Membrane and Virus Filter Trends in the Processes of Biopharmaceutical Production (바이오의약품 제조공정에서 분리막의 역할과 바이러스 필터 동향)

  • Membranes are used in most processes of biopharmaceutical production. It is used for pretreatment of other processes, separation of impurities in the process, virus removal, control of products concentration and buffer solution exchange. Virus filters play an important role in ensuring product efficacy and stability because viral contamination of biopharmaceuticals for humans is a sensitive issue that is directly related to serious clinical outcomes. Virus filters typically have complex multilayer structures made of various polymers such as surface-modified PVDF, PES, CRC. Depending on the manufacturer, filters have different pore structures and shapes, such as symmetric or asymmetric, and is used in the form of pleated membrane, flat sheets or hollow fibers. Virus filters are exclusively supplied by few foreign companies such as Asahi Kasei, Millipore, Pall and Sartorius. Replacing virus filters can be time consuming and expensive, including approval from regulatory agencies through validation. As localization has become important due to Japan's recent export regulations, it is necessary to increase the degree of technical independence.

Baicalin, baicalein and wogonin inhibits high glucose-induced vascular inflammation in vitro and in vivo

  • Vascular inflammatory process has been suggested to play a key role in initiation and progression of atherosclerosis, a major complication of diabetes mellitus. Thus, in this study, we attempted to determine whether three structurally related polyphenols found in the Chinese herb Huang Qui, namely baicalin, baicalein, and wogonin, can suppress vascular inflammatory processes induced by high glucose (HG) in human umbilical vein endothelial cells (HUVECs) and mice. Data showed that HG induced markedly increased vascular permeability, monocyte adhesion, expressions of cell adhesion molecules (CAMs), formation of reactive oxygen species (ROS) and activation of nuclear factor (NF)-κB. Remarkably, all of the above mentioned vascular inflammatory effects of HG were attenuated by pretreatment with baicalin, baicalein, and wogonin. Vascular inflammatory responses induced by HG are critical events underlying development of various diabetic complications, therefore, our results suggest that baicalin, baicalein, and wogonin may have significant therapeutic benefits against diabetic complications and atherosclerosis.   • Because of their simplicity, efficiency, and economy, slow sand filters are appropriate means of water treatment for small water systems. In this study, the effect of filtration velocity and dirty skin (Schmutzdecke) was evaluated on the performance of turbidity removal. Also, removal characteristics of particulate were investigated in the case of the usage of non-woven fabric on the surface of sand and the application of PCF as pretreatment process. Comparative column tests were carried out for the various operation condition. From the result of column tests, filtration velocity had little effect on the turbidity removal rate. The formation of algal biofilm on the surface of media is helpful in turbidity removal, while non-woven fabric is not as effective as expected. The relative contribution of biomass and accumulated particulates to head loss development in slow sand filters requires further study.

Study on Pulp Fibers and Paper Morphology by ESEM and LTSEM

  • The ESEM could be used in investigating the fibrous networks developed during handsheet-forming processes with the exception of the stages relating to the actual dispersion of the fibers and the drying of formed sheets. Also the cross-sectional images of swollen fibers were generated with the ESEM but the information given by the images was rarely fresh compared to the CLSM images. The LTSEM was extremely useful in generating images of the microfibrillar structure of a wet fiber with great resolution. However, pretreatment required in the LTSEM chamber was somewhat tedious due to the time consumed in sublimation of ice and sputter coating. For observation of lamellar structure of a hydrated fiber, the LTSEM exhibited greatly detailed structure with high resolution. Finally ESEM and LTSEM should be used in a finite field such as observation of surface morphology in detail.

