I procollagen in fibroblasts was analyzed, and polydatin significantly induced the production of type I procollagen in a dose-dependent manner. Finally we conformed that topical treatment of polydatin improved wrinkle and induced whitening of human skin in vivo. These data provide evidence that polydatin can be a potent candidate for the improvement of both skin wrinkle and whitening from the point of industry view.

Anti-wrinkle Effect of Safflower (Carthamus tinctorius L.) Seed Extract (II) (홍화씨추출물의 피부 주름개선 효과(II))

  • Phytoestrogens derived from plants and foods, which are diphenolic compounds with structural similarities to natural and synthetic estrogens, have been shown to estrogenic and antiestrogenic actions. Particularly, recent study revealed that phenolic compounds in safflower seed, such as serotonin derivatives, lignans and flavonoids, could be acted as phytoestrogens. Safflower (Carthamus tinctorius L.) seed extract (SID C.SE), therefore, are receiving a renewed interest as potential therapeutic source against skin wrinkles induced by estrogen deficiency. This study was conducted to investigate the anti-wrinkle effect of SID C.SE on normal human fibroblasts through the expression of type I procollagen and UVA-induced MMP-1 in vitro. The SID C.SE increased the type I procollagen expression, comparable to trans-retinol and reduced UVA-induced MMP-1 expression in a dose-dependent manner. The clinical study indicated that cream group treated with $0.1\%$ SID C.SE significantly reduced a skin wrinkles, as compared with a control (non-treated cream group) (p<0.05). These results suggest that the safflower seed extract may be useful as potential source of anti-wrinkle cosmetics.

Anti-aging Effects of Prescription Extracts Containing Forsythia viridissima L. (연교를 함유한 처방단 추출물의 항노화 효과)

  • Forsythia fructus has been shown to have antioxidative, anti-inflammatory, antibacterial, anti-aging and whitening effects. This work was carried out to investigate the anti-aging effects of Forsythia viridissima-prescription extracts (Yeongyoseungma-tang, Gamiyeongyoseungma-tang, Hoechunyangkyeok-san) on skin. Skin anti-aging effect of Forsythia viridissima-prescription extracts (Yeongyoseungma-tang, Gamiyeongyoseungma-tang, Hoechunyangkyeok-san) was evaluated by using antioxidant assay, anti-glycation activity, inhibitory effect of tyrosinase activity, expression of type I procollagen and UVA-induced matrix metalloproteinase-1 on HS68 cells, and reduction of ${\alpha}$-MSH-induced melanin on B16F1 cells. Forsythia viridissima-prescription extracts showed anti-oxidative and anti-glycation activity. The pectinex hydrolysed extract from Yeongyoseungma-tang 75% EtOH extract and Gamiyeongyoseungma-tang 75% EtOH extract increased the type I procollagen synthesis, and decreased the UVA-induced MMP-1 expression on HS68 cells. The pectinex hydrolysed extracts of Hoechunyangkyeok-san 75% EtOH extract had the inhibitory effect of melanin synthesis on B16F1 cells. Based on these results, we suggest that Forsythia viridissima-prescription extracts may be useful as a potential source of functional anti-aging cosmetics.
