Production of Healthy Shrimp(Penaeus chinensis, Penaeus japonicus) Seedling Free from White Spot Baculovirus(WSBV) (White Spot Baculovirus(WSBV) 미감염 새우(Penaeus chinensis, Penaeus japonicus) 종묘 생산)

  • For the production of healthy shrimp(Penaeus chinensis, Penaeus japonicus) seedlings free from the white spot baculovirus(WSBV), we tried to disinfect shrimp eggs with iodine. A relative hatching rate of over 50% has resulted from the treatments of fertilized eggs of fresh shrimps(P. chinensis) with an effective povidon-iodine concentration of 20-200 ppm for 30 seconds and 60 seconds, respectively. The 60 sec treatment group with a 20 ppm effective povidon-iodine concentration showed nearly a 50% relative hatching rate. The over 50 ppm treatment group, however, did not result in hatching show a relative hatching rate. These hatching rates were similar to those in the fertilized eggs of kuruma prawns treated with povidon-iodine solutions. In case of washing fertilized eggs with UV-irradiated seawater 33.3% relative hatching rate in fresh shrimps was much lower than 55.2% in kuruma shrimps. Treatment fertilized eggs of 2 species of shrimp with 200 ppm iodine for 30 sec did not affect the survival rate of the larvae until the early stage of the post larvae even though the hatching rates were low. In order to produce healthy shrimp seedlings non-infected by WSBV, the shrimp eggs should be washed with UV-irradated seawater and sterilized with a low povidon-iodine concentration for a short time.

  • Sex steroids are generally considered as natural sex inducers in fish, and aromatase (cytochrome P450 aromatase) that catalyzes androgens into estrogens in the steroidogenic pathway is also known to be involved in sex differentiation. The timing of aromatase action is, thus, of central importance in the study of fish sex differentiation. We treated sexually undifferentiated tilapia (Oreochromis niloticus) larvae with $Fadrozole^{TM}$, a non-steroidal aromatase inhibitor (AI), by immersing the fish in a solution containing AI during the sex differentiation period to narrow down the critical period of aromatase action. Fish were treated once at 11 or 13 days post fertilization (dpf), or twice at 11 and 13 dpf. The concentrations of AI at each time of the treatment were 0 mg/L (control), 50 mg/L or 100 mg/L. Survival rate was not statistically associated with AI immersion treatment (p>0.25). However, sex ratio was significantly altered by the treatment, with higher concentration and double immersion being more effective in masculinizing genetic females (p<0.05). These results suggest that aromatase action for sex differentiation in this fish species would begin at least from 11 dpf which is much earlier than previously expected, and that only 3 hours of brief immersion in AI solution is powerful enough to alter genetically programed sex.

Feeding Habits of the Larval Ammodytes japonicus in the Coastal Waters of Gujora and Mangchi of Geoje, Korea (거제시 구조라 및 망치 연안에 출현하는 까나리(Ammodytes japonicus) 자어의 식성)

  • The feeding habits of larval Ammodytes japonicus were examined. A total of 208 individuals were collected by RN80 Net from January to February, 2019 in the coastal waters of Gujora and Mangchi, Geoje, Korea. Range of notochord length of larval A. japonicus was 1.46~8.00 mm and fed mainly on copepods that constituted 99.9% in %IRI. The results of analysis in ontogenetic changes, divided with 3 classes (≤2 mm, 3~4 mm, ≥5 mm, Notochord length), was all classes mainly fed copepods. An opposite relationship was found between the calanoida and cyclopoida. When percentage of calanoida of copepods increased, cyclopoida of copepods decreased.

