Identification of new major histocompatibility complex-B Haplotypes in Bangladesh native chickens

  • Objective: The major histocompatibility complex in chicken demonstrates a great range of variations within varities, breeds, populations and that can eventually influence their immuneresponses. The preset study was conducted to understand the major histocompatibility complex-B (MHC-B) variability in five major populations of Bangladesh native chicken: Aseel, Hilly, Junglefowl, Non-descript Deshi, and Naked Neck. Methods: These five major populations of Bangladesh native chicken were analyzed with a subset of 89 single nucleotide polymorphisms (SNPs) in the high-density MHC-B SNP panel and Kompetitive Allele-Specific polymerase chain reaction genotyping was applied. To explore haplotype diversity within these populations, the results were analyzed both manually and computationally using PHASE 2.1 program. The phylogenetic investigations were also performed using MrBayes program. Results: A total of 136 unique haplotypes were identified within these five Bangladesh chicken populations, and only one was shared (between Hilly and Naked Neck). Phylogenetic analysis showed no distinct haplotype clustering among the five populations, although they were shared in distinct clades; notably, the first clade lacked Naked Neck haplotypes. Conclusion: The present study discovered a set of unique MHC-B haplotypes in Bangladesh chickens that could possibly cause varied immune reponses. However, further investigations are required to evaluate their relationships with global chicken populations.

  • In the electronics industry, a lot of research and development is being conducted on electronic component supply, component alignment and insertion, and automation of soldering on the back side of the PCB for automatic PCB assembly. Additionally, as the use of electronic components increases in the automotive component field, there is a growing need to automate the alignment and insertion of components with leads such as transistors, coils, and fuses on PCB. In response to these demands, the types of PCB and parts used have been more various, and as this industrial trend, the quantity and placement of automation equipment that supplies, aligns, inserts, and solders components has become important in PCB manufacturing plants. It is also necessary to reduce the pre-setting time before using each automation equipment. In this study, we propose a method in which a vision system recognizes the type of component and simultaneously corrects alignment errors during the process of aligning and inserting various types of electronic components. The proposed method is effective in manufacturing various types of PCBs by minimizing the amount of automatic equipment inserted after alignment with the component supply device and omitting the preset process depending on the type of component supplied. Also the advantage of the proposed method is that the structure of the existing automatic insertion machine can be easily modified and utilized without major changes.

Comparison Study of Water Tension and Content Characteristics in Differently Textured Soils under Automatic Drip Irrigation (자동점적관수에 의한 토성별 수분함량 및 장력 변화특성 비교 연구)

  • Maintenance of adequate soil tension or content during the period of crop growth is necessary to support optimum plant growth and yields. A better understanding of soil tension and content for precision irrigation would allow optimal soil water condition to crops and minimize the adverse effects of water stress on crop growth and development. This research reports on a comparison of soil water tension and content variations in differently textured soils over time under drip irrigation using two different water management methods, i.e. pulse time and required water irrigation methods. The pulse time-based irrigation was performed by turning the solenoid valve on and off for preset times to allow the wetting front to disperse in root zone before additional water was applied. The required water estimation method was a new water control logic designed by Rural Development Administration that applies the amount of water required based on a conversion of the measured water tension into water content. The use of the pulse time irrigation method under drip irrigation at a high tension of -20 kPa and high temperatures over $30^{\circ}C$ was not successful at maintaining moisture tensions within an appropriate range of 5 kPa because the preset irrigation times used for water control could not compensate for the change in evapotranspiration during day and night. The response time and pattern of water contents for all of the tested soils measured with capacitance-based sensor probes were faster and more direct than those of water tensions measured with porous and ceramic cup-based tensiometers when water was applied, indicating water content would be a better control variable for automatic irrigation. The required water estimation-based irrigation method provided relatively stable control of moisture tension, even though somewhat lower tension values were obtained as compared to the target tension of -20 kPa, indicating that growers could expect to be effective in controlling low tensions ranging from -10 to -20 kPa with the required water estimation system.