  • Carbon dioxide fixation by microalgae has several advantages such as no requirements for the pretreatment over phycal and/or chemical $CO_2$ fixation processes. Chlorella sp. HA-1, fresh algae. and Chlorococcum littorale. marine algae, were used to do $CO_2$ fixation work. The experimental parameters were light intensity and $CO_2$ concentration. Chlorella sp. HA-1 has the maximum growth rate at 8 Klux and 10% $CO_2$ concentration. Chlorococcum littorale showed the maximum growth at similar conditions. The results indicated the feasibility of the Chlorella HA-1 and Chlorococcum littorale for mass cultivation using flue gas.

  • Reverse osmosis(R/O), ultrafiltration(UF) and microfiltration (MF) processes are widely used for water treatment. In the seminar, characteristics of typical membrane modules including tubular, hollow fiber, plate and frame and spiral wound types will be discussed in detail. The design methods based on hydrodynamics for hollow fiber and spiral wound modules will be introduced analytically. Concentration polarization (CP) and membrane fouling mechnism as well as the techniques for CP reduction will be handled. The CP control techniques contain chemically modified membrane surface, pretreatment of feed water, operation of low trans-membrane pressure, chemical or physical cleaning methods and artificial production of various fluid turbulences near the membrane surface, etc. In especially, the recent commercial membrane modules for CP control including module rotation, vibration and Taylor or Dean vortex system will be introduced and discussed in detail.

Development of An Eco-friendly Surface Treatment Process for the Design of the Al Lead Tab in Lithium-ion Batteries

  • With the recent popularity of mobile devices, the demand for lithium-ion batteries is increasing. In this study, the surface treatment process for the development of the Al (aluminum) lead tab for positive electrode, a key component of the pouch-type lithium-ion battery, was investigated. Anodizing and sealing processes were tested as surface treatment techniques. It was found that only a sealing process is needed to obtain sufficient adhesive strength. In the present study, an adhesive strength of 17 N/12 mm was achieved by degreasing and etching pretreatment, followed by a sealing process of 10 min duration. This adhesive strength was greater than that achievable using Cr (chromium) surface treatment. Using various surface analysis techniques, the shape and composition of the surface before and after being subjected to the surface treatment were compared and analyzed. The results of this study are expected to contribute to the development of an eco-friendly lead tab.

  • At the present time the desalination technology of sea water for portable water in islands employs the RO method. The technology which needs complicated pretreatment processes with various chemicals can generate secondary water pollution and the high maintenance costs such as replacements of filters and membranes make islanders nearly impossible to operate. The MVRS technology for desalination of sea water however has several advantages such as constant production of quality portable water and capability of managing broad operating load. The variable-speed turbo-type vapor compressors employed in the system can utilize wind energy which is abundant in most Korean islands. Salt as a by-product can be produced by applying solar energy to the salt-concentrated waste water from the system. This paper discusses the relating topics such as technical and economical viabilities of the new MVRS desalination system for the production of portable water and salt as a by-product using new & renewable sources of energy.

  • Recently, to avoid global warming, reduction of $CO_2$ generation has been demanded. Each car manufacture takes positive actions for improvement in fuel consumption by various kinds of countermeasures, e.g. improving engine efficiency, using alternative fuels, lightening the car body. Regarding lightening of the car body, aluminum alloys and high tensile strength steels has been gradually adopted as lightweight materials. However, such materials are normally not easy to be treated with zinc phosphate system. Focusing the pre-treatment processes, low temperature phosphate system could be demanded for energy saving. This time, new surface conditioning process for lightweight materials and low temperature phosphate system shall be discussed.

  • Plasma membrane blebs are observed in many types of apoptotic cells, but their processes of formation remain to be clarified. In the present study, we investigated whether there is a relationship between change of intracellular phosphotyrosine levels and biochemical apoptotic events in Jurkat T cells undergoing apoptosis by agonistic anti-Fas antibody. When Jurkat cells were treated with Fas-antibody in the presence or absence of pretreatment with sodium orthovanadate ($Na_3${VO}_4$), a phosphotyrosine phosphatase (PTPase) inhibitor, membrane blebs disappeared in orthovanadate-treated cells.