Evolution of a Productive Bivoltine Hybrid of Silkworm Bombyx mori L.(SD7${\times}$SD12) with Shorter Larval Duration

  • With an objective of evolving quantitatively and qualitatively superior bivolitine silkworm hybrid of Bombyxmori L. for tropics with shorter larval duration without compromising on productivity traits, a breeding programme was initiated at this institute during 1997 by utilizing breeding resource material from the institutes Germplasm collection. The breed SD7 is characterized with plain larvae spinning white oval cocoons and SD12 is characterized by sex-limited larval markings spinning white dumb-bell cocoons. After fixation, these breeds along with other newly evolved breeds were subjected for hybrid evaluation in the laboratory. Based on the hybrid studies, the hybrid SD7${\times}$SD12 was selected and evaluated for one year comprising three major sea- sons (6 trials) of tropics viz., Pre-monsoon (characterized by high temperature, low humidity and without any rain fall), Monsoon (characterized by moderate temperature, heavy rain fall with high humidity) and post-monsoon season (characterized by low temperature and low humidity with frequent rain fall) to know the fluctuations in the larval duration. The evaluation studies indicated that the hybrid is having shorter larval duration with productive merits. The hybrid SD7${\times}$SD12 recorded survival percent of 92.74, shell weight of 46.7 cg, shell ratio % of 24.0 and filament length of 1,200 meters with a shorter larval duration of 24 hrs; when compared to productive hybrid CSR2${\times}$CSR5(control). The breeding methodologies and hybrids usefulness for tropics were discussed.

  • Changes of feeding habitat and feeding organ of a land-locked sweetfish, Plecoglossus altivelis related to their growth process were investigated in the Lake Okjeong, Sannae-myeon, Jeongeup-si, Jeollabuk-do, Korea from 2004 to 2005. In the lake, the post-larvae from 25 to 40 mm in standard length fed mainly on animal organisms such as Copepoda, Branchiopoda and Rotatoria. Also, the juveniles from 50 to 60 mm (SL) fed on Chironomidae of Diptera, Copepoda and Branchiopoda. However, the juvenile from 60 to 70 mm (SL) in the Churyeong Stream fed on animal organisms and aquatic algae, the immature fishes more than 70 mm (SL) fed on only periphyton such as Synedra, Cymbella, Navicula and Pinnularia. Also, the feeding organs of the tooth and stomach were changed according to its growth and food change.

Evaluation of Relative Fluoride Toxicity and Its Impact on Growth, Economic Characters and Fecundity of the Silkworm, Bombyx mori L.

  • A detailed evaluation of relative toxicity of fluoride to die 5th instar larvae of PM and NB$_4$D$_2$ races of silkworm and the effects of lethal, sublethal and prevailing levels of fluoride in groundwater on the growth, economic characters and fecundity of the silkworm, Bombyx mori (Lepidoptera: Bombycidae) were studied. The feeding of mulberry, Morus alba leaves treated with lethal and sublethal doses of fluoride to PM and NB$_4$D$_2$ races of silkworms from the beginning of the 5th instar to the end of the feeding period resulted in significant reduction in growth, single cocoon weight, single shell weight, silk index, average filament length, and fecundity when compared to controls. These manges were more pronounced on exposure to lethal dose than sublethal dose and in general, the changes induced by fluoride were more striking in NB$_4$D$_2$ than PM, indicating the greater resistance of PM to higher fluoride levels. Groundwater quality in sericulturally important villages of Karnataka and Andhra Pradesh States has been studied with special reference to the presence of fluoride. On exposure to dose of prevailing levels in the waters of sericulturally important areas, die changes observed on growth, economic characters and fecundity were slightly lower when compared to controls and the decrease was found to be insignificant (P$\geq$0.05). It was concluded that, though minute dose (4.0 ppm) of fluoride did not have any toxic impact, it is toxic at higher concentrations to silkworms.

Copper, Zinc-Superoxide Dismutase (Cu/Zn SOD) Gene During Embryogenesis of Bombyx mori: Molecular Cloning, Characterization and Expression

  • BmCu/Zn SOD was isolated from early embryo of Bombyx mori using microarray analysis. The BmCu/Zn SOD gene was observed during the early embryonic stage with the strongest signal found at the unfertilizaion, fertilization and blastoderm stages. The BmCu/Zn SOD gene encodes a protein of 154 amino acids with a calculated Mr of 15 kDa. The deduced amino acid sequence of BmCu/Zn SOD indicated that the residues that form on the Cu/Zn binding site are conserved and that the sequence is a 60% identity to that of M. domestica. In a phylogenetic tree, Bm SOD was also close to Drosophila SODs rather than other insect SODs. The BmCu/Zn SOD gene exists as a single copy in the genome. Transcripts of BmCu/Zn SOD cDNA were identified by northern blot analysis. The expression of the BmCu/Zn SOD gene was observed weakly in most of larvae, pre-pupae, pupae and adult tissues. Also, the BmCu/Zn SOD gene was observed in early embryonic stage. Although the roles of SODs remains to be further elucidated, the high expression of BmCu/Zn SOD gene at before 24 h post fertilization suggests that this gene is of general importance during early embryogenesis in the Bombyx mod.