  • Proton exchange membrane fuel cell(PEMFC) performance could be affected by various factors such as cell temperature, total pressure, partial pressure of reactants and relative humidity. Hydrogen ion is combined with water to form hydronium ion [$H_3O^+$] and pass through membrane resulting electricity generation. Cooling system is needed to remove heat and other uses on large scale fuel cell. In case that collant conductivity is increased, fuel cell performance could be decreased because produced electricity could be leaked through coolant. In this study, triple distilled water(TDW) and antifreeze solution containing ethylene glycol was used to observe resistance change. Resistance of TDW was taken 28 days to reach preset value, and effect on fuel cell operation was not observed. Resistance of antifreeze solution was not reached to preset value up to 48 days, but performance failure occurred presumably caused by bipolar plate junction resulting stoppage resistance experiment. Generally PEMFC humidification is performed near-saturated operating conditions at various temperatures and pressures, but non-humidifying condition could be applied in small scale fuel cell to improve efficiency and reduce system cost. However, it was difficult to operate large scale fuel cell without humidifying, especially higher than $50{\sim}60^{\circ}C$. In case of small flux such as 0.78 L/min, temperature difference between inlet and outlet was occurred larger than other cases resulting performance decrease. Non-humidifying performance experiments were done at various cell temperature. When both of anode and cathode humidification were removed, cell performance was strongly depended on cell operating temperature.

  • Principles of a novel pulse growing method are described. The method realized in the crystal growing on a seed from melts under raw melt feeding provided a more reliable control of the crystallization process when producing large alkali halide crystals. The slow natural convection of the melt in the crucible at a constant melt level is intensified by rotating the crucible, while the crystal rotation favors a more symmetrical distribution of thermal stresses over the crystal cross-section. Optimum rotation parameters for the crucible and crystal have been determined. The spatial position oi the solid/liquid phase interface relatively to the melt surface, heaters and the crucible elements are considered. Basing on that consideration, a novel criterion is stated, that is, the immersion extent of the crystallization front (CF) convex toward the melt. When the crystal grows at a <> CF immersion, the raised CF may tear off from the melt partially or completely due to its weight. This results in avoid formation in the crystal. Experimental data on the radial crystal growth speed are discussed. This speed defines the formation of a gas phase layer at the crystal surface. The layer thickness il a function of time a temperature at specific values of pressure in the furnace and the free melt surface dimensions in the gap between the crystal and crucible wall. Analytical expressions have been derived for the impurity component mass transfer at the steady-state growth stage describing two independent processes, the impurity mass transfer along the <> path and its transit along the <> one. The heater (and thus the melt) temperature variation is inherent in any control system. It has been shown that when random temperature changes occur causing its lowering at a rate exceeding $0.5^{\circ}C/min$, a kind of the CF decoration by foreign impurities or by gas bubbles takes place. Short-term temperature changes at one heater or both result in local (i.e., at the front) redistribution of the preset axial growth speed.

  • 결핵성 병변의 단순 x-ray 촬영이나 CT, MRI 소견은 매우 다양하며, 결핵과 전이암 혹은 원발성 암과 감별이 어려운 경우가 있어 결핵으로 확진하기 위하여서 조직 생검이나 수술 등 침습적인 진단 방법을 이용하여야 하였다. 그러므로 이러한 결핵 병변을 비 침습적인 방법으로 정확히 감별할 수 있는 방법을 연구하던 바, 결핵균에 대한 항체를 동위원소에 부착시켜 신티그래피로 진단할 수 있는지의 가능성을 동물실험을 통하여 알아보고자 하였다. 15마리의 가토에서 M.tuberculosis H37Rv를 슬관절에 주입시켜 결핵병변을 유발시키고, 대조군으로 2마리의 가토의 고환에 T.pallidum을 주입하여 매독병변을 유발시킨 후 M.bovis BCG에 대한 특이항체 (specific polyclonal antibody)와 정상 가토의 immunoglobulin을 I-131에 부착시켜 각각의 가토에 주입하여 preset time 10분간 감마카메라로 주사한 결과 다음과 같은 결과를 얻었다. (1) 8마리의 결핵에 감염된 가토에 M.bovis BCG에 대한 $F(ab')_2$를 1 mCi의 I -131 labeling 시킨후 주사한 결과 모두에서 주사후 2시간 부터 72시간까지 병소가 hot uptake으로 보였으며 주사후 24 시간에 가장 높은 target/background ratio를 보였다. (2) 2마리의 매독에 감염된 가토에서 anti-BCG $F(ab')_2$를 주사한 결과 2시간에서는 병소에 hot activity를 보였으나 24시간부터 급격히 activity가 감소하였다. (3) $F(ab')_2$ 대신에 intact antibody를 결핵에 감염된 가토에 투여한 결과 specific polyclonal antibody나 정상가토의 immunoglobulin 모두 결핵병소에 96시간까지 hot uptake를 보였다. 그러므로 결핵균에 대한 specific antibody fragment를 이용하면 방사면역 신티그램으로 진단이 가능하리라 사료되었고, intact antibody를 사용할 경우 sensitivity는 높으나 specificity는 적을 것으로 사료되었다.

Evaluation of Renal Uptake Rate in 99mTc-DMSA Scan on Pediatrics (소아 99mTc-DMSA 검사에서 신장 섭취율의 평가)

  • The aims of this study were to evaluate the difference of renal uptake rate in $^{99m}Tc-DMSA$ scan on pediatrics by including the bladder. Phantom and Clinical studies were performed. In the phantom study, we put $^{99m}TcO_4{^-}$ (300uCi, 11 MBq) in 3cups filled with distilled water at the rate 1:1:0, 1:1:0.5, 1:1:1, 1:1:2 and were placed Lt kidney, Rt kidney and bladder position on the table. To acquire the image, we used Symbia-E gamma camera from Siemens with preset count method(400,000 counts). In quantitative analysis, the counts of drawing ROIs on the phantom were analyzed. In clinical studies, we analyzed the 20 pediatrics who were examined by $^{99m}Tc-DMSA$ scan. At first, the images were acquired with both kidney and bladder. Secondly we acquired images after shielding the bladder. And the data were compared using a pared t-test by SPSS(ver.22.0). As a result of renal phantom's experiment, we compared with average of uptake rate(%), 1:1:0 was Lt 43.32%, Rt 45.97%, 1:1:0.5 was Lt 35.79%, Rt 36.89%, 1:1:1 was Lt 29.68%, Rt 31.45% and 1:1:2 was Lt 22.89%, Rt 24.32%. There was no correlation between the zoom and uptake rate. The results of patient were that excluded bladder was $29.83{\pm}8.81%$(Lt), $24.29{\pm}6.66%$(Rt) and included bladder was $26.65{\pm}8.03%$(Lt, $21.78{\pm}6.24%$(Rt). This is deemed statistically significant (p<0.05). Renal uptake rate was undervalued because the counts of bladder were included in the total counts.

In Situ Monitoring of the MBE Growth of AlSb by Spectroscopic Ellipsometry

  • AlSb is a promising material for optical devices, particularly for high-frequency and nonlinear-optical applications. And AlSb offers significant potential for devices such as quantum-well lasers, laser diodes, and heterojunction bipolar transistors. In this work we study molecular beam epitaxy (MBE) growth of an unstrained AISb film on a GaAs substrate and identify the real-time monitoring capabilities of in situ spectroscopic ellipsometry (SE). The samples were fabricated on semi-insulating (0 0 1) GaAs substrates using MBE system. A rotating sample stage ensured uniform film growth. The substrate was first heated to $620^{\circ}C$ under As2 to remove surface oxides. A GaAs buffer layer approximately 200 nm- thick was then grown at $580^{\circ}C$. During the temperature changing process from $580^{\circ}C$ to $530^{\circ}C$, As2 flux is maintained with the shutter for Ga being closed and the reflection high-energy electron diffraction (RHEED) pattern remaining at ($2{\times}4$). Upon reaching the preset temperature of $530^{\circ}C$, As shutter was promptly closed with Sb shutter open, resulting in the change of RHEED pattern from ($2{\times}4$) to ($1{\times}3$). This was followed by the growth of AlSb while using a rotating-compensator SE with a charge-coupled-device (CCD) detector to obtain real-time SE spectra from 0.74 to 6.48 eV. Fig. 1 shows the real time measured SE spectra of AlSb on GaAs in growth process. In the Fig. 1 (a), a change of ellipsometric parameter ${\Delta}$ is observed. The ${\Delta}$ is the parameter which contains thickness information of the sample, and it changes in a periodic from 0 to 180o with growth. The significant change of ${\Delta}$ at~0.4 min means that the growth of AlSb on GaAs has been started. Fig. 1b shows the changes of dielectric function with time over the range 0.74~6.48 eV. These changes mean phase transition from pseudodielectric function of GaAs to AlSb at~0.44 min. Fig. 2 shows the observed RHEED patterns in the growth process. The observed RHEED pattern of GaAs is ($2{\times}4$), and the pattern changes into ($1{\times}3$) with starting the growth of AlSb. This means that the RHEED pattern is in agreement with the result of SE measurements. These data show the importance and sensitivity of SE for real-time monitoring for materials growth by MBE. We performed the real-time monitoring of AlSb growth by using SE measurements, and it is good agreement with the results of RHEED pattern. This fact proves the importance and the sensitivity of SE technique for the real-time monitoring of film growth by using ellipsometry. We believe that these results will be useful in a number of contexts including more accurate optical properties for high speed device engineering.