Effect of Temperature on the Development of Egg and Larvae of Sea Urchin (Strongylocentrotus intermedius) (북쪽말똥성게 (Strongylocentrotus intermedius)의 난발생과 유생의 발달)

  • Effect of temperature (5, 10, 15, 18, 21$^{\circ}C$) on pre-and post-embryonic development of the sea urchin Strongylocentrotus intermedius was studied. The egg, which measured 122.5 ${\mu}{\textrm}{m}$, became globular. At 18$^{\circ}C$, it attained 4 celled stage at 3 hours after fertilization, hatched after 15 hours, 4-armed larval stage after 3 days and 8- armed larval stage after 20 days. The relationships between temperature (WT) and time (1, hour) required for each of the selected developmental stages are: Hatching: 1/t=0.0036WT+0.0088 Pyramid: 1/t=0.0014Wl-0.0016 4- armed: 1/t=0.0009WT-0.0020 6- armed: 1/t=0.0004WT-0.0005 8-armed : 1/t=0.0002WT+0.0002 Biological minimum temperature for the egg and larval development is calculated as 1.61$^{\circ}C$.

Alteration of Cytokine Production during Visceral Larva Migrans by Toxascaris leonina in Mice

  • To determine alteration of immune responses during visceral larva migrans (VLM) caused by Toxascaris leonina at several time points, we experimentally infected mice with embryonated eggs of T. leonina and measured T-helper (Th) cell-related serial cytokine production after infection. At day 5 post infection (PI), most larvae were detected from the lungs, spleen, intestine, and muscle. Expression of thymic stromal lymphopoietin (TSLP) and CCL11 (eotaxin) showed a significant increase in most infected organs, except the intestine. However, expression of the CXCL1 (Gro-${\alpha}$) gene was most highly enhanced in the intestine at day 14 PI. Th1-related cytokine secretion of splenocytes showed increases at day 28 PI, and the level showed a decrease at day 42 PI. Th2-related cytokine secretion of splenocytes also showed an increase after infection; in particular, IL-5 level showed a significant increase at day 14 PI, and the level showed a decrease at day 28 PI. However, levels of Th17-related cytokines, IL-6 and IL-17A, showed gradual increases until day 42 PI. In conclusion, Th1, Th2, and Th17-related cytokine production might be important in immune responses against T. leonina VLM in experimental mice.

The Influence of Challenge on Cathepsin B and D Expression Patterns in the Silkworm Bombyx mori L.

  • Cathepsins are well-characterized proteases that are ubiquitously expressed in lysosomes. Previous work revealed that $Bombyx$ $mori$ cathepsins B and D are expressed in the fat body and undergo decomposition during larval-pupal metamorphosis. Quantitative RT-PCR was performed to detect cathepsin gene expression at the transcription level when challenged by $B.$ $mori$ nuclear polyhedrosis virus (BmNPV), temperature and hormones (20-hydroxyecdysone (20E) and juvenile hormone analogue (JHA)). mRNAs encoding cathepsins B and D were significantly enhanced after the larvae were infected with BmNPV, and the peak of the induction appeared at 1 day before spinning. This attenuated the inducing effect on cathepsin expression caused by infection. Temperature shock induced cathepsin expression at the later stage of the $5^{th}$ instar, and transcription levels varied with development stage and temperature. Cathepsin B and D mRNA expression in the fat body were significantly induced by JHA at the day before spinning, and with 20E, the expression reached a peak at the last day of the $5^{th}$ instar. Cathepsin B and D mRNA expression exhibited detectable changes post-treatment, without significant differences between or among the hormone concentrations.