A Study on the Optimum Field Preparation Procedures for the Proper Working Performances of Rice Transplanters (논 써레질한 후의 경과일수 및 담수심이 수도이앙기의 작업성능에 미치는 영향)

  • This study was conducted in order to find out the performance of rice transplanters in accordance with the change of the trans-planting days after pudding and the water depth flooding the paddy field at the time of transplanting : and thus to select the optimum paddy field preparation procedures for an efficient utilization of rice transplanters. The performance factors of the two different types of rice transplanters were measured during the first 6 consecutive days after puddling and with 3 different levels of water depth flooding the paddy fields. The results of this study were analysed and summarized as follows : 1. Wheel sinkage decreased very rapidly from 0 to 2 days after puddling and slowly from 3 to 5 days after puddling. 2. The depth of the test cone penetration decreased rapidly during the first few days after puddling. It was 17.8cm just after puddling, and decreased to 13.4cm one day after puddling. After 2 days, the rate of decrease was dampened, and after 5 days it kept constant value of 9.2cm. 3. Two days after puddling, the hill interval was 15.8cm (98.75% of the preset value) for broadcasted seedling rice transplanter with 3cm flooding depth : This value was the closest to the pre-adjusted value of 16cm. The general performance of broadcasted-seedling type rice transplanter was better than that of strip-seedling type rice transplanter. 4. Usually the working performance of a rice transplanter is evaluated with uniformity and adjustability of the hill intervals. The hill interval was the most uniform and closest to the pre-set value of 16cm when planted two days after puddling with 3cm of water depth. When it was inavoidable to plant 4 days after puddling with stripseedling type rice transplanter, it is advisable to let the water flooded somewhat deeper. 5. The percentage of missing hills including floating and burried seedlings was the highest just after puddling and ie decreased substancially until 3 days after puddling and then it increased again. Hence, the optimal time transplanting is to be between 2 and 3 days after puddling. 6. Better postures of planted seedlings were found when planter 2 days after puddling than 3 days after puddling. Six cm of flooding water depth always gave the best results with respect to the postures of planted seedlings. Broadcasted-seedling rice transplanter, in general, showed better posture of planted seedlings than did strip-seedling type rice transplanter. 7. Judging from the above results, the optimal conditions will be 3cm of flooding depth and transplanting between 2 and 3 days after puddling.

Photomosaic Algorithm with Adaptive Tilting and Block Matching (적응적 타일링 및 블록 매칭을 통한 포토 모자이크 알고리즘)

  • Mosaic is to make a big image by gathering lots of small materials having various colors. With advance of digital imaging techniques, photomosaic techniques using photos are widely used. In this paper, we presents an automatic photomosaic algorithm based on adaptive tiling and block matching. The proposed algorithm is composed of two processes: photo database generation and photomosaic generation. Photo database is a set of photos (or tiles) used for mosaic, where a tile is divided into $4{\times}4$ regions and the average RGB value of each region is the feature of the tile. Photomosaic generation is composed of 4 steps: feature extraction, adaptive tiling, block matching, and intensity adjustment. In feature extraction, the feature of each block is calculated after the image is splitted into the preset size of blocks. In adaptive tiling, the blocks having similar similarities are merged. Then, the blocks are compared with tiles in photo database by comparing euclidean distance as a similarity measure in block matching. Finally, in intensity adjustment, the intensity of the matched tile is replaced as that of the block to increase the similarity between the tile and the block. Also, a tile redundancy minimization scheme of adjacent blocks is applied to enhance the quality of mosaic photos. In comparison with Andrea mosaic software, the proposed algorithm outperforms in quantitative and qualitative analysis.
